RESUMEN

Environmental risk assessment of contaminated soils should ideally be carried out with complementary approaches (chemical and biological) conducted in situ and ex situ. While biological methods based on the assessment of effect and bioaccumulation in bioindicators exist for soil fauna organisms, such as land snails, the methodology is currently limited in the field to 14 metallic elements (MEs). To provide new relevant tools to the stakeholders of polluted fields, the aim of this work is to determine ex situ threshold guide values (ex situ TGVs), for 15 MEs, 16 polycyclic aromatic hydrocarbons (PAHs) and 7 polychlorinated biphenyls (PCBs). These ex situ TGVs are the usual concentration of contaminants found in the viscera of the bioindicator Cantareus aspersus after 28 days of exposure to uncontaminated soils. The second objective was to assess and validate the relevance of these ex situ TGVs for the interpretation of contamination levels in various European contaminated soils based on global index calculations: i) The sum of the excess of transfers (SETs) and ii) the weighted SETs based on the general toxicity points of each contaminant used to evaluate the risk of transferred MEs, PAHs and PCBs (ERITMEs, ERITPAHs and ERITPCBs, respectively). In addition, the influence of soil physico-chemical properties on accumulation was modelled to better understand their roles in bioavailability. The presented ex situ TGV and the associated indicators (the global sum of the excess of transfers and global ecotoxicological risk) provide a basis by which stakeholders can prioritize the management of polluted soils depending on the risk they may represent. The determination of ex situ TGVs for organic and inorganic compounds provides new tools to characterize excess contaminant transfers, and it will also allow the use of snails for ERAs, notably for common pollutants, such as PAHs and PCBs for which guide values are not available.

Stressors experience early in life by animals may have carry over impacts on life-traits over the life cycle. Accelerated telomere attrition induced by stress during development and growth could play a role in such delayed effects. Among stressors, exposure to chemicals may modify telomere dynamic but, to date, the trends evidenced between exposure and telomere shortening remains inconsistent. Moreover, the role of corticosterone as a possible mediator of chemical impact on telomere is not yet clearly established. Here, we investigated in wild populations of Red kite whether nestling exposure to metals and pesticides was related to corticosterone concentrations in feathers and telomere length measured in 47 individuals. Lead and mercury concentrations in blood ranged from 2.3 to 59.0 µg L-1 and to 1.4 to 115.7 µg L-1, respectively, and were below the toxicity thresholds proposed for wildlife. Rodenticides were detected in 30% of the chicks. Corticosterone increased with mercury and lead in interaction, showing a synergistic effect of these 2 non-essential metals on this stress hormone. Telomere length was not linked to metals and/or rodenticide exposure while it was related negatively to corticosterone. The relationship between telomere and corticosterone was modulated by nestling's age, which suggests that the rate of telomere shortening is higher when corticosterone increases. Our findings propose an effect of low exposure of Red Kite nestlings to mercury and lead mixture to raise baseline corticosterone in feathers. The relationships established suggest the hypothesis that telomere attrition could be an indirect consequence of metal exposure mediated by corticosterone.

Human health risk assessment (HHRA) and ecotoxicological risk assessment (ERA) of contaminated soils are frequently performed separately and based on total soil concentrations without considering the concepts of mobility, bioaccessibility and bioavailability. However, some chemical and biological assays rarely used in combination can be applied to more accurately assess the exposure of organisms to metal(loid)s and thus to better estimate the links between soil contamination and effects. For humans, the unified bioaccessibility method (UBM) assesses oral bioaccessibility, while for soil fauna such as land snails, the bioaccumulation test reflects the bioavailability of contaminants. The aim of this study is to explore the relationship between oral bioaccessibility and the bioavailability of arsenic, cadmium and lead in twenty-nine contaminated soils. The results show a modulation of bioaccumulation and bioaccessibility of metal(loid)s by soil physicochemical parameters (organic matter especially). For the three metal(loid)s studied, strong relationships were modelled between the UBM and snail tests (0.77 < r²adj.<0.95), depending on the parameters of the linear regressions (contaminant and phases of the UBM test). The original models proposed demonstrate the feasibility of linking bioaccessibility to humans and bioavailability to snails and the relevance of their association for an integrative risk assessment of contaminated soils.

The occurrence and fluxes of 18 priority substances and emerging pollutants listed in the European Union Water Framework Directive and a Watch List (trace metals (Cd, Pb and Ni), nonylphenols, octylphenols, 8 polyaromatic hydrocarbons, 4 dioxin-like polychlorinated biphenyls and diclofenac) were investigated in a Ukrainian city and the mass discharge loads of these compounds into EU-transboundary watersheds were estimated. Fluxes of chemicals were calculated per capita and per area of the Ukrainian urban territory and used to estimate mass loading of priority and emerging concern compounds from Lviv, Uzhorod and Chernivtsi (West Ukraine) to neighbouring EU-transboundary rivers. The highest loading was found for trace metals (1.15 t a-1), diclofenac (0.7 t a-1) and nonylphenols (0.4 t a-1). Transboundary water contamination must be considered in order to successfully manage water resources in a manner that fulfils the requirements of EU environmental quality standards.

AIMS: Emergence of azole-resistant Aspergillus fumigatus complicates management of Aspergillus diseases. Currently, selection pressure caused by azole fungicide use in farming is strongly suspected of creating resistance. As sawmills also use azole fungicides, we investigated the presence of azole-resistant strains in this environment and studied the relationship between azole fungicide use and development of resistance. METHODS AND RESULTS: Air (n = 200) and substrate (n = 600) samples were taken in 20 sawmills. Azole-resistant strains (Etest and EUCAST methods) were confirmed by sequencing the cyp51A gene and its promoters. Dosage of propiconazole and tebuconazole was performed by gas chromatography coupled with mass spectrometry. Twenty-four azole-resistant A. fumigatus strains were collected among 20 of the 600 substrate samples (3%). Eighty-three percent of theses strains had TR34 /L98H mutation. A significantly higher number of resistant strains was collected in sawmills using fungicide products made with propiconazole mixed with a high concentration of tebuconazole (P = 0·009). The presence of resistant strains was significantly linked to propiconazole quantities in substrates (P = 0·03). CONCLUSIONS: The outcome of azole-resistant A. fumigatus carrying TR34 /L98H mutation seems to greatly depend on the azole fungicide formulation and quantities of azole. These preliminary results are valuable to propose new approaches limiting the emergence of azole-resistant strains. SIGNIFICANCE AND IMPACT OF THE STUDY: Azole resistance is an emerging problem in A. fumigatus and threatens clinical advances made possible by the use of azole antifungals in the treatment of Aspergillus-related diseases. Azole fungicides are also used in the wood industry, notably in sawmills, to protect wood from wood-destroying fungi. Through our study, we show that sawmills represent another professional environment affected by the presence of azole-resistant A. fumigatus strains carrying the TR34 /L98H mutation. Moreover, this study provides valuable preliminary results to propose some new approaches to limit the emergence of azole-resistant A. fumigatus strains.

An As-contaminated soil (Unt) was amended with either iron grit (Z), a coal fly ash (beringite, B) or B + Z (BZ) and placed in lysimeters in 1997. An uncontaminated soil (R) was also studied. In summer and autumn 2003, lettuces were cultivated in the lysimeters and snails were caged for one month. Lettuce As concentrations were higher during the summer, while no differences occurred in snails between seasons. Snail As concentrations (microg g(-1) DW) ranged from 2.5 to 7.0 in B, Z and BZ, and peaked at 17.5 in Unt. In summer, snail survival was affected in Unt and Z compared to R and B while no mortality was noticed in autumn. Snail growth decreased only in B, BZ and Unt in autumn. Snail As concentrations suggest a risk for their predators even on the remediated soils.

As part of a larger program aiming at assessing transfer and effects of metals in food webs, this work studied the spatial distribution of Cd, Cr, Cu, Pb, and Zn in 101 sub-surface soils, systematically sampled (1 x 1 km regular grid) over a large area around Annaba, the fourth most-populated city of Algeria. Cd and Cr exhibited only one abnormally high value, with all other concentrations being close to pedogeological background. Some places in the centre of the city were polluted by Pb (up to 823 mg kg(-1)), probably due to aerial deposition from gasoline exhausts. Zn never exceeded regulatory limits over the whole sampling area. Cu was the only element for which a spatial autocorrelation occurred. A spatial interpolation by cokriging allowed the identification of agricultural activities as the main Cu pollution source. Our approach revealed various anthropogenic pollution sources, more efficiently for large-scale patterns than for local abnormalities.
