Lakes play a crucial role in the nitrogen (N) cycle, and eutrophication disrupts the balance of the nitrogen cycle within lakes, including both the N removal process and the N supplement process. However, the mechanisms by which different nutrient levels affect seasonal nitrogen variations in the water columns are not clear, especially for long-term and large- scale studies. In this study, we used 206 independent spatial samples from a total of 108 subtropical shallow lakes from four surveys in the middle and lower reaches of the Yangtze River, as well as time-case study data from Lake Taihu and Lake Donghu of up to 23 and 14 years, respectively, to analyze the changes in summer TN compared to spring (delta TN). Delta TN was significantly negatively correlated with initial spring TN concentrations, with similar trends observed in both space and time. Furthermore, the slopes of spring TN vs. delta TN varied little across lakes in both time and space, suggesting a consistent relationship between initial spring TN and summer TN changes. When initial TN or TN: TP ratio was low, N fixation by algae played a significant role in compensating for summer N removal, thus mitigating summer N reductions; when TN was high or TN: TP ratio was high, ammonia stress reduced the compensatory effect of algae and denitrification played a significant role in summer N removal, thus increasing summer N reductions. Our study suggested that no matter what the initial conditions are, lakes tend to evolve towards a common nutrient status through biological regulation.

This single-center, randomized, open, two-preparation, single-dose, two-period, self-crossover trial aimed to assess the bioequivalence and safety of the test (T) preparation compared to the reference (R) preparation following intravenous injection in healthy subjects under fasting conditions. Twenty-four healthy subjects were enrolled in the study and subjects were randomly divided into two groups at a 1:1 ratio and were administered once per period, with an 8-day washout period. During each period, serum drug concentrations were detected for pharmacokinetic analysis and adverse events were recorded for safety analysis. The 90% confidence intervals for the geometric mean ratios (T:R) of maximum serum concentration, area under the serum concentration-time curve from time zero to the last measurable concentration, and area under the serum concentration-time curve from time zero to infinite time fell within the predefined bioequivalence range of 80%-125%, indicating bioequivalence between the T and R preparation under fasting conditions. Additionally, four subjects (16.7%) experienced five instances of adverse events in the T group, while five subjects (21.7%) experienced five instances of adverse events in the R group. This trial indicated the potential bioequivalence between the T and R products under fasting conditions, based on pharmacokinetic and safety profile.

Grapevine (Vitis vinifera L.,) is among the world's leading fruit crops. The production of grapes is severely affected by many diseases including gray mold, caused by the necrotrophic fungus Botrytis cinerea. Although all Vitis species can be hosts for B. cinerea, V. vinifera are particularly susceptible. Accordingly, this disease poses a significant threat to the grape industry and causes substantial economic losses. Development of resistant V. vinifera cultivars has progressed from incidental selection by farmers, to targeted selection through the use of statistics and experimental design, to the employment of genetic and genomic data. Emerging technologies such as marker-assisted selection and genetic engineering have facilitated the development of cultivars that possess resistance to B. cinerea. A promising method involves using the CRISPR/Cas9 system to induce targeted mutagenesis and develop genetically modified non-transgenic crops. Hence, scientists are now engaged in the active pursuit of identifying genes associated with susceptibility and resistance. This review focuses on the known mechanisms of interaction between the B. cinerea pathogen and its grapevine host. It also explores innate immune systems that have evolved in V. vinifera, with the objective of facilitating the rapid development of resistant grapevine cultivars.

The detection rate of children's behavioral problems is on the rise throughout the world, reaching 18.8% in China. Maternal depressive symptoms is one of the main causes of children's behavioral problems. Our study explored the moderating roles and the specific moderating mechanism of children's emotion regulation and teacher-student relationship quality in the association between maternal depressive symptoms and children's behavioral problems based on the resilience framework and the multiple moderating model. A cross-sectional survey on mothers and teachers of Chinese primary school students in grades 1 to 3 in suburban Beijing was conducted to investigate children's behavioral problems, emotion regulation, teacher-student relationship quality, maternal depressive symptoms, and demographic characteristics (n = 300) in this study. Pathway analysis and the Johnson-Neyman method were used to determine the moderating roles and the specific moderating mechanism of emotion regulation and teacher-student relationship quality. Results showed that emotion regulation and teacher-student relationship quality played moderating roles in the association between maternal depressive symptoms and children's behavioral problems and the moderating mechanism was the additive moderating model. To be specific, emotion regulation and teacher-student relationship quality played moderating roles parallelly. Emotion regulation could moderate the negative effect of maternal depressive symptoms on both internalizing and externalizing behavioral problems in children. Meanwhile, teacher-student relationship quality could moderate the negative effect of maternal depressive symptoms on children's externalizing behavioral problems. The study highlighted the value of children's emotion regulation and teacher-student relationship quality against adverse family environments and gave an orientation for intervention.

In this study, we developed a flexible cathode for fabricating high-performance ternary organic solar cells (OSCs). With solvent engineering and acid treatment, the conductivity of the poly(3,4-ethylenedioxythiophene):poly(styrenesulfonate) (PEDOT:PSS) electrode was significantly enhanced with the sheet resistance reduced from 1081 to 83 Ω sq-1. After being coated with polyethylenimine, work function of the PEDOT:PSS electrode was tuned from -5.07 to -4.12 eV, which is beneficial for electron collection in OSCs. With this technique, the OSCs (on glass) showed an average power conversion efficiency (PCE) of 16.3%, which is comparable to that of conventional inverted OSCs with commonly used indium-tin oxide and sol-gel-processed zinc oxide. However, the processing temperature of the inverted OSCs was dramatically lowered from 200 to 120 °C. The flexible OSCs (on polyethylene naphthalate/PEDOT:PSS/PEIE) exhibited a high PCE of 14.1%. After being bended for 300 cycles, the PCE was only degraded by 8.5%, indicating the excellent bendability of the flexible OSCs with the organic cathode.

We develop a general theory of Fermi polarons at nonzero temperature, including particle-hole excitations of the Fermi sea shakeup to arbitrarily high orders. The exact set of equations of the spectral function is derived by using both Chevy ansatz and diagrammatic approach, and their equivalence is clarified to hold in free space only, with an unregularized infinitesimal interaction strength. The correction to the polaron spectral function arising from two-particle-hole excitations is explicitly examined for an exemplary case of Fermi polarons in one-dimensional optical lattices. We find quantitative improvements at low temperatures with the inclusion of two-particle-hole excitations, in both polaron energies and decay rates. Our exact theory of Fermi polarons with arbitrary orders of particle-hole excitations might be used to better understand the intriguing polaron dynamical responses in two or three dimensions, whether in free space or within lattices.

Triple negative breast cancer (TNBC) is a particularly lethal breast cancer (BC) subtype driven by cancer stem cells (CSCs) and an immunosuppressive microenvironment. Our study reveals that nucleus accumbens associated protein 1 (NAC1), a member of the BTB/POZ gene family, plays a crucial role in TNBC by maintaining tumor stemness and influencing myeloid-derived suppressor cells (MDSCs). High NAC1 expression correlates with worse TNBC prognosis. NAC1 knockdown reduced CSC markers and tumor cell proliferation, migration, and invasion. Additionally, NAC1 affects oncogenic pathways such as the CD44-JAK1-STAT3 axis and immunosuppressive signals (TGFß, IL-6). Intriguingly, the impact of NAC1 on tumor growth varies with the host immune status, showing diminished tumorigenicity in natural killer (NK) cell-competent mice but increased tumorigenicity in NK cell-deficient ones. This highlights the important role of the host immune system in TNBC progression. In addition, high NAC1 level in MDSCs also supports TNBC stemness. Together, this study implies NAC1 as a promising therapeutic target able to simultaneously eradicate CSCs and mitigate immune evasion.

Background: Ovarian cancer (OC) is a kind of lethiferous cancer in gynecology, and the development of chemoresistance is the brief reason for treatment failure. The genes which contribute to chemoresistance are often leading to short survival. Thus, this study aims to identify predictive markers for chemoresistance and survival from chemoresistant-related genes. Methods: Coremine was used to retrieve of genes linked to OC chemoresistance. The relationship of genes with patient survival was analyzed in 489 OC patients of The Cancer Genome Atlas (TCGA) cohort, which the subgroup of 90 resistant and 197 sensitive samples was used to determine gene expression. Kaplan-Meier (KM) plotter of 1,816 OC patients with survival data was retrieved for survival analysis. Survival analysis was carried out by the R survival package in R (version 3.3.1). KM and receiver operating characteristic (ROC) curve were respectively used to access the ability of a gene to predict survival and chemoresistance. Results: In this study, a group of genes potentially linked to OC chemoresistance was identified, which dysregulated in 90 chemoresistant tissues compared with 197 sensitive tissues. Of them, thirteen genes could predict chemoresistance in 1,347 patients, especially SOS1, MSH6, STAT5A were excellent for predicting chemoresistance to any drugs, platin and taxane, CASP2 and PARD6B for any drugs and platin, and HSP90AA1 and HSP90B1 for taxane. Meanwhile, 44 genes linked to OC chemoresistance could predict short overall survival (OS) and/or disease-free survival (DFS) in 489 OC patients, and 10 of them could predict short OS in large cohort of up to 1,657 patients. Finally, it is noteworthy that CASP2 was down-regulated in 90 chemoresistant samples, and low expression of the gene predicted chemoresistance in 1,347 patients, short OS and DFS in 489 patients, and short OS and progression-free survival (PFS) in 1,657 patients. Conclusions: The identified genes specifically the CASP2 might be potentially used as predictive marker, prognostic marker and therapeutic target in management of OC.

Soil salinization is a major environmental factor that severely affects global agriculture. Root endophytes can enter root cells, and offer various ecological benefits, such as promoting plant growth, improving soil conditions, and enhancing plant resistance. Su100 is a novel strain of endophytic fungus that was characterized from blueberry roots. In this study, we focused on evaluating the effects of Su100 secretion on maize growth. The results demonstrated that maize treated with Su100 fermentation broth (SFB) exhibited significantly stronger salt tolerance than the control. It is worth mentioning that the treated root system not only had an advantage in terms of biomass but also a change in root structure with a significant increase in lateral roots (LRs) compared to the control. Transcriptome analysis combined with hormone content measurements indicated that SFB upregulated the auxin signaling pathway, and also caused alterations in brassinosteroids (BR) and jasmonic acid (JA) biosynthesis and signaling pathways. Transcriptome analyses also indicated that SFB caused significant changes in the sugar metabolism of maize roots. The major changes included: enhancing the conversion and utilization of sucrose in roots; increasing carbon flow to uridine diphosphate glucose (UDPG), which acted as a precursor for producing more cell wall polysaccharides, mainly pectin and lignin; accelerating the tricarboxylic acid cycle, which were further supported by sugar content determinations. Taken together, our results indicated that the enhanced salt tolerance of maize treated with SFB was due to the modulation of sugar metabolism and phytohormone biosynthesis or signaling pathways. This study provided new insights into the mechanisms of action of endophytic fungi and highlighted the potential application of fungal preparations in agriculture.

For background, Hirsutella sinensis, the only anamorphic fungus considered an effective substitute for Cordyceps sinensis, possesses immunoregulatory properties. However, the specific mechanism underlying the immunoregulatory function of Hirsutella sinensis remains unclear. The purpose is to investigate the therapeutic effects of Hirsutella sinensis alcohol extract (HSAE) on immune dysregulation and elucidate the underlying mechanisms involved. For methods, we established inflammatory and immunosuppression models in vitro and in vivo to evaluate the bidirectional immunoregulatory function of HSAE via qRT-PCR and immunoblotting. We also studied its potential mechanism via RNA sequencing and transcriptional analysis. We further established M1 and M2 cell models to explore the effect of HSAE on M1/M2 polarization using qRT-PCR, immunoblotting, and flow cytometry. For results, our data demonstrated enhanced proliferation, phagocytosis, and antipathogenic activities of macrophages. Treatment with HSAE led to increases in the proportions of CD3+ and CD4+ immune cells in cyclophosphamide-induced immunosuppressed mice. Additionally, HSAE reduced the lipopolysaccharide (LPS)-induced expression of Il1b, Il6, Ifnb1, and Cxcl10 by inhibiting the activation of the NF-κB and MAPK pathways in vitro and improved mouse survival by reducing the proportion of M1/M2 macrophages in septic mice. Finally, we found that HSAE inhibited M1 polarization by decreasing the expression of iNOS and CD86 and promoted M2 polarization by increasing the expression of ARG1 and CD206. For conclusions, our study provides evidence that HSAE has the potential to enhance immune responses and suppress excessive inflammation. These effects were realized by modulating macrophage polarization, providing novel insights into the fundamental mechanism underlying the bidirectional immunomodulatory effect of HSAE.

OBJECTIVE: To assess the efficacy and safety of robotic thyroidectomy (RT) with super-meticulous capsular dissection (SMCD) versus open thyroidectomy (OT) we used a dynamic risk assessment system incorporating 131I-WBS along with radioactive iodine (RAI) efficacy evaluation. BACKGROUND: Currently, the therapeutic efficacy of robotic surgery remains controversial. The 131I whole-body scan (131I-WBS) dynamic risk assessment system can detect small residual thyroid tissues and lesions, which may be used as indicators for the surgical efficacy of RT or OT thyroidectomy in differentiated thyroid cancer (DTC). METHODS: This retrospective cohort study included 2,349 patients who underwent total thyroidectomy followed by RAI therapy in our department between August 2017 and June 2023. Propensity score matching was performed at a ratio of 1:3 based on surgical type and mean follow-up duration to minimize selection bias after excluding those lost to follow-up. The primary outcome was surgical completeness, assessed using a dynamic risk system incorporating 131I-WBS along with RAI efficacy evaluation. RESULTS: There was no significant difference in the number of metastatic lymph nodes removed between the two groups (P=0.45). The incidence rate of parathyroid gland transplantation was 395 (68.7%) in the OT group and 8 (3.8%) in the RT group (P<0.001). There were no differences in the thyroidectomy completeness based on the 3-hour iodine uptake rate and 99mTcO4- thyroid imaging between the two groups. The dynamic risk assessment with and without 131I-WBS showed significant differences (P<0.001). The postoperative and post-RAI dynamic risk scores, evaluated at the time of RAI and 6 months after RAI, did not differ significantly between the two groups (P>0.05). The rates of transient and permanent hypoparathyroidism were higher in the OT group than in the RT group (P<0.05). The local recurrence rates showed no significant difference between the groups. CONCLUSIONS: This study demonstrates that RT with SMCD can achieve outcomes equivalent to those of traditional open surgery when integrating the 131I-WBS dynamic evaluation system and the therapeutic effects of RAI. Additionally, robot surgery demonstrated a notable advantage in protecting parathyroid function.

Intensification of agricultural practices has been pivotal in meeting the nutritional demands of a burgeoning global population. However, the widespread application of nitrogen (N) fertilisers has contributed to environmental pollution. In this study, we quantitatively assessed the role of international crop trade in optimising the productivity of micro-nutrients and its implications for N fertiliser use. Using a comprehensive dataset spanning from 1961 to 2019, we analysed the trade flows of seven key micro-nutrients-vitamin C (VC), vitamin B3 (VB3), vitamin B6 (VB6), calcium (Ca), magnesium (Mg), iron (Fe), and zinc (Zn)-embedded in agricultural products. We developed a novel framework to evaluate trade optimality and functionality based on the concentration-weighted productivity of micro-nutrients per kilogram of N fertiliser. Our findings reveal that while international trade has generally contributed to enhancing micro-nutrient productivity per unit of N fertiliser, trade optimality has shown a decreasing trend. High-productivity countries tend to export less relative to their potential, whereas countries with lower productivity import a larger share of crops. This decoupling suggests the need to re-evaluate trade policies to ensure that they align with sustainable agricultural practices and environmental conservation goals. We also identified potential savings in N fertiliser use through optimised trade practices, with estimated savings of 15-45 Tg of N per year. This could mitigate the negative agricultural impact and demonstrates the significant role that trade can play in achieving global sustainability targets. Overall, our research underscores the importance of aligning international crop trade with sustainable N management strategies to enhance micro-nutrient availability, improve environmental outcomes, and contribute to global efforts in achieving the Sustainable Development Goals.

BACKGROUND: Research dissemination is essential to accelerate the translating of evidence into practice. Little is known about dissemination among Chinese public health researchers. This study aimed to explore the understanding and practices of disseminating research findings and to identify barriers and facilitators that influence dissemination activities to non-research audiences. METHODS: This study deployed an exploratory qualitative design with purposive and snowball sampling. One focus group with 5 participants and 12 in-depth interviews were conducted with participants working in diverse fields from universities (n = 10), the National Chinese Center for Disease Control and Prevention (n = 4), the Chinese National Cancer Center (n = 1), the Chinese National Center for Cardiovascular Disease (n = 1), and China office of a global research institute (n = 1) from May to December 2021 to reach saturation. Data were initially analyzed using inductive thematic analysis. The designing for dissemination (D4D) logic model was then used to organize themes and subthemes. Two coders independently coded all transcripts and discussed disparities to reach a consensus. RESULTS: Out of 17 participants, 12 misunderstood the concept of dissemination; 14 had disseminated to non-research audiences: 10 to the public, 10 to practitioners, and 9 to policymakers. We identified multiple barriers to dissemination to non-research audiences across four phases of the D4D logic model, including low priority of dissemination, limited application of D4D strategies, insufficient support from the research organizations, practice settings, and health systems, and overemphasis on academic publications. CONCLUSIONS: There was a lack of understanding and experience of dissemination, indicating a lack of emphasis on active dissemination in China. We provide implications for raising awareness, building capacity, facilitating multidisciplinary collaboration, providing incentives and infrastructure, changing climate and culture, establishing communication and executive networks, and accelerating systematic shifts in impact focus.

BACKGROUND: Epidermal growth factor receptor (EGFR) tyrosine kinase inhibitors (TKIs) have become the standard treatment for advanced non-small cell lung cancer (NSCLC) with EGFR mutations. However, NSCLC heterogeneity leads to differences in efficacy; thus, potential biomarkers need to be explored to predict the prognosis of patients. Recently, the prognostic importance of pre-treatment malnutrition and systemic inflammatory response in cancer patients has received increasing attention. METHODS: In this study, clinical information from 363 NSCLC patients receiving EGFR-TKI treatment at our clinical center was used for analysis. RESULTS: High nutritional risk index (NRI) and systemic inflammation response index (SIRI) were significantly associated with poor overall survival (OS) and progression-free survival (PFS) in NSCLC patients (P < 0.05). Importantly, NRI and SIRI were the best combination models for predicting clinical outcomes of NSCLC patients and independent OS and PFS predictors. Moreover, a nomogram model was constructed by combining NRI/SIRI, sex, smoking history, EGFR mutation, TNM stage, and surgery treatment to visually and personally predict the 1-, 2-, 3-, 4-, and 5-year OS of patients with NSCLC. Notably, risk stratification based on the nomogram model was better than that based on the TNM stage. CONCLUSION: NRI and SIRI were the best combination models for predicting clinical outcomes of NSCLC patients receiving EGFR-TKI treatment, which may be a novel biomarker for supplement risk stratification in NSCLC patients.

Development of functional recovery therapies is critical to reduce the global impact of stroke as the leading cause of long-term disability. Our previous studies found that acute-phase protein orosomucoid (ORM) could provide an up to 6h therapeutic time window to reduce infarct volume in acute ischemic stroke by improving endothelial function. However, its role in neurons and functional recovery post-stroke remains largely unknown. Here, we showed that exogenous ORM administration with initial injection at 0.5h (early) or 12h (delayed) post-MCAO daily for consecutive 7 days significantly decreased infarct area, improved motor and cognitive functional recovery, and promoted mitochondrial biogenesis after MCAO. While neuron-specific knockout of ORM2, a dominant subtype of ORM in the brain, produced opposite effects which could be rescued by exogenous ORM. In vitro, exogenous ORM protected SH-SY5Y cells from OGD-induced damage and promoted mitochondrial biogenesis, while endogenous ORM2 deficiency worsened these processes. Mechanistically, inactivation of CCR5 or AMPK eliminated the protective effects of ORM on neuronal damage and mitochondrial biogenesis. Taken together, our findings demonstrate that ORM, mainly ORM2, is an endogenous regulator of neuronal mitochondrial biogenesis by activating CCR5/AMPK signaling pathway, and might act as a potential therapeutic target for the functional recovery post-stroke.

RESUMEN

Ancient acupuncture medical cases not only possess historical literary value but also hold significant clinical importance. To enhance the utilization of ancient acupuncture medical cases and to guide the excavation of acupuncture theory and its clinical application, this study constructs an indexing template for "Ancient Acupuncture Medical Cases" based on the knowledge element theory and the fine-grained indexing requirements of ancient acupuncture texts. This template includes two levels, 7 categories, 28 knowledge element data, 31 semantic types, and 15 semantic relationships. These element data construct the interconnections of knowledge in ancient acupuncture medical cases, serving as a basis for fine-grained indexing of acupuncture medical case literature.

OBJECTIVES: To observe the effect of electroacupuncture (EA) on the intestinal flora-short chain fatty acids (SCFAs) metabolism axis in rats with simple obesity, so as to explore the underlying mechanism of EA in reducing obesity. METHODS: Male SD rats were randomly divided into control group, model group and EA group, with 6 rats in each group. The obesity model was established by feeding the rats with high-fat diet. Rats in the EA group were treated with EA at "Quchi" (LI11) and "Zusanli" (ST36) for 15 min, once daily for 21 consecutive days. The changes of body weight were observed every other day. H.E. staining was used to observe the pathological changes of adipose tissue and liver. The blood lipid content was detected by automatic biochemical analyzer. The diversity of intestinal flora in rat feces was analyzed by 16S rRNA high-throughput sequencing. The content of SCFAs in rat feces was detected by ultra-high performance liquid chromatography-electrospray ionization-mass spectrometry (UPLC-ESI-MS/MS). The correlation between the relative abundance of fecal intestinal flora and the content of SCFAs in rats was analyzed by Pearson method. RESULTS: Compared with the control group, the body weight of rats, the serum total cholesterol (TC) and triglyceride (TG) were significantly increased (P<0.01) in the model group. The results of 16S rRNA sequencing showed that, at the genus level the relative abundance of Bacteroides, Butyrivibrio and Roseburia in were decreased significantly(P<0.05), while that of the Lachnospiraceae_NK4A136_group increased(P<0.05). After the intervention, compared with the model group, the body weight, serum TC and TG contents of rats in the EA group were significantly decreased (P<0.05, P<0.01);the results of 16S rRNA sequencing showed that, the relative abundance of the Bacteroidota phylum significantly increased (P<0.01) and Firmicutes decreased (P<0.01) at the phylum level, and at the genus level the relative abundances of Bacteroides, Butyrivibrio and Roseburia significantly increased (P<0.01, P<0.05);the contents of acetic acid and propionic acid in SCFAs significantly increased (P<0.01). H.E. staining showed an increase of the diameter of adipocytes, with obvious lipid droplets and inflammatory infiltration in the model group, which was relatively milder in the EA group. PCoA analysis showed that there were significant differences in the structure of intestinal flora between the control group and the model group, as well as the model group and the EA group. At the phylum level, the relative abundance of Bacteroidota was positively correlated with acetic acid and propionic acid contents, with that of Firmicutes negatively correlated with acetic acid and propionic acid contents (P<0.001). At the genus level, the relative abundances of Bacteroides, Streptococcus and Butyricimonas were positively correlated with acetic acid content (P<0.01, P<0.05), and the relative abundances of Bacteroides, Streptococcus and Roseburia were positively correlated with propionic acid content (P<0.001, (P<0.05). CONCLUSIONS: EA can improve the disorder of lipid metabolism in obese rats by improving the disorder of intestinal flora-SCFAs metabolic axis, thus playing a role in inhibiting obesity.

Monitoring pesticide exposures in honey bees provides fundamental risk information that informs efforts to improve regulatory policy, pesticide use, and beekeeping management so pollinators are protected in realistic field conditions. We investigated pesticide exposures to bee colonies while colonies moved along commercial migratory routes in 2022 and 2023 to pollinate multiple pollinator-dependent, high-value U.S. specialty crops (e.g., almonds in California and apples and cherries in Washington). We found evident pesticide exposure patterns, including increasing exposures (both levels and number of pesticides) to fungicides during almond pollination, higher exposures to insecticides and persistent exposures to fungicides during springtime fruit pollination, and declining exposures in summer. Exposure risk assessment by risk quotient (RQ) model based on residues in bee bread indicates no concern of acute toxicity to adult honey bees during pollination, however, during colony inspections we found severe brood mortality in fields associated with high exposure to buprofezin, an insect growth regulator (IGR) thought to be safe for adult bees, which is permitted for use any time across the season. Our results suggest a need to improve compliance with insecticide label requirements during tree fruit pollination and a need for further research into the negative impacts of IGR on colony health especially on immature bees to inform potential policy changes.
