RESUMEN

Illustrating the population structure and genetic diversity in selected germplasm resources (after three year multi locations trials) plays a key role which directly utilize the selection of lines in a population for accumulative trait breeding in crops. In order to further understand, the structure of population and genetic variability, we explored 100 selected lines, cultivated for three consecutive years (2016-2019) in swat, University of Malakand, Khyber Pakhtunkhwa Pakistan and Provinces of China (Chongqing and Beijing) with 33 mapped SSR markers. The integrated population structure analysis in a core of hundred germplasm with Pakistani origin with three approved commercial barley cultivars have strong stratification that allowed their division into four major subpopulations (i.e. PI, PII, PIII and PIV) and an admixture subpopulation, with 52, 9, 15 and 27 germplasm respectively. A total of 133 alleles were identified with mean value of 0.80 Polymorphic information content. The number of alleles detected by the system varied from two alleles amplified to as six with an average of 4.03 per SSR marker pair. The gene diversity ranged from 0.56 to 0.98 with an average of 0.82 in selected germplasm resources. Based on the SSR data, the 100 selected germplasm with three cultivars were classified into four main phylogenetic Linages (LI, LII, LIII and LIV) which corresponded to the phylogenic grouping in genotypes. We assembled a core set of 20 barley genotypes (~1/5 of original population size) to sustain sufficient mapping of SSR marker with Phenotype, in which we proposed four SSR markers, Bmac0040, Bmac0134, Bmag0125 and Bmag0211 for malt gene and marker (Bmac0399) for tolerance to salinity gene, which will be applicable for marker assisted breeding in barley gene resources.

Mandarins are mostly preferred specie of Citrus genus, and there has been a continuous rise in consumption and global marketing due to having easy-to-peel, attractive flavor, and fresh consumption advantages. However, most of the existing knowledge on quality traits of citrus fruit comes from research conducted on oranges, which are the main products for the citrus juice manufacturing industry. In recent years, mandarin production in Turkey surpassed orange production and took the first place in citrus production. Mandarins are mostly grown in the Mediterranean and Aegean Regions of Turkey. Due to suitable climatic conditions, they are also grown in the microclimatic condition in Rize province located in the Eastern Black Sea region. In this study, we reported the total phenolic content, total antioxidant capacity, and volatiles of 12 Satsuma mandarin genotypes selected from Rize province of Turkey. Considerable differences in the total phenolic content, total antioxidant capacity (2,2-diphenyl-1-picrylhydrazyl assay), and fruit volatile constituent were found among the 12 selected Satsuma mandarin genotypes. The total phenolic content ranged from 3.50 to 22.53 mg of gallic acid equivalent per 100 g of the fruit sample in the selected mandarin genotypes. The total antioxidant capacity was the highest in genotype HA2 as 60.40%, and followed by IB (59.15%) and TEK3 (58.36%), respectively. A total of 30 aroma volatiles were detected from the juice samples of 12 mandarin genotypes by GC/MS, which comprised six alcohols, three aldehydes (including one monoterpene), three esters, one ketone, and one other volatiles. The main volatile compounds were identified in fruits of all Satsuma mandarin genotypes as α-terpineol (0.6-1.88%), linalool (1.1-3.21%), γ-terpinene (4.41-5.5%), ß-myrcene (0.9-1.6%), dl-limonene (79.71-85.12%), α-farnesene (1.1-2.44), and d-germacrene (0.66-1.37%). Limonene accounts for most of the aroma compounds (79.71-85.12%) in fruits of all Satsuma genotypes. The genotypes MP and TEK8 had the highest total phenolic content, and HA2, IB, and TEK 3 had the highest antioxidant capacity. The YU2 genotype was found to contain more aroma compounds than the other genotypes. The genotypes selected on the basis of their high bioactive content could be used to develop new Satsuma mandarin cultivars with high human health promoting contents.

LC-ESI-MS and HPLC were used for the identification of the constituents from G. lutea leaves collected at different localities, as well as for quantification of the main compounds. Seven secoiridoids, five C-glucoflavones and three xanthones, were identified. Swertiamarin derivatives, namely eustomorusside (2), eustomoside (3) and septemfidoside (5), were detected in G. lutea for the first time. Concentrations of five constituents (swertiamarin, gentiopicrin, isovitexin, mangiferin and isogentisin) were determined. The relationship between concentrations of y-pyrones and altitude was observed with statistically significant correlation (r = 0.94). The extracts were also evaluated for their content of total phenolics, and antiradical and cytotoxic activities. The total phenolics content ranged from 7.7 to 12.7 mg GAE/g, and the IC50 values for DPPH radical scavenging activity varied between 0.45 to 2.02 mg/mL. The leaf extract exhibited moderate cytotoxic effects toward HeLa cells with an IC50 value of 41.1 microg/mL, while gentiopicrin, mangiferin and isogentisin exerted strong activity against HeLa cells, with IC50 values ranging from 5.7 to 8.8 microg/mL. The results confirm the traditional usage of G. lutea leaves and also suggest their possible utilization as hepatoprotective, hypoglycemic and anti-inflammatory agents.
