RESUMEN

In this work, an ingenious sensor technology was established by integrating the EBFCs on a flexible paper strip carrier (PE) that was used for simultaneous detection of tumor markers in complex samples. Adopting high performance ultrathin graphdiyne (U-GDY) as the substrate can increase the enzyme load, accelerate the electron transfer rate, and significantly enhance the detection sensitivity. A homologous DNA nanomanager strategy cleverly uses signal switches to recycle and amplify target miRNAs, while the smartphone receives real-time instantaneous current values to realize multivariate detection. Electrochemical data show that the detection limits (LODs) of miRNA-21 and miRNA-155 are 0.09 and 0.15 fM in the wide concentration range. The results confirm that the tailored sensor platform provides a strategy for the early cancer diagnosis and lays the foundation for the construction of a flexible wearable platform.

RESUMEN

Realization of a highly sensitive analysis and sensing platform is important for early-stage tumor diagnosis. In this work, a self-powered biosensor with a novel sandwich graphdiyne (SGDY) combined with an aptamer-specific recognition function was developed to sensitively and accurately detect tumor markers. Results indicated that the detection limits of microRNA (miRNA)-21 and miRNA-141 were 0.15 and 0.30 fM (S/N = 3) in the linear range of 0.05-10000 and 1-10000 fM, respectively. The newly designed platform has great promise for early-stage tumor diagnosis.

We report here for the first time a self-powered biosensing platform based on graphene/graphdiyne/graphene (GDY-Gr) heterostructure substrate material for ultrasensitive hepatocarcinoma marker (microRNA-21) detection in both electrochemical and colorimetric test modes. The dual-mode signal intuitively displayed on a smartphone fundamentally improves the detection accuracy. In electrochemical mode, the calibration curve is established in the linear range of 0.1-10000 fM, and the detection limit is as low as 0.333 fM (S/N = 3). Simultaneously, colorimetric analysis of the miRNA-21 is realized by using ABTS as an indicator. The detection limit is confirmed as 32 fM (S/N = 3), and miRNA-21 of concentration from 0.1 pM to 1 nM exhibit a linear relationship with R2 = 0.9968. Overall, the combination of GDY-Gr and multiple signal amplification strategy significantly improved the sensitivity by 310 times compared with traditional enzymatic biofuel cells (EBFCs) based detection platform, showing broad application prospects for on-site analysis and future mobile medical services.

Currently, the development of polyvalent ions battery systems are still restricted by lacking suitable cathode materials with high energy density and long cycle life attributing to sluggish kinetic mechanism and of polyvalent ions. Herein, an effective inter-layer scaling strategy is proposed by using a simple hydrothermal method. The super layer spacing VS2 (∼1 nm) cathode dramatically improves electrochemical performance of zinc-ion batteries (ZIBs) and magnesium/lithium hybrid ion batteries (MLIBs). The specific discharge capacities of ZIBs and MLIBs are 450.7 and 488.8 mA h g-1 at current density of 0.1 A g-1 which are much higher than the same type of battery systems. Finally, the diffusion mechanism and the corresponding theoretical model is established by adopting first principles. In brief, the work provides an effective strategy for the large scale application of multivalent and hybrid batteries systems.

Background: Angiogenesis is a major promotor of tumor progression and metastasis in gastric adenocarcinoma (STAD). We aimed to develop a novel lncRNA gene signature by identifying angiogenesis-related genes to better predict prognosis in STAD patients. Methods: The expression profiles of angiogenesis-related mRNA and lncRNA genes were collected from The Cancer Genome Atlas (TCGA). Then, the "limma" package was used to identify differentially expressed genes (DEGs). The expression profiles of angiogenesis-related genes were clustered by consumusclusterplus. The Pearson correlation coefficient was further used to identify lncRNAs coexpressed with angiogenesis-related clustere genes. We used Lasso Cox regression analysis to construct the angiogenesis-related lncRNAs signature. Furthermore, the diagnostic accuracy of the prognostic risk signature were validated by the TCGA training set, internal test sets and external test set. We used multifactor Cox analysis to determine that the risk score is an independent prognostic factor different from clinical characteristics. Nomogram has been used to quantitatively determine personal risk in a clinical environment. The ssGSEA method or GSE176307 data were used to evaluate the infiltration state of immune cells or predictive ability for the benefit of immunotherapy by angiogenesis-related lncRNAs signature. Finally, the expression and function of these signature genes were explored by RT-PCR and colony formation assays. Results: Among angiogenesis-related genes clusters, the stable number of clusters was 2. A total of 289 DEGs were identified and 116 lncRNAs were screened to have a significant coexpression relationship with angiogenic DEGs (P value<0.001 and |R| >0.5). A six-gene signature comprising LINC01579, LINC01094, RP11.497E19.1, AC093850.2, RP11.613D13.8, and RP11.384P7.7 was constructed by Lasso Cox regression analysis. The multifactor Cox analysis and Nomogram results showed that our angiogenesis-related lncRNAs signature has good predictive ability for some different clinical factors. For immune, angiogenesis-related lncRNAs signature had the ability to efficiently predict infiltration state of 23 immune cells and immunotherapy. The qPCR analysis showed that the expression levels of the six lncRNA signature genes were all higher in gastric adenocarcinoma tissues than in adjacent tissues. The functional experiment results indicated that downregulation of the expression of these six lncRNA signature genes suppressed the proliferation of ASG and MKN45 cells. Conclusion: Six angiogenesis-related genes were identified and integrated into a novel risk signature that can effectively assess prognosis and provide potential therapeutic targets for STAD patients.

The effect of Ni on microstructure, elemental partition behavior, γ' phase solvus temperature, lattice misfit between γ and γ' phases, and mechanical properties of the Co-8Ti-11V-xNi alloys was investigated. The result shows that the lattice misfit in the alloys decreases from 0.74% to 0.61% as the Ni content increases from 0 to 10%, and the average sizes of the cuboidal γ' phase were measured to be 312.10 nm, 112.86 nm, and 141.84 nm for the Co-8Ti-11V, Co-8Ti-11V-5Ni, and Co-8Ti-11V-10Ni, respectively. Ti, V, and Ni exhibit a slight tendency to partition into the γ' phase, while Co shows a slight tendency to partition into the γ phase. The solvus temperatures of the γ' phase were measured to be 1167°C, 1114°C, and 1108°C for the Co-8Ti-11V, Co-8Ti-11V-5Ni, and Co-8Ti-11V-10Ni alloys, respectively, by using differential scanning calorimetry (DSC). Moreover, the yield strength and ultimate strength of the Co-8Ti-11V, Co-8Ti-11V-5Ni, and Co-8Ti-11V-10Ni alloys were investigated, and the yield strength and ultimate strength of the 10Ni alloy were highest, at 219 MPa and 240 MPa. After compression at 1000°C, the dislocations cannot effectively shear the γ' phase in the 0Ni and 10Ni alloys, resulting in a relatively high compressive strength of the 0Ni and 10Ni alloys. However, the γ' phase of the 5Ni alloy is no longer visible, and its strength is the lowest.

AIM: To investigate the effects and mechanisms of ischemic preconditioning (IPC) on the ischemia/reperfusion (I/R) injury of liver cirrhosis in rats and the effect of IPC on P-selectin expression in hepatocytes. METHODS: Forty male SD rats with liver cirrhosis were randomly divided into sham operation group (SO group), ischemia/reperfusion group (I/R group), ischemic preconditioning group (IPC group), L-Arginine preconditioning group (APC group), L-NAME preconditioning group (NPC group), eight rats in each group. Hepatocellular viability was assessed by hepatic adenine nucleotide level and energy charge (EC) determined by HPLC, ALT, AST and LDH in serum measured by auto- biochemical analyzer and bile output. The expression of P-selectin in the liver tissue was analyzed by immunohistochemical technique. Leukocyte count in ischemic hepatic lobe was calculated. RESULTS: At 120 min after reperfusion, the level of ATP and EC in IPC and APC groups was higher than that in I/R group significantly. The increases in AST, ALT and LDH were prevented in IPC and APC groups. The livers produced more bile in IPC group than in I/R group during 120 min after reperfusion (0.101+/-0.027 versus 0.066+/-0.027 ml/g liver, P=0.002). There was a significant difference between APC and I/R groups, (P=0.001). The leukocyte count in liver tissues significantly increased in I/R group as compared with SO group (P<0.05). The increase in the leukocyte count was prevented in IPC group. Administration of L-arginine resulted in the same effects as in IPC group. However, inhibition of NO synthesis (NPC group) held back the beneficial effects of preconditioning. Significant promotion of P-selectin expression in hepatocytes in the I/R group was observed compared with the SO group (P<0.01). IPC or L-arginine attenuated P-selectin expression remarkably (P<0.01). However, inhibition of NO synthesis enhanced P-selectin expression (P<0.01). The degree of P-selectin expression was positively correlated with the leukocyte counts infiltrating in liver (r=0.602, P=0.000). CONCLUSION: IPC can attenuate the damage induced by I/R in cirrhotic liver and increase the ischemic tolerance of the rats with liver cirrhosis. IPC can abolish I/R induced leukocyte adhesion and infiltration by preventing post-ischemic P-selectin expression in the rats with liver cirrhosis via a NO-initiated pathway.
