RESUMEN

Due to its fibrous structure and high water holding capacity, rock mineral wool (RMW) has boosted the development of hydroponics. Consequently, the amount of waste RMW has also increased tremendously, which has stimulated the research and development of RMW reuse options. In this study, composting and degradability of RMW from hydroponics (gRMW) were tested in combination with different ratios of biowaste compost, including physical and chemical properties of the starting and final materials, and potential ecological hazards of the final product. gRMW had high water holding capacity and low organic matter content, which was easily degradable. Limits of toxic elements according to EU regulation were not exceeded. Degraded gRMW mixtures with compost did not exhibit toxicity to plants or aquatic bacteria and showed intermediate or limited habitat function for earthworms, which preferred the sole gRMW not mixed with compost. Overall, degraded gRMW exhibited parameters of safe soil amendment.

BACKGROUND: Biliary tract cancers (BTCs) are usually diagnosed at an advanced stage, when the disease is incurable. Currently used tumor biomarkers have limited diagnostic value for BTCs, so there is an urgent need for sensitive and specific biomarkers for their earlier diagnosis. Deregulation of the homeostasis of trace elements is involved in the carcinogenesis of different cancers, including BTCs. The objective of the study is to determine/compare the total concentrations of copper (Cu), zinc (Zn) and iron (Fe) and the proportions of free Cu and Cu bound to ceruloplasmin (Cp) and the isotopic ratio of 65Cu/63Cu in serum samples from healthy volunteers and cancer patients using inductively coupled plasma-mass spectrometry-based methods (ICP-MS). PATIENTS AND METHODS: In this prospective, noninterventional, nonrandomized study 20 patients and 20 healthy volunteers will be enrolled to identify serum Cu, Zn and Fe levels, Cu isotopic fractionation as a predictive biomarker of response to systemic therapy of BTCs, which will be evaluated by computed tomography. Newly developed analytical methods based on ICP-MS will be applied to metal-based biomarker research in oncology. CONCLUSIONS: In the study the comparison of the total concentration of selected trace elements, the proportion of free Cu and Cu bound to Cp and the isotopic ratio of 65Cu/63Cu in serum samples from healthy volunteers and cancer patients will be conducted to provide the foundation for the development of a BTC cancer screening methodology and the data on their usability as a potential predictive biomarker for BTCs of response to systemic therapy.

BACKGROUND: Human lung tissue, as an interface with the environment, is susceptible to various environmental pollutants, including trace metals. However, quantitative data on trace metals in human lung tissues remain poorly described. METHODS: This study aimed to characterize the elemental composition of histologically healthy, unaffected parts of human lung tissues, associated with non-infective, non-infiltrative, and non-malignant diseases (n = 60) for essential (Cr, Mn, Fe, Co, Cu, Zn, and Se) and toxic trace elements (Sr, Ni, As, Cd, and Pb). Additionally, we investigated the influence of personal factors (sex, age, and smoking habits) on the examined trace element profiles, as well as between the trace elements correlations in the healthy human lungs. RESULTS: Among the analyzed trace elements, Fe was the most prevalent, while As was the least prevalent in healthy lung tissues. Stratifying by age revealed significantly higher Cr and Co (less Sr, Ni, and Pb) and lower Se levels in older individuals (above 65 years) compared to their younger counterparts. Sex-based differences were also notable, with Cu and Co 1.2- and 2.3-fold higher levels in females than in males. Exploring the impact of smoking habits revealed a striking 10-fold increase in Cd levels in the lung tissues of smokers compared to non-smokers. Correlation analyses showed significant positive associations between concentrations of certain toxic and essential trace elements in healthy lung tissues. CONCLUSIONS: This study could contribute to the establishment of baseline intervals for essential and toxic trace elements, valuable for toxicological and clinical assessment, in healthy, unaffected human lungs, and indicates the influence of sex, age, and smoking. However, further larger-scale studies are needed to make more stable conclusions.

BACKGROUND: Electrochemotherapy (ECT) is a treatment involving the administration of chemotherapeutics drugs followed by the application of 8 square monopolar pulses of 100 µs duration at a repetition frequency of 1 Hz or 5000 Hz. However, there is increasing interest in using alternative types of pulses for ECT. The use of high-frequency short bipolar pulses has been shown to mitigate pain and muscle contractions. Conversely, the use of millisecond pulses is interesting when combining ECT with gene electrotransfer for the uptake of DNA-encoding proteins that stimulate the immune response with the aim of converting ECT from a local to systemic treatment. Therefore, the aim of this study was to investigate how alternative types of pulses affect the efficiency of the ECT. MATERIALS AND METHODS: We performed in vitro experiments, exposing Chinese hamster ovary (CHO) cells to conventional ECT pulses, high-frequency bipolar pulses, and millisecond pulses in the presence of different concentrations of cisplatin. We determined cisplatin uptake by inductively coupled plasma mass spectrometry and cisplatin cytotoxicity by the clonogenic assay. RESULTS: We observed that the three tested types of pulses potentiate the uptake and cytotoxicity of cisplatin in an equivalent manner, provided that the electric field is properly adjusted for each pulse type. Furthermore, we quantified that the number of cisplatin molecules, resulting in the eradication of most cells, was 2-7 × 107 per cell. CONCLUSIONS: High-frequency bipolar pulses and millisecond pulses can potentially be used in ECT to reduce pain and muscle contraction and increase the effect of the immune response in combination with gene electrotransfer, respectively.

Temperature is a hallmark parameter influencing almost all magnetic resonance properties (e.g., T1 , T2 , proton density, and diffusion). In the preclinical setting, temperature has a large influence on animal physiology (e.g., respiration rate, heart rate, metabolism, and oxidative stress) and needs to be carefully regulated, especially when the animal is under anesthesia and thermoregulation is disrupted. We present an open-source heating and cooling system capable of regulating the temperature of the animal. The system was designed using Peltier modules capable of heating or cooling a circulating water bath with active temperature feedback. Feedback was obtained using a commercial thermistor, placed in the animal rectum, and a proportional-integral-derivative controller was used to modulate the temperature. Its operation was demonstrated in a phantom as well as in mouse and rat animal models, where the standard deviation of the temperature of the animal upon convergence was less than a 10th of a degree. An application where brain temperature of a mouse was modulated was demonstrated using an invasive optical probe and noninvasive magnetic resonance spectroscopic thermometry measurements.

We investigated microbial growth in increasing concentrations of hexavalent chromium (Cr(VI)) and its reduction by a microbial community enriched from tannery effluent and by the bacterial strains isolated from the enriched community. The bacterial growth was monitored by measuring the optical cell density (OD650), while the Cr(VI) concentration in the samples was determined using spectrophotometry and liquid chromatography hyphenated to inductively coupled plasma mass spectrometry (HPLC-ICP-MS). At a Cr(VI) concentration of 100 mg/L, the isolates affiliated with Pseudomonas aeruginosa (P. aeruginosa) reached higher optical cell densities, but were in general less effective for Cr(VI) reduction than the isolates affiliated with Mammaliicoccus sciuri (M. sciuri). All three M. sciuri isolates and only one of the seven P. aeruginosa isolates were able to reduce 50% of the Cr(VI) with an initial concentration of 100 mg/L within 24 h (pH 7.1), while the six isolates affiliated with P. aeruginosa were less effective. Compared to the isolated, individual bacterial strains, the enriched microbial community was better adapted to the elevated Cr(VI) concentrations, but needed a longer time (48 h) to reduce the Cr(VI) with the same efficacy as the most efficient individual isolates. The ability of the enriched microbial community and the isolated bacterial strains to reduce the Cr(VI) highlights their potential for use in the rapid bioremediation of wastewaters contaminated with Cr(VI).

BACKGROUND: Nanosecond electric pulses showed promising results in electrochemotherapy, but the underlying mechanisms of action are still unexplored. The aim of this work was to correlate cellular cisplatin amount with cell survival of cells electroporated with nanosecond or standardly used 8 × 100 µs pulses and to investigate the effects of electric pulses on cisplatin structure. MATERIALS AND METHODS: Chinese hamster ovary CHO and mouse melanoma B16F1 cells were exposed to 1 × 200 ns pulse at 12.6 kV/cm or 25 × 400 ns pulses at 3.9 kV/cm, 10 Hz repetition rate or 8 × 100 µs pulses at 1.1 (CHO) or 0.9 (B16F1) kV/cm, 1 Hz repetition rate at three cisplatin concentrations. Cell survival was determined by the clonogenic assay, cellular platinum was measured by inductively coupled plasma mass spectrometry. Effects on the structure of cisplatin were investigated by nuclear magnetic resonance spectroscopy and high-resolution mass spectrometry. RESULTS: Nanosecond pulses equivalent to 8 × 100 µs pulses were established in vitro based on membrane permeabilization and cell survival. Equivalent nanosecond pulses were equally efficient in decreasing the cell survival and accumulating cisplatin intracellularly as 8 × 100 µs pulses after electrochemotherapy. The number of intracellular cisplatin molecules strongly correlates with cell survival for B16F1 cells, but less for CHO cells, implying the possible involvement of other mechanisms in electrochemotherapy. The high-voltage electric pulses did not alter the structure of cisplatin. CONCLUSIONS: Equivalent nanosecond pulses are equally effective in electrochemotherapy as standardly used 8 × 100 µs pulses.

A new analytical procedure for the speciation of chromium (Cr) in plants by high performance liquid chromatography inductively coupled plasma mass spectrometry (HPLC-ICP-MS) was developed using a strong anion-exchange Mono Q column for the separation of the Cr species. To optimize the analytical procedure, Cr complexes were first synthesized from Cr-nitrate with the addition of an excess of ligand (90°C). Cr-oxalate, Cr-malate, Cr-citrate, Cr-aconitate and Cr-quinate complexes and Cr-nitrate (pH 6.5) were chromatographically separated from Cr(VI) by applying linear gradient elution from 100% water to 100% NH4Cl at a flow rate of 1.5 ml min-1 in 10 min. The column recoveries ranged from 100 to 104%. The exception was Cr-aconitate (column recovery 33%), where a quantitative synthesis was not possible. Good repeatability of the measurements (relative standard deviations better than ± 3%) and low limits of detection (below 0.37 ng ml-1 Cr) were achieved for the individual Cr species. The developed analytical procedure was applied to Cr speciation for dandelions (Taraxacum officinale) grown in soil with a high Cr content and a study of the uptake and metabolism of Cr species in dandelions grown in soil with a low Cr content treated with solutions of Cr(VI) or Cr-nitrate (5000 ng ml-1 Cr, pH 6.5) for 48 h. The separated Cr species were quantified by post-column isotope dilution ICP-MS, while the identification was based on retention times and was also supported by mass spectra obtained with high resolution mass spectrometry (HR-MS). The data indicate that for dandelions grown in Cr-rich soil and that treated with Cr-nitrate (pH 6.5), the Cr was mainly accumulated in the roots, while in plants treated with Cr(VI) (pH 6.5), the Cr was evenly distributed between the roots and the leaves. The Cr species found in dandelion roots and leaves were Cr-aconitate, Cr-malate, and Cr-quinate. The results revealed that Cr(VI) was completely reduced and metabolized to Cr(III) complexes. LA-ICP-MS data showed that the Cr in a leaf of dandelion grown in Cr-rich soil was localized mainly at the apex of the leaf.

This review presents integrated literature data on the occurrence and type of dietary fibres present in cereals and pseudocereals, as well as their technological role in production of wheat based dough and bread. The focus on wheat based dough and bread was in order to objectively compare the influence of the fibre and flour fractions of cereals and pseudocereals on the same food matrix. The evident heterogeneity, contradiction of scientific results and the lack of objective comparability were the main reason for compilation of systematic review. Remarkably, the literature is characterized by a large methodology gap that leads to significant knowledge gap due to the lack of the comparability of presented results on dietary fibre content, composition and influence on wheat based dough and bread properties. Based on this, the authors must emphasize the need for standardized presentation of scientific results in the future by scientific community.

BACKGROUND: The pan-European human biomonitoring initiative HBM4EU targets the harmonization of human biomonitoring (HBM) procedures and data for both environmental and occupational exposure, including chromium. The determination of chromium in urine (U-Cr), plasma (P-Cr) and whole blood (WB-Cr) is a common HBM application in employees occupationally exposed to chromium (VI) compounds. METHODS: European laboratories which have registered as candidate laboratories for chromium analysis within HBM4EU were invited to participate in a quality assurance/qualitycontrol (QA/QC) programme comprising interlaboratory comparison investigations (ICI) for the parameters U-Cr, P-Cr and WB-Cr. Participating laboratories received two samples of different concentrations in each of four rounds and were asked to analyse the samples using their standard analytical procedure. The data were evaluated by the Z-score approach and were reported to the participants after each round. RESULTS: The majority of the 29 participating laboratories obtained satisfactory results, although low limits of quantification were required to quantify chromium concentrations in some of the ICI materials. The robust relative standard deviation of the participants' results (study RSDR) obtained from all ICI runs ranged from 6 to 16 % for U-Cr, 7-18 % for P-Cr and 4-47 % for WB-Cr. The application of both inductively coupled plasma mass spectrometry (ICP-MS) and electrothermal atomic absorption spectrometry (EAAS) appeared appropriate for the determination of chromium in urine, plasma and whole blood with regard to occupational exposure levels. CONCLUSION: This QA/QC programme succeeded in establishing a network of laboratories with high analytical comparability and accuracy for the analysis of chromium across Europe.

To achieve good performance, athletes need to synchronize a series of movements in an optimal manner. One of the indicators used to monitor this is the order of occurrence of relevant events in the movement timeline. However, monitoring of this characteristic of rapid movement is practically limited to the laboratory settings, in which motion tracking systems can be used to acquire relevant data. Our motivation is to implement a simple-to-use and robust IMU-based solution suitable for everyday praxis. In this way, repetitive execution of technique can be constantly monitored. This provides augmented feedback to coaches and athletes and is relevant in the context of prevention of stabilization of errors, as well as monitoring for the effects of fatigue. In this research, acceleration and rotational speed signal acquired from a pair of IMUs (Inertial Measurement Unit) is used for detection of the time of occurrence of events. The research included 165 individual strikes performed by 14 elite and national-level karate competitors. All strikes were classified as slow, average, or fast based on the achieved maximal velocity of the hand. A Kruskal-Wallis test revealed significant general differences in the order of occurrence of hand acceleration start, maximal hand velocity, maximal body velocity, maximal hand acceleration, maximal body acceleration, and vertical movement onset between the groups. Partial differences were determined using a Mann-Whitney test. This paper determines the differences in the temporal structure of the reverse punch in relation to the achieved maximal velocity of the hand as a performance indicator. Detecting the time of occurrence of events using IMUs is a new method for measuring motion synchronization that provides a new insight into the coordination of articulated human movements. Such application of IMU can provide additional information about the studied structure of rapid discrete movements in various sporting activities that are otherwise imperceptible to human senses.

PURPOSE: Deuterium metabolic imaging (DMI) maps the uptake of deuterated precursors and their conversion into lactate and other markers of tumor metabolism. Even after leveraging 2 H's short T1 s, DMI's signal-to-noise ratio (SNR) is limited. We hypothesize that a multi-echo balanced steady-state free precession (ME-bSSFP) approach would increase SNR compared to chemical shift imaging (CSI), while achieving spectral isolation of the metabolic precursors and products. METHODS: Suitably tuned 2 H ME-bSSFP (five echo times [TEs], ΔTE = 2.2 ms, repetition time [TR]/flip-angle = 12 ms/60°) was implemented at 15.2T and compared to CSI (TR/flip-angle = 95 ms/90°) regarding SNR and spectral isolation, in simulations, in deuterated phantoms and for the in vivo diagnosis of a mouse tumor model of pancreatic adenocarcinoma (N = 10). RESULTS: Simulations predicted an SNR increase vs. CSI of 3-5, and that the peaks of 2 H-water, 2 H6,6' -glucose, and 2 H3,3' -lactate can be well isolated by ME-bSSFP; phantoms confirmed this. In vivo, at equal spatial resolution (1.25 × 1.25 mm2 ) and scan time (10 min), 2 H6,6' -glucose's and 2 H3,3' -lactate's SNR were indeed higher for bSSFP than for CSI, three-fold for glucose (57 ± 30 vs. 19 ± 11, P < .001), doubled for water (13 ± 5 vs. 7 ± 3, P = .005). The time courses and overall localization of all metabolites agreed well, comparing CSI against ME-bSSFP. However, a clearer localization of glucose in kidneys and bladder, the detection of glucose-avid rims in certain tumors, and a heterogeneous pattern of intra-tumor lactate production could only be observed using ME-bSSFP's higher resolution. CONCLUSIONS: ME-bSSFP provides greater SNR per unit time than CSI, providing for higher spatial resolution mapping of glucose uptake and lactate production in tumors.

Recent magnetic resonance studies in healthy and cancerous organs have concluded that deuterated metabolites possess highly desirable properties for mapping non-invasively and, as they happen, characterizing glycolysis and other biochemical processes in animals and humans. A promising avenue of this deuterium metabolic imaging (DMI) approach involves looking at the fate of externally administered 2H6,6'-glucose, as it is taken up and metabolized into different products as a function of time. This study employs deuterium magnetic resonance to follow the metabolism of wildtype and preeclamptic pregnant mice models, focusing on maternal and fetoplacental organs over ≈2 h post-injection. 2H6,6'-glucose uptake was observed in the placenta and in specific downstream organs such as the fetal heart and liver. Main metabolic products included 2H3,3'-lactate and 2H-water, which were produced in individual fetoplacental organs with distinct time traces. Glucose uptake in the organs of most preeclamptic animals appeared more elevated than in the control mice (p = 0.02); also higher was the production of 2H-water arising from this glucose. However, the most notable differences arose in the 2H3,3'-lactate concentration, which was ca. two-fold more abundant in the placenta (p = 0.005) and in the fetal (p = 0.01) organs of preeclamptic-like animals, than in control mice. This is consistent with literature reports about hypoxic conditions arising in preeclamptic and growth-restricted pregnancies, which could lead to an enhancement in anaerobic glycolysis. Overall, the present measurements suggest that DMI, a minimally invasive approach, may offer new ways of studying and characterizing health and disease in mammalian pregnancies, including humans.

Heat stress is one of the most important environmental factors that influences wheat growth and development, leading to significant losses in grain yield and has become a significant detrimental factor for worldwide wheat production. In recent years, several studies suggested that eukaryotic elongation factor 1A (eEF1A), may contribute to heat tolerance in plants, therefore the aim of this study was: to investigate the accumulation of eEF1A in wheat under conditions of moderate and high air temperatures; to determine the amount of photosynthetic pigments and to determine the yield traits; and to examine whether there is a correlation between eEF1A accumulation, photosynthetic pigments, and yield in different wheat varieties. The results showed that heat stress induced accumulation of eEF1A significantly different among wheat varieties and showed that varieties with a higher accumulation of eEF1A under heat stress are characterized by a smaller decrease in the photosynthetic pigments. A correlation between higher accumulation of eEF1A under heat stress and yield traits was found. Analyzed parameters from two growing seasons, indicated that the higher accumulation of eEF1A and a smaller decrease in photosynthetic pigments distinguishes the varieties more resistant to heat stress. The analysis of the molecular mechanisms by immunoblot, under conditions of high and moderate air temperatures in two growing seasons, aims to develop agricultural strategy and develop wheat varieties tolerant to heat stress.

Detecting and mapping metabolism in tissues represents a major step in detecting, characterizing, treating and understanding cancers. Recently introduced deuterium metabolic imaging techniques could offer a noninvasive route for the metabolic imaging of animals and humans, based on using 2 H magnetic resonance spectroscopic imaging (MRSI) to detect the uptake of deuterated glucose and the fate of its metabolic products. In this study, 2 H6,6' -glucose was administered to mice cohorts that had been orthotopically implanted with two different models of pancreatic ductal adenocarcinoma (PDAC), involving PAN-02 and KPC cell lines. As the tumors grew, 2 H6,6' -glucose was administered as bolii into the animals' tail veins, and 2 H MRSI images were recorded at 15.2 T. 2D phase-encoded chemical shift imaging experiments could detect a signal from this deuterated glucose immediately after the bolus injection for both the PDAC models, reaching a maximum in the animals' tumors ~ 20 min following administration, and nearly total decay after ~ 40 min. The main metabolic reporter of the cancers was the 2 H3,3' -lactate signal, which MRSI could detect and localize on the tumors when these were 5 mm or more in diameter. Lactate production time traces varied slightly with the animal and tumor model, but in general lactate peaked at times of 60 min or longer following injection, reaching concentrations that were ~ 10-fold lower than those of the initial glucose injection. This 2 H3,3' -lactate signal was only visible inside the tumors. 2 H-water could also be detected as deuterated glucose's metabolic product, increasing throughout the entire time course of the experiment from its ≈10 mM natural abundance background. This water resonance could be imaged throughout the entire abdomen of the animals, including an enhanced presence in the tumor, but also in other organs like the kidney and bladder. These results suggest that deuterium MRSI may serve as a robust, minimally invasive tool for the monitoring of metabolic activity in pancreatic tumors, capable of undergoing clinical translation and supporting decisions concerning treatment strategies. Comparisons with in vivo metabolic MRI experiments that have been carried out in other animal models are presented and their differences/similarities are discussed.

Oncology research uses different imaging techniques to provide information about the spatial distribution of the chemotherapy drugs used for the targeted tissues. Among them, laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) is increasingly being used to track the spatial distribution of metal-based chemotherapeutics in different tissue samples. In this investigation, instrumental parameters were optimized for the bioimaging of Pt in HT29 tumour spheroids treated with cisplatin (CDDP) or Texas Red cisplatin (TR-CDDP) using LA-ICP-MS. A high spatial resolution, using pixel dimensions of 2.0 µm × 2.5 µm, and a high sensitivity, with the limits of detection (LOD) better than 0.78 mg kg-1 Pt, was achieved. Matrix-matched gelatine standards and/or isotope dilution (ID) analyses were used to quantify the amount of Pt. Differences between the results of the Pt concentrations determined by the two quantification were less than 4%. The results of the LA analysis revealed that the Pt in the CDDP-treated tumour spheroids was localized primarily in the outer rim of the spheroids and to a lesser extent in the intermediary layer and the necrotic core. Due to the steric effects, significantly lower Pt concentrations were accumulated in the spheroids treated with TR-CDDP (2.2 times lower than in CDDP-treated spheroids, normalized to the spheroid volume), while the Pt was mostly distributed in the areas of the outer rim. Finally, imaging with confocal fluorescence microscopy, which is commonly used in oncology research, was compared with that by LA-ICP-MS. The results of the two complementary techniques demonstrated good agreement in terms of the spatial distribution of the TR-CDDP, while the intensity of the fluorescence matched well with the concentrations of Pt determined with LA-ICP-MS.

Nanosecond electric pulses have several potential advantages in electroporation-based procedures over the conventional micro- and millisecond pulses including low level of heating, reduced electrochemical reactions and reduced muscle contractions making them alluring for use in biomedicine and food industry. The aim of this study was to evaluate if nanosecond pulses can enhance the cytotoxicity of chemotherapeutics bleomycin and cisplatin in vitro and to quantify metal release from electrodes in comparison to 100 µs pulses commonly used in electrochemotherapy. The effects of nanosecond pulse parameters (voltage, pulse duration, number of pulses) on cell membrane permeabilization, resealing and on cell survival after electroporation only and after electrochemotherapy with bleomycin and cisplatin were evaluated on Chinese hamster ovary cells. Application of permeabilizing nanosecond pulses in combination with chemotherapeutics resulted in successful cell kill. Higher extracellular concentrations of bleomycin - but not cisplatin - were needed to achieve the same decrease in cell survival with nanosecond pulses as with eight 100 µs pulses, however, the tested bleomycin concentrations were still considerably lower compared to doses used in clinical practice. Decreasing the pulse duration from microseconds to nanoseconds and concomitantly increasing the amplitude to achieve the same biological effect resulted in reduced release of aluminum ions from electroporation cuvettes.

This study explored the usefulness of multiple quantitative MRI approaches to detect pancreatic ductal adenocarcinomas in two murine models, PAN-02 and KPC. Methods assayed included 1 H T1 and T2 measurements, quantitative diffusivity mapping, magnetization transfer (MT) 1 H MRI throughout the abdomen and hyperpolarized 13 C spectroscopic imaging. The progress of the disease was followed as a function of its development; studies were also conducted for wildtype control mice and for mice with induced mild acute pancreatitis. Customized methods developed for scanning the motion- and artifact-prone mice abdomens allowed us to obtain quality 1 H images for these targeted regions. Contrasts between tumors and surrounding tissues, however, were significantly different. Anatomical images, T2 maps and MT did not yield significant contrast unless tumors were large. By contrast, tumors showed statistically lower diffusivities than their surroundings (≈8.3 ± 0.4 x 10-4 for PAN-02 and ≈10.2 ± 0.6 x 10-4 for KPC vs 13 ± 1 x 10-3 mm2 s-1 for surroundings), longer T1 relaxation times (≈1.44 ± 0.05 for PAN-02 and ≈1.45 ± 0.05 for KPC vs 0.95 ± 0.10 seconds for surroundings) and significantly higher lactate/pyruvate ratios by hyperpolarized 13 C MR (0.53 ± 0.2 for PAN-02 and 0.78 ± 0.2 for KPC vs 0.11 ± 0.04 for control and 0.31 ± 0.04 for pancreatitis-bearing mice). Although the latter could also distinguish early-stage tumors from healthy animal controls, their response was similar to that in our pancreatitis model. Still, this ambiguity could be lifted using the 1 H-based reporters. If confirmed for other kinds of pancreatic tumors this means that these approaches, combined, can provide a route to an early detection of pancreatic cancer.
