RESUMEN

OBJECTIVES: Fourier Transform Infrared microspectroscopy let characterize the macromolecular composition and distribution of tissues and cells, by studying the interaction between infrared radiation and matter. Therefore, we hypothesize to exploit this analytical tool in the analysis of inflamed pulps, to detect the different biochemical features related to various degrees of inflammation. MATERIALS AND METHODS: IR maps of 13 irreversible and 12 hyperplastic pulpitis, together with 10 normal pulps, were acquired, compared with histological findings and submitted to multivariate (HCA, PCA, SIMCA) and statistical (one-way ANOVA) analysis. The fit of convoluted bands let calculate meaningful band area ratios (means ± s.d., P < 0.05). RESULTS: The infrared imaging analysis pin-pointed higher amounts of water and lower quantities of type I collagen in all inflamed pulps. Specific vibrational markers were defined for irreversible pulpitis (Lipids/Total Biomass, PhII/Total Biomass, CH2 /CH3 , and Ty/AII) and hyperplastic ones (OH/Total Biomass, Collagen/Total Biomass, and CH3 Collagen/Total Biomass). CONCLUSION: The study confirmed that FTIR microspectroscopy let discriminate tissues' biological features. The infrared imaging analysis evidenced, in inflamed pulps, alterations in tissues' structure and composition. Changes in lipid metabolism, increasing amounts of tyrosine, and the occurrence of phosphorylative processes were highlighted in irreversible pulpitis, while high amounts of water and low quantities of type I collagen were detected in hyperplastic samples.

RESUMEN

Fourier transform infrared (FTIR) microspectroscopy is considered a useful tool in the biomedical field, for analysing in situ and at cellular level, very small areas of tissues and cells, with minimal sample preparation and without the use of stains or probes. This spectroscopic technique has been successfully applied to analyse biological samples from patients affected by tumoral pathologies, with particular attention to oral cavity lesions. In this study, we describe the application of FTIR microspectroscopy to characterize and discriminate the most recurrent benign and malignant diseases of oral cavity compartment. Infrared maps were acquired on tissues affected by the following pathologies: squamous cell carcinoma, adenoid cystic carcinoma, polymorphous low-grade adenocarcinoma, squamous dysplasia, keratocystic odontogenic tumor, radicular cyst, residual cyst, unicystic ameloblastoma, and ameloblastic fibroma, together with healthy tissue samples (used as control group). The epithelial and connective components of all samples were distinguished and submitted to multivariate analysis. The results were in agreement with histological suggestions.

RESUMEN

The primitive gut originates at week 3 of gestation from the endoderm, with posterior incorporation of the remaining embryo layers. Wnt, Notch and TLR4 pathways have been shown to play central roles in the correct development of the intestine. The classical hypothesis for intestinal atresia development consists of failure in bowel recanalization or a vascular accident with secondary bowel reabsorption. These have been challenged due to the high frequency of associated malformations, and furthermore, with the discovery of molecular pathways and genes involved in bowel formation and correlated defects producing atresia. Necrotizing enterocolitis (NEC) has a multifactorial pathogenesis with prematurity being the most important risk factor; therefore, bowel immaturity plays a central role in NEC. Some of the same molecular pathways involved in gut maturation have been found to correlate with the predisposition of the immature bowel to develop the pathological findings seen in NEC.

RESUMEN

The paper deals with effects of two different widespread extraction methods (conventional extraction and Soxhlet extraction) and four different pre-treatments (homogenization with pressure and with blades, sonication, and impact with glass spheres) on the extraction yields and properties of polyhydroxyalkanoate (PHA) extracted from biomass coming from an innovative process (short-cut enhanced phosphorus and PHA recovery) applied in a real wastewater treatment plant. The results show that the two different extraction processes affected the crystallization degree and the chemical composition of the polymer. On the other hand, the extractive yield was highly influenced by pre-treatments: homogenization provided a 15% more extractive yield than the others. Homogenization, especially at high pressure, proved to be the best pre-treatment also in terms of the purity, visual appearance (transparency and clearness), thermal stability, and mechanical performances of the obtained PHA films. All the PHA films begin to melt long before their degradation temperature (Td > 200 °C): this allows their use in the fields of extrusion or compression moulding. SYNOPSIS: Optimizing the extraction of PHAs from municipal wastewater gives a double beneficial environmental impact: wastewater treatment and circular bio-based carbon upgrade to biopolymers for the production of bioplastics and other intersectoral applications.

RESUMEN

OBJECTIVE: Compulsive buying disorder was first described as a psychiatric syndrome in the early twentieth century. Its classification remains elusive, and investigators have debated its potential relationship to mood, substance use, obsessive-compulsive, and impulse control disorders. The objective of this study is to present a review of compulsive buying disorder and present a case vignette. METHOD: Two databases were reviewed (Medline and PsycINFO) in search for articles published in the last 40 years. Selected terms included oniomania, compulsive buying, and compulsive shopping. Other relevant articles were also identified through reference lists. RESULTS: Compulsive buying disorder is a prevalent and chronic condition that is found worldwide, sharing commonalities with impulse control disorders. In clinical samples, women make up more than 80% of subjects. Its etiology is unknown, but neurobiologic and genetic mechanisms have been proposed. The disorder is highly comorbid with mood, substance use, eating and impulse control disorders. CONCLUSIONS: Treatment recommendations derived from the literature and clinical experience suggest that problem shoppers can benefit from psychosocial interventions. Cognitive-behavioral group models appear promising. Medication trials have reported mixed results. The identification and treatment of psychiatric comorbidity is also a key aspect of treatment. In order to determine the validity of compulsive buying disorder, future work should focus on psychopathology and neurobiological findings unique to the syndrome.

RESUMEN

Previously we demonstrated that the treatment with live Saccharomyces cerevisiae exerts beneficial therapeutic effects against vaginal candidiasis. Here, we address potential mechanisms particularly examining the probiotic capacity to modulate both fungus and host-related factors. We show that the S. cerevisiae-based probiotic markedly affects the expression of virulence traits of Candida albicans such as aspartyl proteinases (SAPs) as well as hyphae-associated proteins Hwp1 and Ece1 in the vaginal cavity. On the host side, the probiotic suppression of the influx of neutrophils caused by the fungus into the vaginas of the mice is likely related to: (1) lower production of interleukin-8; and (2) inhibition of SAPs expression. However, these neutrophils displayed reactive oxygen species hyperproduction and increased killing activity as compared to the neutrophils of placebo-treated mice. There was no evidence of any cytotoxic effect by the probiotic, either when used in vivo on vaginal epithelial cell and organ architecture, or in in vitro in human vaginal epithelium. Inactivated yeast cells did not affect any of the factors above. In summary, the data suggest that the beneficial effect exerted by this S. cerevisiae-based probiotic is the result of its interference with the expression of fungus virulence factors coupled with the modulation of the inflammatory response of the host.

RESUMEN

OBJECT: The aim of the present study was the evaluation of the effect of different polishing and finishing procedures on Filtek Z250 FZ ESPE restorative material. Particularly, the consequence of artificial aging (UV-irradiation) on this resin-based dental material was investigated determining also its outcome on cell behavior. METHODS: 96 specimens of restorative material were prepared using a light emitting diode curing unit and randomly divided into four finishing and polishing groups: (I) No treatment (FZ); (II) Identoflex rubbers (ID); (III) Enhance System (EN) and (IV) Sof-Lex Pop-on XT discs (SF). The surface morphology of native and artificially aged materials was assessed with Atomic Force Microscopy (AFM) and Scanning Electron Microscopy (SEM). FTIR and biological (biocompatibility and bacterial adhesion) analyses were also performed. RESULTS: Among all, the ID procedure represented an acceptable compromise for efficiency of polymerization and biocompatibility both before and after artificial ageing. SF and EN techniques showed better interactions with the biological environment. CONCLUSION: UV artificial ageing of the tested specimens has shown an acceleration of the surface degrading processes, favoring a possible decrease in the mechanical properties and the release of toxic free radicals. Finishing and polishing procedure seemed to affect the photodegrading pathways, even though no differences among the techniques were observed. As the cytotoxicity of materials undergoing accelerated aging is relevant, further improvement of dental restorative materials are required to limit the long-term biological damage.
