RESUMEN

This article describes an optical computed tomography (OCT) for polymer gel dosimetry, focusing on two systems recently constructed by us. The first OCT system, which is categorized as a first-generation system, comprised a single He-Ne laser, photodiode, and mechanical stages for moving and turning gel dosimeter. Projection data per angle are acquired from turned gel dosimeter. In this system, the reconstructed image is obtained using filtered back-projection (FBP) method from projection data. The second OCT system is a 2D-OCT scanner that utilizes a light panel and a camera detector. The dose-response relationship between the optical density and radiation dose reconstructed from the scanning images is comparable to that obtained using magnetic resonance imaging. Although there is much room for improvement in the image artifacts due to reflection and refraction of light and so on, OCT is expected as a modality for the future polymer gel dosimetry as the rapid, high-resolution, highly accurate evaluation tool.

Sweetpotato peptide (SPP) was prepared by enzyme digestion of sweetpotato protein from starch wastewater. Animal experiments assessed the effect of SPP on body weight, abdominal adipose tissue mass, serum lipids and adipocytokines. Body and liver weight and epididymal and mesenteric fat of mice fed a high-fat diet containing 0.5% or 5% SPP for 28 days were significantly lower than control mice. Triglyceride and cholesterol in VLDL and LDL and leptin levels were significantly lower in the serum of SPP-administered mice compared to control mice. Biomarker arrays showed that adiponectin, melanocyte-stimulating-hormone-alpha and neuromedin U were more than 1.5 times higher, while TNF-alpha was about 1.5 times lower in the livers of SPP-administered mice compared to control mice. These results suggest SPP mitigated leptin resistance in mice administered a high-fat diet, and maintained anorexigenic peptide levels. SPP administration may suppress lipogenesis by increasing adiponectin levels and decreasing TNF-alpha levels in adipocytes.

RESUMEN

Equol is metabolized from daidzein, a soy isoflavone, by the gut microflora. In this study, we identified a novel dihydrodaidzein racemase (L-DDRC) that is involved in equol biosynthesis in a lactic acid bacterium, Lactococcus sp. strain 20-92, and confirmed that histidine-tagged recombinant L-DDRC (L-DDRC-His) was able to convert both the (R)- and (S)-enantiomers of dihydrodaidzein to the racemate. Moreover, we showed that recombinant L-DDRC-His was essential for in vitro equol production from daidzein by a recombinant enzyme mixture and that efficient in vitro equol production from daidzein was possible using at least four enzymes, including L-DDRC. We also proposed a model of the metabolic pathway from daidzein to equol in Lactococcus strain 20-92.

PURPOSE: T1WI (T1 weighted image) was acquired in order to grade bone fusion following the studies by FIFA (Federation Internationale de Football Associations). Research using images other than T1WI has not been reported. The aim of this study is to evaluate the grade of epiphyseal fusion by T2* weighted images (T2*WI) and to investigate new findings on T2*WI as compared with T1WI. METHODS: A total of 87 subjects, all junior football players between the ages of 12 and 17 years old, were examined. T1 and T2* WI were obtained using a 1.2T Open type MR system. The T1WI and T2*WI were rated twice randomly by four radiologists using the FIFA grading system. RESULTS: The intra-rater reliability for grading was higher in T1WI (The Intraclass Correlation Coefficient (ICC)=0.949-0.985) than in T2*WI (ICC=0.917-0.943). The inter-rater reliability for grading was also higher in T1WI (ICC=0.923) than in T2*WI (ICC=0.867). CONCLUSIONS: This research showed that T1WI is a better sequence than T2*WI to evaluate bone fusion following FIFA protocol. It was speculated that the reason for this is that T1WI has higher tissue contrast resolution and enables clearer images of the epiphyseal fusion than T2*WI and the grading system by T1WI was not suitable for T2*WI.

PURPOSE: This study was performed to introduce and evaluate the potential of kinematic magnetic resonance imaging (KMRI) using a high-field open-magnet magnetic resonance (MR) system. METHODS: We attempted to perform KMRI of healthy volunteers' lumbar spine and knee in the lateral position and ankle in the supine position utilizing the superconducting, horizontally opened, 1.2 T MR system (OASIS, HITACHI, Tokyo, Japan). For the KMRI of the lumbar spine, the volunteer had to lie on one side while maintaining maximally anteflexed, neutral, and maximally retroflexed positions and remain still for the duration of the acquisition time for each posture. In the same way, KMRI of the knee was performed with the volunteer's knee flexed at 0°, 30°, 60°, 90°, and 120° in the lateral position, and KMRI of the ankle was performed with the volunteer's ankle in maximally dorsiflexed, neutral, and maximally plantarflexed positions while lying in the supine position. RESULTS: We could acquire higher quality kinematic MR images than those acquired using low-field MR systems. The spinal canal, intervertebral discs and foramina, and facet joints in lumbar spine KMRI; the ligaments, menisci and patellofemoral joint in knee KMRI; and the tibiotalar articulation and peroneal tendon in ankle KMRI were clearly depicted. CONCLUSION: The results of our pilot trial indicated that a superconducting horizontally opened, 1.2 T MR system offers high-quality KMRI images and can be utilized for the kinematic diagnosis and evaluation of sports injuries.

Lactococcus strain 20-92 is a bacterium that produces equol directly from daidzein under anaerobic conditions. In this study, we reveal that the transcription of the gene encoding daidzein reductase in Lactococcus strain 20-92 (L-DZNR), which is responsible for the first stage of the biosynthesis of equol from daidzein, is regulated by the presence of daidzein. We analyzed the sequence surrounding the L-DZNR gene and found six novel genes, termed orf-US4, orf-US3, orf-US2, orf-US1, orf-DS1 and orf-DS2. These genes were expressed in Escherichia coli, and the resulting gene products were assayed for dihydrodaidzein reductase (DHDR) and tetrahydrodaidzein reductase (THDR) activity. The results showed that orf-US2 and orf-US3 encoded DHDR and THDR, respectively. DHDR in Lactococcus strain 20-92 (L-DHDR) was similar to the 3-oxoacyl-acyl-carrier-protein reductases of several bacteria and belonged to the short chain dehydrogenase/reductase family. THDR in Lactococcus strain 20-92 (L-THDR) was similar to several putative fumarate reductase/succinate dehydrogenase flavoprotein domain proteins. L-DHDR required NAD(P)H for its activity, whereas L-THDR required neither NADPH nor NADH. Thus, we succeeded in identifying two novel enzymes that are related to the second and third stages of the biosynthetic pathway that converts daidzein to equol.

Equol is a metabolite produced from daidzein by enteric microflora, and it has attracted a great deal of attention because of its protective or ameliorative ability against several sex hormone-dependent diseases (e.g., menopausal disorder and lower bone density), which is more potent than that of other isoflavonoids. We purified a novel NADP(H)-dependent daidzein reductase (L-DZNR) from Lactococcus strain 20-92 (Lactococcus 20-92; S. Uchiyama, T. Ueno, and T. Suzuki, international patent WO2005/000042) that is involved in the metabolism of soy isoflavones and equol production and converts daidzein to dihydrodaidzein. Partial amino acid sequences were determined from purified L-DZNR, and the gene encoding L-DZNR was cloned. The nucleotide sequence of this gene consists of an open reading frame of 1,935 nucleotides, and the deduced amino acid sequence consists of 644 amino acids. L-DZNR contains two cofactor binding motifs and an 4Fe-4S cluster. It was further suggested that L-DZNR was an NAD(H)/NADP(H):flavin oxidoreductase belonging to the old yellow enzyme (OYE) family. Recombinant histidine-tagged L-DZNR was expressed in Escherichia coli. The recombinant protein converted daidzein to (S)-dihydrodaidzein with enantioselectivity. This is the first report of the isolation of an enzyme related to daidzein metabolism and equol production in enteric bacteria.

Mint-1, which is also called as X11 or mammalian Lin10, protein has been implicated in the synaptic vesicle exocytosis and the targeting and localization of synaptic membrane proteins. Here, we established mint-1 gene knockout (mint-1 KO) mice and investigated vesicular and transporter-mediated dopamine (DA) release evoked by high K(+) and methamphetamine (METH), respectively. Compared with wild-type control, high K(+)-evoked striatal DA release was attenuated, but not significantly, in the KO mice as measured by microdialysis method. The METH-induced DA release was significantly attenuated in the KO mice. In addition, METH-induced stereotypy was also significantly attenuated in the KO mice. Mint-1 KO mice showed more sensitive and prominent behavioral response to an approaching object as compared with wild-type mice. These results suggest that mint-1 protein is involved in transporter-mediated DA release induced by METH.
