RESUMEN

In this study, Mucuna pruriens extracts were used to verify their application as a natural-based raw material with anti-inflammatory function. A nitric oxide inhibition activity assay showed that M. pruriens extracted with hot water (MW), M. pruriens extracted with 70% ethanol (ME), and M. pruriens extracted with 70% acetone (MA) presented NO inhibition activity; among them, MW and ME demonstrated the best activity and were selected for Western blot analysis. After identifying the expression patterns of inflammation-related proteins, such as inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), c-jun N-terminal kinase (JNK), extracellular signal-regulated kinase (ERK), and nuclear factor kappa-light-chain-enhancer of activated B cell (NF-κB), through Western blots, both MW and ME showed inhibition patterns. As a result of analyzing L-DOPA contained in M. pruriens through ultra-performance liquid chromatography (UPLC), high L-DOPA content was detected in MW, ME, and MA. Therefore, it can be concluded that M. pruriens extracts have the potential for use as an anti-inflammatory material.

Pectin was dissolved in deionized distilled water (2%, vol/vol) and irradiated at 20 kGy using a Co-60 gamma ray irradiator. The resulting solution was dialyzed and lyophilized. The samples were separated into three groups to estimate their antioxidant and cancer cell proliferation effects: non-irradiated (0 kGy), irradiated (20 kGy), and dialyzed (20 kGy-F, mol wt <10,000) samples. Antioxidant properties of each treatment was tested by a beta-carotene-linoleic acid bleaching assay and electron donating ability and compared for antioxidant index, which indicated that the activity was higher in the order of 20 kGy-F > 20 kGy > 0 kGy. Spleen cell survival effect of the irradiated pectin (20 kGy) and dialyzed (20 kGy-F) samples was higher than the non-irradiated control (0 kGy). The pectins inhibited growth of the cancer cell in the order of 20 kGy- F > 20 kGy > 0 kGy. The Ames test revealed that none of the fractions was mutagenic, and there was no indication of a dose-dependent response for any of the samples. These results suggest that a functional pectin oligosaccharide can be produced by irradiation for the food industry without any chemical treatment.

Aruncus dioicus var. kamtschaticus HARA, also known as goat's beard, is a native plant in Ulleung-do, South Korea. It has been used as a remedy in skin care, detoxification, blood stanching, tonsillitis. High performance liquid chromatography with diode array detection was used for partial validation of bioactive chemicals in A. dioicus ethyl acetate (EtOAc) extract, and EtOAC extract was examined for its effect on ultraviolet (UV)-induced cell aging using CCD-986sk-human skin fibroblast cells. Cells were exposed to UV-B for 1 min before extract treatment. An established viability assay was performed to test cell toxicity of A. dioicus at 5, 10, or 50 µg/ml concentrations, and activities of matrix metalloproteinase (MMP) 1, 2, 3, phosphorylated-p38 (p-p38, an activated form of p38), p38, and c-fos transcription factors were evaluated. A. dioicus extract decreased the amount of mRNA transcripts and total proteins of MMP1, 2, 3 as well as p-p38 and c-fos. The c-fos expression was also confirmed by in vivo fluorescent staining of CCD-986sk cells after UV-B exposure followed by EtOAc extract treatment. The results showed that expression of skin aging related genes encoding MMP1, 2, and 3 was inhibited by reduced transcription factor expression of p-p38 and c-fos by A. dioicus EtOAc extract. The results suggest that A. dioicus extract can be used to reduce UV-B-induced skin aging and is a potential candidate for cosmedical materials.
