Here, we report the use of an amphiphilic Pt(II) complex, K[Pt{(O3SCH2CH2CH2)2bzimpy}Cl] (PtB), as a model to elucidate the key role of Pt···Pt interactions in directing self-assembly by combining temperature-dependent ultraviolet-visible (UV-Vis) spectroscopy, stopped-flow kinetic experiments, quantum mechanics (QM) calculations, and molecular dynamics (MD) simulations. Interestingly, we found that the self-assembly mechanism of PtB in aqueous solution follows a nucleation-free isodesmic model, as revealed by the temperature-dependent UV-Vis experiments. In contrast, a cooperative growth is found for the self-assembly of PtB in acetone­water (7:1, vol/vol) solution, which is further verified by the stopped-flow experiments, which clearly indicates the existence of a nucleation phase in the acetone­water (7:1, vol/vol) solution. To reveal the underlying reasons and driving forces for these self-assembly processes, we performed QM calculations and show that the Pt···Pt interactions arising from the interaction between the pz and dz2 orbitals play a crucial role in determining the formation of ordered self-assembled structures. In subsequent oligomer MD simulations, we demonstrate that this directional Pt···Pt interaction can indeed facilitate the formation of linear structures packed in a helix-like fashion. Our results suggest that the self-assembly of PtB in acetone­water (7:1, vol/vol) solution is predominantly driven by the directional noncovalent Pt···Pt interaction, leading to the cooperative growth and the formation of fibrous nanostructures. On the contrary, the self-assembly in aqueous solution forms spherical nanostructures of PtB, which is primarily due to the predominant contribution from the less directional hydrophobic interactions over the directional Pt···Pt and π−π interactions that result in an isodesmic growth.

Aldosterone (ALD) is excreted in urine mainly as glucuronide conjugates of ALD and tetrahydroaldosterone. Measuring these urinary metabolites might be an alternative screening test to plasma ALD for primary aldosteronism. We report a validated LC-MS/MS method to measure both analytes simultaneously. Urine samples underwent enzymatic hydrolysis to release the analytes from their glucuronide conjugates followed by organic solvent extraction and LC-MS/MS. The analytical performance of this method was evaluated. The within-batch and between-batch coefficients of variation for urine ALD and urine THA were all ≤5.2 and ≤3.7%. The lower limit of quantification was 0.5 nmol/L, and the linearity was up to at least 2770 nmol/L for both analytes. No significant matrix interference and carryover were observed. Both analytes in urine were stable for at least 48 h at 10°C and at least 18 months at -80°C. Local reference intervals were established from 126 healthy normotensive Chinese subjects (53% women, age: 20-65 years). Reference intervals for urine ALD and tetrahydroaldosterone were 2-38 and 9-139 nmol/day, respectively. This validated method can be applied to screening and diagnosing primary aldosteronism.

Delayed parenthood is constantly increasing worldwide due to various socio-economic factors. In the last decade, a growing number of epidemiological studies have suggested a link between advanced parental age and an increased risk of diseases in the offspring. Also, poor reproductive outcome has been described in pregnancies conceived by aged parents. Similarly, animal studies showed that aging negatively affects gametes, early embryonic development, pregnancy progression, and the postnatal phenotype of resulting offspring. However, how and to what extent parental age is a risk factor for the health of future generations is still a subject of debate. Notwithstanding the limitation of an animal model, the mouse model represents a useful tool to understand not only the influence of parental age on offspring phenotype but also the biological mechanisms underlying the poor reproductive outcome and the occurrence of diseases in the descendants. The present review aims at i) providing an overview of the current knowledge from mouse model about the risks associated with conception at advanced age (e.g. neurodevelopmental and metabolic disorders), ii) highlighting the candidate biological mechanisms underlying this phenomenon, and iii) discussing on how murine-derived data can be relevant to humans.

OBJECTIVES: The renin-angiotensin-aldosterone system (RAAS) regulates blood pressure. Plasma renin activities (PRA) and plasma aldosterone concentrations (PAC) are biomarkers related to RAAS. Liquid chromatography-tandem mass spectrometry (LC-MS/MS)-based measurements for PRA and PAC have become popular. Method-specific reference intervals (RIs) are required. METHODS: Routine PRA and PAC services in a Hong Kong teaching hospital were based on LC-MS/MS methods. PRA and PAC RIs were developed for normotensive subjects and essential hypertensive (EH) patients. Healthy volunteers were recruited to establish normotensive RIs. PRA and PAC results of hypertensive patients with urine aldosterone tests for primary aldosteronism (PA) screening were retrieved from the laboratory information system. Patients without PA were included. Patients with secondary hypertension and patients on medications affecting the RAAS were excluded. The central 95% RIs were established based on the recommendations of the Clinical and Laboratory Standards Institute guideline C28-A3. RESULTS: PRA and PAC of 170 normotensive volunteers and 362 EH patients were analysed. There was no sex-specific difference in PRA and PAC for normotensive and EH reference subjects. Differences for PRA and PAC were noted between normotensive subjects aged below 45 and their older counterparts. However, such a difference was only identified for PRA but not PAC in EH patients. Age-specific RIs were established accordingly. CONCLUSIONS: This study presented age-specific LC-MS/MS RIs of PRA and PAC for both normotensive and EH populations for local Chinese in Hong Kong.

With the rapidly growing exploration of artificial molecular machines and their applications, there is a strong demand to develop molecular machines that can have their motional states and configuration/conformation changes detectable by more sensitive and innovative methods. A visual artificial molecular hinge with phosphorescence behavior changes is designed and synthesized using square-planar cyclometalated platinum(II) complex and rigid aromatic alkynyl groups as the building blocks to construct the wings/flaps and axis, respectively. The molecular motions of this single molecular hinge and its reversible processes can be powered by both solvent and temperature changes. The rotary motion can be conveniently observed by the visual phosphorescence changes from deep-red to green emission in real time.

A shortage of physiotherapist (PT) manpower is a barrier for providing better rehabilitation service in Hong Kong. Quality training can benefit patients with better recovery, on the contrary, insufficient training may cause a longer length of stay, readmission, and thus the burden of healthcare system. The estimated cost for PT services in Hospital Authority was HK$7.0 Billion in 2020. A novel Danish robot with a 7-joint robotic arm became popular in Denmark and Germany in the last two years. The robot is designed for lower limb patient rehabilitation. It can enhance the mobility of patients. Based on the experience of a university hospital in Denmark, this robotic rehabilitation was well accepted by both patients and PTs. Function-wise, the robot provides many clinical benefits to patients, especially stroke ones. A physiotherapist's time can be saved when the robot is being used. The cost-effectiveness of ROBERT® is better than PT performing repetitive exercises for lower limbs. The robot potentially provides a cost-effective solution to the Hong Kong healthcare system.

BACKGROUND: The aim of this study was to evaluate the role of AT1 and AT2 receptors in a periodontal inflammation experimental model. METHODS: Periodontal inflammation was induced by LPS/Porphyromonas gingivalis. Maxillae, femur, and vertebra were scanned using Micro-CT. Maxillae were analyzed histopathologically, immunohistochemically, and by RT-PCR. RESULTS: The vertebra showed decreased BMD in AT1 H compared with WT H (p < 0.05). The femur showed increased Tb.Sp for AT1 H and AT2 H, p < 0.01 and p < 0.05, respectively. The Tb.N was decreased in the vertebra (WT H-AT1 H: p < 0.05; WT H-AT2 H: p < 0.05) and in the femur (WT H-AT1 H: p < 0.01; WT H-AT2 H: p < 0.05). AT1 PD increased linear bone loss (p < 0.05) and decreased osteoblast cells (p < 0.05). RANKL immunostaining was intense for AT1 PD and WT PD (p < 0.001). OPG was intense in the WT H, WT PD, and AT2 PD when compared to AT1 PD (p < 0.001). AT1 PD showed weak immunostaining for osteocalcin compared with WT H, WT PD, and AT2 PD (p < 0.001). AT1 H showed significantly stronger immunostaining for osteonectin in fibroblasts compared to AT2 H (p < 0.01). CONCLUSION: AT1 receptor knockout changed bone density, the quality and number of bone trabeculae, decreased the number of osteoblast cells, and increased osteonectin in fibroblasts.

Calcium phosphate cements have the advantage that they can be prepared as a paste that sets in a few minutes and can be easily adapted to the shape of the bone defect, which facilitates its clinical application. In this research, six formulations of brushite (dicalcium phosphate dihydrated) cement were obtained and the effect of the addition of sodium alginate was analyzed, such as its capacity as a tetracycline release system. The samples that contain sodium alginate set in 4 or 5 min and showed a high percentage of injectability (93%). The cements exhibit compression resistance values between 1.6 and 2.6 MPa. The drug was released in a range between 12.6 and 13.2% after 7 days. The antimicrobial activity of all the cements containing antibiotics was proven. All samples reached values of cell viability above 70 percent. We also observed that the addition of the sodium alginate and tetracycline improved the cell viability.

A discrete four-choice response task with auditory signal presentation and a joystick-controlled visual tracking task was used to investigate how spatial compatibility influences the dual-task performance of different display-control settings. It was found that the more incompatible the stimulus-response mapping, the longer the delay for both tasks, presumably because of the longer stimulus encoding time required for the incompatible conditions. A comparison of the findings of this study with those of past experiments on visual visual setting shows that the dual-task performance in a cross-modality (auditory visual) setting was significantly better than that in an intra-modality (visual visual) setting because of visual scanning required in the intra-modal dual tasks. However, when the locations of visual visual tasks were close enough such that ambient and focal vision was concurrently used for information processing, the dual-task performance of intra-modality (visual visual) configuration was slightly better than that of the cross-modality (auditory visual) configuration. Practitioner Summary: The effect of spatial compatibility with auditory signal presentation in multiple display-control configurations was examined in a dual-task paradigm. The results provided important and useful ergonomics design implications and consequent recommendations for intra- and cross- modal interface design. The results should facilitate human-machine system design and improve overall system performance.

Traumatic brain injury (TBI) is a devastating event for which current therapies are limited. Stem cell transplantation may lead to recovery of function via different mechanisms, such as cell replacement through differentiation, stimulation of angiogenesis and support to the microenvironment. Adult hair follicle bulge-derived stem cells (HFBSCs) possess neuronal differentiation capacity, are easy to harvest and are relatively immune-privileged, which makes them potential candidates for autologous stem cell-based therapy. In this study, we apply in vivo multimodal, optical and magnetic resonance imaging techniques to investigate the behavior of mouse HFBSCs in a mouse model of TBI. HFBSCs expressed Luc2 and copGFP and were examined for their differentiation capacity in vitro. Subsequently, transduced HFBSCs, preloaded with ferumoxytol, were transplanted next to the TBI lesion (cortical region) in nude mice, 2 days after injury. Brains were fixed for immunohistochemistry 58 days after transplantation. Luc2- and copGFP-expressing, ferumoxytol-loaded HFBSCs showed adequate neuronal differentiation potential in vitro. Bioluminescence of the lesioned brain revealed survival of HFBSCs and magnetic resonance imaging identified their localization in the area of transplantation. Immunohistochemistry showed that transplanted cells stained for nestin and neurofilament protein (NF-Pan). Cells also expressed laminin and fibronectin but extracellular matrix masses were not detected. After 58 days, ferumoxytol could be detected in HFBSCs in brain tissue sections. These results show that HFBSCs are able to survive after brain transplantation and suggest that cells may undergo differentiation towards a neuronal cell lineage, which supports their potential use for cell-based therapy for TBI.

The photophysical properties of a series of T-shaped coinage d10 metal complexes, supported by a bis(mesoionic carbene)carbazolide (CNC) pincer ligand, are explored. The series includes a rare new example of a tridentate T-shaped AgI complex. Post-complexation modification of the AuI complex provides access to a linear cationic AuI complex following ligand alkylation, or the first example of a cationic square planar AuIII -F complex from electrophilic attack on the metal centre. Emissions ranging from blue (CuI ) to orange (AgI ) are obtained, with variable contributions of thermally-dependent fluorescence and phosphorescence to the observed photoluminescence. Green emissions are observed for all three gold complexes (neutral T-shaped AuI , cationic linear AuI and square planar cationic AuIII ). The higher quantum yield and longer decay lifetime of the linear gold(I) complex are indicative of increased phosphorescence contribution.

Accumulating evidence indicates that ceramide (Cer) and palmitic acid (PA) possess the ability to modulate switching of macrophage phenotypes and possess anti-tumorigenic effects; however, the underlying molecular mechanisms are largely unknown. The aim of the present study was to investigate whether Cer and PA could induce switching of macrophage polarization from the tumorigenic M2- towards the pro-inflammatory M1-phenotype, and whether this consequently altered the potential of colorectal cancer cells to undergo epithelial-mesenchymal transition (EMT), a hallmark of tumor progression. Our study showed that Cer- and PA-treated macrophages increased expression of the macrophage 1 (M1)-marker CD68 and secretion of IL-12 and attenuated expression of the macrophage 2 (M2)-marker CD163 and IL-10 secretion. Moreover, Cer and PA abolished M2 macrophage-induced EMT and migration of colorectal cancer cells. At the molecular level, this coincided with inhibition of SNAI1 and vimentin expression and upregulation of E-cadherin. Furthermore, Cer and PA attenuated expression levels of IL-10 in colorectal cancer cells co-cultured with M2 macrophages and downregulated STAT3 and NF-κB expression. For the first time, our findings suggest the presence of an IL-10-STAT3-NF-κB signaling axis in colorectal cancer cells co-cultured with M2 macrophages, mimicking the tumor microenvironment. Importantly, PA and Cer were powerful inhibitors of this signaling axis and, consequently, EMT of colorectal cancer cells. These results contribute to our understanding of the immunological mechanisms that underlie the anti-tumorigenic effects of lipids for future combination with drugs in the therapy of colorectal carcinoma.

(1) Background: doxorubicin is a potent chemotherapeutic agent, but it has limitations regarding its side effects and therapy resistance. Hydrogels potentially deal with these problems, but several characterizations need to be optimized to better understand how hydrogel assisted chemotherapy works. Poloxamer 407 (P407) hydrogels were mixed with doxorubicin and physico-chemical, biological, and pharmacological characterizations were considered. (2) Methods: hydrogels were prepared by mixing P407 in PBS at 4 °C. Doxorubicin was added upon solutions became clear. Time-to-gelation, hydrogel morphology, and micelles were studied first. The effects of P407-doxorubicin were evaluated on MC-38 colon cancer cells. Furthermore, doxorubicin release was assessed and contrasted with non-invasive in vivo whole body fluorescence imaging. (3) Results: 25% P407 had favorable gelation properties with pore sizes of 30-180 µm. P407 micelles were approximately 5 nm in size. Doxorubicin was fully released in vitro from 25% P407 hydrogel within 120 h. Furthermore, P407 micelles strongly enhanced the anti-neoplastic effects of doxorubicin on MC-38 cells. In vivo fluorescence imaging revealed that hydrogels retained fluorescence signals at the injection site for 168 h. (4) Conclusions: non-invasive imaging showed how P407 gels retained drug at the injection site. Doxorubicin P407 micelles strongly enhanced the anti-tumor effects.

High-quality data are the foundation to monitor the progress and evaluate the effects of road traffic injury prevention measures. Unfortunately, official road traffic injury statistics delivered by governments worldwide, are often believed somewhat unreliable and invalid. We summarized the reported problems concerning the road traffic injury statistics through systematically searching and reviewing the literature. The problems include absence of regular data, under-reporting, low specificity, distorted cause spectrum of road traffic injury, inconsistency, inaccessibility, and delay of data release. We also explored the mechanisms behind the problematic data and proposed the solutions to the addressed challenges for road traffic statistics.

Recovering heavy and precious metals from wastewater is both economically and environmentally attractive. However, a challenge still remains in how to realize the highly selective removal and recovery of a particular metal of interest, such as platinum. Here we experimentally demonstrate a two-dimensional (2D) supramolecular polymer that can serve as a host for the highly selective capture of anionic platinum(II) (PtII) species including its metal-organic complexes and water-soluble ions. This host polymer possesses a 2D honeycomb-like nanostructure noncovalently bridged by cationic alkynylplatinum(II) terpyridine dimers. Anionic PtII guests readily bind to the dimeric PtII building blocks of the internal cavities via electrostatic and specific PtII···PtII interactions. Such unique host-guest interactions have endowed thin membranes comprising highly oriented supramolecular polymers with a versatile ability to selectively recognize and adsorb anionic PtII species over other metal ions in water upon simple filtration.

A new class of amphiphilic tridentate cyclometalated gold(III) complexes has been designed and synthesized as luminescent supramolecular building blocks. Positively charged trimethylammonium (-CH2NMe3+) containing alkynyl ligands have been incorporated to introduce the electrostatic interactions. The X-ray crystal structures of two of the complexes have been determined, and the existence of π-π interactions between molecules has been observed. Steady-state and time-resolved absorption and emission studies have been carried out to investigate the nature of the excited states. The complexes are found to exhibit self-assembly properties with the assistance of π-π stacking and hydrophobic interactions and possibly weak Au···Au interaction, resulting in notable emergence of low-energy absorption bands and luminescence changes. The presence of a large hydrophobic moiety is found to be crucial for the formation of aggregates, especially in polar media where hydrophobic interactions play an important role. The nature of the counterion has been shown to have a significant effect on the extent of self-assembly in different media. Upon aggregation, nanofibers are formed in polar media, while nanorods are observed in nonpolar media in one of the representative complexes. Interestingly, a small modification on the alkynyl ligand resulted in the formation of nanoribbons instead. Intriguing luminescence mechanochromic properties have also been observed. This orthogonal and rational molecular design strategy has been shown to be effective in the construction of gold(III)-based smart and multiresponsive materials.

Alkynylplatinum(II) terpyridine complexes have been increasingly explored since the previous decades, mainly arising from their intriguing photophysical properties and aggregation affinities associated with their extensive Pt(II)···Pt(II) and π-π stacking interactions. Through molecular engineering, one can modulate their fundamental properties and assembly behavior by introduction of various functional groups and structural features. They can therefore serve as ideal candidates to construct metal complex-based molecular architectures to provide an alternative to organic compounds. The metal-based framework can be simultaneously built from predetermined building blocks, giving rise to their well-defined, unique, and discrete natures for molecular recognition. The individual constituents can contribute to molecular architectures with their integrated properties, allowing the manipulation of the various noncovalent intermolecular forces and interactions for selective guest capture. In this Account, our recent progress in the development of these metallomolecular frameworks based on the alkynylplatinum(II) terpyridine system and their recognition properties toward different guest molecules will be presented. Phosphorescent molecular tweezers have been constructed from the alkynylplatinum(II) terpyridine moiety to demonstrate host-guest interactions with cationic, charge-neutral and anionic platinum(II), palladium(II), gold(I), and gold(III) complexes and their binding affinities were found to be perturbed by different metal···metal, π-π and electrostatic interactions. The host-guest assembly process has also resulted in dramatic color changes, together with the turning on of near-IR (NIR) emissions as a result of extensive Pt(II)···Pt(II) interactions. Further work has also been performed to demonstrate that the tweezers can selectively recognize π-surfaces of different planar π-conjugated organic guests. The framework of molecular tweezers has been extended to a double-decker tweezers structure, or a triple-decker structure, which can bind two equivalents of square-planar platinum(II) guests cooperatively to induce a significant color change in solution, representing rare examples of discrete Magnus' green-like salts. By the approaches of structural modifications, we have further modulated the host architecture from molecular tweezers to molecular rectangles. The rectangles have been found to show selective encapsulation of different transition metal complex guests based on the size and steric environment of the host cavity. The molecular rectangles also exhibit reversible host-guest association, in which guest capture and ejection processes can be manipulated by the pH environment, illustrating a potential approach for precise and smart delivery of therapeutic reagents to the slightly more acidic cancer cells.

An l-glutamine-derived functional group was introduced to the bis(arylalkynyl)platinum(II) bipyridine complexes 1-4. The emission could be switched between the 3 MLCT excited state and the triplet excimeric state through solvent or temperature changes, which is attributed to the formation and disruption of hydrogen-bonding, π-π stacking, and metal-metal interactions. Different architectures with various morphologies, such as honeycomb nanostructures and nanospheres, were formed upon solvent variations, and these changes were accompanied by 1 H NMR and distinct emission changes. Additionally, yellow and red emissive metallogels were formed at room temperature due to the different aggregation behaviors introduced by the substituent groups on bipyridine. The thermoresponsive metallogel showed emission behavior with tunable colors by controlling the temperature. The negative Gibbs free-energy change (ΔG) and the large association constant for excimer formation have suggested that the molecules undergo aggregation through hydrogen-bonding, π-π, and metal-metal interactions, resulting in triplet excimeric emission.
