RESUMEN

Chalcogel represents a unique class of meso- to macroporous nanomaterials that offer applications in energy and environmental pursuits. Here, the synthesis of an ion-exchangeable amorphous chalcogel using a nominal composition of K2CoMo2S10 (KCMS) at room temperature is reported. Synchrotron X-ray pair distribution function (PDF), X-ray absorption near-edge structure (XANES), and extended X-ray absorption fine structure (EXAFS) reveal a plausible local structure of KCMS gel consisting of Mo5+ 2 and Mo4+ 3 clusters in the vicinity of di/polysulfides which are covalently linked by Co2+ ions. The ionically bound K+ ions remain in the percolating pores of the Co-Mo-S covalent network. XANES of Co K-edge shows multiple electronic transitions, including quadrupole (1s→3d), shakedown (1s→4p + MLCT), and dipole allowed 1s→4p transitions. Remarkably, despite a lack of regular channels as in some crystalline solids, the amorphous KCMS gel shows ion-exchange properties with UO2 2+ ions. Additionally, it also presents surface sorption via [S∙∙∙∙UO2 2+] covalent interactions. Overall, this study underscores the synthesis of quaternary chalcogels incorporating alkali metals and their potential to advance separation science for cations and oxo-cationic species by integrating a synergy of surface sorption and ion-exchange.

An efficient synthesis of 3-amino-2-formyl-functionalized benzothiophenes by a domino reaction protocol and their use to synthesize a library of novel scaffolds have been reported. Reactions of ketones and 1,3-diones with these amino aldehyde derivatives formed a series of benzothieno[3,2-b]pyridine and 3,4-dihydro-2H-benzothiopheno[3,2-b]quinolin-1-one, respectively. A plausible mechanism for the formation of fused pyridine derivatives by the Friedlander reaction has been elucidated by density functional theory (DFT) calculations. Furthermore, hydrazones were obtained by reacting the aldehyde functional group of benzothiophenes with different hydrazine derivatives. Preliminary screening of these compounds against several bacterial strains and cancer cell lines led to the discovery of several hit molecules. Hydrazone and benzothieno[3,2-b]pyridine derivatives are potent cytotoxic and antibacterial agents, respectively. One of the potent compounds effected ∼97% growth inhibition of the LOX IMVI cell line at 10 µM concentration.

Effective removal of chemically toxic selenium oxoanions at high-capacity and trace levels from contaminated water remains a challenge in current scientific pursuits. Here, we report the functionalization of the MgAl layered double hydroxide with molybdenum-oxysulfide (MoO2S2) anion, referred to as LDH-MoO2S2, and its potential to sequester SeVIO42- and SeIVO32- from aqueous solution. LDH-MoO2S2 nanosheets were synthesized by an ion exchange method in solution. Synchrotron X-ray pair distribution function (PDF) and extended X-ray absorption fine structure (EXAFS) revealed an unexpected transformation of the MoO2S22- to Mo2O2S62- like species during the intercalation process. LDH-MoO2S2 is remarkably efficient in removing SeO42- and SeO32- ions from the ppm to trace level (≤10 ppb), with distribution constant (Kd) ranging from 104 to 105 mL/g. This material showed exceptionally high sorption capacities of 237 and 358 mg/g for SeO42- and SeO32-, respectively. Furthermore, LDH-MoO2S2 demonstrates substantial affinity and efficiency to remove SeO32-/SeO42- even in the presence of competitive ions from contaminated water. Hence, the removal of selenium (VI/IV) oxoanions collectively occurs through reductive precipitation and ion exchange mechanisms. This work provides significant insights into the chemical structure of the MoO2S2 anion into LDH and emphasizes its exceptional potential for high-capacity selenium removal and positioning it as a premier sorbent for selenium oxoanions.

Dopamine receptors are therapeutic targets for the treatment of various neurological and psychiatric disorders, including Parkinson's and Alzheimer's. Previously, PF-06649751 (tavapadon), PF-2562 and PW0464 have been discovered as potent and selective G protein-biased D1/D5 receptor agonists with optimal pharmacokinetic properties. However, no selective D5R agonist has been reported yet. In this context, we designed and synthesized forty non-catecholamines-based pyrimidine derivatives and identified four pyrimidine derivatives as selective D5R partial agonists. Using cAMP-based GloSensor assay in transiently transfected HEK293T cells with human D1 or D5 receptors, we discovered that compound 5c (4-(4-bromophenyl)-6-(2,4,5-trimethoxyphenyl)pyrimidin-2-amine) exhibited modest D5R agonist activity. This leads us to explore various modifications of this scaffold to improve the D5 agonist potency and efficacy. Using molecular docking, and rational design followed by their evaluation at D1 and D5 receptors for agonist activity, we identified three new derivatives, 5j, 5h, and 5e. The most potent compound of this series 5j (4-(4-iodophenyl)-6-(2,4,5-trimethoxyphenyl)pyrimidin-2-amine), exhibited EC50 of 269.7 ± 6.6 nM. Mice microsomal stability studies revealed that 5j is quite stable (>70 % at 1 hr). Furthermore, pharmacokinetic analysis of 5j (20 mg/kg, p.o) in C57BL/6j mice showed that 5j is readily absorbed via oral route of dosing and also enters into the brain (plasma Tmax: 1 h, Cmax: 51.10 ± 13.51 ng/ml; Brain Tmax: 0.5 h, Cmax: 22.54 ± 4.08 ng/ml). We further determined the in-vivo effect of 5j on cognition in scopolamine-induced amnesia in C57BL/6j mice. We observed that 5j (10 mg/kg, p.o) alleviated scopolamine-induced impairment in short-term memory and social recognition, which were blocked by D1/D5 antagonist SCH23390 (0.1 mg/kg, i.p.). Furthermore, 5j did not exhibit any cytotoxicity (up to 10 µM) or in vivo acute toxicity up to 200 mg/kg (p.o). These results strongly suggest that 5j could be further developed for treating neurological disorders wherein the D5 receptors play pivotal roles.

A barrier to meeting the goal of universal health coverage in India is the inequality in utilisation of health services between indigenous and non-indigenous people. This study aimed to explore the determinants of utilisation, or non-utilisation, of public healthcare services among the Santals, an indigenous community living in West Bengal, India. The study holistically explored the utilisation of public healthcare facilities using a framework that conceptualised service coverage to be dependent on a set of determinants - viz. the nature and severity of the ailment, availability, accessibility (geographical and financial), and acceptability of the healthcare options and decision-making around these further depends on background characteristics of the individual or their family/household. This cross-sectional study adopts ethnographic approach for detailed insight into the issue and interviewed 422 adult members of Santals living in both rural (Bankura) and urban (Howrah) areas of West Bengal for demographic, socio-economic characteristics and healthcare utilisation behaviour using pre-tested data collection schedule. The findings revealed that utilisation of the public healthcare facilities was low, especially in urban areas. Residence in urban areas, being female, having higher education, engaging in salaried occupation and having availability of private allopathic and homoeopathic doctors in the locality had higher odds of not utilising public healthcare services. Issues like misbehaviour from the health personnel, unavailability of medicine, poor quality of care, and high patient load were reported as the major reasons for non-utilisation of public health services. The finding highlights the importance of improving the availability and quality of care of healthcare services for marginalised populations because these communities live in geographically isolated places and have low affordability of private healthcare. The health programme needs to address these issues to improve the utilisation and reduce the inequality in healthcare utilisation, which would be beneficial for all segments of Indian population.

Dust pollution is common in Indian roads and several industrial settings (including mines) that affects human health. Identification and characterization of the dust particles in the mining area is essential for knowing the properties of the dust that effectively causes ailments to humans, particularly among workers those who are working in unorganized industrial settings. The present study aimed to determine the level of dust pollution and to know the size and characterize the dust particles in the Pachami-Hatgacha stone mine areas of Birbhum district, West Bengal, India. Dust samples were collected and analysed for Dynamic Light Scattering (DLS) to determine the size and shape of the particles, Fourier Transform Infrared Spectroscopy (FT-IR) to determine the free silica content, and X-ray Florence (XRF) analysis for quantitative estimation of components in the sample. All the analyses were done following standard instrumentation and techniques. The size of the dust particles was much less (ranges 101-298 nm) than the size of respirable particles (2500 nm). Those were mostly generated as well as precipitated during peak working hours of the day. Presence of considerable amounts of silica was confirmed by the FT-IR (strong and broad band at 1000 cm-1) and XRF analysis (76.85% SiO2). Exposure to these dust particles may cause severe health impairments. Therefore, interventions like wet drilling and blasting, sprinkling of water during peak working hours, and awareness of use of personal protective devices among workers are required to reduce the risk and hazards associated with dust pollution to the health of miners and inhabitants around the mines.

Two thermodynamically metastable polymorphs of vanadium(III) phosphate, VIIIPO4-m1 and VPO4-m2, have been obtained via reduction of ß-VVOPO4 by moist hydrogen. The XRPD pattern of VPO4-m1 can be assigned based on the crystal structure of ß-VVOPO4, though with distinctly different lattice parameters (VPO4-m1/ß-VOPO4: Pnma, a = 7.3453(12)/7.7863(5) Å, b = 6.4001(12)/6.1329(3) Å, c = 7.3196(13)/6.9673(5) Å). The XRPD pattern of VPO4-m2 was found to be very similar to that of Fe2(VO)(P2O7)(PO4) (VPO4-m2: P21/m, Z = 2, a = 8.792(4) Å, b = 5.269(2) Å, c = 10.398(6) Å, ß = 112.60(4)°). The crystal structure models for VPO4-m1 and VPO4-m2 have been optimized by DFT calculations. Polymorph m1 contains the unprecedented butterfly shaped [VIIIO4] chromophore and has been further characterized by magnetic measurements, by powder reflectance spectroscopy (NIR/vis/UV), and IR spectroscopy. For six polymorphic forms of VPO4 (m1', m1'', m2, m3, m4, and m5), DFT calculations have been performed. For the existence of VPO4-m1', -m1'', and -m2, our experiments provide evidence. VPO4-m3, -m4, and -m5 were obtained by structure optimization based on reduced ß-VOPO4. Their stability is predicted by the DFT calculations.

Removal of chromate (CrO42-) and pertechnetate (TcO4-) from the Hanford Low Activity Waste (LAW) is beneficial as it impacts the cost, life cycle, operational complexity of the Waste Treatment and Immobilization Plant (WTP), and integrity of vitrified glass for nuclear waste disposal. Here, we report the application of [MoIV3S13]2- intercalated layer double hydroxides (LDH-Mo3S13) for the removal of CrO42- as a surrogate for TcO4-, from ppm to ppb levels from water and a simulated LAW off-gas condensate of Hanford's WTP. LDH-Mo3S13 removes CrO42- from the LAW condensate stream, having a pH of 7.5, from ppm (∼9.086 × 104 ppb of Cr6+) to below 1 ppb levels with distribution constant (Kd) values of up to ∼107 mL/g. Analysis of postadsorbed solids indicates that CrO42- removal mainly proceeds by reduction of Cr6+ to Cr3+. This study sets the first example of a metal sulfide intercalated LDH for the removal of CrO42-, as relevant to TcO4-, from the simulated off-gas condensate streams of Hanford's LAW melter which contains highly concentrated competitive anions, namely F-, Cl-, CO32-, NO3-, BO33-, NO2-, SO42-, and B4O72-. LDH-Mo3S13's remarkable removal efficiency makes it a promising sorbent to remediate CrO42-/TcO4- from surface water and an off-gas condensate of nuclear waste.

Enterococci and methicillin-resistant S. aureus (MRSA) are among the menacing bacterial pathogens. Novel antibiotics are urgently needed to tackle these antibiotic-resistant bacterial infections. This article reports the design, synthesis, and antimicrobial studies of 30 novel pyrazole derivatives. Most of the synthesized compounds are potent growth inhibitors of planktonic Gram-positive bacteria with minimum inhibitory concertation (MIC) values as low as 0.25 µg/mL. Further studies led to the discovery of several lead compounds, which are bactericidal and potent against MRSA persisters. Compounds 11, 28, and 29 are potent against S. aureus biofilms with minimum biofilm eradication concentration (MBEC) values as low as 1 µg/mL.

This work describes disposable plasma generators made from metallized paper. The fabricated plasma generators with layered and patterned sheets of paper provide a simple and flexible format for dielectric barrier discharge to create atmospheric plasma without an applied vacuum. The porosity of paper allows gas to permeate its bulk volume and fuel plasma, while plasma-induced forced convection cools the substrate. When electrically driven with oscillating peak-to-peak potentials of ±1 to ±10 kV, the paper-based devices produced both volume and surface plasmas capable of killing microbes. The plasma sanitizers deactivated greater than 99% of Saccharomyces cerevisiae and greater than 99.9% of Escherichia coli cells with 30 s of noncontact treatment. Characterization of plasma generated from the sanitizers revealed a detectable level of UV-C (1.9 nW⋅cm-2⋅nm-1), modest surface temperature (60 °C with 60 s of activation), and a high level of ozone (13 ppm with 60 s of activation). These results deliver insights into the mechanisms and suitability of paper-based substrates for active antimicrobial sanitization with scalable, flexible sheets. In addition, this work shows how paper-based generators are conformable to curved surfaces, appropriate for kirigami-like "stretchy" structures, compatible with user interfaces, and suitable for sanitization of microbes aerosolized onto a surface. In general, these disposable plasma generators represent progress toward biodegradable devices based on flexible renewable materials, which may impact the future design of protective garments, skin-like sensors for robots or prosthetics, and user interfaces in contaminated environments.

BACKGROUND: Undernutrition is a serious health problem and highly prevalent in developing countries. There is no as such confirmatory test to measure undernutrition. The objective of the present study is to determine a new Composite Score using anthropometric measurements. Composite Score was then compared with other methods like body mass index (BMI) and mid-upper arm circumference (MUAC) classification, to test the significance of the method. METHODS: Anthropometric data were collected from 780 adult Oraon (Male = 387, Female = 393) labourers of Alipurduar district of West Bengal, India, following standard instruments, and protocols. Nutritional status of the study participants were assessed by conventional methods, BMI and MUAC. Confirmatory factor analysis was carried out to reduce 12 anthropometric variables into a single Composite Score (C) and classification of nutritional status was done on the basis of the score. Furthermore, all the methods (BMI, MUAC and C) were compared and discriminant function analysis was adopted to find out the percentage of correctly classified individuals by each of the three methods. RESULT: The frequency of undernutrition was 45.9% according to BMI category, 56.7% according to MUAC category and 51.8% according to newly computed Composite Score. Further analysis showed that Composite Score has a higher strength of correct classification (98.7%), compared to BMI (95.9%) and MUAC (96.2%). CONCLUSION: Therefore, anthropometric measurements can be used to identify nutritional status in the population more correctly by calculating Composite Score of the measurements and it is a non-invasive and relatively correct way of identification.

Physics-informed neural networks (PINNs) are an emerging technology that can be used both in place of and in conjunction with conventional simulation methods. In this paper, we used PINNs to perform a forward simulation without leveraging known data. Our simulation was of a 2D natural convection-driven cavity using the vorticity-stream function formulation of the Navier-Stokes equations. We used both 2D simulations across the x and z domains at constant Rayleigh (Ra) numbers and 3D simulations across the x, z and Ra domains. The 3D simulation was tested for a PINN's ability to learn solutions in a higher-dimensional space than standard simulations. The results were validated against published solutions at Ra values of 10 3 , 10 4 , 10 5 , and 10 6 . Both the 2D simulations and 3D simulations successfully matched the expected results. For the 2D cases, more training iterations were needed for the model to converge at higher Ra values (10 5 and 10 6 ) than at lower Ra (10 3 and 10 4 ) indicating increased nonlinear fluid-thermal coupling. The 3D case was also able to converge but, but it required more training than any of the 2D cases due to the curse of dimensionality. These results showed the validity of standard simulations via PINNs and the feasibility of higher-order parameter space solutions that are not possible using conventional methods. They also showcased the additional computational demand associated with increasing the dimensionality of the learned parameter space.

Water constitutes an indispensable resource for global life but remains susceptible to pollution from diverse human activities. To mitigate this issue, researchers are committed to purifying water using a variety of materials to remove harmful chemicals, such as heavy metals. Layered double hydroxides (LDHs), with their intriguing, layered structure and chemical behavior, have attained substantial attention for their effectiveness in removing heavy metal cations and various inorganic oxoanions from water. To enhance the efficiency, considerable endeavors have focused on functionalizing LDHs with different chemical species. Intercalation with metal sulfides has proven to be particularly effective, facilitating heavy metal absorption through multiple mechanisms, including ion-exchange, reductive precipitation, and surface sorption. This review concentrates on the synthesis and performance of polysulfide (Sx, x = 2-5), Mo-S, and Sn-S anion intercalated LDHs for heavy metal cations and inorganic oxoanion sorption, along with their mechanisms. Furthermore, the discussion includes prospects for expanding the chemistry of metal sulfide intercalated LDHs, with existing challenges and future outlooks.

We report the inactivation of SARS CoV-2 and its surrogate-Human coronavirus OC43 (HCoV-OC43), on representative porous (KN95 mask material) and nonporous materials (aluminum and polycarbonate) using a Compact Portable Plasma Reactor (CPPR). The CPPR is a compact (48 cm3), lightweight, portable and scalable device that forms Dielectric Barrier Discharge which generates ozone using surrounding atmosphere as input gas, eliminating the need of source gas tanks. Iterative CPPR exposure time experiments were performed on inoculated material samples in 3 operating volumes. Minimum CPPR exposure times of 5-15 min resulted in 4-5 log reduction of SARS CoV-2 and its surrogate on representative material samples. Ozone concentration and CPPR energy requirements for virus inactivation are documented. Difference in disinfection requirements in porous and non-porous material samples is discussed along with initial scaling studies using the CPPR in 3 operating volumes. The results of this feasibility study, along with existing literature on ozone and CPPR decontamination, show the potential of the CPPR as a powerful technology to reduce fomite transmission of enveloped respiratory virus-induced infectious diseases such as COVID-19. The CPPR can overcome limitations of high temperatures, long exposure times, bulky equipment, and toxic residuals related to conventional decontamination technologies.

Thiazole derivatives are known for a wide range of therapeutic properties. Bisnoralcohol is an inexpensive natural product obtained by the biodegradation of sterols. This article describes an efficient synthesis of a library of thiazole-fused bisnoralcohol derivatives. These novel compounds have been studied for their antineoplastic and antibacterial properties, which led to the discovery of hit compounds with therapeutic potential. The antibacterial compound is noncytotoxic and nonhemolytic against cancer cell lines and sheep red blood cells, respectively. Several of the antineoplastic compounds showed activity against human cancer cell lines with growth inhibition at submicromolar concentration.

CRISPR-based transcriptional activation (CRISPRa) has extensive research and clinical potential. Here, we show that commonly used CRISPRa systems can exhibit pronounced cytotoxicity. We demonstrate the toxicity of published and new CRISPRa vectors expressing the activation domains (ADs) of the transcription factors p65 and HSF1, components of the synergistic activation mediator (SAM) CRISPRa system. Based on our findings for the SAM system, we extended our studies to additional ADs and the p300 acetyltransferase core domain. We show that the expression of potent transcriptional activators in lentiviral producer cells leads to low lentiviral titers, while their expression in the transduced target cells leads to cell death. Using inducible lentiviral vectors, we could not identify an activator expression window for effective SAM-based CRISPRa without measurable toxicity. The toxicity of current SAM-based CRISPRa systems hinders their wide adoption in biomedical research and introduces selection bottlenecks that may confound genetic screens. Our results suggest that the further development of CRISPRa technology should consider both the efficiency of gene activation and activator toxicity.

Despite large theoretical energy densities, metal-sulfide electrodes for energy storage systems face several limitations that impact the practical realization. Here, we present the solution-processable, room temperature (RT) synthesis, local structures, and application of a sulfur-rich Mo3S13 chalcogel as a conversion-based electrode for lithium-sulfide batteries (LiSBs). The structure of the amorphous Mo3S13 chalcogel is derived through operando Raman spectroscopy, synchrotron X-ray pair distribution function (PDF), X-ray absorption near edge structure (XANES), and extended X-ray absorption fine structure (EXAFS) analysis, along with ab initio molecular dynamics (AIMD) simulations. A key feature of the three-dimensional (3D) network is the connection of Mo3S13 units through S-S bonds. Li/Mo3S13 half-cells deliver initial capacity of 1013 mAh g-1 during the first discharge. After the activation cycles, the capacity stabilizes and maintains 312 mAh g-1 at a C/3 rate after 140 cycles, demonstrating sustained performance over subsequent cycling. Such high-capacity and stability are attributed to the high density of (poly)sulfide bonds and the stable Mo-S coordination in Mo3S13 chalcogel. These findings showcase the potential of Mo3S13 chalcogels as metal-sulfide electrode materials for LiSBs.

This paper presents a proof-of-concept study establishing effectiveness of the Active Plasma Sterilizer (APS) for decontamination in planetary protection. The APS uses Compact Portable Plasma Reactors (CPPRs) to produce surface dielectric barrier discharge, a type of cold plasma, using ambient air to generate and distribute reactive species like ozone used for decontamination. Decontamination tests were performed with pathogenic bacteria (Escherichia coli and Bacillus subtilis) on materials (Aluminum, Polycarbonate, Kevlar and Orthofabric) relevant to space missions. Results show that the APS can achieve 4 to 5 log reductions of pathogenic bacteria on four selected materials, simultaneously at 11 points within 30 min, using power of 13.2 ± 2.22 W. Spatial decontamination data shows the APS can uniformly sterilize several areas of a contaminated surface within 30 min. Ozone penetration through Kevlar and Orthofabric layers was achieved using the CPPR with no external agent assisting penetration. Preliminary material compatibility tests with SEM analysis of the APS exposed materials showed no significant material damage. Thus, this study shows the potential of the APS as a light-weight sustainable decontamination technology for planetary protection with advantages of uniform spatial decontamination, low processing temperatures, low exposure times, material compatibility and the ability to disinfect porous surfaces.

A number of novel pyrazole derivatives have been synthesized, and several of these compounds are potent antibacterial agents with minimum inhibitory concentrations as low as 0.5 µg/mL. Human cell lines were tolerant to these lead compounds, and they showed negligible hemolytic effects at high concentrations. These bactericidal compounds are very effective against bacterial growth in both planktonic and biofilm contexts. Various techniques were applied to show the inhibition of biofilm growth and eradication of preformed biofilms by lead compounds. Potent compounds are more effective against persisters than positive controls. In vivo studies revealed that lead compounds are effective in rescuing C. elegans from bacterial infections. Several methods were applied to determine the mode of action including membrane permeability assay and SEM micrograph studies. Furthermore, CRISPRi studies led to the determination of these compounds as fatty acid biosynthesis (FAB) inhibitors.

Herpes simplex virus (HSV) usually produces cytopathic effect (CPE) within 24-72 h post-infection (P.I.). Clinical isolates from recurrent HSV infections in patients on Acyclovir therapy were collected between 2016 and 2019 and tested in cell cultures for cytopathic effects and further in-depth characterization. Fourteen such isolates did not show any CPE in A549 or Vero cell lines even at 120 h P.I. However, these cultures remained positive for HSV-DNA after several passages. Sequence analysis revealed that the non-CPE isolates were all HSV-1. Analysis of the thymidine kinase gene from the isolates revealed several previously reported and two novel ACV-resistant mutations. Immunofluorescence and Western blot data revealed a low-level expression of the immediate early protein, ICP4. Late proteins like ICP5 or capsid protein, VP16 were almost undetectable in these isolates. AFM imaging revealed that the non-CPE viruses had structural deformities compared to wild-type HSV-1. Our findings suggest that these strains are manifesting an unusual phenomenon of being non-CPE herpesviruses with low level of virus protein expressions over several passages. Probably these HSV-1 isolates are evolving towards a more "cryptic" form to establish chronic infection in the host thereby unraveling yet another strategy of herpesviruses to evade the host immune system.
