RESUMEN

Induced pluripotent stem cells (iPSCs) are a promising cell source for regenerative medicine. However, their feeder-free maintenance in undifferentiated states remains challenging. In recent past extensive studies have been directed using pristine or functionalized carbon nanotube in tissue engineering. Here we proposed thin films of functionalized carbon nanotubes (OH-single-walled CNTs [SWCNTs] and OH-multiwalled CNTs [MWCNTs]), as alternatives for the feeder-free in vitro culture of canine iPSCs (ciPSCs), considered as the cellular model. The ciPSC colonies could maintain their dome-shaped compactness and other characteristics when propagated on CNT films. Concomitantly, high cell viability and upregulation of pluripotency-associated genes and cell adhesion molecules were observed, further supported by molecular docking. Moreover, CNTs did not have profound toxic effects compared to feeder cultures as evident by cytocompatibility studies. Further, cardiac and neuronal differentiation of ciPSCs was induced on these films to determine their influence on the differentiation process. The cells retained differentiation potential and the nanotopographical features of the substrates provided positive cues to enhance differentiation to both lineages as evident by immunocytochemical staining and marker gene expression. Overall, OH-SWCNT provided better cues, maintained pluripotency, and induced the differentiation of ciPSCs. These results indicate that OH-functionalized CNT films could be used as alternatives for the feeder-free maintenance of ciPSCs towards prospective utilization in regenerative medicine.

With the rampant usage of antibiotics as growth promoters (AGPs) in poultry sector, there has been alarming concerns of antimicrobial resistant microbes such as Escherichia coli. Diversification of poultry farming due to consumer demand for safer products with higher protein content, turkey production is gaining popularity. Feed additives such as formic acid (FA) and thymol (TH) are effectively replacing AGPs due to their antimicrobial action. This directed the researchers to find alternatives to antibiotics such as thymol and formic acid because of their strong antimicrobial, anti-oxidative, digestive-stimulating properties. To assess the efficacy of FA and TH as growth promoters and their effect on the antimicrobial resistance (AMR) load, the current study (0-12 weeks) was conducted in CARI VIRAT turkey poults (n = 256; unsexed) those were randomly distributed into eight treatment groups: control(T1), AGP (T2), graded levels of FA (T3 to T5) @ 2.5, 5 and 7.5 ml/kg and TH (T6 to T8) @ 120, 240 and 350 mg/kg. Cloacal swab samples were collected at 0, 4th, 8th and 12th week interval and processed further for isolation, identification and assessment of resistance profile of E. coli. The final body weight, cumulative gain and FCR were significantly (p < 0.05) better for birds under supplementation. The Total plate count (TPC) and coliforms showcased a significant (p < 0.001) reduction in the FA and TH supplement groups as compared to control and AGP group. The resistance profile indicated E. coli isolates from AGP group with significantly (p < 0.001) highest resistivity against antibiotics (viz. chloramphenicol, tetracycline, nalidixic acid, chlortetracycline) while isolates from FA (T5) and TH (T8) groups were the least resistant. blaAmpC gene was significantly (p < 0.001) harbored in T2 isolates whereas least detected in T5 and T8. It was inferred that formic acid (7.5 ml/kg) and thymol (360 mg/kg) can effectively replace AGPs and lower AMR burden in poultry.

This study was conducted to record the ideal source and level of alkali treatment to treat jatropha meal (JM) and to determine the effect of inclusion of variously processed JM (pJM) on nutrient intake, digestibility, blood metabolites and hormonal status in goats. The JM was treated with 10 g/kg sodium chloride and 5 g/kg calcium hydroxide. The content of phorbol ester and hemagglutination (HA) activity of JM and pJM were assessed. A feeding trial for 90 days was conducted in short-haired multipurpose goats (n = 15; five per group). The experimental animals were offered oat (Avena sativa) straw ad libitum throughout the experimental period of 90 days. Each group was assigned to one of the three diets, viz. R1--soybean meal, R2--sodium chloride (10 g/kg dry matter, DM), and R3--calcium hydroxide (5 g/kg DM), with pJM substituting 250 g/kg DM of crude protein (CP) of control (R1). At the end of the feeding period, digestion trial of 7 days was conducted. Blood samples were collected at the end of the experimental period to assess the blood metabolites and hormonal status. The phorbol ester and HA activity were reduced considerably in pJM. The intake of DM, organic matter, CP, and nitrogen-free extract were comparable among all the groups. However, the intake of ether extract was significantly higher in pJM-fed groups. The hemoglobin, packed cell volume, serum urea, triiodothyronine and testosterone contents decreased in R2 and R3 as compared to R1. Concentration of glucose and activity of serum glutamic pyruvic transaminase and lactate dehydrogenase increased (P < 0.01) in goats fed pJM. It was concluded that phorbol ester content and HA activity markedly decreased by processing JM with sodium chloride and calcium hydroxide. However, they were not reduced to the levels of safe feeding, as reflected in unusual values of blood metabolites among the experimental animals.

AIM: To explore the possibilities of feeding unconventional agro-industrial byproduct for livestock production. Sugarcane press-mud (SPM), is a byproduct derived from sugarcane industry, which is rich in protein as well as minerals. The effects of dietary inclusion of SPM at different levels on the carcass characteristics of lambs were evaluated. MATERIALS AND METHODS: SPM was incorporated in concentrate mixture at different levels 0% (SP0 - concentrate mixture without SPM [Control diet]), 10% (SP10 - concentrate mixture containing 10% SPM) and 20% (SP20 - concentrate mixture containing 20% SPM). The concentrate mixtures were fed along with wheat straw for 180 days. At the end of the experimental period, six lambs per group were slaughtered to evaluate carcass and meat quality characteristics. RESULTS: No significant difference was observed in dressing percentage on pre-slaughter weight or empty body weight basis of lambs fed different levels of SPM incorporated diets. Likewise carcass weight, carcass length, and wholesale cuts appeared to have similar values among groups. The yield of visceral organs, chemical composition, and sensory attributes were not statistically affected by inclusion of SPM in the diets except juiciness of control group meat was significantly (p<0.05) higher than treatment group (SP20). CONCLUSION: The SPM can be incorporated in the diet of lambs up to the level of 20% without affecting the carcass characteristics of lambs.
