RESUMEN

Microplastic particles (MP) and nanoplastic particles (NP) as persistent anthropogenic pollutants may impact environmental and human health. A relevant potential source of primary MP and NP is water-based dispersion paint which are commonly used in any household. Given the worldwide high application volume of dispersion paint and their diverse material composition MP and NP may enter the environment with unforeseeable consequences. In order to understand the relevance of these MP and NP from paint dispersion we investigated the components of two representative wall paints and analyzed their composition in detail. The different paint components were then investigated for their impact on the model organism Daphnia magna and on a murine cell line. Plastic NP, dissolved polymers, titanium dioxide NPs, and calcium carbonate MPs demonstrated adverse effects in both biological test systems, indicating detrimental consequences of several typical components of wall paints upon release into the environment. The outcome of this study may form the basis for the evaluation of impact on other organisms, environmental transport and impact, other related technical materials and for the development of strategies for the prevention of potential detrimental effects on organisms.

OBJECTIVE: Research documents that even subtle changes in visible skin condition affect perceptions of age, health, and attractiveness. There is evidence that hair quality also affects the assessment of physical appearance, as variations in hair diameter, hair density, and hair style have systematic effects on perception. Here, we consider combined effects of hair color and skin color on the perception of female physical appearance. METHODS: In two experiments, we digitally manipulated facial skin color of lightly-pigmented, young women, both between-subjects (Experiment 1) and within-subjects (Experiment 2), and investigated possible interactions with hair color in regard to age, health, and attractiveness perception. RESULTS: In both experiments, we detected hair color and skin color interaction effects on men's and women's assessments. For between-subjects comparisons, participants with lighter hair color were judged to be younger than those with darker shades; this effect was more pronounced in women with light skin color. No such effect was observed for within-subjects variation in skin color. Both experiments showed that smaller perceived contrast between hair color and skin color resulted in more positive responses. CONCLUSION: We conclude that hair color and facial skin color together have an effect on perceptions of female age, health, and attractiveness in young women, and we discuss these findings with reference to the literature on the role of hair and skin in the assessment of female physical appearance. Fistulas between the upper intestinal tract and the airway following esophagectomy are a rare and severe complication with significant mortality. Treatment and therapy are difficult and require a multidisciplinary approach. The objective of this retrospective study was to identify risk factors for these fistulas following esophagetcomy, and to assess their impact on the further clinical course and outcome. 211 patients undergoing Ivor-Lewis esophagectomy for esophageal cancer between 2005 and 2012 were included. The preoperative risk factors including the risk score according to Schröder et al. and the O-Physiological and Operative Severity Score (POSSUM) score, operative and postoperative parameters and the outcome were evaluated. 65% of all patients developed postoperative complications, including 12 patients that developed fistulas between the upper intestinal tract and the airway (airway fistulas [AF]; 5.6%). Neither patient related risk factors nor esophagus-specific risk scores correlated with occurrence of AF. Furthermore, surgical treatment and neoadjuvant treatment did not show any effect on development of AF in our patients. However, we could demonstrate that AF significantly impacted on length of hospital stay (AF 52 days vs. No-AF group 16 days, P < 0.001), incidence of major pulmonary complications (83.3% vs. 17.1%, P < 0.001), 90-day mortality (42% vs. 7.5%, P = 0.002) and overall survival (133 days vs. 636 days, P=0.029). With the current study, we could not identify any patient related risk factors, esophagus-specific risk scores or treatment related details that might be useful as predictors of AF after Ivor-Lewis esophagectomy. However, we confirmed that AF significantly impacted on outcomes. This highlights the urgent need for further studies on this rare but devastating complication after esophagectomy.

Electrospun nanofibrous membranes (ENM) possess many advantages over commonly utilized water purification systems. They provide high porosity with interconnected pores and a high surface to volume ratio, facilitating particle adsorption. Affinity separation moves into a promising future for application, for example, nanoparticle adsorption with excellent filtration efficiency, because of its highly specific adsorption mechanism. However, not all effects on filtration performance are entirely understood. In this paper, we investigate significant filtration parameters, such as pore size, mechanical stability, and hydrophilicity, and determine a sequence of importance for an optimal pressure drop. Copolymers with various hydrophilic functional groups such as acid, amide, pyridine, and quaternary amine were utilized. Effects on the pressure drop or nanoparticle filtration efficiency can then easily be attributed to the corresponding functional group. UV-light was used to induce cross-linking in the membranes, which subsequently surpassed the mechanical stability of commonly used hydrophobic membranes. A maximum tensile-stress of up to 11.6 MPa was obtained, whereby an optimization of at least 22% was achieved. Moreover, these cross-links reduce fiber swelling by a maximum of 26%. The membrane potential depends on the different functional groups and their incorporation number from 10 to 50 mol %. Successful gold nanoparticle (AuNP) filtration in flow mode was demonstrated and highlighted the outstanding membrane properties and selectivity. The Nplus membrane achieved 100% filtration efficiency over a duration of 6 min, surpassing the Pyr membrane's performance. This was attributed to the ionic interaction of the Nplus membrane, in contrast with the physical adsorption of the Pyr membrane.

In nature, photoreceptor proteins undergo molecular responses to light, that exhibit supreme fidelity in time and space and generally occur under mild reaction conditions. To unlock these traits for material science, the light-induced homodimerization of light-oxygen-voltage (LOV) photoreceptors is leveraged to control the assembly of gold nanoparticles. Conjugated to genetically encodable LOV proteins, the nanoparticles are monodispersed in darkness but rapidly assemble into large aggregates upon blue-light exposure. The study establishes a new modality for reaction control in macromolecular chemistry and thus augurs enhanced precision in space and time in diverse applications of gold nanoparticles.

In order to circumvent the limited access and donor variability of human primary alveolar cells, directed differentiation of human pluripotent stem cells (hiPSCs) into alveolar-like cells, provides a promising tool for respiratory disease modeling and drug discovery assays. In this work, a unique, miniaturized 96-Transwell microplate system is described where hiPSC-derived alveolar-like cells were cultured at an air-liquid interface (ALI). To this end, hiPSCs were differentiated into lung epithelial progenitor cells (LPCs) and subsequently matured into a functional alveolar type 2 (AT2)-like epithelium with monolayer-like morphology. AT2-like cells cultured at the physiological ALI conditions displayed characteristics of AT2 cells with classical alveolar surfactant protein expressions and lamellar-body like structures. The integrity of the epithelial barriers between the AT2-like cells was confirmed by applying a custom-made device for 96-parallelized transepithelial electric resistance (TEER) measurements. In order to generate an IPF disease-like phenotype in vitro, the functional AT2-like cells were stimulated with cytokines and growth factors present in the alveolar tissue of IPF patients. The cytokines stimulated the secretion of pro-fibrotic biomarker proteins both on the mRNA (messenger ribonucleic acid) and protein level. Thus, the hiPSC-derived and cellular model system enables the recapitulation of certain IPF hallmarks, while paving the route towards a miniaturized medium throughput approach of pharmaceutical drug discovery.

Massive declines in insect biodiversity and biomass are reported from many regions and habitats. In urban areas, creation of native wildflower meadows is one option to support insects and reduce maintenance costs of urban green spaces. However, benefits for insect conservation may depend on previous land use, and the size and location of new wildflower meadows. We show effects of conversion of roadside plantings-from exotic shrubs into wildflower meadows-on (1) the abundance of 13 arthropod taxa-Opiliones, Araneae, Isopoda, Collembola, Orthoptera, Aphidoidea, Auchenorrhyncha, Heteroptera, Coleoptera, Nematocera, Brachycera, Apocrita, Formicidae-and (2) changes in maintenance costs. We assessed the influence of vegetation type (meadow vs. woody), meadow age, size, location (distance to city boundary), and mowing regime. We found many, but not all, arthropod taxa profiting from meadows in terms of arthropod activity abundance in pitfall traps and arthropod density in standardized suction samples. Arthropod number in meadows was 212% higher in pitfall traps and 260% higher in suction samples compared to woody vegetation. The increased arthropod number in meadows was independent of the size and isolation of green spaces for most taxa. However, mowing regime strongly affected several arthropod taxa, with an increase of 63% of total arthropod density in unmown compared to mown meadow spots. Costs of green space maintenance were fivefold lower for meadows than for woody vegetation. Our study shows that (1) many different arthropod taxa occur in roadside vegetation in urban areas, (2) replacement of exotic woody vegetation by native wildflower meadows can significantly increase arthropod abundance, especially if meadow management permits temporarily unmown areas, and (3) maintenance costs can be considerably reduced by converting woody plantings into wildflower meadows. Considering many groups of arthropods, our study provides new insights into possible measures to support arthropods in urban environments.

Patients with metastatic prostate cancer (PCa) have a poorer prognosis than patients with organ-confined tumors. We strove to uncover the proteome signature of primary PCa and associated lymph node metastases (LNMs) in order to identify proteins that may indicate or potentially promote metastases formation. We performed a proteomic comparative profiling of PCa tissue from radical prostatectomy (RPE) of patients without nodal metastases or relapse at the time of surgical resection (n=5) to PCa tissue from RPE of patients who suffered from nodal relapse (n=5). For the latter group, we also included patient-matched tissue of the nodal metastases. All samples were formalin fixed and paraffin embedded. We identified and quantified more than 1200 proteins by liquid chromatography tandem mass spectrometry with subsequent label-free quantification. An increase of ribosomal or proteasomal proteins in LNM (compared to corresponding PCa) became apparent, while extracellular matrix components rather decreased. Immunohistochemistry (IHC) corroborated accumulation of poly-(ADP-ribose)-polymerase 1 and N-myc-downstream-regulated-gene 3, alpha/beta hydrolase domain-containing protein 11, and protein phosphatase slingshot homolog 3 in LNM. These findings strengthen the present interest in examining PARP inhibitors for the treatment of aggressive PCa. IHC also corroborated increased abundance of retinol dehydrogenase 11 in metastasized primary PCa compared to organ-confined PCa. Generally, metastasizing primary tumors were characterized by an enrichment of proteins involved in cellular lipid metabolic processes with concomitant decrease of cell adhesion proteins. This study highlights the usefulness of a combined proteomic-IHC approach to explore novel aspects in tumor biology. Our initial results open novel opportunities for follow-up studies.
