RESUMEN

Osteosarcoma is rare but is the most common bone tumor. Diagnostic tools such as magnetic resonance imaging development of chemotherapeutic agents have increased the survival rate in osteosarcoma patients, although 5-year survival has plateaued at 70%. Thus, development of new treatment approaches is needed. Here, we report that IL-17, a proinflammatory cytokine, increases osteosarcoma mortality in a mouse model with AX osteosarcoma cells. AX cell transplantation into wild-type mice resulted in 100% mortality due to ectopic ossification and multi-organ metastasis. However, AX cell transplantation into IL-17-deficient mice significantly prolonged survival relative to controls. CD4-positive cells adjacent to osteosarcoma cells express IL-17, while osteosarcoma cells express the IL-17 receptor IL-17RA. Although AX cells can undergo osteoblast differentiation, as can patient osteosarcoma cells, IL-17 significantly inhibited that differentiation, indicating that IL-17 maintains AX cells in the undifferentiated state seen in malignant tumors. By contrast, IL-17RA-deficient mice transplanted with AX cells showed survival comparable to wild-type mice transplanted with AX cells. Biopsy specimens collected from osteosarcoma patients showed higher expression of IL-17RA compared to IL-17. These findings suggest that IL-17 is essential to maintain osteosarcoma cells in an undifferentiated state and could be a therapeutic target for suppressing tumorigenesis.

Lumbar spinal stenosis (LSS) is a degenerative disease characterized by intermittent claudication and numbness in the lower extremities. These symptoms are caused by the compression of nerve tissue in the lumbar spinal canal. Ligamentum flavum (LF) hypertrophy and spinal epidural lipomatosis in the spinal canal are known to contribute to stenosis of the spinal canal: however, detailed mechanisms underlying LSS are still not fully understood. Here, we show that surgically harvested LFs from LSS patients exhibited significantly increased thickness when transthyretin (TTR), the protein responsible for amyloidosis, was deposited in LFs, compared to those without TTR deposition. Multiple regression analysis, which considered age and BMI, revealed a significant association between LF hypertrophy and TTR deposition in LFs. Moreover, TTR deposition in LF was also significantly correlated with epidural fat (EF) thickness based on multiple regression analyses. Mesenchymal cell differentiation into adipocytes was significantly stimulated by TTR in vitro. These results suggest that TTR deposition in LFs is significantly associated with increased LF hypertrophy and EF thickness, and that TTR promotes adipogenesis of mesenchymal cells. Therapeutic agents to prevent TTR deposition in tissues are currently available or under development, and targeting TTR could be a potential therapeutic approach to inhibit LSS development and progression.

When ruptured, ligaments and tendons have limited self-repair capacity and rarely heal spontaneously. In the knee, the Anterior Cruciate Ligament (ACL) often ruptures during sports activities, causing functional impairment and requiring surgery using tendon grafts. Patients with insufficient time to recover before resuming sports risk re-injury. To develop more effective treatment, it is necessary to define mechanisms underlying ligament repair. For this, animal models can be useful, but mice are too small to create an ACL reconstruction model. Thus, we developed a transgenic rat model using control elements of Scleraxis (Scx), a transcription factor essential for ligament and tendon development, to drive GFP expression in order to localize Scx-expressing cells. As anticipated, Tg rats exhibited Scx-GFP in ACL during developmental but not adult stages. Interestingly, when we transplanted the flexor digitorum longus (FDP) tendon derived from adult Scx-GFP+ rats into WT adults, Scx-GFP was not expressed in transplanted tendons. However, tendons transplanted from adult WT rats into Scx-GFP rats showed upregulated Scx expression in tendon, suggesting that Scx-GFP+ cells are mobilized from tissues outside the tendon. Importantly, at 4 weeks post-surgery, Scx-GFP-expressing cells were more frequent within the grafted tendon when an ACL remnant was preserved (P group) relative to when it was not (R group) (P vs R groups (both n = 5), p<0.05), and by 6 weeks, biomechanical strength of the transplanted tendon was significantly increased if the remnant was preserved (P vsR groups (both n = 14), p<0.05). Scx-GFP+ cells increased in remnant tissue after surgery, suggesting remnant tissue is a source of Scx+ cells in grafted tendons. We conclude that the novel Scx-GFP Tg rat is useful to monitor emergence of Scx-positive cells, which likely contribute to increased graft strength after ACL reconstruction.

Detailed observations of animal reactions to a collapsed individual in wild are rare but essential to debates about the perception of death by nonhuman animals, including chimpanzees. A male juvenile chimpanzee named Volta, a member of the M group in the Mahale Mountains National Park, fell from a tall tree and was temporarily incapacitated, suffering a severe concussion and nasal bone fracture. However, Volta showed signs of gradual recovery. We compared the behavior of other chimpanzees towards Volta with the previous reports on the behavior towards collapsed or recently dead group members. We found that behaviors towards Volta were similar to those observed towards collapsed or dead members. These included other-regarding behaviors and aggressive behaviors, and notably, licking of Volta's blood, which has not been previously reported. Adult males tended to be in close proximity to Volta for longer periods than adult females. The social situation with adult males including alpha male, surrounding Volta likely influenced the behavior of other individuals. Exploring the state of recovery of the injured individual, by closely approaching, directing various behaviors, and observing the reactions of the victim, and demonstrate tolerance and consideration towards the victim.

Although knowledge of the functions of the gut microbiome has increased greatly over the past few decades, our understanding of the mechanisms governing its ecology and evolution remains obscure. While host genetic distance is a strong predictor of the gut microbiome in large-scale studies and captive settings, its influence has not always been evident at finer taxonomic scales, especially when considering among the recently diverged animals in natural settings. Comparing the gut microbiome of 19 populations of Japanese macaques Macaca fuscata across the Japanese archipelago, we assessed the relative roles of host genetic distance, geographic distance and dietary factors in influencing the macaque gut microbiome. Our results suggested that the macaques may maintain a core gut microbiome, while each population may have acquired some microbes from its specific habitat/diet. Diet-related factors such as season, forest, and reliance on anthropogenic foods played a stronger role in shaping the macaque gut microbiome. Among closely related mammalian hosts, host genetics may have limited effects on the gut microbiome since the hosts generally have smaller physiological differences. This study contributes to our understanding of the relative roles of host phylogeography and dietary factors in shaping the gut microbiome of closely related mammalian hosts.

Hip fractures are fragility fractures frequently seen in persons over 80-years-old. Although various factors, including decreased bone mineral density and a history of falls, are reported as hip fracture risks, few large-scale studies have confirmed their relevance to individuals older than 80, and tools to assess contributions of various risks to fracture development and the degree of risk are lacking. We recruited 1395 fresh hip fracture patients and 1075 controls without hip fractures and comprehensively evaluated various reported risk factors and their association with hip fracture development. We initially constructed a predictive model using Extreme Gradient Boosting (XGBoost), a machine learning algorithm, incorporating all 40 variables and evaluated the model's performance using the area under the receiver operating characteristic curve (AUC), yielding a value of 0.87. We also employed SHapley Additive exPlanation (SHAP) values to evaluate each feature importance and ranked the top 20. We then used a stepwise selection method to determine key factors sequentially until the AUC reached a plateau nearly equal to that of all variables and identified the top 10 sufficient to evaluate hip fracture risk. For each, we determined the cutoff value for hip fracture occurrence and calculated scores of each variable based on the respective feature importance. Individual scores were: serum 25(OH)D levels (<10 ng/ml, score 7), femoral neck T-score (<-3, score 5), Barthel index score (<100, score 3), maximal handgrip strength (<18 kg, score 3), GLFS-25 score (≥24, score 2), number of falls in previous 12 months (≥3, score 2), serum IGF-1 levels (<50 ng/ml, score 2), cups of tea/day (≥5, score -2), use of anti-osteoporosis drugs (yes, score -2), and BMI (<18.5 kg/m2, score 1). Using these scores, we performed receiver operating characteristic (ROC) analysis and the resultant optimal cutoff value was 7, with a specificity of 0.78, sensitivity of 0.75, and AUC of 0.85. These ten factors and the scoring system may represent tools useful to predict hip fracture.

Drawings have been widely used as a window to the mind; as such, they can reveal some aspects of the cognitive and emotional worlds of other animals that can produce them. The study of non-human drawings, however, is limited by human perception, which can bias the methodology and interpretation of the results. Artificial intelligence can circumvent this issue by allowing automated, objective selection of features used to analyze drawings. In this study, we use artificial intelligence to investigate seasonal variations in drawings made by Molly, a female orangutan who produced more than 1299 drawings between 2006 and 2011 at the Tama Zoological Park in Japan. We train the VGG19 model to first classify the drawings according to the season in which they are produced. The results show that deep learning is able to identify subtle but significant seasonal variations in Molly's drawings, with a classification accuracy of 41.6%. We use VGG19 to investigate the features that influence this seasonal variation. We analyze separate features, both simple and complex, related to color and patterning, and to drawing content and style. Content and style classification show maximum performance for moderately complex, highly complex, and holistic features, respectively. We also show that both color and patterning drive seasonal variation, with the latter being more important than the former. This study demonstrates how deep learning can be used to objectively analyze non-figurative drawings and calls for applications to non-primate species and scribbles made by human toddlers.

Studies have suggested that the brain morphology and flight ability of Aves are interrelated; however, such a relationship has not been thoroughly investigated. This study aimed to examine whether flight ability, volant or flightless, affects brain morphology (size and shape) in the Rallidae, which has independently evolved to adapt secondary flightlessness multiple times within a single taxonomic group. Brain endocasts were extracted from computed tomography images of the crania, measured by 3D geometric morphometrics, and were analyzed using principal component analysis. The results of phylogenetic ANCOVA showed that flightless rails have brain sizes and shapes that are significantly larger than and different from those of volant rails, even after considering the effects of body mass and brain size respectively. Flightless rails tended to have a wider telencephalon and more inferiorly positioned foramen magnum than volant rails. Although the brain is an organ that requires a large amount of metabolic energy, reduced selective pressure for a lower body weight may have allowed flightless rails to have larger brains. The evolution of flightlessness may have changed the position of the foramen magnum downward, which would have allowed the support of the heavier cranium. The larger brain may have facilitated the acquisition of cognitively advanced behavior, such as tool-using behavior, among rails.

This study analyses 749 drawings by five female Bornean orang-utans (Pongo pygmaeus) at Tama Zoological Park in Japan. We searched for differences between individuals but also tried to identify possible temporal changes among the drawings of one individual, Molly, who drew almost 1300 drawings from 2006 to 2011. An analysis of the drawings was carried out after collecting quantitative and qualitative variables. Our findings reveal evidence of differences in the drawing style of the five individuals as well as creative changes in Molly's drawing style throughout her lifetime. Individuals differed in terms of the colours used, the space they filled, and the shapes (fan patterns, circles, or loops) they drew. Molly drew less and less as she grew older, and we found a significant difference between drawings produced in winter, when orang-utans were kept inside and had less activity, and those produced during other seasons. Our results suggest that the drawing behaviour of these five orang-utans is not random and that differences among individuals might reflect differences of styles, states of mind, and motivation to draw.

Although herd size, structure, stability, and social rank among Misaki feral horses have been reported, no studies have been conducted on the affiliative relationships and interactions among members in a Misaki horse herd. The validity of three hypotheses regarding the function of social grooming, the affiliative relationship strengthening hypothesis, the worsened relationship restoring hypothesis, and the grooming parasite removal hypothesis, were tested in a Misaki feral horse (Equus caballus) herd in Cape Toi, Japan. All the nine horses in the "6m" herd were investigated in terms of kinship, grooming, aggression, proximity, social rank, and social network. Mutual grooming occurred only in pairs and was almost perfectly symmetrical. For each member, there was a significant negative correlation between total grooming received from other individuals and self-grooming. Controlling for kinship, there were significant positive partial correlations between mutual grooming and proximity and between aggression and proximity. No correlation was observed between aggression and mutual grooming. The results suggest that mutual grooming symmetry may contribute that both participants simultaneously benefit from parasite removal and strengthen affiliative relationships between seasonally changing herd members; however, mutual grooming did not foster restoring the worsened relationship following aggression promoted by physical proximity. The findings of this study may elucidate the mechanisms by which interactions between herd members are maintained or strengthened.

Chimpanzees in zoos with sufficient and appropriate environmental enrichment devices are expected to exhibit behaviors, interactions, and societies similar to those in the wild. In this study, we compared the activity budgets of each observed behavior, characteristics of social grooming, and social networks of captive chimpanzees at Tama Zoological Park (Tama) with those of wild chimpanzees at Mahale Mountains National Park, Tanzania (Mahale), and tested our predictions. We surveyed 16 chimpanzees in both Tama and Mahale and recorded the behaviors and individuals in proximity of each focal individual and social grooming the focal individuals participated in. The proportion of time spent collecting foraging was significantly lower in Tama than in Mahale. Additionally, the percentage of mutual grooming was much higher in Tama than in Mahale. All focal individuals in Mahale performed mutual grooming interactions, including grooming handclasp (GHC) but this was not observed in Tama. The result of a high rate of mutual grooming in chimpanzees in Tama without GHC and the finding that individuals forming the core of their social network are sex independent suggest that chimpanzees placed in an appropriate environmental enrichment have idiosyncratic grooming or social features, even in captivity.

This study reports the first observed case of wild chimpanzees (Pan troglodytes) obtaining animal prey freshly killed by a sympatric leopard (Panthera pardus) and scavenging it with the leopard still nearby. This observation has important implications for the emergence of confrontational scavenging, which may have played a significant role in human evolution. Many scholars agree that eating meat became important during human evolution, and hominins first obtained meat by scavenging. However, it is debatable whether scavenging behavior was "passive" or "confrontational (power)." The latter is more dangerous, as it requires facing the original predator, and it is thus considered to have been important for the evolution of several human traits, including cooperation and language. Chimpanzees do scavenge meat, although rarely, but no previous evidence of confrontational scavenging has hitherto emerged. Thus, it was assumed that they are averse to confrontation with even leopard-sized predators. However, in the observed case the chimpanzees frequently emitted waa barks, which indicated that they were aware of the leopard's presence but they nevertheless continued to eat the scavenged meat. In addition, we compiled and reviewed 49 cases of chimpanzee encounters with animal carcasses in the Mahale Mountains of Tanzania in 1980-2017. Chimpanzees scavenged meat in 36.7% of these cases, and tended to eat the meat when it was fresh or if the animal species was usually hunted by chimpanzees. However, no evidence indicated that carcasses were avoided when leopard involvement was likely. These results suggest that chimpanzee-sized hominins could potentially confront and deprive leopard-size carnivores of meat.

A series of disilane-linked donor‒acceptor‒donor triads (D‒Si‒Si‒A‒Si‒Si‒D) was synthesized to investigate the effects of substituents on the photophysical properties. The triads were prepared by metal-catalyzed diiodosilylation of aryl iodides using a Pd(P(t-Bu)3)2/(i-Pr)2EtN/toluene system that we previously developed. Optical measurements, X-ray diffraction analysis, and density functional theory calculations revealed relationships between the photophysical properties and molecular structures of these triads in solution and in the solid state. The compounds emitted blue to green fluorescence in CH2Cl2 solution and in the solid state. Notably, compound 2 showed fluorescence with an absolute quantum yield of 0.17 in the solid state but showed no fluorescence in CH2Cl2. Our findings confirmed that the substituent adjacent to the disilane moiety affects the conformations and emission efficiencies of compounds in solution and in the solid state.

Six novel donor-acceptor-donor organic dyes containing a Si-Si moiety based on triarylamine functionalities as donor units were prepared by Pd-catalyzed arylation of hydrosilanes. Their photophysical, electrochemical, and structural properties were studied in detail. Most of the compounds showed attractive photoluminescence (PL) and electrochemical properties both in solution and in the solid state because of intramolecular charge transfer (ICT), suggesting these compounds could be useful for electroluminescence (EL) applications. The aggregation-induced emission enhancement (AIEE) characteristics of 1 and 3 were examined in mixed water/THF solutions. The fluorescence intensity in THF/water was stronger in the solution with the highest ratio of water because of the suppression of molecular vibration and rotation in the aggregated state. Single-crystal X-ray diffraction of 4 showed that the reduction of intermolecular π-π interaction led to intense emission in the solid state and restricted intramolecular rotation of the donor and acceptor moieties, thereby indicating that the intense emission in the solid state is due to AIEE. An electroluminescence device employing 1 as an emitter exhibited an external quantum efficiency of up to 0.65% with green light emission. The emission comes solely from 1 because the EL spectrum is identical to that of the PL of 1. The observed luminescence was sufficiently bright for application in practical devices. Theoretical calculations and electrochemical measurements were carried out to aid in understanding the optical and electrochemical properties of these molecules.

Social play and grooming are typical affiliative interactions for many primate species, and are thought to have similar biological functions. However, grooming increases with age, whereas social play decreases. We proposed the hypothesis that both social grooming and social play in juveniles strengthen their social bonds in daily activities. We carried out field research on the social relationships among juvenile wild Japanese macaques in a troop in Kinkazan, Miyagi Prefecture, Japan, from fall 2007 to spring 2008 to investigate this hypothesis. We evaluated three relationships among juveniles, play indices (PI), grooming indices (GI), and 3-m-proximity indices (3mI) of each dyad (i.e., interacting pair), and compared these social networks based on the matrices of the indices. The play and grooming networks were correlated with the association network throughout the two research periods. The multiple network level measurements of the play network, but not the grooming network, resembled those of the association network. Using a causal step approach, we showed that social play and grooming interactions in fall seem to predict associations in the following spring, controlling for the PI and GI matrix in spring, respectively. Social play and grooming for each juvenile were negatively correlated. The results partially support our predictions; therefore, the hypothesis that the biological function of social play among immature Japanese macaques is to strengthen their social bonds in the near future and develop their social life appears to be correct. For juvenile macaques, social play, rather than grooming, functions as an important social mechanism to strengthen affiliative relationships.

Both symmetrical and unsymmetrical cyclophanes containing disilane units, tetrasila[2.2]cyclophanes 1-9, were synthesized. The syn and anti conformations and the kinetics of inversion between two anti-isomers were investigated by X-ray diffraction and variable-temperature NMR analysis, respectively. The flipping motion of two aromatic rings was affected by the bulkiness of the aromatic moiety (1 vs 6), the phase (solid vs solution), and the inclusion by host molecules (1 vs 1⊂[Ag2L]2+). The photophysical, electrochemical, and structural properties of the compounds were thoroughly investigated. Unsymmetrical tetrasila[2.2]cyclophanes 5-8 displayed blue-green emission arising from intramolecular charge transfer. Compound 6 emitted a brilliant green light in the solid state under 365 nm irradiation and showed a higher fluorescence quantum yield in the solid state (Φ = 0.49) than in solution (Φ = 0.05). We also obtained planar chiral tetrasila[2.2]cyclophane 9, which showed interesting chiroptical properties, such as a circularly polarized luminescence (CPL) with a dissymmetry factor of |glum| = ca. 2 × 10-3 at 500 nm. Moreover, an organic green light-emitting diode that showed a maximum external quantum efficiency (ηext) of ca. 0.4% was fabricated by doping 4,4'-bis(2,2'-diphenylvinyl)-1,1'-biphenyl with 6.

Imine-linked BODIPY oligomers show fluorescence quantum yields of up to 0.93. The azine linker may mediate π-conjugation that extends over six BODIPY units with redshifts up to 125 nm. BODIPYs are immobilized on silica gel through heterogeneous imine-bond formation, being fluorescent and robust.

RESUMEN

Asymmetric arylation of secondary silanes catalyzed by a Pd-chiral phosphoramidite complex was developed for application to low-molecular-weight circularly polarized luminescence (CPL) materials. The asymmetric arylation provided a convenient, efficient synthetic method for a variety of chiral tertiary silanes (2-21), which were key intermediates for preparing the quaternary silicon center. A stepwise, one-pot procedure was used to transform the appropriate aryl iodide to the quaternary silane (22) with good yield and enantioselectivity. Among compounds synthesized in this work, four optically pure tertiary silanes (18-21) were selected to investigate the relationship between the structure and optical properties. Optically pure (S,S)-21 displayed the highest CPL emission with a high fluorescence quantum yield (glum: +0.008, ΦF: 0.42). This simple molecular design provides new strategies for developing small organic CPL dyes.

A paramagnetic, luminescent organometallic gold(I) complex AuI(C6F5)(PyBTM), where PyBTM is a photostable fluorescent polychlorinated diphenyl(4-pyridyl)methyl radical, was prepared, and its crystal and electronic structures and magnetic and optical properties were investigated. Magnetic studies using electron spin resonance spectroscopy and a superconducting quantum interference device magnetometer indicated the existence of S = 1/2 spin per molecule, with the spin density distributed mainly on the PyBTM ligand. The complex exhibited fluorescence in CHCl3 with emission peak wavelength (λem) of 619 nm and the absolute fluorescence quantum yield (ϕem) of 0.04, confirming that AuI(C6F5)(PyBTM) is the first luminescent organometallic complex with a coordinated luminescent radical. Solvent-dependent unique luminescent characteristics were observed in halogenated solvents (CCl4, CHCl3, CH2Cl2, and ClCH2CH2Cl). ϕem decreased, and λem shifted to longer wavelengths as the polarity (dielectric constant) of the solvent increased. Notably, the complex in CCl4 displayed fluorescence with ϕem = 0.23, which was quite high in radicals, while showed dual fluorescence in CH2Cl2 and ClCH2CH2Cl with lifetimes of around 1 and 7 ns for two emissive components. Density functional theory (DFT) and time-dependent (TD)-DFT calculations indicated that the fluorescence occurred from an interligand charge transfer (CT) excited state in CCl4, in which the C6F5 and PyBTM moieties acted as electron donor and acceptor, respectively, while the fluorescence was centered at the PyBTM ligand in the other three solvents. This method, i.e., the formation of an interligand CT state, to enhance ϕem is distinctly different from the methods reported previously. The present study revealed that a coordination bond is available for forming emissive CT excited states that lead to high ϕem, providing a novel method with greater capability for realizing highly emissive radicals.
