In a fog-enabled IoT environment, a fog node is regarded as the proxy between end users and cloud servers to reduce the latency of data transmission, so as to fulfill the requirement of more real-time applications. A data storage scheme utilizing fog computing architecture allows a user to share cloud data with other users via the assistance of fog nodes. In particular, a fog node obtaining a re-encryption key of the data owner is able to convert a cloud ciphertext into the one which is decryptable by another designated user. In such a scheme, a proxy should not learn any information about the plaintext during the transmission and re-encryption processes. In 2020, an ID-based data storage scheme utilizing anonymous key generation in fog computing was proposed by some researchers. Although their protocol is provably secure in a proof model of random oracles, we will point out that there are some security flaws inherited in their protocol. On the basis of their work, we further present an improved variant, which not only eliminates their security weaknesses, but also preserves the functionalities of anonymous key generation and user revocation mechanism. Additionally, under the Decisional Bilinear Diffie-Hellman (DBDH) assumption, we demonstrate that our enhanced construction is also provably secure in the security notion of IND-PrID-CPA.

This study examined the effect of the Flos Lonicerae Japonicae water extract (FLJWE), chlorogenic acid, and luteolin on pseudorabies virus (PRV)-induced inflammation in RAW264.7 cells and elucidated related molecular mechanisms. The results revealed that FLJWE and luteolin, but not chlorogenic acid, inhibited the production of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), and inflammatory cytokines in PRV-infected RAW 264.7 cells. We found that the FLJWE and luteolin suppressed nuclear factor (NF)-κB activation by inhibiting the phosphorylation of signal transducer and activator of transcription 1 and 3 (STAT1 and STAT3, respectively). Moreover, the FLJWE significantly upregulated the expression of pNrf2 and its downstream target gene heme oxygenase-1 (HO-1). Our data indicated that FLJWE and luteolin reduced the expression of proinflammatory mediators and inflammatory cytokines, such as COX-2 and iNOS, through the suppression of the JAK/STAT1/3-dependent NF-κB pathway and the induction of HO-1 expression in PRV-infected RAW264.7 cells. The findings indicate that the FLJWE can be used as a potential antiviral agent.

Age-related macular degeneration (AMD) leads to gradual central vision loss and is the third leading cause of irreversible blindness worldwide. The underlying mechanisms for this progressive neurodegenerative disease remain unclear and there is currently no preventive treatment for dry AMD. Sodium iodate (NaIO3) has been reported to induce AMD-like retinal pathology in mice. We established a mouse model for AMD to evaluate the effects of quercetin on NaIO3-induced retinal apoptosis, and to investigate the pertinent underlying mechanisms. Our in vitro results indicated that quercetin protected human retinal pigment epithelium (ARPE-19) cells from NaIO3-induced apoptosis by inhibiting reactive oxygen species production and loss of mitochondrial membrane potential as detected by Annexin V-FITC/PI flow cytometry. We also evaluated the relative expression of proteins in the apoptosis pathway. Quercetin downregulated the protein expressions of Bax, cleaved caspase-3, and cleaved PARP and upregulated the expression of Bcl-2 through reduced PI3K and pAKT expressions. Furthermore, our in vivo results indicated that quercetin improved retinal deformation and increased the thickness of both the outer nuclear layer and inner nuclear layer, whereas the expression of caspase-3 was inhibited. Taken together, these results demonstrate that quercetin could protect retinal pigment epithelium and the retina from NaIO3-induced cell apoptosis via reactive oxygen species-mediated mitochondrial dysfunction, involving the PI3K/AKT signaling pathway. This suggests that quercetin has the potential to prevent and delay AMD and other retinal diseases involving NaIO3-mediated apoptosis.

Prospective cohort studies have indicated that a highly nickel-polluted environment may severely affect human health, resulting in such conditions as respiratory tract cancers. Such exposure can trigger vascular endothelial growth factor (VEGF) expression. However, the signal transduction pathways leading to VEGF induction by nickel compounds are not well understood. This study revealed the occurrence of VEGF induction in human non-small-cell lung cancer H460 cells exposed to NiCl2 . Moreover, exposing H460 cells to NiCl2 activated extracellular signal-regulated protein kinase (ERK), nuclear factor kappa B (NFκB), and protein kinase B (Akt) as well as downregulated AMP activated protein kinase (AMPK) expression. The mitogen-activated protein kinase (MAPK) and ERK inhibitor significantly blocked NiCl2 -induced ERK activation and VEGF production. Pretreating H460 cells with a PI3K/Akt inhibitor substantially inhibited NiCl2 -induced VEGF expression and reduced Akt, ERK, and NFκB phosphorylation. Furthermore, 5-aminoimidazole-4-carboxamide ribonucleoside-induced AMPK activation improved VEGF expression in NiCl2 -treated H460 cells significantly. These results indicate that NiCl2 induces VEGF production through Akt, ERK, NFκB activation and AMPK suppression and mediates various types of pathophysiological angiogenesis.

A novel water resistant antifog (AF) coating for plastic substrates was developed, which has a special hydrophilic/hydrophobic bilayer structure. The bottom layer, acting both as a mechanical support and a hydrophobic barrier against water penetration, is an organic-inorganic composite comprising colloidal silica embedded in a cross-linked network of dipentaethritol hexaacrylate (DPHA). Atop this layer, an AF coating is applied, which incorporates a superhydrophilic species synthesized from Tween-20 (surfactant), isophorone diisocyanate (coupling agent), and 2-hydroxyethyl methacrylate (monomer). Various methods, e.g., FTIR, SEM, AFM, contact angle, and steam test, were employed to characterize the prepared AF coatings. The results indicated that the size and the continuity of the hydrophilic domains on the top surface increased with increasing added amount of T20, however, at the expense of hardness, adhesiveness, and water resistivity. The optimal T20 content was found to be 10 wt %, at which capacity the resultant AF coating was transparent and wearable (5H, hardness) and could be soaked in water for 7 days at 25 °C without downgrading of its AF capability.

PURPOSE: According to previous studies, the prevalence rate of cataracts has increased in recent years. This study aims to investigate and analyze the risk factors of early-onset cataracts in Taiwan. METHODS: A total of 71 subjects aged between 20 and 55 were diagnosed with cataracts in a medical center. Participants were divided into three groups: control, early-onset cataract (EOC), and combined (EOC combined with dry eye) groups. Eye examinations including autorefraction, best-corrected visual acuity (BCVA), subjective refraction, axial length, fundus, slit lamp, and reactive oxygen species (ROS, including total antioxidative capacity, TAC; C-reactive protein, CRP; and glutathione peroxidase, GPx) were performed. In addition, a questionnaire on patient information, history, habits, family history, and Depression Anxiety Stress Scales (DASS) was completed before the examination. RESULTS: 27 non-EOC (control group), 20 EOC, and 24 combined patients participated in the study. Compared with the control group, Body Mass Index (BMI), gender, educational level, hypertension, diabetes, hyperlipidemia, chronic pain, and body-related diseases were significantly different between the three groups. Family history was also significantly different: family heart disease, hypertension, asthma, allergies, stroke, and immune system were also significantly different. In addition, subjects who took hypertensive drugs, antihistamines, and other medications were also significantly different. Statistical analysis indicated that best corrective visual acuity and the spherical equivalent were significantly different between the three groups. Similar results were found in CRP blood analysis. DISCUSSION AND CONCLUSION: According to the results, EOC may result from systemic diseases. The risk corresponded to an increase in ROS blood analysis. Furthermore, eye drops and medicine intake significantly influenced EOC patients. To prevent or defer early-onset cataracts, monitoring physical health, CRP, and GPx analysis may be worth considering in the future.

INTRODUCTION: Vision is critical for children's development. However, prevalence of visual impairment (VI) is high in students with special educational needs (SEN). Other than VI, SEN students are prone to having functional deficits. Whether visual problems relate to these functional deficits is unclear. This study aimed to assess the impact of vision on visual processing functions and balance in SEN students through a community service. METHODS: Visual acuity (VA) and contrast sensitivity were measured in a total of 104 (aged 14.3±4.3) SEN students as the visual outcomes, followed by retinoscopy. Visual processing function assessment included facial expression recognition by card matching examiner's facial expression matching, and visual orientation recognition. Dynamic balance, by Timed Up and Go test, and static standing balance (postural sway in double-legged standing with feet-together and tandem-stance for open-eye and closed-eye conditions) were assessed. Static balance was presented in terms of the maximal medial-lateral and antero-posterior sways. RESULTS: Of the 104 students, 62 (59.6%) were classified as visually impaired according to WHO classification of visual impairment based on presenting distance acuity. Ocular problems (e.g. optic nerve anomaly, uncorrected/ under-corrected refractive errors) and neurological anomalies were the major causes of vision loss. VA was positively associated with visual processing functions (all p ≤ 0.01), as SEN students with better vision tended to perform better in visual orientation and facial expression recognition tasks, as well as dynamic balance function (p = 0.04). For the static balance, postural sway and VA showed a positive relationship under open-eye and tandem stance conditions. However, the relationship between postural sway and VA became negative under closed-eye and tandem stance conditions. CONCLUSION: This study found a high prevalence of SEN students with visual impairment, in which many of them were undetected. Optometric examination is important to improve their visual function to minimize the effect of vision on functional performance. Vision is critical in visual processing as well as playing an important role in maintaining balance in SEN students.

Phalaenopsis orchids are popular ornamentals all over the world. A tospovirus, capsicum chlorosis virus (CaCV-Ph) had been identified as the cause of chlorotic ringspots on leaves of Phalaenopsis orchids in Taiwan. The tripartite genome of CaCV-Ph was found to contain 3608, 4848 and 8916 nt of S, M and L RNAs, respectively. Phylogenetic analysis of the nucleocapsid (N) protein confirmed that CaCV-Ph is a member of the watermelon silver mottle virus (WSMoV) serogroup in the genus Tospovirus. Based on the relations among the nonstructural protein (NSs), glycoprotein (GnGc), thrips genera, host and geographical distribution, tospoviruses and thrips could be classified into two major types: WSMoV-Thrips-Asian and Tomato spotted wilt virus (TSWV)-Frankliniella-EuroAmerican. The proline (P(459)) of all tospoviral Gn proteins was indispensable for thrips transmission, but the RGD motif, which is maintained by only six tospoviruses, may not be required for thrips transmission. An RdRp catalytic domain found in the conserved region of the L protein may recognize the typically conserved sequences on the 5' and 3' terminal regions (5' AGAGCAAU 3').

Manual material handling (MMH) tasks create a burden for workers which could result in musculoskeletal injuries. Assessments of the decrease of muscular strength and the maximum endurance time (MET) for MMH tasks are essential in studying the ergonomic risk of MMH tasks. A backpacking experiment was conducted for measuring the MET for MMH tasks. Human participants carried a load on their back and walked on a treadmill under various load, walking speed, and ramp angle conditions until they coud no longer do so. It was found that the participants were able to walk for approximately 15 min to two hours before they needed to have a pause. Their back and leg strengths declined moderately due to performing the tasks. These tasks resulted in an increase in heart rate and elevated perceived physical exertion. The rating of perceived exertion (RPE)/heart rate ratio in our backpacking tasks was 31% higher than that in the literature, implying the calibration of the RPE may be required for such tasks. A MET model incorporating the fMVC_back, body weight, walking speed, and ramp angle was established. This model may be used to determine the work/rest allowance for backpacking tasks under conditions similar to this study.

Recent investigations have demonstrated that carotenoid extract of Dunaliella salina alga (Alga) contains abundant ß-carotene and has good anti-inflammatory activities. Murine macrophage (RAW264.7 cells) was used to establish as an in vitro model of pseudorabies virus-induced reactive oxygen species (ROS) response. In this study, antioxidant activities of Alga were measured based on 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging, trolox equivalent antioxidant capacity assays, reducing power, and virus-induced ROS formation in RAW264.7 cells. Anti-inflammatory activities of Alga were assessed by its ability to inhibit the production of interleukin-6 and nitric oxide (NO) using enzyme-linked immunosorbent assay, then the Janus kinase/signal transducer and activator of transcription (JAK/STAT) pathway was investigated by measuring the inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), nuclear factor-κB (p50 and p65), JAK, STAT-1/3, and suppressor of cytokine signaling 3 (SOCS3) by Western blotting. In addition, Alga inhibited virus replication by plaque assay. Our results showed that the Alga had high antioxidant activity, significantly reduced the virus-induced accumulation of ROS, and inhibited the levels of nitric oxide and interleukin-6. Further studies revealed that Alga also downregulated the gene and protein expressions of iNOS, COX-2, nuclear factor-κB (p50 and p65), and the JAK/STAT pathway. The inhibitory effects of Alga were similar to pretreatment with specific inhibitors of JAK and STAT-3 in pseudorabies virus -infected RAW264.7 cells. Alga enhanced the expression of SOCS3 to suppress the activity of the JAK/STAT signaling pathway in pseudorabies virus-infected RAW264.7 cells. In addition, Alga has decreased viral replication (p < 0.005) at an early stage. Therefore, our results demonstrate that Alga inhibits ROS, interleukin6, and nitric oxide production via suppression of the JAK/STAT pathways and enhanced the expression of SOCS3 in virus-infected RAW264.7 cells.

ABSTRACT Calla lily chlorotic spot virus (CCSV) isolated from central Taiwan was recently identified as a tospovirus serologically but distantly related to Watermelon silver mottle virus (WSMoV). To clarify the serological relationship between the two viruses, rabbit polyclonal antibody (PAb) to CCSV and mouse monoclonal antibodies (MAbs) to WSMoV NP or CCSV NP were produced in this investigation, using purified nucleocapsid protein (NP) as immunogens. The PAb to CCSV NP reacted stronger with the homologous antigen than with the heterologous antigen, with much lower A(405) readings in indirect enzyme-linked immunosorbent assay (ELISA) and low-intensity banding in immunoblotting. MAbs produced to CCSV NP or WSMoV NP reacted specifically with the homologous antigens but not with the heterologous antigens in both ELISA and immunoblot analyses. The CCSV S RNA was determined to be 3,172 nucleotides in length, with an inverted repeat at the 5' and 3' ends and two open reading frames encoding the NP and a nonstructural (NSs) protein in an ambisense arrangement. A typical 3'-terminal sequence (5'-AUUGCUCU-3') that is shared by all members of the genus Tospovirus also is present in the CCSV S RNA. The CCSV NP and NSs protein share low amino acid identities of 20.1 to 65.1% and 19.9 to 66.1%, respectively, with those of reported tospoviruses. Phylogenetic dendrogram analysis indicates that CCSV is a distinct member in the genus Tospovirus. The results provide evidence that CCSV is a new species in the genus Tospovirus and belongs to WSMoV serogroup.

Slip and fall incidences are common in our daily lives. They are not only important environmental safety issues but also important occupational safety and health problems. The purpose of this study was to use the Brungraber Mark II to measure the friction so as to investigate the effects of the shoe sole, surface condition and the inclined angle of the floor and their interactions on friction coefficient. The results of the study showed the effects of all the main factors and their interactions were significant (p<0.001). Engineering designs & ergonomic interventions in slip & fall prevention should take these factors in full consideration.

AtMAPR5/MSBP1 and its homologs can be ubiquitinated in the absence of E3 ligase in in vitro ubiquitination assays. Ubiquitinated AtMAPR3, AtMAPR5/MSBP1, and AtMAPR2 were identified using LC-MS/MS. Analysis of trypsin-released signature peptides showed that this E3-independent ubiquitination of AtMAPR3, AtMAPR5/MSBP1, and AtMAPR2 was dominated by mono-ubiquitination at multiple sites. Unlike AtUBC8-type E2s, AtUBC36 was not able to transfer ubiquitin to AtMAPR2. The truncated mutants AtMAPR2Δ1-10, AtMAPR2Δ1-30, and AtMAPR2_1-73 could also be ubiquitinated. The presence of a ubiquitin-binding domain (UBD) allows proteins to be ubiquitinated independently of E3 ligases. However, AtMAPRs do not contain any known UBD. In vitro ubiquitination of AtMAPR2 observed in this study will be further studied in biochemical and physiological aspects.

Chitosan/poly(acrylic acid)/poly(ethylene glycol) diacrylate (PEGDA) composite membranes with a bi-layer configuration were prepared and their potential application as an antibacterial material was examined. A two-step process was adopted. A dope consisting of PEGDA, acrylic acid (AA) and a photoinitiator was cast and then UV-cured on a glass substrate to form a mechanically strong, dense membrane. Subsequently, the membrane was coated with a layer of solution composed of chitosan, AA and water. As the majority of AA diffused downwards into the supporting layer underneath, chitosan coagulated with residual AA to form a nano-layer on the top surface by means of UV irradiation. Low-voltage field-emission scanning electron microscopy was used to observe the membrane morphology and to measure the thickness of the top layer. Contact angle measurements indicated a top layer mixed with chitosan and poly(acrylic acid), which was confirmed by chemical composition analysis of X-ray photon spectroscopy. The antibacterial activities of the formed membranes were tested both with respect to a Gram-negative (Escherichia coli) and a Gram-positive (Staphylococcus aureus) bacteria.
