RESUMEN

As an endosymbiont, Wolbachia exerts significant effects on the host, including on reproduction, immunity, and metabolism. However, the study of Wolbachia in Thysanopteran insects, such as flower thrips Frankliniella intonsa, remains limited. Here, we assembled a gap-free looped genome assembly of Wolbachia strain wFI in a length of 1,463,884 bp (GC content 33.80%), using Nanopore long reads and Illumina short reads. The annotation of wFI identified a total of 1838 protein-coding genes (including 85 pseudogenes), 3 ribosomal RNAs (rRNAs), 35 transfer RNAs (tRNAs), and 1 transfer-messenger RNA (tmRNA). Beyond this basic description, we identified mobile genetic elements, such as prophage and insertion sequences (ISs), which make up 17% of the entire wFI genome, as well as genes involved in riboflavin and biotin synthesis and metabolism. This research lays the foundation for understanding the nutritional mutualism between Wolbachia and flower thrips. It also serves as a valuable resource for future studies delving into the intricate interactions between Wolbachia and its host.

Megalurothrips usitatus (Bagnall) is a destructive pest of legumes, such as cowpea. The biology, population dynamics and control strategies of this pest have been well studied. However, the lack of a high-quality reference genome for M. usitatus has hindered the understanding of key biological questions, such as the mechanism of adaptation to feed preferentially on high-protein host plants and the resistance to proteinase inhibitors (PIs). In this study, we generated a high-resolution chromosome-level reference genome assembly (247.82 Mb, 16 chromosomes) of M. usitatus by combining Oxford Nanopore Technologies (ONT) and Hi-C sequencing. The genome assembly showed higher proportions of GC and repeat content compared to other Thripinae species. Genome annotation revealed 18,624 protein-coding genes, including 4613 paralogs that were preferentially located in TE-rich regions. GO and KEGG enrichment analyses of the paralogs revealed significant enrichment in digestion-related genes. Genome-wide identification uncovered 506 putative digestion-related enzymes; of those, proteases, especially their subgroup serine proteases (SPs), are significantly enriched in paralogs. We hypothesized that the diversity and expansion of the digestion-related genes, especially SPs, could be driven by mobile elements (TEs), which promote the adaptive evolution of M. usitatus to high-protein host plants with high serine protease inhibitors (SPIs). The current study provides a valuable genomic resource for understanding the genetic variation among different pest species adapting to different plant hosts.

Vitrimerization of thermoset polymers plays an important role in addressing resource recovery and reuse. Vitrimer elastomers with good mechanical properties often require well-designed crosslinking agents or fillers, but this increases processing complexity or reduces vitrimer dynamic properties. In this report, a simple green strategy to build a strong vitrimer elastomer is designed. Commercially available epoxidized natural rubber (ENR) is cross-linked with biomass-derived D-Fructose 1,6-bisphosphoric acid to get a vitrimer elastomer cross-linked by ß-hydroxy phosphate ester bonds and has abundant hydrogen bonds. Hydrogen bonds can preferentially break and dissipate energy under external forces, which makes the sample robust. The topological network can be reformed at high temperatures through the dynamic exchange of ß-hydroxy phosphate ester bonds, which gives the material malleability and recyclability. In addition, through the strategy of combining reprocessing and welding, multiple shape memory effects can be achieved in one postprocessing step. Considering that a variety of commercially available epoxy polymers are easily available, it is believed that this strategy can be a simple and versatile way to enable commercial epoxy polymers to achieve green crosslinking through biomass crosslink agents, which results in robust and recyclable vitrimers based on ß-hydroxy phosphate bonds.

Patients with colorectal cancer (CRC) are at high risk of frailty, leading to reduced quality of life and survival. Diet is associated with frailty in the elderly through regulating inflammation. Thus, we hypothesized that dietary inflammatory potential (as assessed by dietary inflammatory index [DII]) might be associated with frailty in patients with CRC through regulating inflammatory biomarkers. A total of 231 patients with CRC were included in this cross-sectional study. Dietary intake was evaluated by 3-day, 24-hour dietary recalls, and frailty status was assessed in accordance with the Fried frailty criteria. Plasma inflammatory cytokines were determined in 126 blood samples. A total of 67 patients (29.0%) were frail, with significantly higher DII scores than nonfrail patients, accompanied with significantly increased interleukin-6 (IL-6) and decreased interleukin-10 (IL-10) concentrations. Each 1-point increase of DII was related to a 25.0% increased risk of frailty. IL-6 was positively correlated with frailty and DII, whereas IL-10 was negatively correlated. After adjusting for age, sex, body mass index, education level, smoking status, and energy, mediation analysis revealed that the association between DII and frailty was significantly mediated by IL-6 (average causal mediation effect [ACME], 0.052; 95% confidence interval, 0.020-0.087; P = .002) and IL-10 (ACME, 0.025; 95% confidence interval, 0.004-0.063; P = .016). The ρ values for the sensitivity measure at which estimated ACMEs were zero were 0.3 and -0.2 for IL-6 and IL-10, respectively. Therefore, a pro-inflammatory diet was associated with frailty in patients with CRC possibly in part by affecting circulating IL-6 and IL-10 concentrations.

Several trivalent and pentavalent vanadium complexes bearing 8-anilide-5,6,7-trihydroquinoline ligands were synthesized. These vanadium complexes were identified by elemental analysis, FTIR spectroscopy and NMR. Single crystals of trivalent vanadium complexes V2, V3', and V4 and pentavalent vanadium complexes V5 and V7 were further obtained and identified by X-ray single crystal diffraction. In addition, the catalytic performance of these catalysts was adjusted by controlling the electronic and steric effects of substituents in the ligands. In the presence of diethylaluminium chloride, complexes V5-V7 displayed high activity (up to 82.8 × 106 g molV-1 h-1) and good thermal stability toward ethylene polymerization. In addition, the copolymerization ability of complexes V5-V7 was evaluated, and complexes V5-V7 displayed high activity (up to 105.6 × 106 g molV-1 h-1) and high copolymerization ability toward ethylene/norbornene copolymerization. By adjusting the polymerization conditions, copolymers with norbornene insertion ratios of 8.1%-30.9% can be obtained. Complex V7 was further studied for ethylene/1-hexene copolymerization, and the obtained copolymer displayed a moderate 1-hexene insertion ratio of 1.2%. Complex V7 displayed high activity and high copolymerization ability while having thermal stability. The results showed that 8-anilide-5,6,7-trihydroquinoline ligands with fused rigid-flexible rings were beneficial for vanadium catalysts.

Increasing evidence indicates that tryptophan (Trp) metabolism disturbance controls hippocampal 5-hydroxytryptamine (5-HT) and thereby affecting depression-like behavior, in which the gut microbiota (GM) might be involved. This study investigated the effect of Trp-rich whey protein isolate (WPI) on depressive-like behavior in 4T1 tumor-bearing mice. Female BALB/c mice were subcutaneously inoculated with murine 4T1 mammary carcinoma cells and received 2 g/kg of WPI by gavage daily for 21 days. The results showed that WPI exerted no significant effects on tumor weight and volume, but abrogated tumor-induced depression-like behavior, as evidenced by remarkably increased time and distance in the center of the open-field test, decreased immobility time in the tail suspension test, increased time and number of entries to the open arms in the elevated plus maze and sucrose preference. Moreover, WPI promoted the hippocampal Trp, 5-hydroxytryptophan (5-HTP), 5-HT, 5-hydroxyindoleacetic acid (5-HIAA) levels and inhibited kynurenine (Kyn) through up-regulating serotonin transporter (SERT) and down-regulating indoleamine 2, 3-dioxygenase (IDO). WPI showed an enriched microbial diversity indicated by increased Shannon index and decreased Simpson index, reduced the abundances of Proteobacteria, Rikenellaceae_RC9_gut_group, Alloprevotella and Prevotellaceae_UCG-001, and increased the abundance of unclassified_k__norank_d__Bacteria in tumor-bearing mice (P < 0.05). At level 3, WPI enhanced the function of microbial gene related to Trp metabolism in the KEGG pathways (P < 0.05). Our results suggest that WPI exhibits a potent antidepressant-like effect via the regulation of hippocampal Trp metabolism and alteration of GM composition and function, and it may be an effective prevention for cancer-related depression.

This study investigated the associations between diet and frailty in lung cancer patients and the potential role of the gut microbiota involved. We assessed dietary intake and frailty status in 231 lung cancer patients by 3-day, 24-h dietary recalls and Fried frailty criteria, respectively, and collected 50 fecal samples for next-generation sequencing. A total of 75 (32.5%) patients were frail, which might be related to significantly lower intake of energy, protein, carbohydrate, dietary fiber, niacin, leucine, some minerals, and a poorer dietary quality as indicated by the Chinese Healthy Eating Index (p < 0.05). Among these, carbohydrate (OR = 0.98; 95% CI 0.96-0.99; p = 0.010), calcium (OR = 0.99; 95% CI 0.99-1.00; p = 0.025), and selenium (OR = 1.03; 95% CI 1.00-1.06; p = 0.022) were all significantly associated with frailty. A multivariate logistic regression analysis showed that the mean risk of frailty was 0.94 times lower (95% CI 0.90-0.99; p = 0.009) among participants with higher CHEI scores. Additionally, the frail patients demonstrated significantly lower gut microbiota ß diversity (p = 0.001) and higher relative abundance of Actinobacteriota (p = 0.033). Frailty in lung cancer patients might be associated with insufficient nutrients intake and a poor dietary quality through gut microbiota regulation.

As an economically important insect pest, the flower thrips Frankliniella intonsa (Trybom) causes great damage to host plants by directly feeding and indirectly transmitting various pathogenic viruses. The lack of a well-assembled genomic resource has hindered our understanding of the genetic basis and evolution of F. intonsa. In this study, we used Oxford Nanopore Technology (ONT) long reads and High-through chromosome conformation capture (Hi-C) linked reads to construct a high-quality reference genome assembly of F. intonsa, with a total size of 225.5 Mb and a contig N50 of 3.37 Mb. By performing the Hi-C analysis, we anchored 91.68% of the contigs into 15 pseudochromosomes. Genomic annotation uncovered 17,581 protein-coding genes and identified 20.09% of the sequences as repeat elements. BUSCO analysis estimated over 98% of genome completeness. Our study is at the first time to report the chromosome-scale genome for the species of the genus Frankliniella. It provides a valuable genomic resource for further biological research and pest management of the thrips.

Bisphenol A (BPA) is a synthetic estrogen compound, which widely exists in the environment, interferes with mammalian endocrine and affects the function of reproductive system of males. Taking fresh sperm of boar, 17 ℃ preservation boar sperm, and mouse sperm as test materials, we examined the effects of BPA (0, 0.1,1,10,100 µmol∙L-1) on proteins tyrosine phosphorylation in sperm and the molecular mechanism by using wes-tern blot (WB) and immunofluorescence techniques coupled to in vitro culture method. The results showed that low BPA concentration (0.1, 1 µmol∙L-1) markedly accelerated the protein tyrosine phosphorylation of fresh boar capacitated sperm. However, the tyrosine phosphorylation of boar sperm decreased in high BPA concentration (10, 100 µmol∙L-1). The tyrosine phosphorylation of the mouse sperm raised with the increases of BPA concentration. Moreover, BPA affected different kinds of proteins related to tyrosine phosphorylation modification of porcine and mouse sperm capacitation, suggesting that the effect of BPA exposure on mammalian sperm was species-specific. Furthermore, the results of immunofluorescence showed that the effects of BPA on protein tyrosine phosphorylation in sperm mainly occurred in the middle and principal piece of flagellum.

A covalent adaptable network can endow rubber materials with recyclability and reprocessability and is expected to alleviate black pollution caused by end-of-life rubber. However, the loss of traditional vulcanization systems severely sacrifices their strength, and the tensile strength in the current study rarely exceeds 10 MPa unless fillers are added. In this work, we proposed a self-strengthening process based on dual-dynamic units (imine and disulfide), briefly, under heating, phenylsulfur radicals generated from aromatic disulfide bonds can react with double bonds (mostly vinyl) and/or couple with allyl sites, thus reforming a stronger cross-linked network. The neighboring imine unit is not affected and provides excellent thermal reprocessability and chemical recyclability. The result shows that the tensile strength can reach 19.27 MPa via self-strengthening without adding fillers or any other additives, and this ultra-high-strength is much higher than those of all known recyclable polybutadiene-based rubber materials. In addition, the material also has malleability, shape memory, and self-welding properties. By doping carbon nanotubes, a recyclable conductive composite can also be achieved. In general, we envision that this enhanced strategy has great potential to be generalized for all elastomers containing double bonds (such as styrene-butadiene rubber, nitrile rubber, isoprene rubber, and their derivatives). The reprocessability and self-welding are practical for on-site assembly or repair of composite parts and extend the service life of materials.

The vast majority of traditional vulcanized rubber products are insoluble and infusible, which is difficult to reprocess and biodegrade, resulting in black pollution. In addition, although most rubber materials based on covalent adaptive networks (CANs) can achieve structural reconstruction, the lack of traditional vulcanization system leads to a decline in strength. In this study, biobased vanillin derivatives (PV) were synthesized to cross-link the commercially available 1,2-polybutadiene rubber precursor to construct imine-based CANs, thereby fabricating a resource-renewable, recyclable, and degradable high-performance rubber material. Due to the rigid tripod structure of the PV, the tensile strength of the material can achieve as high as 16.24 MPa, ranking among the best in the field of recyclable polybutadiene-based materials. Benefiting from the dynamic imine unit, the "dynamic covalent bridge" can be re-established to repair the damaged network and endow the material with excellent weldability. And, shape memory faculty of the material was proved and depicted. Moreover, this material displayed excellent antibacterial property originates from the introduced Schiff-base structure. By mixing with graphene, the application of action sensors can also be achieved.

In this work, a series of oxovanadium complexes bearing the ligands (S,E)-(+)-2, 6-dialkyl-N-(2-((2-(4-isopropyl-4,5-dihydrooxazole-2-yl)phenyl)amino)benzylidene)aniline (dialkyl = dimethyl (V1), diethyl (V2), and isopropyl (V3)) have been synthesized and characterized by FTIR spectroscopy and elemental analysis. Moreover, the molecular structures of complexes V2 and V3 were defined by X-ray diffraction. On activation with ethylaluminium sesquichloride (Al2Et3Cl3), these complexes exhibited high activity towards ethylene polymerization (up to 1.39 × 107 g molv-1 h-1) and showed excellent thermal stability (up to 60 °C). The obtained polyethylene had a moderate molecular weight (21.9 × 104 to 66.4 × 104 g mol-1) and exhibited narrow distribution (1.91 to 2.86) and unimodal features. The effect of the substituents on the ligands was also investigated in detail. The compound bearing the diisopropyl group showed the highest activity toward ethylene polymerization as the bimolecular deactivation of the catalyst can be effectively inhibited by the steric hindrance of the ortho-substituent on aniline. The complex V2 with moderate steric hindrance was also evaluated as a catalyst for the copolymerization of ethylene with norbornene and showed moderate to high activity.

PURPOSE: Normal weight obesity (NWO), defined as normal body mass index (BMI) and excessive body fat percentage (BF%), has been shown to be associated with a significantly higher risk of developing metabolic syndrome, cardiometabolic dysfunction and with higher mortality. However, there is limited literature regarding the potential associations between NWO and lifestyles. This study aimed to investigate the associations of lifestyles with NWO in Chinese university students. PARTICIPANTS AND METHODS: A total of 279 university students with normal BMI were recruited and divided into NWO and normal weight non-obesity (NWNO) groups by BF%. Body composition and anthropometrics were measured, and participants were asked to finish the Healthy Lifestyle Scale for University Students (HLSUS) questionnaire. RESULTS: A total of 26 male (25.5%) and 71 female (40.1%) students were identified as NWO. Compared to NWNO students, body weight, BMI, body fat mass, visceral fat area, waist circumference and hip circumference of NWO students were all significantly higher both in male and female students (P < 0.05). The body fat mass, BF% and visceral fat area were significantly negatively correlated with the total HLSUS, physical exercise behavior, and dietary nutrition behavior scores in NWNO males, NWO and NWNO females (P < 0.05). The risk of NWO was lower in those students with higher scores in physical exercise behavior in both males (OR = 0.298, 95% CI = 0.121~0.733) and females (OR = 0.653, 95% CI = 0.505~0.843), while dietary nutrition behavior (OR = 0.759, 95% CI = 0.584~0.986) and stress management behavior (OR = 0.503, 95% CI = 0.335~0.755) decreased the risk of NWO only in females. CONCLUSION: The incidence of NWO was high among university students, especially in females, which might be related with unhealthy lifestyles. NWO university students should pay attention to lifestyle adjustments, especially physical exercise, dietary nutrition and stress management, for preventing the health risk in NWO.

Vitrimers endow cross-linked polymers with malleability and reprocessability via exchange reactions. However, designing of reprocessable, shape-memory polymer materials with high strength via a catalyst-free method remains a challenge under mild conditions. Here, we propose a facile strategy to address this dilemma by introducing the exchangeable imine bond and N-coordinated boroxine into a polybutadiene (PB)-based network. Specifically, PB grafted with 2-aminoethanethiol is reacted with the formyl group of phenylboronic acid and dehydrated to form a dual-dynamic covalently cross-linked network at room temperature. The dynamic network draws on the advantage of imine (toughness) and N-coordinated boroxine (strength), making the PB-based materials exhibit favorable malleability, mechanical property, reprocessability, and thermal-induced shape-memory behavior. We can obtain customized high mechanical properties by tuning the cross-linking density, and the tensile strength reaches a high value (12.35 MPa) without fillers or any other additives. Meanwhile, the unique network framework makes the material recycle over several times without sacrificing its property. This work presents a facile and effective approach to achieve a multifunctional polymer with customized attributes. Besides, this strategy can recycle end-of-life rubber to alleviate environmental pollution and provide inspiration for fabricating targeted materials by uniting the dynamic covalent or noncovalent bonds.

Changes in expression levels of serum interleukin-4 (IL-4), IL-10 and adiponectin (APN) in patients with postoperative infection of colorectal cancer were studied. The clinical data of 159 patients receiving radical surgery for colorectal cancer in Xiangyang No. 1 People's Hospital, Hubei University of Medicine from January 2014 to December 2017, were retrospectively analyzed. A total of 67 patients with postoperative infection were enrolled into the infection group, while the remaining 92 patients without infection were enrolled into the non-infection group. The expression levels of serum IL-4, IL-10 and APN of patients were detected via enzyme-linked immunosorbent assay. The correlation of IL-4, IL-10 and APN levels with stage of colorectal cancer were explored by the Spearmans correlation analysis. The expression levels of IL-4 and IL-10 in the infection group were significantly higher than those in the non-infection group at day 3 after surgery (P<0.05). The expression level of APN in the infection group was lower than that in the non-infection group at day 3 after surgery (P<0.05). The serum IL-4 and IL-10 levels in pulmonary infection was higher and the serum IL-10 level in pulmonary infection was higher than those in incision infection and abdominal infection (P<0.05). The IL-4 and IL-10 levels in patients with colorectal cancer in the infection group at day 3 after surgery had a significant positive correlation with the stage of colorectal cancer (r=0.9357, P<0.001; r=0.9717, P<0.001), and the APN level in patients with colorectal cancer in the infection group at day 3 after surgery had a significant negative correlation with the stage of colorectal cancer (r=-0.9736, P<0.001). The serum IL-4 and IL-10 levels in patients with postoperative infection of colorectal cancer are positively correlated with the stage of cancer, while the serum APN level was negatively correlated with the stage of cancer.

This study estimated the attributable direct medical economic burden of health care-associated infections (HAIs) in China. Data were extracted from hospitals' information systems. Inpatient cases with HAIs and non-HAIs were grouped by the propensity score matching (PSM) method. Attributable hospitalization expenditures and length of hospital stay were measured to estimate the direct medical economic burden of HAIs. STATA 12.0 was used to conduct descriptive analysis, bivariate χ2 test, paired Z test, PSM ( r = 0.25σ, nearest neighbor 1:1 matching), and logistic regress analysis. The statistically significant level was set at .05. The HAIs group had statistically significant higher expenditures and longer hospitalization stay than the non-HAIs group during 2013 to 2015 ( P < .001). The annual average HAI attributable total expenditure, medicines expenditure, out-of-pocket expenditure, and number of hospitalization days per inpatient were (2015 US$) 6173.02, 2257.98, and 1958.25 and 25 days during 2013 to 2015. The direct medical cost savings was estimated at more than 2015 US$12 billion per year in Chinese tertiary hospitals across the country. The significant attributable direct medical economic burden of HAIs calls for more effective HAI surveillance and better control with appropriate incentives.
