The Chinese pangolin (Manis pentadactyla) is a critically endangered mammal with a highly specialized diet. To enhance nutritional knowledge of its diet, we analyzed the colony composition of a nest of Polyrhachis dives ants, which is the key natural prey in the Chinese pangolin's diet. In addition, we determined the nutrient composition of the total colony compared with adult ants. Nutrients quantified in this study included: crude protein, crude fat, carbohydrate, and amino acids, minerals, and vitamins, as well as formic acid and chitin, which have not been reported in previous diet studies. Our results showed that the colony consisted of adults (82%), pupae (10%), larvae (7%), and eggs (1%) (fresh mass). Both the total colony and adult ants, respectively, contained high concentrations of crude protein (62.97% and 64.68%), chitin (49.25% and 60.40%), crude fat (10.12% and 9.91%) (dry matter basis), and formic acid (2.06% and 3.07%) (fresh mass). This implies that Chinese pangolin might prefer prey with high protein, high chitin, low fat, and low formic acid content. Colony and adult ants differed in chemical composition in many aspects, thus it might be unsuitable to feed Chinese pangolin with only adult ants. Chitin and formic acid may play important roles in the diet and selectivity of Chinese pangolin. This study provides reference information that may be useful for developing better artificial diets with more comprehensive nutrient compositional data to meet the nutritional requirements of the Chinese pangolin under managed feeding programs.

Two new dopamine derivatives divesamides A (1) and B (2), along with six known N-containing compounds were isolated from the Chinese black ant (Polyrhachis dives). Their structures were determined on the basis of spectroscopic methods. Compound 1 is a racemate, and chiral HPLC separation yielded a pair of antipodes. The absolute configuration of (+)-1 was assigned by a computational method. The double signals in the (1)H and (13)C NMR spectra of 2 that resulted from the presence of a formamide group were discussed. The T- and B-lymphocytes proliferation assay showed that 2 has moderate immunosuppressive activity toward T- and B-lymphocytes proliferation at a concentration of 20 µM.

In the study, specific primers were designed based on the CO Ⅰ gene sequence of Polyrhachis dives. By optimizing the genomic DNA extraction method and amplification conditions, we established an efficient, specific, and accurate DNA molecular identification method for Polyrhachis dives. In this method, the length of the target fragment was 294-308 bp, and the other counterfeits had no target bands. In this paper, the specific identification method of the origin of Polyrhachis dives established can be used to identify the medicinal materials of Polyrhachis dives accurately.

Chinese black ant, Polyrhachis dives Smith, also known as P. vicina Roger has been used as a traditional edible insect and Chinese medicine in China and some southeast countries for thousands of years. In this study, Mitogenome of this species was assembled with high coverage using Illumina sequencing data and is 15,806 bp long in size. The base composition is 37.8% for A, 12.5% for C, 6.4% for G and 43.3% for T. The mitogenome contains 13 protein-coding genes, 22 transfer RNA genes and 2 ribosomal RNA genes. The phylogeny showed that it was closely related to Camponotus atrox (Hymenoptera: Formicidae) with high bootstrap value supported. The mitogenome of P. dives can provide essential DNA molecular data for further phylogenetic and evolutionary analysis.
