RESUMEN

Mutant cycle analysis has been used in previous studies to constrain possible docking orientations for various toxins. As an independent test of the bound orientation of µ-conotoxin PIIIA, a selectively targeted sodium channel pore blocker, we determined the contributions to binding voltage dependence of specific residues on the surface of the toxin. A change in the "apparent valence" (zδ) of the block, which is associated with a change of a specific toxin charge, reflects a change in the charge movement within the transmembrane electric field as the toxin binds. Toxin derivatives with charge-conserving mutations (R12K, R14K, and K17R) showed zδ values similar to those of wild type (0.61 ± 0.01, mean ± S.E.M.). Charge-changing mutations produced a range of responses. Neutralizing substitutions for Arg14 and Lys17 showed the largest reductions in zδ values, to 0.18 ± 0.06 and 0.20 ± 0.06, respectively, whereas unit charge-changing substitutions for Arg12, Ser13, and Arg20 gave intermediate values (0.24 ± 0.07, 0.33 ± 0.04, and 0.32 ± 0.05), which suggests that each of these residues contributes to the dependence of binding on the transmembrane voltage. Two mutations, R2A and G6K, yielded no significant change in zδ. These observations suggest that the toxin binds with Arg2 and Gly6 facing the extracellular solution, and Arg14 and Lys17 positioned most deeply in the pore. In this study, we used molecular dynamics to simulate toxin docking and performed Poisson-Boltzmann calculations to estimate the changes in local electrostatic potential when individual charges were substituted on the toxin's surface. Consideration of two limiting possibilities suggests that most of the charge movement associated with toxin binding reflects sodium redistribution within the narrow part of the pore.

RESUMEN

Antarctic permafrost soils have not received as much geocryological and biological study as has been devoted to the ice sheet, though the permafrost is more stable and older and inhabited by more microbes. This makes these soils potentially more informative and a more significant microbial repository than ice sheets. Due to the stability of the subsurface physicochemical regime, Antarctic permafrost is not an extreme environment but a balanced natural one. Up to 10(4) viable cells/g, whose age presumably corresponds to the longevity of the permanently frozen state of the sediments, have been isolated from Antarctic permafrost. Along with the microbes, metabolic by-products are preserved. This presumed natural cryopreservation makes it possible to observe what may be the oldest microbial communities on Earth. Here, we describe the Antarctic permafrost habitat and biodiversity and provide a model for martian ecosystems.

RESUMEN

We report on efficient lasing of Pr-doped fluoride materials with cw output powers up to 600 mW in the visible spectral range. Praseodymium doped LiYF(4) and LiLuF(4) crystals were pumped either by an intracavity frequency doubled optically pumped semiconductor laser with output powers up to 1.6 W and nearly diffraction limited beam quality or by a multimode GaN-laser diode with an output power of about 370 mW. Furthermore, intracavity frequency doubling of the red Pr-laser radiation to 320 nm reaching output powers of more than 360 mW with a conversion efficiency of 61% and an optical-to-optical efficiency of 22% are presented.

RESUMEN

The search for proteins with immunochemical affinity to plant stress proteins in endemic Baikal fishes shows the presence of proteins, immunochemically related to plant heat-stabile proteins and plant uncoupling protein CSP 310. Western blotting showed that among the native cytoplasmic proteins of endemic Baikal fishes there are proteins immunochemically related to heat-stabile plant proteins with molecular weights about 480, 200-290, 150, 140 and about 90-100kD. SDS-electrophoresis showed the presence of polypeptides with molecular weights 23, 17 and 14kD in all species investigated and an additional 35kD polypeptide in Cottocomephorus grewingki. The search for polypeptides with immunochemical affinity to plant stress uncoupling protein CSP 310 in endemic Baikal fishes shows the presence of a 14kD polypeptide, immunochemically related to it.

We demonstrate longitudinally diode-pumped operation of Nd:YAG and Nd:YAP lasers at the long-wavelength end of the (4)F(3/2) ? (4)I(13/2) transition in the eye-safe spectral region at 1444 and 1430 nm, respectively. Special crystal coatings were required for achievement of lasing at these wavelengths. Output powers of up to 4.9 and 2.2 W, with slope efficiencies of up to 22% and 8%, respectively, were achieved. Polarized operation of the Nd:YAG laser yielded output powers of 76% as compared with unpolarized operation. Because of thermal stress, both YAG and YAP crystals were fractured at roughly 25 W of absorbed longitudinal pump power.

Data from experiments with model samples show that ion transfer coefficients in the water-rich permafrost on Mars must be three orders of magnitude less than those of terrestrial permafrost. The effects of low temperatures and of carbon dioxide have been accounted for. Exchange between cells and the environment is impeded in Martian permafrost. The microscopic distributional heterogeneity of concentration, pH, Eh, and other physicochemical parameters may be more pronounced in the permafrost of Mars. We present a classification of unfrozen water types in the permafrost that is based on the structures of unfrozen water films and on their functions with respect to cells. Any viable microorganisms on Mars probably exist with minimum metabolism in compact zones with energy carriers and high transfer coefficients. These zones may be microvolumes of unfrozen water in which cells accumulate.

RESUMEN

As many as 24 patients suffering from essential hypertension (EH) were examined. The patients were subjected to Holter ECG monitoring, echocardiography, coronary angiography, exercise scintigraphy of the myocardium with transesophageal pacing of the atria and the dipyridamole test. The patients manifested defects of thallium accumulation during exercise scintigraphy of the myocardium. They were transitory defects of accumulation with clearance impairment recorded in EH patients with atherosclerosis of the coronary arteries; transitory defects of accumulation without clearance impairment recorded in EH patients with the angiographically unchanged coronary arteries. In Holter ECG monitoring, the patients with a silent depression of the ST segment demonstrated transitory defects of thallium accumulation by the myocardium in all the cases during exercise scintigraphy of the myocardium.

RESUMEN

A nuclear photographic emulsion method was used to study the charge-state, ionization, and angular characteristics of secondaries produced in inelastic interactions of 56Fe nuclei at 1.8 GeV/nucleon with H, CNO, and AgBr nuclei. The data obtained are compared with the results of calculations made in terms of the Dubna version of the cascade evaporation model (DCM). The DCM has been shown to satisfactorily describe most of the interaction characteristics for two nuclei in the studied reactions. At the same time, quantitative differences are observed in some cases.
