Background Dengue, the mosquito-borne febrile disease caused by the dengue virus, has become one of the major concerns of public health. It may present with only fever, or there may be a hemorrhagic manifestation or septic shock. As there is no specific treatment for dengue, early detection of the disease, assessment of progression, and prediction of outcome by studying the laboratory markers will help guide the management of cases and lower morbidity and mortality. Methodology This clinico-observational study was conducted at the Department of Microbiology in a tertiary care hospital in Kolkata, India, from February 2020 to August 2022 to determine the outcome of dengue patients in correlation with viral load, NS1 antigen, IgM and IgG antibodies, ferritin level, platelet count, and other laboratory parameters. Results Out of 316 samples from fever patients, 103 (32.5%) were NS1 antigen reactive. We followed up the dengue patients (n = 103) for 15 days and divided them into three groups according to their duration of symptoms (group A suffered for ≤5 days, group B for 5 to 10 days, and group C for >10 days) and per the WHO classification of disease severity, namely dengue without warning signs (DOS), dengue with warning signs (DWS), and severe dengue (SD). Based on severity, 65 (63.1%) patients had DOS, whereas 31 (30.09%) patients had DWS, and seven (6.79%) patients had SD. Secondary infection was present in 83.33% of patients in group C, 71% of DWS cases, and 57% of SD cases, which positively correlates with liver enzymes, viral load (mean value 102195 in secondary infection vs. 1195 copies/10 µl in primary infection), and negatively correlates with platelet counts (mean value 60,213 in secondary infection vs. 1,25,516 in primary infection). Patients in group C had higher liver enzymes, a lower platelet count, and a higher initial viral load than groups A and B. Similarly, SD cases had a higher ferritin level (9215 ug/l), a lower platelet count (mean value 23,250), and a higher initial viral load (mean value 2,74,257 copies/10 µl). An increase in hematocrit value considering the peak value and its baseline value is an important marker for disease severity rather than its absolute value. Conclusion Poor outcome of dengue infection, i.e., an increase in the duration of symptoms and disease severity depends on concurrent associations between high serum ferritin, increased hematocrit level, thrombocytopenia, secondary infection, increasing liver enzymes, and increased initial viral load.

The present study explores the use of periphyton to ameliorate toxic properties of arsenic (As) to Labeo rohita and also assesses the human food safety aspects. Fish were introduced to arsenite [As(III)] contaminated water (0.3 and 3 mg/L) along with periphyton. Biochemical, physiological and immunological parameters, including gene expression, were assessed after 30 days of exposure. Periphyton incorporation significantly improved (p < 0.05) the adverse effects of As on respiration, NH3 excretion and brain AChE activity by reducing oxidative stress and As bioaccumulation. The presence of periphyton in As(III) exposed fish (3 mg/L) increased the immune response (Immunoglobulin M and Complement C3) in the serum and the regulation of the respective immune genes in the anterior kidney was found to be similar to the control. A speciation study using LC-ICP-MS confirmed the high accumulation of As by periphyton (5.0-31.9 µg/g) as arsenate [As (V)], resulting in a lower amount of As in fish muscle. The calculated human health risk indices, Target Hazard Quotient (THQ) and Target Cancer risk (TCR) indicate that fish grown in periphyton-treated water may lower the human health risks associated with As. The study signifies the importance of periphyton-based aquaculture systems in As contaminated regions for safe fish production with enhanced yield.

Blood-borne infections caused by the carbapenem-resistant Enterobacter cloacae complex (CR-ECC) are major public threats with respect to the challenges encountered during treatment. This study describes the whole genome sequencing-based molecular characteristics of blood isolates (n = 70) of CR-ECC from patients admitted to the intensive care unit of tertiary care hospitals in Kolkata, India, during 2017-2022 with respect to species identification, antimicrobial resistance (AMR) profiling, mechanism of drug resistance, and molecular subtypes. Vitek2 MALDI and species-specific PCR identified Enterobacter hormaechei subsp. xiangfangensis (47.14%) as the emerging CR-ECC subspecies in Kolkata. The predominating carbapenemase and extended-spectrum ß-lactamase genes found were blaNDM-1 (51.42%) and blaCTX-M-15 (27%), respectively. Besides, blaNDM-4, blaNDM-5, blaNDM-7, blaCMH-3, blaSFO-1, blaOXA-181, blaOXA-232, blaKPC-3, and blaDHA-7 genes were also detected, which were not previously reported from India. A multitude of Class 1 integrons (including In180, In4874, In4887, and In4888, which were novel) and plasmid replicon types (IncFIB, IncFII, IncX3, IncHI1-HI2, IncC, and IncR) involved in AMR dissemination were identified. Reverse transcription-PCR and western blot revealed that carbapenem resistance in non-carbapenemase-producing CR-ECC isolates was contributed by elevated levels of ampC, overexpression of acrAB, and loss of ompF. A total of 30 distinct sequence types (STs) were ascertained by multi-locus sequence typing; of which, ST2011, ST2018, ST2055, ST2721, and ST2722 were novel STs. Pulsed-field gel electrophoresis analysis showed heterogeneity (69 pulsotypes with a similarity coefficient of 48.40%) among the circulating isolates, suggesting multiple reservoirs of infections in humans. Phylogenetically and genetically diverse CR-ECC with multiple AMR mechanisms mandates close monitoring of nosocomial infections caused by these isolates to forestall the transmission and dissemination of AMR.IMPORTANCEThe emergence and extensive dissemination of the carbapenem-resistant Enterobacter cloacae complex (CR-ECC) have positioned it as a critical nosocomial global pathogen. The dearth of a comprehensive molecular study pertaining to CR-ECC necessitated this study, which is the first of its kind from India. Characterization of blood isolates of CR-ECC over the last 6 years revealed Enterobacter hormaechei subsp. xiangfangensis as the most prevalent subsp., exhibiting resistance to almost all antibiotics currently in use and harboring diverse transmissible carbapenemase genes. Besides the predominating blaNDM-1 and blaCTX-M-15, we document diverse carbapenemase and AmpC genes, such as blaNDM-4, blaNDM-7, blaOXA-181, blaOXA-232, blaKPC-3, blaCMH-3, blaSFO-1, and blaDHA-7, in CR-ECC, which were not previously reported from India. Furthermore, novel integrons and sequence types were identified. Our findings emphasize the need for strengthened vigilance for molecular epidemiological surveillance of CR-ECC due to the presence of epidemic clones with a phylogenetically diverse and wide array of antimicrobial resistance genes in vulnerable populations.

Arsenic (As) is a toxic environmental contaminant with global public health concern. In aquatic ecosystems, the quantification of total As is restricted chiefly to the individual organisms. The present study has quantified the total As in different trophic layers (sediment-water-phytoplankton-periphyton-zooplankton-fish-gastropod-hydrophytes) of lentic freshwater ecosystems. As transfer pathways quantifying the transmission rate across trophic-level compartmental route were delineated using a novel model-based approach along with its potential contamination risk to humans. Lentic water bodies from Indo-Gangetic region, a core area of groundwater As, were selected for the present investigation. The study revealed that among the lower biota, zooplankton were the highest accumulator of total As (5554-11,564 µg kg-1) with magnification (rate = 1.129) of the metalloid, followed by phytoplankton (2579-6865 µg kg-1) and periphytic biofilm (1075 to 4382 µg kg -1). Muscle tissue of zooplanktivore Labeo catla is found to store higher As (80-115 µg kg-1 w.w.) compared to bottom-dwelling omnivore Cirrhinus mrigala (58-92 µg kg-1 w.w.). Whereas, Amblypharyngodon mola has accumulated higher As (203-319 µg kg-1 w.w.) than Puntius sophore (30-98 µg kg-1 w.w.) that raised further concern. The hepatic concentration indicated arsenic-mediated stress based on As stress index (threshold value = 1). Mrigal and Mola showed significant biomagnification among fishes while biodiminution was observed in Catla, Bata, Rohu and Punti. All the studied fishes were under the arsenic mediated stress. In the 'sediment-water-periphytic biofilm-gastropod' compartment, the direct grazing accumulation was higher (rate = 0.618) than the indirect path (rate = 0.587). Stems of edible freshwater macrophytes accumulated lesser As (32-190 µg kg-1 d.w.) than roots (292-946 µg kg-1 d.w.) and leaves (62-231 µg kg-1 d.w.). The target cancer risk (TCR) revealed a greater concern for adults consuming edible macrophyte regularly. Similarly, the varied level of target hazard quotient and TCR for adults consuming fishes from these waterbodies further speculated significant health concerns. The trophic transfer rate of environmental As in soil-water-biota level at an increasing trophic guild and consumer risk analysis have been unravelled for the first time in the Indo-Gangetic plains, which will be helpful for the strategic mitigation of As contamination.

Carbapenem-resistant Enterobacter cloacae complex (CRECC) constitutes a global public health threat challenging clinical treatment and infection control, especially in low- and middle-income countries such as India. We analyzed the antimicrobial susceptibility, major ß-lactamase genes, plasmid profiles, and genetic relatedness to understand the molecular epidemiology of CRECC clinical isolates (n = 44) in West Bengal, India, during 2021-2022. The majority (> 55%) of the isolates were resistant to fluoroquinolones, aminoglycosides, and co-trimoxazole, even > 20% for tigecycline and > 35% were extensively drug-resistant. Co-ß-lactamase production was categorized into twenty-seven types, importantly NDM (84%), OXA-48 (40%), TEM (61%), CTX-M (46%), OXA-1 (55%), and MIR (27%). The NDM-1 and OXA-181 were major variants with the first observations of NDM-24 and -29 variants in India. Wide-range of plasmids (2 to > 212 kb) were harbored by the ß-lactamase-producing isolates: small (91%), medium (27%), large (9%), and mega (71%). IncX3, ColE1, and HI2 were noted in about 30% of isolates, while IncF and R were carried by < 20% of isolates. The clonally diverse CRECC isolates were noted to cause cross-infections, especially at superficial site, bloodstream, and urinary-tract. This is the first molecular surveillance on CRECC in India. The study isolates serve as the dockyard of NDM, TEM, and CTX-M harboring a wide range of plasmids. The outcomes of the study may strengthen local and national policies for infection prevention and control practices, clarifying the genetic diversity among CRECC. Extensive genomic study may further intersect the relationships between these different plasmids, especially with their sizes, types, and antibiotic resistance markers.

Growing human population and climate change are leading reasons for water quality deterioration globally; and ecologically important waterbodies including freshwater wetlands are in a vulnerable state due to increasing concentrations of pollutants like heavy metals. Given the declining health of these valuable resources, the present study was conducted to evaluate the effect of natural floating island in the form of fish aggregating devices (FADs) made of native weed mass on the distribution of heavy metals in the abiotic and bio compartments of a freshwater wetland. Lower concentrations of surface water heavy metals were observed inside the FADs with a reduction of 73.91%, 65.22% and 40.57-49.16% for Cd, Pb and other metals (viz. Co, Cr, Cu, Ni and Zn), respectively as compared to outside FAD. These led to 14.72-55.39% reduction in the heavy metal pollution indices inside the FAD surface water. The fish species inside the FADs were also found less contaminated (24.07-25.07% reduction) with lower health risk indices. The study signifies the valuable contribution of natural floating island as FADs in ameliorating the effect of heavy metals pollution emphasizing the tremendous role of the natural floating islands in sustainable maintenance of freshwater wetlands for better human health and livelihood.

Rivers get polluted with diverse types of hazardous and toxic substances, pesticides being one of them. The water and sediment of rivers get contaminated with pesticide residues coming through the run-off of vast agricultural fields along the catchment area and also from domestic sewage water. The residues get bio-concentrated and bio-accumulated in different aquatic organisms and animals including fishes along the food chain. Fish, one of the important and chief sources of proteins, are consumed by humans. The presence of toxic substances like pesticides in any food item is undesirable for the fear of health hazards. We have monitored the status of pesticide residue in river Gomti, a tributary of River Ganga that passes through the Uttar Pradesh state of India. Water, sediment, and fish samples collected from the different locations along the river stretch were analyzed for 34 targeted pesticide compounds belonging to organochlorines (OC), organophosphates (OP), and synthetic pyrethroids (SP) groups. In 52% of water, 30% of sediment, and 43% of fish samples residues of OCs were detected while the OPs were present in 33%, 25%, and 39% of samples respectively. However, none of the SPs could be recorded in any sample. The concentrations of the pesticides in water indicate stress conditions to some extent to aquatic life, but based on the human health risk assessment it can be concluded that consumption of fishes from the river contaminated with different OC or OP residues would not pose any direct risk to the consumers.

The acute crisis of carbapenem resistance impedes the empirical use of carbapenems in medical emergencies, especially, bloodstream infections. Carbapenemase-producing carbapenem-resistant organisms (CP-CROs) attribute high case-fatality, necessitating rapid diagnostics to initiate early targeted antibiotics. Expensive diagnostics are the major driver of antibiotic misuse, neglecting evidence-based treatment in India. One in-house molecular diagnostics assay was customized for rapid detection of CP-CROs using positive blood-culture (BC) broths at a low-cost. The assay was validated using a known-set of isolates and evaluated on positive BC broths. DNA was extracted from positive BC broths using a modified alkali-wash/heat-lysis method. One end-point multiplex-PCR was customized targeting five carbapenemases (KPC, NDM, VIM, OXA-48-, and OXA-23-type) with 16S-rDNA as internal extraction control. Carbapenem resistance due to other carbapenemases, efflux-pump activity, and loss of porins was not under the scope of the assay. Promising analytical performances (sensitivity and specificity, >90%; kappa = 0.87), encouraged to assess diagnostic value, qualified the assay for the WHO minimal requirements (both≥95%) for a multiplex-PCR. Higher LR+ (>10) and lower LR- (<0.1) indicate a good diagnostic tool for ruling in or ruling out CRO bloodstream infections. Inclusion of OXA-23-type improved assay positivity. Multiple carbapenemases were detected in>30% of samples. Good concordance was found (kappa = 0.91) with twenty-six discrepant results. The results were available in 3 hours. The running cost of the assay was US$10 per sample. Fast and reliable detection of carbapenemase(s) allows clinicians and infection-control practitioners to execute early-directed therapy and containment measures. This convenient approach facilitates implementing the assay in resource-limited healthcare settings.

INTRODUCTION: There is increasing development of antibiotic resistance among the Enterococcus species. OBJECTIVES: This study was performed to determine prevalence and characterize the vancomycin-resistant and linezolid-resistant enterococcus isolates from a tertiary care center. Moreover, the antimicrobial susceptibility pattern of these isolates was also determined. MATERIALS AND METHODS: A prospective study was performed in Medical College, Kolkata, India, over a period of two years (from January 2018 to December 2019). After obtaining clearance from the Institutional Ethics Committee, Enterococcus isolates from various samples were included in the present investigation. In addition to the various conventional biochemical tests, the VITEK 2 Compact system was used to identify the Enterococcus species. The isolates were tested for antimicrobial susceptibility to different antibiotics using the Kirby-Bauer disk diffusion method and VITEK 2 Compact to determine the minimum inhibitory concentration (MIC). The Clinical and Laboratory Standards Institute (CLSI) 2017 guidelines were used to interpret susceptibility. Multiplex PCR was performed for genetic characterization of the vancomycin-resistant Enterococcus isolates and sequencing was performed for characterization of the linezolid-resistant Enterococcus isolates. RESULTS: During the period of two years, 371 isolates of Enterococcus spp. were obtained from 4934 clinical isolates showing a prevalence of 7.52%. Among these isolates, 239 (64.42%) were Enterococcus faecalis, 114 (30.72%) Enterococcus faecium, and others were Enterococcus durans, Enterococcus casseliflavus, Enterococcus gallinarum, and Enterococcus avium. Among these, 24 (6.47%) were VRE (Vancomycin-Resistant Enterococcus) of which 18 isolates were Van A type and six isolates of Enterococcus casseliflavus and Enterococcus gallinarum were resistant VanC type. There were two linezolid-resistant Enterococcus, and they were found to have the G2576T mutation. Among the 371 isolates, 252 (67.92%) were multi-drug resistant. CONCLUSION: This study found an increasing prevalence of vancomycin-resistant Enterococcus isolates. There is also an alarming prevalence of multidrug resistance among these isolates.

Herein we have discussed such important modified rhodamine compounds which have been used as chemosensors for the last 7-8 years. This review covered some chemosensors for the detection of metal ions like Al(III), Cu(II), Hg(II), Co(II), Fe(III), Au(III), Cr(III), and some anion like CN-. The selectivity, sensitivity, photophysical properties (i.e., UV-Vis spectral studies, fluorescence studies giving special emphasis to absorption wavelength in UV-Vis spectra and excitation and emission wavelength in fluorescence spectra), binding affinity, the limit of detection, and the application of those chemosensors are described clearly. Here we have also discussed some functionalized rhodamine-based chemosensors that emit in the near-infrared region (NIR) and can target lysosomes and detect lysosomal pH. Their versatile applicability in the medicinal ground is also delineated. We have focused on the photophysical properties of spirolactam rhodamine photoswitches and applications in single-molecule localization microscopy and volumetric 3D light photoactivable dye displays. The real-time detection of radical intermediates has also been exemplified.

The zooplankton community is a widely used bioindicator for the biological assessment of riverine aquatic ecosystems. Phyto-zooplankton interaction and spatially varying river environment parameters perceivably govern their spatial distribution in a large river. This invites the challenge of predicting zooplankton abundance along the river channel. The present article has proposed a geostatistical framework to predict zooplankton abundance along the river course while decoupling phyto-zooplankton relationship from spatial dependency. The strength of secondary data on the river Narmada-a large tropical river in India-has been utilised to accomplish the goal. The nonlinear logistic regression kriging has been found to be the most effective framework. The phyto-zooplankton relationship captured 66% of zooplankton variability, having moderate (37%) residual spatial dependence. The results have shown longitudinally fluctuating spatial variability, which supports the river serial discontinuity concept. The proposed framework has generated smooth zooplankton abundance and sustainability predictive maps that have allowed detection of the change point locations of zooplankton abundance. The map has precisely identified the most productive zone of zooplankton sustainability. The study also has appraised obtaining approximate data in the areas where sampling is infeasible, which will be helpful for location-specific management strategies on a lower spatial scale.

Background Pyogenic skin infection (pyoderma) is a bacterial infection of the skin and its appendages. Primary pyoderma is caused by the direct invasion of healthy skin, whereas secondary pyoderma originates in diseased skin as superimposed conditions, such as scabies, pediculosis, wounds, insect bites, and eczema. This study aimed to identify the clinical patterns and risk factors of pyoderma in a pediatric population and to isolate various causative bacteria and determine their susceptibility patterns. Methodology A prospective study was performed at the Medical College and Hospital, Kolkata, India, for one year (from August 2016 to July 2017), which included all children younger than 12 years with pyoderma attending the outpatient dermatology department (as the study was conducted among the pediatric population, only children below 12 years of age were included). Sterile cotton swabs were used to aseptically collect exudates or pus from lesions and anterior nares, which were used for culture, identification, and antibiotic susceptibility testing of the causative organisms. Results During the study period, a total of 182 patients were included, 121 (66.48%) of whom had primary pyoderma and 61 (33.52%) of whom had secondary pyoderma. Of the 182 patients, 161 showed bacterial growth on culture: 126 (78.26%) were Staphylococcus aureus, 18 (11.18%) were coagulase-negative staphylococci, 16 (9.94%) were Streptococcus pyogenes, and 1 (0.62%) was Pseudomonas aeruginosa. All staphylococci were susceptible to vancomycin and linezolid. Conclusions The most common cause of pyoderma in the pediatric age group is S. aureus, although the prevalence of methicillin-resistant S. aureus was low in this hospital. Proper identification and antibiogram are required for managing these cases.

Biodiesel production from microalgae has gained significant interest recently due to the growing energy demand and non-renewable nature of petroleum. However, high cost of production and environmental health related issues like excess use of inorganic fertilizers, eutrophication are the major constraints in commercial-scale biodiesel production. Besides this, solid wastes (garden-based) management is also a global concern. In the present study, to overcome these limitations vermicompost extract was tested as nutrient source to enhance growth performance and lipid production from a freshwater microalga (Graesiella emersonii MN877773). Garden wastes were first converted into vermicompost manure and its extract (aerobic and anaerobically digested) was prepared. The efficacy of the extract was then tested in combination with BG11 medium. The mixotrophic cultivation of microalgae in anaerobically digested vermicompost extract at 50:50 combination with BG11 medium enhanced the cell biomass (0.64 g d. wt. L-1) and lipid productivity (3.18 mg L-1 day-1) of microalgae by two times. Moreover, the combination also improved the saturated (methyl palmitate) and monounsaturated fatty acids (oleic acid) content in the test algae. The quality of biodiesel also complies with all the properties of biodiesel standard provided by India, the USA, and Europe except the cold filter plugging property. The combination was also found to improve the cell biomass (0.041 g L-1) as compared to BG11 medium in mass-scale cultivation. Hence, the study proved that G. emersonii grown in media supplemented with garden waste-based vermicompost extract had significant potential for mass-scale biodiesel and bioproduct production.

INTRODUCTION: Herpes simplex virus (HSV) Type 2 primarily causes genital herpes, while HSV Type 1 is responsible for oral and facial lesions. The objective of this study was to isolate and characterize HSV from herpetic lesions among human immunodeficiency virus (HIV) infected patients and to evaluate their acyclovir susceptibility pattern. MATERIALS AND METHODS: Blister fluid and swabs from ulcers were collected from patients with clinical diagnosis of HSV infection among patients attending the HIV clinic of two tertiary care centers - Medical College, Kolkata, and School of Tropical Medicine, Kolkata. These samples were cultured in the Vero cell line. Growth of virus was noted by observing the characteristic cytopathic effect of HSV, which was further confirmed by immunofluorescence and polymerase chain reaction (PCR). These isolates were then subjected to the Vero cells with serial dilutions of acyclovir for determining the susceptibility pattern. RESULTS: Among the 52 samples received, 8 (15.38%) showed growth of HSV. After confirmation by immunofluorescence and PCR, all seven isolates from genital samples were identified as HSV-2 and the lone isolate from oral lesion was confirmed as HSV 1. Out of the eight isolates, 25% showed resistance to acyclovir. The overall isolation rate was more from genital blister than genital ulcer which was 46.15% and 2.86%, respectively. CONCLUSION: HSV was isolated in 15.38% of cases of clinical herpes. There was a higher isolation rate of virus from blister fluid as compared to ulcer scrapings. Acyclovir resistance in 25% of cases is alarmingly high.

BACKGROUND: Enterococcus is an important cause of infection in the hospital as well as in the community. METHODS: A prospective study was done in Medical College, Kolkata for a period of 2 years (from January 2018 to December 2019). After obtaining clearance from the Institutional Ethics Committee, Enterococcus isolates from cases of vaginitis were included in the study. Identification of Enterococcus species was done by Gram stain and conventional biochemical tests along with automated identification by VITEK 2 Compact. These isolates were tested for antimicrobial susceptibility to different antibiotics by Kirby Bauer disc diffusion method and minimum inhibitory concentration (MIC) by VITEK 2 Compact. Interpretation of susceptibility was done according to the Clinical and Laboratory Standards Institute (CLSI) 2017 guidelines. Biofilm detection for Enterococcus species was done. RESULTS: During the period of 2 years, 39 isolates of Enterococcus spp. were obtained from vaginitis cases. Among these, 27 were Enterococcus faecalis and 12 Enterococcus faecium. All isolates were highly susceptible to vancomycin, teicoplanin, and linezolid. Biofilm was detected in eight isolates of which five were strong biofilm producer and three moderate biofilm producers. CONCLUSION: Biofilm production is an important virulence factor in Enterococcus isolates from vaginitis.

The ultimate role of prokaryote (bacteria and archaea), the decomposer of the wetland ecosystem, depends on its community structure and its interaction with the environment. The present study has used three universal prokaryote primers to compare prokaryote community structure and diversity of three distinctly different wetlands. The study results revealed that α-diversity indices and phylogenetic differential abundance patterns did not differ significantly among primers, but they did differ significantly across wetlands. Microbial community composition revealed a distinct pattern for each primer in each wetland. Overall comparison of prokaryote communities in sediments of three wetlands revealed the highest prokaryote richness and diversity in Bhomra (freshwater wetland) followed by Malencho (brackish-water wetland) and East Kolkata wetland (EKW) (sewage-fed wetland). Indicator genus analysis identified 21, 4, and 29 unique indicator genera, having preferential abode for Bhomra, EKW, and Malencho, respectively. Prediction of potential roles of these microbes revealed a preference for sulfate-reducing microbes in Malencho and methanogens in Bhomra. The distinct phylogenetic differential abundance pattern, microbial abode preference, and their potential functional role predict ecosystem variables shaping microbial diversity. The variation in community composition of prokaryotes in response to ecosystem variables can serve as the most sensitive bioindicator of wetland ecosystem assessment and management.

The adverse effect of barge movement on the river's aquatic ecosystem is of global concern. The phytoplankton community, a bioindicator, is possibly the foremost victim of the barge movement. This study hypothesized phytoplankton diversity loss induced by barge movement in a large river. This article presents a novel risk assessment framework to evaluate the hypothesis-with a goal to uncoupling phytoplankton diversity loss due to barge movement over a spatiotemporal scale. For this purpose, a study was conducted in the Bhagirathi-Hooghly stretch of Inland National Waterway 1 of India. This study has proposed a new index of diversity loss and its inferential framework based on full Bayesian Generalized Linear Mixed Model. The results have diagnosed significant barge-induced impact on the phytoplankton diversity and identified ten most impacted species. The proposed framework has successfully disentangled barge-induced phytoplankton diversity loss from the biological process and predicted a substantive overall risk of phytoplankton loss of 31.44%. Besides, it has uncoupled spatiotemporal differential estimates, suggesting a risk of diversity loss in order of 'During vs After' (38.0%) > 'Before vs After' (30.7%) > 'Before vs During' (24%) barge movement in temporal scale and increasing diversity loss along downstream. Finally, the instant study has highlighted the utility of these results to facilitate better water framework directive for inland waterways.

Microplastics are recognized as ubiquitous pollutants in aquatic environments; however, very little study is done on their occurrence and fate at drinking water treatment plants (DWTPs). Though, the toxic effect of microplastics on human health is not yet well established; there is global concern about their possible ill effect on the human. Hence, the present study evaluates the occurrence of microplastics at different treatment stages of a typical DWTP with pulse clarification and its removal efficiency. In the test DWTP, raw water, sourced from river Ganga, was found to contain microplastics 17.88 items/L. Cumulative microplastic removal at key treatment stages viz. pulse clarification and sand filtration was found to be 63% and 85%, respectively. The study also revealed higher microplastic abundance on the sand filter bed due to the screening effect. The most frequently occurring microplastics were fibers and films/fragments with polyethylene terephthalate and polyethylene as a major chemical type. The t-distributed stochastic neighbor embedding machine learning algorithm revealed a strong association between microplastic abundance with turbidity, phosphate and nitrate. The test DWTP with a pulse clarification system was having comparable microplastics removal efficiency with previously reported advanced DWTPs.

Rapid damage assessment of natural disasters is essential for the fast recovery and strategic post-disaster reconstructions. In the present study, National Polar-Orbiting Partnership Visible Infrared Imaging Radiometer Suite (NPP-VIIRS) day/night band (DNB)-extracted night-time lights (NTL) data were explored for damage assessment caused by extremely severe cyclonic storm 'AMPHAN' that struck one of the most populous regions in India. The disaster impact was measured on two parameters: population and crop land area, where NTL density and population density were found to be strongly correlated (r 2 > 0.8). From power outage intensity, three 'crisis zones' indicating the severity of cyclone damage were identified. Finally, the assessment found that the total affected population and crop land area were nearly 70% and 66%, respectively, of the study area. Therefore, NPP-VIIRS DNB image-based rapid damage assessment is potentially a useful tool for generating first hand information about the physical damages caused by extreme events.
