Sulfate radical-based advanced oxidation processes (SR-AOPs) are gaining popularity as a feasible alternative for removing recalcitrant pollutants in an aqueous environment. Persulfates, namely peroxydisulfate (PDS) and peroxymonosulfate (PMS) are the most common sulfate radical donors. Persulfates activation by ultraviolet (UV) irradiation is considered feasible due to the high concentration of radicals produced as well as the lack of catalysts leaching. The research focuses on determining the impact of activated PDS and PMS on the degradation of anthraquinone dye, i.e., Acid Blue 129 (AB129). UV-activated PDS and PMS can quickly degrade the AB129 as well as restrict the formation of by-products. This could explain the reduced ecotoxicity levels of the treated water after degradation, using an aquatic plant (Lemna minor) and a crustacean (Daphnia magna). This, on the other hand, can ensure that the sulfate radical-based processes can be an environmentally friendly technology.

Japanese encephalitis (JE) is a re-emerging mosquito borne disease, for which equines are most susceptible amongst all animals. Detection of specific immunoglobulin 'M' (IgM) is considered as an ideal way to diagnose recent JE virus infection in equines due to low virus load and short-term viremia. The present study was undertaken to develop a sensitive and specific recombinant NS1 protein based indirect IgM-ELISA and IgM capture (MAC) ELISA to diagnose recent infection of JEV in equines. Indirect IgM ELISA was standardized with relative diagnostic sensitivity and specificity of 100% and 88.5%, respectively. The validation of indirect IgM-ELISA in different laboratories revealed excellent reproducibility with Cohen's kappa value ranging between 0.84 and 1. The standardization of MAC ELISA was attempted using checker board titration method and non-specific binding of polyclonal anti-equine IgM capture antibody with anti-porcine IgG conjugate and with hyperimmune serum raised in swine against the antigen was observed. Hence, the MAC ELISA was standardized with monoclonal capture antibody; however, its diagnostic performance could not meet the satisfactory limit. Due to better sensitivity and less turnaround time, indirect IgM-ELISA was employed to screen 821 equine serum samples revealing 33.73% positivity of IgM antibodies against JEV in equine population of India. The high JEV sero-positivity warrants the need for vaccination in Indian equine population along with the demand for research focused towards anti-viral therapy. The indirect IgM-ELISA developed in the present study could be useful to diagnose acute or recent infection of JEV in equines as well as in sero-epidemiological studies.

BACKGROUND & AIMS: Esophageal adenocarcinomas (EACs) are heterogeneous and often preceded by Barrett's esophagus (BE). Many genomic changes have been associated with development of BE and EAC, but little is known about epigenetic alterations. We performed epigenetic analyses of BE and EAC tissues and combined these data with transcriptome and genomic data to identify mechanisms that control gene expression and genome integrity. METHODS: In a retrospective cohort study, we collected tissue samples and clinical data from 150 BE and 285 EAC cases from the Oesophageal Cancer Classification and Molecular Stratification consortium in the United Kingdom. We analyzed methylation profiles of all BE and EAC tissues and assigned them to subgroups using non-negative matrix factorization with k-means clustering. Data from whole-genome sequencing and transcriptome studies were then incorporated; we performed integrative methylation and RNA-sequencing analyses to identify genes that were suppressed with increased methylation in promoter regions. Levels of different immune cell types were computed using single-sample gene set enrichment methods. We derived 8 organoids from 8 EAC tissues and tested their sensitivity to different drugs. RESULTS: BE and EAC samples shared genome-wide methylation features, compared with normal tissues (esophageal, gastric, and duodenum; controls) from the same patients and grouped into 4 subtypes. Subtype 1 was characterized by DNA hypermethylation with a high mutation burden and multiple mutations in genes in cell cycle and receptor tyrosine signaling pathways. Subtype 2 was characterized by a gene expression pattern associated with metabolic processes (ATP synthesis and fatty acid oxidation) and lack methylation at specific binding sites for transcription factors; 83% of samples of this subtype were BE and 17% were EAC. The third subtype did not have changes in methylation pattern, compared with control tissue, but had a gene expression pattern that indicated immune cell infiltration; this tumor type was associated with the shortest time of patient survival. The fourth subtype was characterized by DNA hypomethylation associated with structure rearrangements, copy number alterations, with preferential amplification of CCNE1 (cells with this gene amplification have been reported to be sensitive to CDK2 inhibitors). Organoids with reduced levels of MGMT and CHFR expression were sensitive to temozolomide and taxane drugs. CONCLUSIONS: In a comprehensive integrated analysis of methylation, transcriptome, and genome profiles of more than 400 BE and EAC tissues, along with clinical data, we identified 4 subtypes that were associated with patient outcomes and potential responses to therapy.

Candida species are opportunistic human fungal pathogens that cause acute and chronic infections against which only few antifungal agents are available. Here we have elucidated the antifungal effect of Syzygium samarangense leaf extracts (SSLE). Antifungal activity of SSLE was studied against Candida albicans, C. krusei, C. parapsilosis, C. glabrata, C. auris and C. tropicalis. Following experiments were performed: minimum fungicidal concentration (MFC) determination, agar well disc diffusion assays, fungal morphology analysis using scanning electron microscope (SEM), ex vivo fungal survival assays on porcine tongue and skin and in vivo fungal survival assays using Drosophila melanogaster fly model. Results demonstrated MFC of SSLE ranges between 100 and 125 mg ml-1 . SEM images showed cell wall degradation of C. albicans when treated with SSLE. Around 75% decrease in C. albicans viability was observed when infected porcine tongue and skin were treated using SSLE. The C. albicans infected D. melanogaster when fed with SSLE showed significant decrease (around 80%) of fungal count than the infected control. Furthermore, agar plate disc diffusion assays demonstrated that the antifungal activity of SSLE could be due to chalcone, which is one of the active constituents in SSLE. Our study demonstrated that SSLE could be used for the topical treatment of Candida infections.

This study investigates an environmentally benign approach to generate platinum nanoparticles (Pt NP) supported on the reduced graphene oxide (RGO) by non-edible gum waste of gum kondagogu (GK). The reaction adheres to the green chemistry approach by using an aqueous medium and a nontoxic natural reductant-GK-whose abundant hydroxyl groups facilitate in the reduction process of platinum salt and helps as well in the homogenous distribution of ensued Pt NP on RGO sheets. Scanning Electron Microscopy (SEM) confirmed the formation of kondagogu gum/reduced graphene oxide framed spherical platinum nanoparticles (RGO-Pt) with an average particle size of 3.3 ± 0.6 nm, as affirmed by Transmission Electron Microscopy (TEM). X-ray Diffraction (XRD) results indicated that the Pt NPs formed are crystalline with a face-centered cubic structure, while morphological analysis by XRD and Raman spectroscopy revealed a simultaneous reduction of GO and Pt. The hydrogenation of 4-nitrophenol could be accomplished in the superior catalytic performance of RGO-Pt. The current strategy emphasizes a simple, fast and environmentally benign technique to generate low-cost gum waste supported nanoparticles with a commendable catalytic activity that can be exploited in environmental applications.

BACKGROUND: The severe forms of thalassemia are the most common inherited anemias managed with regular blood transfusion therapy. Transfusion policies and complications are critical to quality of life and survival, but there is a lack of standardized care. STUDY DESIGN AND METHODS: A survey of 58 items was completed in 2016 by 11 centers in California, Washington, Oregon, Nevada, and Arizona providing long-term care for thalassemia. The questionnaire addressed demographic information, transfusion practices and complications, and educational needs. RESULTS: The centers followed 717 patients with ß-thalassemia (314, 43.8%) or α-thalassemia (394, 55%). One-third (34.7%) of patients were transfusion-dependent. Indications and goals of transfusion therapy differed between centers. Prestorage leukoreduction was universal, while routine irradiation of units was limited to one site. Red blood cell antigen phenotype was determined before the first transfusion and patients received Rh/Kell-matched units. However, more than half of the transfused patients had received blood at multiple hospitals within or outside the United States. Alloantibodies were seen in 16.9% of transfused group, but management of such patients was variable. Unusual or emerging transfusion-transmitted pathogens were not observed. Multiple educational needs were recognized, with iron overload as the biggest challenge; the approach to iron chelation varied within the group. CONCLUSION: This study identified many patients not included in earlier surveys limited to major national centers, suggesting that the thalassemia population in the United States is vastly underestimated. Lack of evidence-based guidelines is a barrier to optimal care, which should be addressed through regional consortia of thalassemia centers.

BACKGROUND: Dentigerous cyst is the second most common type of odontogenic cysts that encloses the crown of an unerupted tooth by expansion of its follicle due to the collection of cystic fluid. In view of the capability of these lesions attaining a marked size if not diagnosed early and treated properly, the present study was done based on the clinical and radiographic presentation of the dentigerous cyst in addition to the analysis of the cystic contents. OBJECTIVE: The present study reported 14 cases of dentigerous cysts (DC) with their incidence and relative distribution of the maxillofacial region along with the clinical and radiographic features and biochemical analysis of cystic fluid. METHOD: The study was conducted at Sri Govind Tricentenary Dental College, Hospital and Research Institute, Gurgaon, Haryana for a period of one year and six months. A detailed case history and thorough clinical examination was done for the patients who were provisionally found to have odontogenic cysts. Necessary radiographs, aspiration of the cystic fluid and incisional biopsy were performed to the 14 patients who were provisionally diagnosed with dentigerous cysts (DC) after obtaining the informed consent and the cystic fluid was subjected to biochemical analysis. RESULTS: Majority of the affected patients with DC were in their second decade and showed more predilection for mandible with a male predominance. The area of predilection was third molar region in the mandible and it is the canine region in maxilla. All the cases were associated with impacted teeth and majority showed expansion of the buccal/labial cortical plate. The various biochemical variables (total protein content, albumin, globulin and albumin: globulin ratio) were also assessed in the present study. CONCLUSION: The awareness of protean features of DC evident through this study is essential for the general as well as specialty practitioners for the accurate diagnosis and proper treatment planning of these non cancerous but potentially destructive lesions and also opens new avenues for further research.

The strategy for interpreting the role of autophagy on the basis of evidence obtained through autophagic inhibition sounds logical, but is beset with practical constraints. The knock down of autophagy-related (ATG) gene(s) or blockage of class III PI3-Kinase are the most common approaches for inhibiting autophagy. However, during stressful conditions, autophagy may operate in synchrony with other processes such as apoptosis; autophagy-related genes, unlike what their name implies, exert their regulation on apoptosis as well. Knocking down such genes not only blocks autophagy but also renders apoptosis defective, making the interpretation of autophagic roles unreliable. Similarly, class III PI3-Kinase aids in initiating autophagy but it is not a quintessential autophagic regulator. Class III PI3-Kinase also has a role in regulating almost all membrane transport in cells. Blocking it not only inhibits autophagy, but also hampers all the membrane trades, including endosomal transport. The pharmacological inhibitors used to block autophagy by blocking class III PI3-Kinase further compound these limitations with their off-target effects. Knowing the limitations involved in blocking a target or using an autophagy-blocking tool is a prerequisite for designing the experiments meant for analyzing autophagic functions. This review attempts to provide a detailed overview about the practical constraints involved in using autophagic inhibition as a strategy to understand autophagy.

AIM: To isolate, fermentatively evaluate and identify black pepper (Piper nigrum L.)-associated bacteria for the microbial decortication of fresh ripened berries and dried black pepper for preparation of off-odour-free white pepper. METHODS AND RESULTS: Among 45 bacterial isolates obtained from black pepper, seven of them were found to decorticate black pepper (>60%) and fresh pepper berries (98-100%) into white pepper within 5 days of immersion in bacterial suspension. The 16S rRNA genes (1500-bp amplicon) of these bacteria were sequenced, and species identity was established by closest match in GenBank. Superior-quality white pepper was obtained with Bacillus subtilis (IISR WP 33, 34, 38), Bacillus licheniformis (IISR WP 43), Acinetobacter baumanii (IISR WP 35), Klebsiella pneumoniae (IISR WP 19) and Microbacterium barkeri (IISR WP25). The bacterial isolates were found to secrete multiple hydrolytic enzymes such as cellulase, pectinase, amylase, protease and xylanase. Bacterial cultures were deposited with International Depository Authority at Microbial Type Culture Collection, India, as patent deposits as prescribed in Budapest Treaty for microbial deposits. The white pepper, thus obtained from bacterial decortication process, was free from off-odour compound, especially skatole. Other biochemical constituents such as oleoresin, piperine and essential oils were found in the acceptable range. The bacterial decortication did not affect inherent constituents of pepper such as essential oil constituents, oleoresin and piperine content. CONCLUSION: One of the most significant findings of the work is identification of specific bacterial species for decortication of fresh berries or black pepper berries into value-added white pepper. SIGNIFICANCE AND IMPACT OF THE STUDY: This work paved way for developing a technological process for microbial decortication of fresh/black pepper for the production of superior-quality white pepper.

BACKGROUND: The aim of this study was to determine the incidence of different Kennedy's classes of partial edentulism during 18 months period. MATERIALS AND METHODS: Patients were clinically examined for various Kennedy's classes of partial edentulism in the outpatient department (OPD), prosthodontics, GPRDCH, Kurnool (Andhra Pradesh). RESULTS: Of the total 1,420 OPD patients, Kennedy's class III was the most frequent classification encountered (62%) and followed by Kennedy's class I (18%), class II (11%), and class IV (9%) in decreasing order. CONCLUSION: The patients with various Kennedy's classes of partial edentulism can be offered various treatment modalities like removable cast partial dentures, fixed partial dentures, over dentures and implant supported dentures. This study can be crucial for screening the population for incidence of tooth loss as a factor of gender and age. Clinical significance: Tooth loss appears to have an important role in the loss of esthetics and mastication. Study of incidence of various classes of partial edentulism provides clinically useful information for dental training and continuing education.

One of the most prevalent types of odontogenic cysts is a dentigerous cyst, which is usually connected to the crown of an immature tooth. We report the case of an 11-year-old boy, who had a swelling over his left cheek, which was determined to be a dentigerous cyst by radiological imaging and clinical examination. Over the course of two months, the peanut-sized mass grew to 3x2 cm. A massive, well-defined cystic lesion connected to an unerupted premolar tooth was found on a CT scan of the left maxillary alveolar arch and sinus floor. Under general anesthesia, the patient had a Caldwell-Luc surgery to remove the cyst. In order to avoid difficulties related to cyst formation, which can invade surrounding tissues and even result in cancer if left untreated, early detection using radiological imaging is essential. Complete excision of the cyst is the treatment, particularly for big lesions, in order to limit morbidity and lower the likelihood of aggressive behavior. This case emphasizes the necessity of thorough examination and surgical intervention when necessary, underscoring the significance of early identification and adequate therapy to minimize potential problems related to dentigerous cysts. In cases of dentigerous cysts, early intervention, and appropriate surgical procedures are critical to reducing morbidity and improving patient outcomes.

Kondagogu (Cochlospermum gossypium) gum (KG), a natural tree exudate, was investigated for its morphological, adsorption and metal interaction behavior with various toxic heavy metals (Pb, Cd, Ni, Cr and Fe). SEM, AFM and TEM techniques were used to study the morphological changes occurring after metal adsorption onto the biopolymer structure. The degree of biosorption of metals on KG biopolymer surfaces was assessed by small-angle X-ray scattering analysis. EDXA spectrum revealed that the ion-exchange mechanism plays a major role in the binding process between KG and metal ions. The higher electron density observed in the KG-Cd complex suggests that Cd is strongly bound to KG compared to the other metals. This work provides a potential platform for developing a hydrocolloid-based nanogel for bioremediation of environmental contaminants.

Proper management of the occlusal plane is an essential consideration when multiple long span posterior restorations are designed. When restorations are added to an existing tooth arrangement characterized by rotated, tipped or extruded teeth, excursive interferences may be incorporated. The curve of Spee which exists in natural dentition, allows harmony to exist between the anterior teeth and the condylar guidance. Broadrick fag or occlusal plane analyzer is used to assist in the reproduction of tooth morphology that is commensurate with the curve of Spee when posterior restorations are designed; its use prevents the introduction of protrusive interferences. The current study determines the relationship of the center of anteroposterior curve of occlusion in the cranium and its relationship to other cephalometric landmarks and also evaluates the relationship of anteroposterior curve of occlusion to the condyle.

UNLABELLED: Patients frequently report sensitivity of prepared abutment teeth during the temporization period and after the fnal cementation of full coverage restoration. Purpose of this clinical investigation was to evaluate the effectiveness of desensitizing agents in reducing the pre- and postcementation sensitivity for full coverage restorations and to compare the relative effcacy of three in offce applied desensitizing agents in relieving the postcementation sensitivity with the use of glass ionomer luting cement. MATERIALS AND METHODS: This study consisted of 30 patients requiring either full coverage restoration or 3 unit fxed partial denture. Total of 40 restorations (n = 40) were made and were randomly assigned into four groups comprising 10 restoration (n = 10) in each group. Group C control where no desensitizer application was done, group BB applied with BisBlock dentin desensitizer (Bisco Inc.), group ST applied with Systemp desensitizer (Ivoclar Vivadent), group GC applied with GC Tooth Mousse desensitizer (GC Asia). Desensitizer application was done immediately after the tooth preparation. Sensitivity of the tested abutment was determined by the patient response to cold, hot and bite stimuli and were recorded on visual analog scale (VAS). Sensitivity level scores was evaluated at 4 time intervals, i.e. 1 week after desensitizer application at baseline precementation appointment and others at 5 minutes, 1 day and 1 week postcementation appointment. VAS score data was statistically analyzed using one-way ANOVA followed by post hoc Tukey's test. RESULTS: BisBlock and GC Tooth Mousse desensitizer resulted in statistically signifcant (p < 0.01) reduction in postcementation sensitivity of glass ionomer cement in comparison to Systemp desensitizer at 5 minutes, 1 week postcementation time interval with no statistical difference was seen between all desensitizer groups at 1 day postcementation. Application of BisBlock and GC Tooth Mousse desensitizer resulted in highly signifcant (p < 0.01) reduction in sensitivity level at the end of 1 week. CLINICAL SIGNIFICANCE: Desensitizer's application on the prepared abutment teeth is considerably effective in relieving both pre- and postcementation sensitivity for full coverage restoration over the short duration of time. Immediate reduction in postoperative sensitivity relatively in a short time period may be benefcial in terms of patient's comfort. Nonetheless, multicenter long-term clinical trials should be conducted to confrm the results. CONCLUSION: Efficacy of BisBlock and GC Tooth Mousse desensitizer was more in relieving the postcementation sensitivity of glass ionomer cement at various time intervals in comparison to Systemp desensitizer. In conclusion, application of desensitizers was beneficial to reduce the pre- and postcementation abutment sensitivity.

Several dental materials have been used for core build-up procedures. Most of these materials were not specifically developed for this purpose, but as a consequence of their properties, have found application in core build-up procedures. Improvements in composites and the development of nanocomposites have led to their use as a core build up material due to their superior mechanical properties, optical properties and ease of handling. However it is not clear if they have better mechanical properties than the conventional core build up materials like amalgam, GIC and dual cure composite core build up material. The strength of the core material is very important and this study was undertaken to compare the mechanical properties of materials used for direct core foundations. The differences between the compressive strength and flexural strength of Filtek Z350 nanocomposite with conventional core build up materials like Amalgam, Vitremer GIC and Fluorocore were tested. Cylindrical plexi glass split molds of dimension 6 ± 1 mm [height] x4 ± 1 mm [diameter] were used to fabricate 15 samples of each core material for testing the compressive strength and rectangular plexi glass split molds of dimension 25 ± 1 mm [length] x 2 ± 1 mm[height] x2 ± 1 mm [width] used for fabricating samples for flexural strength. The samples were stored a water bath at 250 °C for 24 h before testing. The samples were tested using a Universal Instron testing machine. The results of the study showed that Fluorocore had the highest compressive strength and flexural strength followed by Filtek Z350 [nanocomposite] Amalgam had the least flexural strength and Vitremer GIC had the least compressive strength. Thus flurocore and nanocomposite are stronger than other core build up materials and hence should be preferred over other conventional core build up materials in extensively damaged teeth.

Use of domestic microwave oven has been suggested as a method of disinfecting a number of dental materials used in dental practice. This study was done to analyse the effect of microwave irradiation on vinyl polysiloxane putty impression material (3M ESPE, Express™ STD) contaminated with test organisms (Staphylococcus aureus, Pseudomonas aeruginosa, Candida albicans. 180 square shaped specimens of addition silicon putty material were prepared and divided into 3 groups for three test organisms. The 3 groups were subdivided into 4 subgroups (n = 15) for different exposure parameters (control group 5, 6 and 7 min exposure at 650 W. The specimens were contaminated using standard inoculums of test organism and then were irradiated using domestic microwaves. Broth cultures of the control and test group specimens were plated on selective media culture plates. Colonies formed were counted. Data analyses included Kruskal-Walli's ANOVA and Mann-Whitney's tests. Nil values shows complete elimination of C. albicans and P. aeruginosa after 5, 6 and 7 min exposure. Staphylococcus aureus showed colonies with the mean value of 7.6 × 10(3) ± 2.3 × 10(3), 4.6 × 10(3) ± 2.6 × 10(3) after 5 and 6 min respectively and nil values after 7 min exposure. 5 min exposure caused complete elimination of C. albicans and P. aeruginosa strains, while 7 min exposure eliminated S. aureus completely.

The major connector is the most vital component critically subjected to maximal stress concentration due to various forces acting on it. The main requirement of a major connector is its resistance to deformation by occlusal stresses. This resistance to deformation is a direct consequence of the rigidity of the major connector. Thus rigidity of the major connector is paramount to resist flexing and torquing forces that could be transmitted to the abutment teeth and other structures as destructive forces. The commonly used major connectors for the mandibular arch are lingual bar and lingual plate. In the present study, the deflection of various major connector designs due to occlusal load is assessed by finite element method. They have been analyzed through finite element models. The differences in the deflection behaviour of mandibular major connector used in Kennedy's Class I, Class II, Class III, and Class IV edentulous situations have been compared. A CT scan of human edentulous mandible was taken and each section from symphysis to condylar region was projected on a graph paper and three-dimensional volumes were created from connected successive profiles to define the final solid geometry of cortical bone. Six framework models with different mandibular major connectors, lingual bar and lingual plate for Kennedy's Class I, Class II, Class III, and Class IV situations were created. The three dimensional finite element models corresponding to the geometric model were generated using Ansys's pre-processor. The model was assigned material properties. A vertical biting force of 20 N was applied. The results showed that the maximum deflection was seen in the saddle area when compared to other areas, i.e., major connector and the occlusal rest regions. The lingual bar in Kennedy's Class III situation and lingual plate in Kennedy's Class IV situation showed the least deflection when compared to Class I and Class II (distal extensions) situations. Lingual plate is more rigid major connector than lingual bar.

OBJECTIVES: TLPL/AY/03/2008 is a polyherbal formulation intended for treatment of osteoarthritis, rheumatoid arthritis, lumbago, spondylitis etc., Acute and repeated dose 90-days studies were conducted to evaluate the safety profile of TLPL/AY/03/2008 in rats. MATERIALS AND METHODS: In acute study, TLPL/AY/03/2008 was orally administered to Sprague Dawley rats at 2000 mg/kg. In repeated dose study, TLPL/AY/03/2008 was administered to rats at 200, 500 and 1000 mg/kg through oral gavage for 90 days and assessed for treatment related changes in body weight, feed consumption, hematological, biochemical and pathological parameters. Histopathological examination was conducted for tissues from control and the high dose groups and was extended to target organs from the lower dose and recovery groups. RESULTS: In acute study, the test item did not produce any mortality or adverse clinical signs. In the 90-days oral toxicity study, animals did not exhibit any toxicity symptoms and no deaths were observed. No significant changes were found in hematological and biochemical endpoints. Also, toxicologically significant alterations in relative organ weights were not observed. Microscopic findings of mild to marked, diffuse hepatocellular degeneration (vacuolar changes with granular of cytoplasm and pyknotic nuclei of hepatocytes) was noticed in males at 1000 mg/kg body weight. Animals of recovery group (1000 mg/kg) did not show any changes when compared with control group animals indicating the complete reversal. CONCLUSIONS: Based on the findings of the study, the median lethal dose of TLPL/AY/03/2008 was found to be more than 2000 mg/kg. The No Observed Adverse Effect Level (NOAEL) of TLPL/AY/03/2008 can be considered as 1000 mg/kg in both male and female rats, under the experimental conditions and doses employed.

Aim: The current study evaluates and compares the percentage distribution of different microorganisms according to their strains and occurrence among the three studied groups. Method and Methodology: Sample of 30 removable dental prosthesis wearing patients was selected: wearing either complete dentures or partial dentures and without any significant medical history or on prescription medication for the past 3-6 months. Samples were obtained in three subcategories based on the duration of prostheses worn by the patient. A sterile swab made up of cotton moistened with phosphate buffer saline (PBS) was scrubbed on the dental prosthesis at the fitting surfaces and the denture-bearing area of the oral cavity. Within two hours, the collected swab sample was infused in the sterile tube containing 1 ml of 0.84% PBS solution maintained at pH 7-7.2 and sent for microbiological analysis. The samples were then inoculated into different medias. Microbial growth was checked after incubating the culture plates for 48 h at 37°C. Microorganisms were recognized and counted by calibrated colony counter. Gram's stain was used to stain the colony smear and biochemical tests such as coagulase, catalase, oxidase, sugar fermentation with acid and gas production (triple sugar iron), methyl red test, test for indole production, hydrogen sulphide (H2S) production, citrate utilization, urease test, germ tube tests were performed. Statistical Analysis: Descriptive statistics included calculation of means and standard deviation using multivariate analysis. All values were considered statistically significant for a value of P < 0.05. Results: Streptococcus species, Coagulase-negative staphylococcus, Staphylococcus aureus, Candida albicans and Klebsiella pneumoniae showed the maximum positive culture among the secluded microorganisms in all three groups. Conclusion: A progressive increase in the microbial contamination was directly proportional to the duration of removable prosthesis usage.

AIMS: Maternal hypercholesterolemia (MHC) is a pathological condition that may cause atherosclerosis in the adulthood of the offspring. The study aims to identify the role of in-utero programming by the placenta in atherogenesis and associated liver pathology in offspring. MAIN METHODS: Female New Zealand white rabbits with normal lipid profiles were fed a 0.3 % HFD after mating. Lipid levels were monitored, and pregnant rabbits were sacrificed at the end of trimester 1, trimester 2, and trimester 3. Placental histology and expression of lipid metabolism genes were studied. Lipid levels, aortic lesions, and mRNA expression of cholesterol synthesis genes were investigated in fetuses at the end of gestation. A group of fetuses was allowed to attain early adulthood to investigate the liver lipid metabolism and atherogenesis with and without an HFD. KEY FINDINGS: Elevated maternal lipid levels and placental gene expression were differentially modulated in HFD-fed mothers. HFD-fed rabbits demonstrated differential expression of the placental genes involved in receptor-mediated endocytosis of cholesterol, lipogenesis, and lipolysis in all three trimesters. It resulted in significant lipid depositions in the placenta, hyperlipidemia, and a decrease in hepatic cholesterol synthesis in fetuses at the end of gestation. There was no atherogenesis in the aorta of offspring at trimester 3, but such offspring of HFD-fed mothers developed atherosclerosis and non-alcoholic fatty liver (NAFL) with profound steatosis in their early adulthood with and without HFD. SIGNIFICANCE: Diet-induced MHC differentially expressed placental lipid genes that may program the offspring to develop atherosclerosis and associated NAFL in early adulthood.
