RESUMEN

OBJECTIVE: To investigate the efficacy and safety of daratumumab based regimens in relapse and/or refractory multiple myeloma (RRMM) in the real world, as well as the impact of daratumumab on stem cell collection and engraftment. METHODS: The clinical data of patients with RRMM who received daratumumab in hematology department of the First Affiliated Hospital of Xiamen University from February 2019 to March 2023 and had evaluable efficacy were retrospective analysis. RESULTS: All 43 RRMM patients were treated with daratumumab-based combination regimens, including Dd, DVd, DRd, Dkd, DId, and Dara-DECP. With median follow-up time 10.1 (2.1-36.6) months, the best overall response rate (ORR) was 74.4% and a best complete response rate (CR) was 25.6%. 1-year overall survival rate (OS) was 84.5%. The most common severe hematologic adverse events (Grade>3) are 3/4 grade leukopenia(18.6%), and the most common severe non-hematologic adverse events were infusion-related reactions (IRRs, 20.9%) and infections(7.0%). Multivariate prognostic analysis showed that extramedullary infiltration was an independent adverse prognostic factor affecting OS (P =0.004). The use of daratumumab has no effect on stem cell collection, or engraftment. CONCLUSION: Daratumumab is safe and effective in RRMM.

RESUMEN

The antioxidant N-(1,3-Dimethylbutyl)-N'-phenyl-p-phenylenediamine (6PPD) and its oxidized quinone product 6PPD-quinone (6PPD-Q) in rubber have attracted attention due to the ecological risk that they pose. Both 6PPD and 6PPD-Q have been detected in various environments that humans cohabit. However, to date, a clear understanding of the biotransformation of 6PPD-Q and a potential biomarker for exposure in humans are lacking. To address this issue, this study presents a comprehensive analysis of the extensive biotransformation of 6PPD-Q across species, encompassing both in vitro and in vivo models. We have tentatively identified 17 biotransformation metabolites in vitro, 15 in mice in vivo, and confirmed the presence of two metabolites in human urine samples. Interestingly, different biotransformation patterns were observed across species. Through semiquantitative analysis based on peak areas, we found that almost all 6PPD-Q underwent biotransformation within 24 h of exposure in mice, primarily via hydroxylation and subsequent glucuronidation. This suggests a rapid metabolic processing of 6PPD-Q in mammals, underscoring the importance of identifying effective biomarkers for exposure. Notably, monohydroxy 6PPD-Q and 6PPD-Q-O-glucuronide were consistently the most predominant metabolites across our studies, highlighting monohydroxy 6PPD-Q as a potential key biomarker for epidemiological research. These findings represent the first comprehensive data set on 6PPD-Q biotransformation in mammalian systems, offering insights into the metabolic pathways involved and possible exposure biomarkers.

The establishment of heterojunctions was considered as an exceptional strategy to obtain high-efficiency charge separation and enhanced photocatalytic performance. Herein, a series of FePMo/MIL-53(Fe) (FeM-53) heterojunctions were successfully constructed through in-situ growth of FePMo onto MIL-53(Fe) surface and their photocatalytic capacity were examined by visible-light-induced Cr(VI) reduction. Interestingly, the as-fabricated composites offered various photocatalytic activities controllably relying on the mass ratio of FePMo to MIL-53(Fe). Particularly, the one with the 10% ratio displayed the highest Cr(VI) reduction rate (100%) within 75 min, which was respectively over 4 and 2 folds higher than pure FePMo and MIL-53(Fe). The boosted photoactivity might be ascribed to the establishment of S-scheme heterojunction with suitable band alignment between FePMo and MIL-53(Fe), which broadened the light absorption range and improved charge separation. Further mechanism investigations implied both •O2- and e- were the key reactive species for Cr(VI) removal. Besides, the composite preserved excellent stability after 4 consecutive tests, and performed well in the presence of organic dyes. Such a S-scheme heterojunction may promise for highly efficient environmental mitigation.

AIMS: The aim of this study is to explore the anti-depressant mechanism of Chaihu- Shugan San based on serum medicinal chemistry and network pharmacology methods. BACKGROUND: Depression lacks effective treatments, with current anti-depressants ineffective in 40% of patients. Chaihu-Shugan San (CHSGS) is a well-known traditional Chinese medicine compound to treat depression. However, the chemical components and the underlying mechanisms targeting the liver and brain in the anti-depressant effects of CHSGS need to be elucidated. METHODS: The chemical components of CHSGS in most current network pharmacology studies are screened from TCMSP and TCMID databases. In this study, we investigated the mechanism and material basis of soothing the liver and relieving depression in the treatment of depression by CHSGS based on serum pharmacochemistry. The anti-depressant mechanism of CHSGS was further verified by proteomics and high-throughput data. RESULTS: Through serum medicinal chemistry, we obtained 9 bioactive substances of CHSGS. These ingredients have good human oral bioavailability and are non-toxic. Based on liver ChIPseq data, CHSGS acts on 8 targets specifically localized in the liver, such as FGA, FGB, and FGG. The main contributors to CHSGS soothing the liver qi targets are hesperetin, nobiletin, ferulic acid, naringin and albiflorin. In addition, network pharmacology analysis identified 9 blood components of CHSGS that corresponded to 63 anti-depressant targets in the brain. Among them, nobiletin has the largest number of anti-depressant targets, followed by glycyrrhizic acid, ferulic acid, albiflorin and hesperetin. We also validated the anti-depressant mechanism of CHSGS based on hippocampal proteomics. CHSGS exerts anti-depressant effects on synaptic structure and neuronal function by targeting multiple synapse related proteins. CONCLUSION: This study not only provides a theoretical basis for further expanding the clinical application of CHSGS, but also provides a series of potential lead compounds for the development of depression drugs.

Sporadic or late-onset Alzheimer's disease (LOAD) accounts for more than 95% of Alzheimer's disease (AD) cases without any family history. Although genome-wide association studies have identified associated risk genes and loci for LOAD, numerous studies suggest that many adverse environmental factors, such as social isolation, are associated with an increased risk of dementia. However, the underlying mechanisms of social isolation in AD progression remain elusive. In the current study, we found that 7 days of social isolation could trigger pattern separation impairments and presynaptic abnormalities of the mossy fibre-CA3 circuit in AD mice. We also revealed that social isolation disrupted histone acetylation and resulted in the downregulation of 2 dentate gyrus (DG)-enriched miRNAs, which simultaneously target reticulon 3 (RTN3), an endoplasmic reticulum protein that aggregates in presynaptic regions to disturb the formation of functional mossy fibre boutons (MFBs) by recruiting multiple mitochondrial and vesicle-related proteins. Interestingly, the aggregation of RTN3 also recruits the PP2A B subunits to suppress PP2A activity and induce tau hyperphosphorylation, which, in turn, further elevates RTN3 and forms a vicious cycle. Finally, using an artificial intelligence-assisted molecular docking approach, we determined that senktide, a selective agonist of neurokinin3 receptors (NK3R), could reduce the binding of RTN3 with its partners. Moreover, application of senktide in vivo effectively restored DG circuit disorders in socially isolated AD mice. Taken together, our findings not only demonstrate the epigenetic regulatory mechanism underlying mossy fibre synaptic disorders orchestrated by social isolation and tau pathology but also reveal a novel potential therapeutic strategy for AD.

PURPOSE: The purpose of our study was to evaluate the interocular symmetry and distribution of peripapillary vessel density in young myopic eyes. METHODS: A cross-sectional observational study was designed. A total of 174 eyes of 87 young myopic patients were recruited in this study. According to spherical equivalent (SE), 48 eyes were classified as mild myopia with a mean SE of - 2.12D (SD 0.66D), 66 as moderate myopia with a mean SE of - 4.50D (SD 0.87D), and 60 as high myopia with a mean SE of - 7.39D (SD 1.30D). Optical coherence tomography angiography (OCTA) was used to measure the vessel density. The distribution and interocular symmetry of peripapillary vessel densities were analyzed. RESULTS: The vessel densities in the whole image, peripapillary, superior and inferior sectors were significantly lower in the high myopia group than in the mild or moderate myopia group (All P < 0.001), and the density in the nasal sector was significantly lower in the high myopia group than in the mild group. And most interesting, the vessel densities in the inside disc and temporal sector showed no difference among the three myopic groups (All P > 0.05). By Pearson correlation analysis, the vessel densities in the whole image, peripapillary, superior, inferior and nasal sectors were negatively correlated with axial length (AL) and SE (All P < 0.001), but vessel densities in the inside disc and temporal sector did not show this correlation (All P > 0.05). Interocular symmetry was observed in all the vascular parameters through paired-samples t-tests (All P > 0.05), intraclass correlation coefficient (ICC) and Pearson correlation analysis (All P < 0.001). CONCLUSION: The density of radial peripapillary capillaries decreased in the myopic eye with axial elongation, and optical vascular parameters showed significant interocular symmetry among young myopic eyes.

The semiconductors have exhibited great potential in the field of photocatalytic energy production, environmental remediation and bactericidal. Nevertheless, those inorganic semiconductors are still restricted in commercial application due to the drawbacks of easy agglomeration and low solar energy conversion efficiency. Herein, ellagic acid (EA) based metal-organic complexes (MOCs) were synthesized through a facile stirring process at room temperature with Fe3+, Bi3+ and Ce3+ as the metal center. The EA-Fe photocatalyst exhibited superior photocatalytic activity toward Cr(VI) reduction, where Cr(VI) were completely removed within 20 min. Meanwhile, EA-Fe also displayed good photocatalytic degradation of organic contaminants and photocatalytic bactericidal performance. The photodegradation rates of TC and RhB by EA-Fe were 15 and 5 times that by bare EA, respectively. Moreover, EA-Fe was capable of effectively eliminating both E. coli and S. aureus bacteria. It was found that EA-Fe was capable of generating superoxide radicals, which could participate in the reduction of heavy metals, degradation of organic contaminants and inactivation of bacteria. A photocatalysis-self-Fenton system could be established by EA-Fe solely. This work would provide a new insight for designing multifunctional MOCs with high photocatalytic efficiency.

The neuron-glia cross-talk is critical to brain homeostasis and is particularly affected by neurodegenerative diseases. How neurons manipulate the neuron-astrocyte interaction under pathological conditions, such as hyperphosphorylated tau, a pathological hallmark in Alzheimer's disease (AD), remains elusive. In this study, we identified excessively elevated neuronal expression of adenosine receptor 1 (Adora1 or A1R) in 3×Tg mice, MAPT P301L (rTg4510) mice, patients with AD, and patient-derived neurons. The up-regulation of A1R was found to be tau pathology dependent and posttranscriptionally regulated by Mef2c via miR-133a-3p. Rebuilding the miR-133a-3p/A1R signal effectively rescued synaptic and memory impairments in AD mice. Furthermore, neuronal A1R promoted the release of lipocalin 2 (Lcn2) and resulted in astrocyte activation. Last, silencing neuronal Lcn2 in AD mice ameliorated astrocyte activation and restored synaptic plasticity and learning/memory. Our findings reveal that the tau pathology remodels neuron-glial cross-talk and promotes neurodegenerative progression. Approaches targeting A1R and modulating this signaling pathway might be a potential therapeutic strategy for AD.

A novel polyimide (PI) with broad-spectra response, high photocatalytic activity and stability under super acidic conditions (pH = 0) was synthesized via polymerizing method. Two types of perylene-cored materials (PDIAN and PTCDA) with anhydride and diamine respectively, were applied as precursors for PI polymerization. The as-prepared PI was optimized at 1:1 initial molar ratio of PDIAN to PTCDA. Using common PI (synthesized from melamine and pyromellitic dianhydride) as comparison, the Cr(VI) reduction rate was boosted from 25.4% to 96.6% within 120 min light irradiation. The corresponding rate constant by PI(PDIAN/PTCDA) was estimated to be ca. 11.7 times relative to that by common PI. The boosted performance was ascribed to the strong π-π conjugation from diperylene cores, which can decrease the photoluminescence intensity and electrochemical impedance, so as to promote the separation and transfer of photogenerated electron-hole pairs. In addition, the optimized PI(PDIAN/PTCDA) displayed wide-spectra response, which can still work under 730 nm light. The influencing factors toward Cr(VI) reduction were also clarified to be beneficial at lower pH and increased concentration of hole scavenger. After five cycles at pH 0, the PI still maintained excellent redox activity and structural stability.

Assessment of human exposure to mixtures of non-persistent chemicals from food matrices and consumer products requires accurate characterization and estimation of their preceding exposure levels, and assessment sampling approaches for these varying chemicals remain disputable. Here, we used high-throughput targeted method to quantify urinary concentrations of 59 most common non-persistent chemicals (6 parabens, 14 bisphenols, 1 triclosan, 7 benzophenones, 2 dichlorophenols, 13 phthalate metabolites and 16 antioxidants) in 158 consecutive spot samples from 11 participants over three consecutive days, 33 samples of which were first morning voids (FMVs). We found 49 chemicals with detection frequencies over 70 % in all urine samples. Principal component analyses showed greater inter-person variations than each person's inter-day variations. Intraclass correlation coefficient (ICC) to assess the reproducibility of targeted chemicals demonstrated that regardless of sampling approaches, dichlorophenols, most parabens, benzophenones and triclosan showed moderate to high reproducibility (0.445 < ICC < 0.969), with relatively high predictive power of FMVs for 24-h collections. Notably, most phthalates, bisphenols and antioxidants showed low ICC values. Together, our work demonstrates that FMV samples may be adequate for assessing human exposure to parabens, benzophenones, triclosan and dichlorophenols, whereas multiple consecutive urine collections may be advantageous for evaluating exposure to most phthalates, bisphenols and antioxidants.

BACKGROUND: We carried out a systematic review and meta-analysis to assess the safety and effectiveness of hepatic arterial infusion chemotherapy (HAIC) compared with transarterial chemoembolization (TACE) for patients with unresectable hepatocellular carcinoma (uHCC). METHODS: Eligible studies were searched by MEDLINE, the Cochrane Library, Embase, and Web of Science from January 1995 to January 2022, investigating eligible literature comparing HAIC and TACE for patients with HCC. The main outcome measures included progression-free survival (PFS), overall survival (OS), adverse events (AEs), objective response rate (ORR), and diseases control rate (DCR). RESULTS: Eight literature and 1028 patients were enrolled in this meta-analysis. The pooled PFS, OS, ORR, and DCR were HR = 0.89 (95% CI, 0.81-0.98), HR = 0.84 (95% CI, 0.75-0.93), OR = 2.77 (95% CI, 2.01-3.80), and OR = 4.64 (95% CI, 2.40-8.99), respectively. The adverse events of HAIC were lower than TACE. CONCLUSION: Our meta-analysis revealed that HAIC can achieve a better effect and survival benefits than TACE in patients with uHCC.

Three-dimensional (3D) tumor cell culture offers a more tissue-recapitulating model in cancer treatment evaluation. However, conventional models based on cell-substrate adhesion deprivation are still of insufficient real tumor mimic. In this work, a novel method is proposed for inducing multicellular spheroids (MCSs) formation based on hydrogel with tunable microenvironmental properties. Colon tumor cells DLD1 cultured on hydrogel substrate with proper physical stimulation form MCSs via self-organization. Chemotherapy based on clinical drug and far-infrared photothermal therapy is evaluated with DLD1 MCSs obtained by this method. The synergism of chemotherapy and noninvasive photothermal therapy based on graphene device is further verified in MCSs model and it is believed this method holds potential in in vitro anti-tumor strategies evaluation for precision medicine.

The spatial distribution, homologue patterns, and ecological risks of chlorinated paraffins (CPs) were investigated in sediments from sixteen mangrove wetlands along the South China Coast (SCS). The total concentrations of CPs in mangrove sediments from Guangdong, Fujian, Guangxi, and Hainan were in the range of 933-4760, 619-2300, 375-1550, and 271-658 ng/g dry weight, respectively. The contamination levels and spatial distribution of short-chain and medium-chain CPs (SCCPs and MCCPs, respectively) in mangrove sediments were mainly affected by local population scale and CP industries. The dominant CP patterns in sediments were C10-11Cl6-8 and C14Cl7-9 for SCCPs and MCCPs, respectively. Redundancy analysis, based on CP levels and several potential influencing factors showed that MCCPs/SCCPs ratio was the main factor affecting the accumulation of CPs in mangrove sediments. Additionally, MCCP concentrations were significantly correlated with total organic carbon (TOC), indicating that TOC might affect MCCP accumulation in mangrove sediments. Risk assessments indicated that CPs would pose medium ecological risks to sediment dwelling organisms in nearly one-third of the sampling sites. This is the first comprehensive report of the sedimentary SCCPs and MCCPs in mangrove wetlands along the SCS and highlights the need for more sediment toxicity data for CPs.

Most of the studies on short- and medium-chain chlorinated paraffins (SCCPs and MCCPs) in human tissues have focused on human milk and blood. However, little is known about the occurrence of CPs in human hair and nails. In this study, SCCPs and MCCPs were analyzed in 62 pairs of human hair and nails from North China. Median concentrations (range) of SCCPs and MCCPs in human hair were 239 (19.2-877) and 325 (16.9-893) ng/g dw, respectively, all of which were significantly higher than 154 (57.7-355) and 233 (61.0-476) ng/g dw, respectively, in nail samples (p < 0.05). The homologue profiles of CPs in human hair were similar to those in nails, where SCCPs and MCCPs were dominated by C10Cl6-7 and C14Cl7-8, respectively. A significant positive relationship was observed between CP levels and age of people for hair, whereas negative linear correlations were observed for nails. The redundancy analysis indicated that age of people might be the main influencing factor on the accumulation of CPs in hair and nails. The present study performed comprehensive evaluation of CP exposure levels in human hair and nail and highlights the need for more data on relationship between internal and external exposure to CPs.

Coronavirus disease 2019 (COVID-19) combined with liver injury has become a very prominent clinical problem. Due to the lack of a clear definition of liver injury in patients with COVID-19, the different selection of evaluation parameters and statistical time points, there are the conflicting conclusions about the incidence rate in different studies. The mechanism of COVID-19 combined with liver injury is complicated, including the direct injury of liver cells caused by severe acute respiratory syndrome coronavirus 2 replication and liver injury caused by cytokines, ischemia and hypoxia, and drugs. In addition, underlying diseases, especially chronic liver disease, can aggravate COVID-19 liver injury. In the treatment of COVID-19 combined with liver injury, the primary and basic treatment is to treat the etiology and pathogenesis, followed by support, liver protection, and symptomatic treatment according to the clinical classification and severity of liver injury. Blast furnace slag loaded with sulfide nano zero valent iron (S-nZVI@BFS) was applied to remove oxytetracycline (OTC) from water. S-nZVI@BFS was synthesized via liquid reduction and characterized using scanning electron microscopy (SEM), X-ray diffraction (XRD), and Brunauer, Emmett and Teller (BET) theory. The effect of reaction time, initial concentration of OTC, initial pH, and coexisting hexavalent chromium[Cr(Ⅵ)] were investigated. The results show that Fe and S were successfully immobilized on the surface of S-nZVI@BFS, the specific surface area and pore volume of which increased to 141.986 m2·g-1 and 0.388 cm3·g-1, respectively, following the loading of nZVI and sulfurization. The utilization rate of the surface active sites of S-nZVI@BFS was improved with an increase of the initial concentration of OTC; the removal rate increased from 20.12 mg·g-1 to 202.74 mg·g-1 when the initial concentration of OTC was increased from 10 mg·L-1 to 100 mg·L-1. The removal rate decreased with pH, declining from 99.78 mg·g-1 to 41.12 mg·g-1 when pH was increased from 3 to 11 due to the switch from Fendon oxidation to electrostatic adsorption. There was notable competition between OTC and Cr(Ⅵ) meaning that Cr(Ⅵ) can inhibit the removal of OTC, which is dose dependent.

This study first discovered the prevalence of phthalate (PAE) alternatives and PAE monoesters alongside traditional PAEs with elevated concentrations in indoor dust from typical e-waste recycling industrial park and adjacent communities. Among nine PAEs, high-molecular-weight (HMW) PAEs dominated over low-molecular-weight (LMW) PAEs in e-waste dust, with total concentrations (∑9PAEs) ranging from 170 to 5300 µg g-1. The diisononyl phthalate (DiNP) was identified as the most abundant PAE in e-waste dust, with over 10 times higher median concentration than that measured in home dust. Total concentrations of three PAE alternatives ranged from 20 to 1600 µg g-1 in e-waste dust, which were 3-10 times higher than the measured levels in home dust. A total of 13 monoesters were all identified in all samples with total concentrations of 4.7-59 µg g-1, and biodegradation of diesters was recognized as the major source of monoesters present in indoor dust. Significant correlations between the concentrations of PAE alternatives and the HMW PAEs were observed (p < 0.05), indicating that they are being simultaneously used in electronic and electrical products. The occupationally high co-exposure of e-waste dismantling workers to multiple PAEs and PAE alternatives as well as their monoesters should be of concern.

Food intake has been identified as a major route for infants to short- and medium-chain chlorinated paraffins (SCCPs and MCCPs, respectively), but evaluations of SCCPs and MCCPs exposure for infants via dietary sources are limited. Here, SCCPs and MCCPs were analyzed in three categories of commonly used infant foods from the Chinese market. The mean concentrations (range) of SCCPs in infant formulas, cereals, and purees were 7.95 (2.32-54.2), 4.26 (2.73-8.81), and 4.66 (1.33-8.43) ng g-1 fresh weight, respectively, and the mean (range) MCCP concentrations were 4.77 (1.67-20.9), 2.91 (1.21-8.24), and 1.89 (0.53-5.41) ng g-1 fresh weight, respectively. Chlorinated paraffins (CP) concentrations in whole milk samples were significantly higher than those detected in the other infant foods (p < 0.01). This study is the first to determine the occurrence of CPs in baby purees. The estimated margin of exposure (MOE) values (SCCPs-MOE and MCCPs-MOE) in this study were all greater than 1000, indicating that there were no health risks posed by CP contamination in infant foods. Principal component analysis indicated that foods of animal origin were more influenced by MCCPs than those of plant-origin, indicating different contamination patterns of CPs between food groups.

Numerous studies have reported the environmental contamination with traditional organophosphate triesters (tri-OPEs), but there is very little information on emerging tri-OPEs and organophosphate diesters (di-OPEs), especially in e-waste recycling areas. In this study, we conducted a comprehensive survey to monitor a broad suite of 11 traditional tri-OPEs, 12 emerging OPEs, and 10 di-OPEs in indoor dust collected from the workshops of (n = 42) and residential homes adjacent to (n = 24) a mega e-waste recycling industrial park in South China. In addition to traditional tri-OPEs, all of the emerging OPEs and di-OPEs were frequently detected in the dust samples. Total concentrations of emerging tri-OPEs and di-OPEs were in the range of 1210-62 900 and 2010-55 600 ng/g in the workshop dust and 435-23 700 and 186-4350 ng/g in the local home dust, respectively, which were comparable to those of traditional tri-OPEs (1160-61 500 and 370-13 900 ng/g, respectively). Most OPEs exhibited significantly higher concentrations in workshop dust versus local home dust (p < 0.05), indicating that e-waste dismantling activities contributed to the high residues of OPEs in indoor dust. Correlation analysis revealed that tri-OPEs have some common emission sources, i.e., e-waste and household products, while di-OPEs could originate from different sources, e.g., tri-OPE degradation, direct commercial application, and impurities in tri-OPE formulas. For both occupational workers and local adults, the median estimated daily intake values of emerging tri-OPEs (7.5 and 1.7 ng/kg bw/day, respectively) and di-OPEs (3.9 and 0.2 ng/kg bw/day, respectively) were comparable to that of traditional tri-OPEs (4.3 and 1.0 ng/kg bw/day, respectively), which suggests the important contribution of the emerging tri-OPEs and di-OPEs to the overall risks of human external exposure to OPE chemicals.
