RESUMEN

Pyrrolizidine alkaloids (PAs) are secondary plant metabolites playing an important role as phytotoxins in the plant defense mechanisms and can be present as contaminant in the food of humans and animals. The PA monocrotaline (MCT), one of the major plant derived toxin that affect humans and animals, is present in a high concentration in Crotalaria spp. (Leguminosae) seeds and can induce toxicity after consumption, characterized mainly by hepatotoxicity and pneumotoxicity. However, the effects of the ingestion of MCT in the central nervous system (CNS) are still poorly elucidated. Here we investigated the effects of MCT oral acute administration on the behavior and CNS toxicity in rats. Male adult Wistar were treated with MCT (109 mg/Kg, oral gavage) and three days later the Elevated Pluz Maze test demonstrated that MCT induced an anxiolytic-like effect, without changes in novelty habituation and in operational and spatial memory profiles. Histopathology revealed that the brain of MCT-intoxicated animals presented hyperemic vascular structures in the hippocampus, parahippocampal cortex and neocortex, mild perivascular edema in the neocortex, hemorrhagic focal area in the brain stem, hemorrhage and edema in the thalamus. MCT also induced neurotoxicity in the cortex and hippocampus, as revealed by Fluoro Jade-B and Cresyl Violet staining, as well astrocyte reactivity, revealed by immunocytochemistry for glial fibrillary acidic protein. Additionally, it was demonstrated by RT-qPCR that MCT induced up-regulation on mRNA expression of neuroinflammatory mediator, especially IL1ß and CCL2 in the hippocampus and cortex, and down-regulation on mRNA expression of neurotrophins HGDF and BDNF in the cortex. Together, these results demonstrate that the ingestion of MCT induces cerebrovascular lesions and toxicity to neurons that are associated to astroglial cell response and neuroinflammation in the cortex and hippocampus of rats, highlighting CNS damages after acute intoxication, also putting in perspective it uses as a model for cerebrovascular damage.

Alterações neurológicas em cães são comuns na rotina da clínica médica, sendo necessário identificar o sítio de lesão quando da ocorrência de neuropatias por compressão em estruturas do Sistema Nervoso Central (SNC). O objetivo deste trabalho foi realizar um estudo epidemiológico das alterações neurológicas compressivas em medula espinal de cães domiciliados em Salvador e região metropolitana: Discopatias, Neoplasias e Traumas identificadas através da Tomografia Computadorizada (TC). Foram avaliados 17 cães de diversas raças com idade entre 2 e 10 anos e de ambos os sexos, atendidos na rotina de dois serviços médicos-veterinários e encaminhados para realização do exame conforme indicação clínica. Imagens em cortes com espessura variando de 0,5 a 2mm em rotação foram produzidas com o Tomógrafo. Dos animais que apresentaram discopatia 12/17 (70,58 por cento), por mielopatia extradural 6/12 (50 por cento), foram da raça Dachshund com idade média 7,33±1,97 anos e para todos os outros a ocorrência foi pontual; 23,53 por cento de osteopatias vertebrais, neoplasias ósseas e fraturas, e 5,89 por cento correspondente a neoplasia de medula. Houve uma maior frequência de lesões nos segmentos C4-C6 3/12 (25 por cento), T12-T13 (25 por cento) e L2-L3 (25 por cento) dos casos.


Neurological disorders in dogs are common in veterinary medical practice routine, being necessary to identify the site of injury in the event of compression on structures of the central nervous system (CNS). The aim of this work was an epidemiological study of neurological changes in compressive spinal cord of pet dogs in Salvador and the metropolitan area. Discopathies, neoplasms and trauma were identified by Computed Tomography (CT). We evaluated 17 dogs of various breeds, aged between 2 and 10 years old and of both sexes, from two veterinary medical routine services and forwarded to the examination as clinically indicated. Slice images with a thickness ranging from 0.5 to 2mm were produced in rotation with the tomograph. Animals that showed disc disease (70.58 percent) for extradural myelopathy 6/12 (50 percent) were the breed Dachshund, mean age 7.33±1.97 years and for all other occurrence was timely; 23.53 percent of vertebral bone disease, bone tumors and fractures, and 5.89 percent corresponding to neoplasm marrow. There was a higher incidence of injuries segments L2-L3 (25 percent), followed by C4-C6 (16.67 percent), T12-T13 (16.67 percent), L7-S1 (16.67 percent) of cases.

O objetivo do trabalho foi descrever os parâmetros hematológicos e as alterações histopatológicas em bijupirás infectados por Amyloodinium ocellatum. Um grupo de 27 peixes foi anestesiado para coleta de amostras de sangue e eutanasiados para coleta de muco e fragmentos de tecido cutâneo e branquial. Foram avaliadas a prevalência e a intensidade parasitária da infecção, assim como os valores de parâmetros hematológicos e alterações histopatológicas. A prevalência parasitária nas brânquias foi de 100% e no muco foi de 80,8% e as intensidades parasitárias médias foram de 683,5 nas brânquias, e 67,1 no muco cutâneo. Os valores médios dos parâmetros hematológicos foram: eritrócitos 4,3x10(6)µL; VG 26%; VGM 64,2fL; proteína plasmática 5,8mg/dL; trombócitos 5,2 x10³/µL e leucócitos 3,6 x10³/µL. Além disso, foram verificadas hiperplasia do epitélio respiratório acompanhada de fusão lamelar, descolamento do epitélio, dilatação do seio venoso, formação de aneurisma, ruptura do epitélio lamelar, hemorragia, necrose, reação inflamatória linfocítica. O parasito foi observado nas lamelas branquiais, o VMA variou do grau discreto ao severo e o IAH foi de 76,8. A pesquisa assume importância por se tratar dos primeiros estudos em Rachycentron canadum, um peixe que se destaca com potencial ao cultivo.


The aim of this study was to describe the hematological parameters and histopathologic alterations in cobia infected by Amyloodinium ocellatum. A group of 27 fish were anesthetized to collect blood samples and euthanatized to collect mucus and tissue fragments of skin and gills. The prevalence and parasitic intensity of the infection, besides hematologic parameters and histopathologic alterations, was measured. Parasite prevalence in the gills was 100% and in the mucus 80.8%, and the average intensity of infection in gills and skin was 683.5 e 67.1 respectively. The mean values of hematological parameters were: erythrocytes 4.3x10(6)μL; PCV 26%, MCV 64.2 fL, plasma protein 5.8mg/dL, thrombocytes 5.2x10³/μL and leukocytes 3.6x10³/μL . Furthermore was found hyperplasia of the respiratory epithelium accompanied by lamellar fusion, detachment of the epithelium, venous sinus dilatation, aneurysm formation and rupture of the lamellar epithelium, hemorrhage, necrosis and lymphocytic inflammatory reaction. The parasite was observed between the gills lamellae, the AMV ranged from mild to severe and AHI values were 76.8. The study assumes importance because it is the first study in Rachycentron canadum, a fish that stands out with potential for growing.
