RESUMEN

In analogy to a wavelength selective switch in wavelength-division multiplexing (WDM) optical fiber communication systems, a spatial and optical mode selective switch (SMSS) would be an important component in future ultrahigh capacity optical fiber communication systems based on space and mode division multiplexing. In this work, a free-space SMSS for orbital angular momentum (OAM) mode-division multiplexing (MDM) is proposed and experimentally demonstrated. The SMSS consists of a separating part for transforming OAM modes to spatial modes and a recombining part for selecting and recombining the modes to any spatial channel. The SMSS is able to implement strictly non-blocking switching between a total of 36 SDM/MDM channels configured as four spatial channels each supporting nine OAM mode channels.

The multiplication of orbital angular momentum (OAM) modes using optical coordinate transformation is useful for OAM optical networks, but the scalability of this scheme is limited by the ray model. Here, we propose an alternative scheme for the scalable multiplication of OAM modes based on modified multi-plane light conversion (MPLC) that can extend azimuthal and radial indices of OAM modes supported by the multipliers and unlock a new degree of freedom for radial high-order OAM states that has been restricted in the zero order. The multiplication for 20 OAM modes with radial index p = 0 and 10 OAM modes with radial index p = 1 is performed in simulation and experiment. The 3-dB optical bandwidth corresponding to the purity of OAM modes covers the entire C-band experimentally. This novel, to the best of our knowledge, approach to manipulating OAM states provides valuable insights and flexible strategies for high-capacity OAM optical communication and high-dimensional optical quantum information processing.

A series of restoration measures such as municipal wastewater treatment and aquaculture closures have been implemented in Wuhan City during recent years. In order to explore the impact of restoration measures and climate change on lake water quality, long-term (2005-2021) water quality data of 47 lakes were explored to reveal spatiotemporal changes in lake water quality. Percentages of polluted lakes were calculated according to six water-quality parameters, including total phosphorus (TP), ammonia nitrogen (NH3-N), chemical oxygen demand (COD), biological oxygen demand (BOD), chemical oxygen demand using potassium permanganate as oxidant (CODMn) and petroleum contamination (PET), at interannual and monthly timescales. At the interannual timescale, percentages of COD, BOD, CODMn and PET pollution decreased significantly, suggestive of water quality improvement during recent years. At the monthly timescale, low percentages of NH3-N and BOD pollution in March 2020 probably resulted from the sharp reduction in human activities during the COVID-19 lockdown. At the monthly timescale, temperature was positively correlated with percentage of CODMn pollution, but negatively correlated with percentage of NH3-N pollution; precipitation was negatively correlated with percentage of BOD pollution. The similarity of water-quality parameters generally decreased with an increase in geographical distance between each pair of lakes. However, the regression coefficients between the similarity of lake water quality and the geographical distance decreased with time, probably resulting from enhanced similarity of water quality parameters among all lakes with rapid urbanization. Our results highlight the importance of active restoration measures for sustainable management of lakes in Wuhan City, as well as in similar developing regions.

Herpes simplex virus 1 (HSV-1) latent infection entails repression of viral lytic genes in neurons. By functional screening using luciferase-expressing HSV-1, we identify ten neuron-specific microRNAs potentially repressing HSV-1 neuronal replication. Transfection of miR-9, the most active candidate from the screen, decreases HSV-1 replication and gene expression in Neuro-2a cells. Ectopic expression of miR-9 from lentivirus or recombinant HSV-1 suppresses HSV-1 replication in male primary mouse neurons in culture and mouse trigeminal ganglia in vivo, and reactivation from latency in the primary neurons. Target prediction and validation identify transcription factors Oct-1, a known co-activator of HSV transcription, and all three Onecut family members as miR-9 targets. Knockdown of ONECUT2 decreases HSV-1 yields in Neuro-2a cells. Overexpression of each ONECUT protein increases HSV-1 replication in Neuro-2a cells, human induced pluripotent stem cell-derived neurons, and primary mouse neurons, and accelerates reactivation from latency in the mouse neurons. Mutagenesis, ChIP-seq, RNA-seq, ChIP-qPCR and ATAC-seq results suggest that ONECUT2 can nonspecifically bind to viral genes via its CUT domain, globally stimulate viral gene transcription, reduce viral heterochromatin and enhance the accessibility of viral chromatin. Thus, neuronal miR-9 promotes viral epigenetic silencing and latency by targeting multiple host transcription factors important for lytic gene activation.
