RESUMEN

A novel bacterial strain, CDC141T, was isolated from sputum samples of a patient with pulmonary infection in Hainan Province, PR China. We performed a polyphasic study to assess the taxonomic position of the new species. Based on the results of 16S rRNA gene sequence analyses, strain CDC141T belonged to the genus Nocardia with the highest sequence similarity to Nocardia nova NBRC 15556T (98.84 %) and Nocardia macrotermitis RB20T (98.54 %). The dapb1 gene sequence-based phylogenetic and phylogenomic trees further showed that the novel strain was clustered in a distinct clade adjacent to Nocardia pseudobrasiliensis DSM 44290T. The DNA G+C content of strain CDC141T was 68.57 mol%. The genomic diversity analysis revealed low average nucleotide identity and in silico DNA‒DNA hybridization values (<84.7 and <28.9 %, respectively) with its closest relative. Growth occurred at 20-40 °C, pH 6.0-9.0 and with NaCl concentrations of 0.5-2.5 % (w/v). The main fatty acids of strain CDC141T were C16 : 0, C18 : 0 10-methyl, TBSA, C16 : 1 ω6c/C16 : 1 ω7c, C18 : 1 ω9c, C18 : 0, C17 : 1 iso I/anteiso B and C17 : 0. The polar lipid profile was dominated by diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylinositol, phosphatidylinositol mannoside, unidentified glycolipids, unidentified phospholipids and unidentified lipids. MK8 (H4ω-cycl) and MK8 (H4) were the major respiratory quinones. These characteristics were consistent with the typical chemotaxonomic properties of members of the genus Nocardia. Based on the results of phenotypic and genetic analyses, strain CDC141T was identified as representing a new species of the genus Nocardia, with the proposed name Nocardia pulmonis sp. nov. (CDC141T=JCM 34955T=GDMCC 4.207T).

OBJECTIVE: To study the relationship between the copy numbers of repetitive units at variable number tandem repeat (VNTR) loci of Mycobacterium tuberculosis complex (MTBC) with its diversity of protein profiles. METHODS: The MTBC strains were subjected to genotyping using multiple locus variable number tandem repeat analysis (MLVA). Also, the principal component analysis (PCA) was performed for bacterial protein profiles of MTBC using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS). The relationship between the polymorphism of VNTR loci and PCA clustering was analyzed. RESULTS: A total of 157 MTBC strains were collected. 146 MTBC strains (MS identification score values ≥1.700) were performed PCA and three clusters, clusterⅠ(61 strains), clusterⅡ(26 strains) and cluster Ⅲ(59 strains), were generated. Polymorphic diversities were observed in 24 VNTR loci, among them, 7 were highly various, 7 were moderately, and 10 were low various. The polymorphism of Mtub39, QUB26 and QUB4156 loci were correlated with the results of MALDI-TOF MS clustering (P=0.000, P=0.035, P=0.017). CONCLUSION: The polymorphism of Mtub39, QUB26 and QUB4156 loci in MTBC was correlated with the difference of MALDI-TOF MS protein profiles, suggesting that these loci may play a role in regulating the composition of protein profiles of MTBC strains.

RESUMEN

A new composite organic oscillating reaction system based on BrO3-Ce(SO4)2-H2SO4-malonic acid/tartaric acid was proposed in this paper. On the basis of the influence of the concentration of each component on the stability and characteristic parameters of the blank system, the electrochemical fingerprints of 30 kinds of traditional Chinese medicines (TCM) were obtained. The results showed that the electrochemical fingerprint can be used for the identification of TCMs, the distinguishment of different parts and the appraisal of genuineness, which is fast, sensitive and accurate. At the same time, we explored and verified the mechanism of oscillation and the formation mechanism of TCM fingerprint.

Nickel-based hydroxides and their derivatives exhibit relatively low capacities and unsatisfactory durability as cathode materials for rechargeable alkaline batteries. In this work, a hybrid NiCo-B nanosheet cathode, integrating electrolyte ion-shuttling channels and electron-transferring networks into a metal-organic framework (MOF), was devised delicately. In the structure, the hybrid ion/electron dual pathways were constructed by NiCo-MOF frameworks and NiCo-B interpenetration networks. It revealed that nano-phase electron-transferring pathways in the MOF obviously boosted ion intercalation kinetics. The as-obtained hybrid NiCo-B nanosheets as cathode materials exhibited reversible capacity as high as 280 mA h g-1 at a current density of 1 A g-1 and excellent rate capability with a capacity retention of 78% from 1 to 10 A g-1. After 2000 charge/discharge cycles at 4 A g-1, the capacity still remained at 94% of the initial one. A full battery assembled with a hybrid NiCo-B cathode and a Fe2O3 anode showed a high capacity of 250 mA h g-1 and a considerable stability of 89% after 1000 cycles. Ragone plots indicated the highest energy density of 409 W h kg-1 and the lowest power density of 1.5 kW kg-1, outperforming other aqueous batteries. It revealed that a syngenetic structure of ion/electron hybrid dual pathways integrated into an MOF could be a potential strategy to optimize ion intercalation electrode materials for alkaline batteries.

Twenty-Five Wei'er Tea Pills (TFP), a traditional Tibetan medicine, has shown to have a promising therapeutic effect in patients with Rheumatoid arthritis (RA), as well as being safe. Nonetheless, there have been limited pharmacological studies that have explored this therapeutic option. As gut microbiota has been proven to have a critical role in the pathogenesis of RA, this study aims to explore and reveal relevant ways by which TFP interacts with the chemical crosstalk between the gut microbiome and its host. 16S rRNA sequencing, combined with un-targeted metabolomics, were conducted on collagen-induced arthritis (CIA) rats. CIA model rats treated with TFP showed significant improvement in weight gain, pathological phenomena in joints, as well as decreased serum levels of TNF-α, IL-6 and increased level of IL-4 and IL-10. Significant dysfunction in the gut microbiome and alteration in serum metabolites were observed in CIA model rats, which were restored by TFP treatment. Coherence analysis indicated that TFP modulated the pathways of histidine metabolism, phenylalanine metabolism, alanine, aspartate, glutamate metabolism, amino sugar and nucleotide sugar metabolism owing to the abundances of Lactobacillus, Bacteroides, Prevotellaceae_UCG-001 and Christensenellaceae_R-7_group in the gut microflora. The corresponding metabolites involved L-histidine, histamine, phenylethylamine, asparagine, L-aspartic acid, D-fructose 1-phosphate, D-Mannose 6-phosphate, D-Glucose 6-phosphate, and Glucose 1-phosphate. In conclusion, this study reveals the ameliorative effects of TFP on RA through the chemical crosstalk that exists between the gut microbiota and its host, and also further enriches our understandings of the pathogenesis of RA.

OBJECTIVE: To investigate the effect of the lysogenic phage Ppa3094 on the biofilm formation of PA3094. METHODS: The modified plate culture method was used to established the biofilm model in vitro. The viable counts of bacteria in biofilm were detected by MTT method; The real-time RT-PCR was applied to measure the expression level of algC and algD during the biofilm formtion of PA3094 and PA3094-L. RESULTS: Biofilm of both strains were mature at 5th to 7th day. The structures of the biofilms were both like pellicle. There was a significant difference in the viable counts of bacteria during biofilm development between PA3094 and PA3094-L. The expression of algC and algD genes was upregulated during biofilm formation. However, the expression level of PA3094-L was lower than PA3094, especially algC at 12 h. CONCLUSION: The lysogenic phage Ppa3094 could influence the biofilm formation during its development through changing the expressing level of the alginate biosynthetic genes.

A total of 50 clinical imipenem-resistant isolates of Pseudomonas aeruginosa and Acinetobacter baumannii were subjected to the ceftazidime-2- mercaptoethanol -double-disk synergy test and to the PCR assays with primers specific for bla(IMP-1). After the process of sequencing the positive one to identify the results, PCR analysis was conducted with primers specific for class 1 integrons. For synergy test, 28 isolates gave positive results, among which were 27 Pseudomonas aeruginosa and Acinetobacter baumannii. Only one Pseudomonas aeruginosa was found to carry bla(IMP-1), and bla(Int1) at the same time. This is the first ascertainment of IMP-1 producing Pseudomonas aeruginosa isolate carrying bla(IntI1) in West China, which is of significance to the research on the clinical spread of these drug-resisitant genes.

AIM: Pulmonary tuberculosis (PTB) is an infectious disease with a high incidence worldwide. Previous genome-wide association studies have identified multiple susceptibility loci for pulmonary tuberculosis (PTB); however, validation of these findings is still needed. METHODS: For this study, we recruited 300 subjects with PTB and 300 healthy subjects from a Tibetan population living in near or in Xi'an, China. Association analyses of single-nucleotide polymorphisms (SNPs) in TAP2 and SEC14L2 were performed with SPSS Statistics (version 17.0), SNPStats, Haploview (version 4.2), and SHEsis software. RESULTS: We found a correction between one SNP (rs1061660) and PTB based on Chi-square or Fisher's exact tests. In the allelic model analysis, the SNPs rs1061660 in SEC14L2 gene increased PTB 1.32-fold risk (OR = 1.32, CI = 1.05-1.66, P = 0.017). In the genetic model analysis, the rs3819721 in TAP2 gene was associated with increased 1.65-fold risk in the co-dominant model and 1.67-fold risk in the over-dominant model, respectively. For the rs1061660 in SEC14L2 gene, we found it was associated with a 1.49-fold increase the risk of PTB in the dominant model and a 1.37-fold increase the risk of PTB in the log-additive model, respectively. CONCLUSION: We found that two SNPs are associated with increased PTB risk in the Chinese Tibetan population.

OBJECTIVE: To inquire about the molecular characteristics of rhlR, a Quorum Sensing gene in Pseudomonas aeruginosa (P. aeruginosa) PAO1, and to explore the immunogenicity of RhlR protein in mouse. METHODS: The rhlR gene of PAO1 was amplified by PCR and cloned into pGEX4T-1 plasmid. The recombination was expressed in E. coli BL21 (DE3) and analyzed by SDS-PAGE and Western blotting. The fusion protein (GST-RhlR) was purified by GST purification Kit and the purified protein was used to immunize mice. P. aeruginosa PA0315 was injected into mouse lung to explore the immuno-protection of the protein. RESULTS: The 726 bp DNA fragment of rhlR was amplified from PAO1 general DNA. The restriction enzyme map showed that the inserted part of rhlR-pGEX4T-1 was successfully constructed and the gene was 100% homologous to rhlR in GenBank. The recombinant plasmid expressed a 54 kDa fusion protein (rhlR-GST) in E. coli BL21 (DE3) after induction by IPTG. The fusion protein could be recognized by mouse polyvalent antiserum against P. aeruginosa. The results showed that the bacterial clearance rate in mouse lung was 86. 92% in rhlR groups and 49.44% in the control group. CONCLUSION: A 54 kDa protein (RhlR-GST) has been successfully expressed in E. coli BL21 (DE3). The RhlR could increase the bacterial clearance rate in mouse lung and may serve as immunoprotective antigen to develop the genetic engineering vaccine.

RESUMEN

A new diterpene alkaloid named delphatisine C (1) has been isolated from aerial parts of Delphinium chrysotrichum along with three known norditerpenoid alkaloids delpheline (2), delbrunine (3), and delectinine (4). Their structures were characterized on the basis of their spectral data. All of them were determined by SRB assay for their cytotoxicity, and compound (1) showed significant cytotoxic activities (IC(50)=2.36 µmol/L) against the A549 cell line.

OBJECTIVE: To investigate class I integrons and integrated gene cassettes in metalloenzyme-producing Pseudomonas aeruginosa. METHODS: A total of 68 isolated clinical strains of Pseudomonas aeruginosa were subjected to PCR analysis with primers specific for bla(IMP-1) and bla(VIM). The positive strains then underwent examination for class I integrons and integrated gene cassettes with PCR with primers specific to class I integrase ((IntI)1) and integrated gene cassettes, followed by sequence analysis for some of the positive strains. RESULTS: Only 1 isolated strain showed positive results for both bla(IMP-1) and bla IntI1 detection. Fifty-five strains were positive for bla(VIM), including 26 positive for bla (IntI)1. Of the 26 bla (IntI)1-positive strains, only 18 contained integrated gene cassettes, which were classified into 5 types according to agarose gel electrophoresis. CONCLUSION: It is the first time to identify IMP-1-producing Pseudomonas aeruginosa carring bla(Int)1 in West China. The class I integrons were widespread in these Pseudomonas aeruginosa and 69.2% of them carry the gene cassettes. These findings provide useful insights into the clinical spread of these drug-resistant genes.
