RESUMEN

IL-13 plays a critical role in mediating many biological processes responsible for allergic inflammation. Mast cells express Il13 mRNA and produce IL-13 protein in response to antigenic stimulation. Enhancers are essential in promoting gene transcription and are thought to activate transcription by delivering essential accessory cofactors to the promoter to potentiate gene transcription. However, enhancers mediating Il13 have not been identified. Furthermore, which Il13 enhancers detect signals triggered by antigenic stimulation have not yet been defined. In this study, we identified potential mouse Il13 enhancers using histone modification monomethylation at lysine residue 4 on histone 3 (H3K4me1) chromatin immunoprecipitation sequencing and acetylation at lysine residue 27 on histone 3 (H3K27ac) chromatin immunoprecipitation sequencing. We used Omni-assay for transposase-accessible chromatin sequencing to determine which accessible regions within the potential Il13 enhancers that responded to IgE receptor crosslinking. We also demonstrated that the transcription factor cluster consisting of the NFATC2, STAT5, GATA2, AP1, and RUNX1 binding sites at the proximal Il13 enhancer and the transcription factor cluster consisting of the EGR2 binding site at the distal Il13 E+6.5 enhancer are critical in sensing the signals triggered by antigenic stimulation. Those enhancers, which are responsive to antigenic stimulation and are constitutively active, cooperate to generate greater transcriptional outputs. Our study reveals a novel mechanism underlying how antigenic stimulation induces robust Il13 mRNA expression in mouse mast cells.

Background and objective Body dysmorphic disorder (BDD), which affects 1.7% to 2.4% of people worldwide, is usually encountered for the first time by nonpsychiatric physicians. Up to 37% of cases have been documented in dermatology clinics. This study aims to estimate the prevalence of BDD among Saudis attending dermatology clinics because the literature is lacking in this field, especially in the Eastern Province. Methods This is a cross-sectional study, conducted in 2023. A total of 412 Saudi Eastern Province residents, aged 18 years and older, were included in the study and given a self-administered web-based questionnaire. The study uses the Body Dysmorphic Disorder Questionnaire as one of its three primary measurements, together with sociodemographic data, and dermatological and previous psychological histories. Results A total of 412 participants were enrolled in this study. Of the total sample, 64.5% had more than one skin condition, with the rest having only one one. The most received cosmetic treatment in this study was topical agents. It was estimated that the prevalence of BDD is 9.5% among the studied population. However, it was found that there are no significantly associated factors with the prevalence of BDD. Conclusions This study reports a prevalence of 9.5% among people visiting dermatological clinics. The prevalence is alarming, which emphasizes the importance of enhancing the awareness of BDD among dermatologists and developing certain guidelines to identify and refer these patients to mental health professionals.

We report herein the dielectric properties response of two ionic chitosan derivatives. More specifically, we report here chitosan derivatives bearing guanidinium groups, prepared through the reaction between chitosan and cyanamide when scandium (III) triflate is present. The anionic counter ions of the guanidinium groups could be changed by working either in acetic acid or hydrochloric acid. In this way, two chitosan derivatives, namely N-guanidinium chitosan acetate (GChAc) and N-guanidinium chitosan chloride (GChCl), were produced. The materials were investigated by 13C solid state NMR, FT-IR spectroscopy, SEM and TEM analysis and compared with the parent chitosan. Dielectric spectroscopy has been used to study the relaxation behavior of the parent chitosan and the new chitosan derivatives over wide ranges of temperature and frequency, indicating that local motion is affected by counter ions. Due to these features, these materials are interesting candidates as high-power electrical materials for green technological applications.
