RESUMEN

Background: The tibia is one of the most vulnerable bones in the human body, accounting for 13.7% of the total fractures. Most tibial fractures (distal articular surface) are caused by high-violence trauma. In recent years, with the rapid development of China's industry, the incidence of tibial fractures has shown an increasing trend. Aim: To investigate the effect of internal fixation of tibial fractures per suprapatellar approach on fracture union and knee function recovery. Methods: A total of 100 patients with tibial shaft fractures who underwent operations in our hospital were selected as the subjects. They were divided into a suprapatellar group (suprapatellar approach for intramedullary nail fixation) and a subpatellar group (subpatellar approach for intramedullary nail fixation) according to a prospective randomized study, with 50 cases in each group. The operative time, blood loss, X-ray irradiation times, fracture healing time, postoperative knee pain score, knee Lysholm score, and surgical complication rate were compared between the two groups. Results: There were no significant differences in operative time, blood loss, and fracture healing time between the suprapatellar and subpatellar groups (P > .05). The number of X-ray irradiations needed and visual analog scale (VAS) scores were lower in the suprapatellar group than those in the subpatellar group (P < .05). The Lysholm score was used to evaluate knee function 6 months postoperatively, and swelling and pain scores were higher in the subpatellar group than in the suprapatellar group (P < .05). However, there were no significant differences in the knee Lysholm total score between the two groups (P > .05). There were also no significant differences in postoperative complications between the two groups (P > .05). Conclusion: Suprapatellar intramedullary nailing reduced the number of intraoperative X-ray irradiations. Postoperative knee joint pain caused by intramedullary nailing was less, which was beneficial to the early functional knee joint exercise.

BACKGROUND: Osteosarcoma (OS) is the most common primary bone malignancy. It has an aggressive nature and produces drug resistance in diseased patients, which in turn causes obstacles in treating cancer with chemotherapy. The objective of our investigation was to analyze the function and hsa_circ_0010220 mechanism in doxorubicin (DOX) resistance to OS. METHODS: The hsa_circ_0010220, IL-6, and miR-574-3p levels in OS diseased tissues and cell resistance towards DOX drug were elucidated by qRT-PCR and Elisa assay. The DOX half-inhibitory concentration (IC50) was quantified by Cell Counting Kit-8. For this study, we used RNA pull-down, RNA immunoprecipitation, and a dual-luciferase reporter experiment to identify the proteins that interacted with has_circ_0010220, IL-6, and miR-574-3p in OS cells that have developed resistance towards DOX. RESULTS: The results indicated upregulated Hsa_circ_0010220 and IL-6 expression, However, DOX-resistant OS tissues and cells showed a downregulation of miR-574-3p. Reducing DOX resistance in vitro was achieved by silencing Has_circ_0010220. Further, by sponging miR-574-3p, increasing has_circ_0010220 boosted DOX resistance. However, miR-574-3p bound to IL-6 and inhibited DOX resistance. Additionally, it was discovered that hsa_circ_0010220 sponged miR-574-3p for upregulating IL-6 expression. CONCLUSIONS: Hsa_circ_0010220 encouraged OS resistance to DOX by miR-574-3p/IL-6 axis regulation, suggesting its potency as a promising biomarker for treating OS.

LncRNAs play a regulatory role in osteoarthritis (OA); however, the detailed mechanism remains to be elucidated. This study aimed to investigate the role of lncRNA zinc finger NFX1-type containing 1 (ZNFX1) antisense 1 (ZFAS1) in OA progression and explore its possible mechanismsagainst oxidative stress. Human cartilage specimens were obtained from 10 patients without OA who underwent traumatic amputation and 25 patients with OA who underwent total knee replacement surgery. Chondrocytes were prepared from harvested articular cartilage. ZFAS1, nuclear factor erythroid 2-related factor 2 (Nrf2), and heme oxygenase 1 (HO-1) expression levels were analyzed using quantitative reverse transcription PCR and WB. The chondrocyte growth was indicated by MTT and colony formation assays. Chondrocyte apoptosis, reactive oxygen species generation, and anti-oxidative enzymes activities were also measured. ZFAS1 expression was reduced in OA samples and lipopolysaccharide (LPS)-treated chondrocytes used as an OA cell model mimic. ZFAS1 overexpression facilitated proliferation and repressed oxidative stress, inflammation, and apoptosis in LPS-induced chondrocytes. ZFAS1 also activated the anti-oxidative Nrf2-HO-1 pathway. ZFAS1 directly targeted miR-1323, which partially reversed the effects of ZFAS1 on chondrocyte proliferation, oxidative stress, inflammation, and apoptosis. Furthermore, Nrf2 was negatively regulated by miR-1323. The effect of miR-1323 inhibition was partly abrogated by the administration of brusatol, an Nrf2 inhibitor. Collectively, the results showed that ZFAS1 promoted chondrocyte proliferation and repressed oxidative stress, possibly by regulating the novel miR-1323-Nrf2 axis of the inflammation and apoptosis triggered by LPS, indicating that ZFAS1 is a promising therapeutic target for OA.

The satellites structures of doubling harmonic spectrum and tripling harmonic spectrum were measured at the reflection direction of laser wave from the front side of targets, employing OMA optical multi-channel spectrometer during the interactions of femtosecond laser with solid targets. The satellites structures of red-shift harmonic spectrum of 2omega0 and 3omega0 were observed. Under the intensity of approximately 10(18) W x cm(-2), the result of measurement indicates that the spontaneous magnetic field was all less than 1 MGs level by analyzing the satellites structures of doubling harmonic spectrum and tripling harmonic spectrum during the laser-plasma interaction. With increasing the intensity of the laser pulse, the peak of the red-shift moves further to the long wavelength side causing the broadening of the spectrum. The measurements of the spontaneous magnetic field provide a basis of diagnosing the movement of the critical surface in the laser-plasma interaction process.

Using flat-field grating Spectrometer, the ions lines with wavelength between 5 and 60 nm were measured, which were produced by the interaction of circularly polarized 35 femtosecond ultraintense and ultrashort laser-pulse with 5 mm length xenon at the pressure 2 and 3 kPa respectively. The highest transition is the XeVIII: 4d10 5s(2 S1/2)--4d9 5s5p('P3/2) line at wavelength 17.0856 nm at 2 kPa and 3 kPa, the highest transition is 11.343 nm line of XeVII 4d10 5s2(1S0)--4d9 5s5f(3P1) transition. The xenon is ionized to XeVII, XeVIII and XeIX at both pressure.

The integrated image spectrum and scattering light spectrum of optical emission at normal direction from rear-side of a metallic foil were measured, employing optical CCD camera and OMA optical multi-channel spectrometer. The integrated image spectrum shows that it presents a ring-shape, and in the near margin of the ring-shape a bright localized signal is shown, which is optical transition radiation (OTR) generated by hot electrons transport through solid targets. The scattering spectrum shows that it presents a series of nonperiodic sharp spikes between 300-500 nm, and the sharp spike is ascribed to the coherent transition radiation (CTR) generated by bunches of hot electron beams generated in v x B acceleration mechanism near 400 nm (2 omega). The intensity of transition radiation decreases with the increase of the target thickness.
