RESUMEN

Metal-free photocatalysis that produces reactive oxygen species (ROS) shows significant promising applications for environmental remediation. Herein, we constructed iodine-doped carbon nitride (I-CN) for applications in the photocatalytic inactivation of bacteria and the heterogeneous Fenton reaction. Our findings revealed that I-CN demonstrates superior photocatalytic activity compared to pure CN, due to enhanced light adsorption and a narrowed band gap. Antibacterial tests confirmed that I-CN exhibits exceptional antibacterial activity against both Escherichia coli and Staphylococcus aureus. The results showed that I-CN effectively generates superoxide radicals and hydroxyl radicals under light irradiation, resulting in enhanced antibacterial activity. In addition, I-CN can also be applied for a heterogeneous photo-Fenton-like reaction, achieving a high performance for the degradation of sulfamethoxazole (SMX), a typical antibiotic, via the photocatalytic activation of peroxymonosulfate (PMS). These results shed new light on the fabrication of metal-free nanozymes and their applications for disinfection and water decontamination.

RESUMEN

The utilization of byproducts from foxtail millet polishing can reduce food loss and waste. Thus, it is necessary to know the chemical compounds from the millet and the segregation of the layers. The nutrients including minerals were compared among the husk, bran, and millet, and a LC-MS metabolomics analysis was also performed among them. The results showed that the protein, crude fat and 4 fatty acids, seven minerals, the nitrogen-containing compounds and phenolic acids were at much higher levels in the bran part than the husk and millet, whereas the husk only contained higher levels of dietary fibre, and some minerals. The millet section, as the edible part, contained the lowest level of chemical constituents. It illustrated that the bran part contained more functional and nutritional components than the millet and husk part. Therefore, the bran of the foxtail millet should be a food resources instead of wasting.

In this study, the gene monascus-5162 from Monascus purpureus LQ-6, identified as chitin synthase gene VI (chs6), was knocked out to disrupt the chitin biosynthetic pathway and regulate the biosynthesis of Monascus pigments (MPs) and citrinin. The results showed that the aerial hyphae on a solid medium were short and sparse after the deletion of chs6 in M. purpureus LQ-6, significantly reducing the germination percentage of active spores to approximately 22%, but the colony diameter was almost unaffected. Additionally, the deletion of chs6 changed the mycelial morphology of M. purpureus LQ-6 during submerged fermentation and increased its sensitivity to environmental factors. MP and citrinin biosynthesis was dramatically inhibited in the recombinant strain. Furthermore, comparative transcriptome analysis revealed that the pathways related to spore development and growth, including the MAPK signaling pathway, chitin biosynthetic pathway, and regulatory factors LaeA and WetA genes, were significantly downregulated in the early phase of fermentation. The mRNA expression levels of genes in the cluster of secondary metabolites were significantly downregulated, especially those related to citrinin biosynthesis. This is the first detailed study to reveal that chs6 plays a vital role in regulating the cell growth and secondary metabolism of the Monascus genus.

Our previous work revealed that the anabolism of Monascus secondary metabolites is closely related to cofactor metabolism. In this study, we have further investigated the regulation mechanisms of respiratory complex I in response to the cell growth and secondary metabolite biosynthesis of M. purpureus. The results showed that downregulating the mRNA level of gene ndufs8 in M. purpureus sharply increased the secondary metabolites biosynthesis, cell growth and glucose consumption rates at the fermentation metaphase; slightly increased the colony diameter and biomass, and dramatically changed the mycelia morphology; and decreased the tolerances to environmental factors (especially H2O2). It also significantly inhibited the enzymes activities of respiratory complex I, III and superoxide dismutase, but stimulated that of complex II, IV and peroxidase, leading to an increase in reactive oxygen species (ROS) level and a decrease in ATP concentration. Furthermore, transcriptome analysis revealed that the mRNA levels of genes involved in respiratory chain, tricarboxylic acid cycle, and fatty acid degradation were downregulated, but those in the citrinin and monascus pigment biosynthesis and related pathways were upregulated. These data revealed that complex I plays a vital role in regulating the cell growth and secondary metabolism of Monascus via changing the intracellular ROS and ATP levels.

The mechanical strength of the stalk affects the lodging resistance and digestibility of the stalk in maize. The molecular mechanisms regulating the brittleness of stalks in maize remain undefined. In this study, we constructed the maize brittle stalk mutant (bk5) by crossing the W22:Mu line with the Zheng 58 line. The brittle phenotype of the mutant bk5 existed in all of the plant organs after the five-leaf stage. Compared to wild-type (WT) plants, the sclerenchyma cells of bk5 stalks had a looser cell arrangement and thinner cell wall. Determination of cell wall composition showed that obvious differences in cellulose content, lignin content, starch content, and total soluble sugar were found between bk5 and WT stalks. Furthermore, we identified 226 differentially expressed genes (DEGs), with 164 genes significantly upregulated and 62 genes significantly downregulated in RNA-seq analysis. Some pathways related to cellulose and lignin synthesis, such as endocytosis and glycosylphosphatidylinositol (GPI)-anchored biosynthesis, were identified by the Kyoto Encyclopedia of Gene and Genomes (KEGG) and gene ontology (GO) analysis. In bulked-segregant sequence analysis (BSA-seq), we detected 2,931,692 high-quality Single Nucleotide Polymorphisms (SNPs) and identified five overlapped regions (11.2 Mb) containing 17 candidate genes with missense mutations or premature termination codons using the SNP-index methods. Some genes were involved in the cellulose synthesis-related genes such as ENTH/ANTH/VHS superfamily protein gene (endocytosis-related gene) and the lignin synthesis-related genes such as the cytochrome p450 gene. Some of these candidate genes identified from BSA-seq also existed with differential expression in RNA-seq analysis. These findings increase our understanding of the molecular mechanisms regulating the brittle stalk phenotype in maize.

Blue-fluorescent blood-CDs were synthesized through a one-pot hydrothermal method using a mixture of chicken blood and trisodium citrate and then explored as a fluorescent probe for detecting Fe3+ and Hg2+. The probe showed excellent selectivity and sensitivity towards Fe3+ and Hg2+ with a dramatic "on-off" fluorescence response. F- recovered the fluorescence quenching by Fe3+, and Al3+ recovered the fluorescence quenching by Hg2+, showing an "off-on" fluorescence response. The blood-CDs were used as an "on-off-on" dual-channel fluorescent sensor for the detection and discrimination of Fe3+ and Hg2+ ions. The probe showed wide linear ranges for determination of Fe3+ (0-100 µM) and Hg2+ (0-120 µM) with low detection limits of 0.23 µM for Fe3+ and 0.17 µM for Hg2+. This probe was practically applied for the determination of Fe3+ and Hg2+ in piggery feed and wastewater with good recoveries. This work provides a fluorescent probe for the quantification of Fe3+ and Hg2+ in livestock feed and environmental water samples.

RESUMEN

The Landrevus species group includes four Velarifictorus species that are related to Landrevinae crickets (Velarifictoruselephas Gorochov, 1992, Velarifictorusbubalus Gorochov, 1992, Velarifictorusgradifrons Ingrisch, 1998, and Velarifictoruslandrevus Ma, Qiao & Zhang, 2019). A new species of the group is discovered in the Yunnan Province of China, and it is described and illustrated here. Velarifictorusyunnanensis Liu & Yin, 1993 is recognized as a junior synonym of Velarifictorusflavifrons Chopard, 1966. The morphological variety of V.flavifrons ectoparamere is documented and studied.

This study investigated the effects of cofactor metabolism on secondary metabolite production in M. purpureus through the application of different cofactor engineering strategies. Total pigment production dramatically increased by 39.08% and 40.89%, and yellow pigment production increased by 74.62% and 114.06% after the addition of 1.0 mg/L of the exogenous cofactor reagents methyl viologen and rotenone, respectively, in submerged batch-fermentation. The extracellular red pigment tone changed to yellow with the application of electrolytic stimulation at 800 mV/cm2, but almost no citrinin production was detected. In addition, the total pigment, yellow pigment and citrinin production increased by 35.46%, 54.89% and 6.27% after disruption of the nuoⅠ gene that encodes NADH-quinone oxidoreductase, respectively. Thus, cofactor metabolic engineering strategies could be extended to the industrial production of Monascus pigment or high yellow pigment with free citrinin production.
