Lactobacillus salivarius was encapsulated by ionic gelation using high polymerization degree agave fructans and sodium alginate to obtain symbiotic microspheres stabilized by different drying methods. The microbeads were characterized by physicochemical, reconstitution, microstructure, thermal, radiographic and infrared properties. The viability of L. salivarius was evaluated for one month at 4 °C and under in vitro gastrointestinal conditions. The ionic gelation-drying technique allowed an encapsulation efficiency greater than 90 %. Oven-dried microbeads showed higher viability under in vitro gastrointestinal conditions. Freeze-drying microbeads showed higher viability during storage at 4 °C, and spray-dried microbeads showed a more notable survival at 40 °C after one month of storage. All samples showed up to 7 log CFU/g of viability after in vitro digestion, which is a benefit for consumer health. It was concluded that the ionic gelation-drying process allows obtaining dry microbeads with the potential to be added to various food products.

Transparent cellulose papers have emerged as a promising substrate and/or functional component for next-generation sustainable flexible electronics. However, obtaining transparent paper with folding endurance comparable to that of plastic films such as polyethylene terephthalate (PET) remains a major challenge, which was addressed in this study by modulating the multiscale structure (from molecular structure, aggregation structure to fibrous morphology) of wood fibers. Compared to the natural wood fibers, the modified fibers not only retained their length and cellulose degree of polymerization (DP) to a large extent, but also had lower crystallinity and improved swelling capability, which well preserved the fiber strength and dense intertwined fiber network, and led to stronger inter-fiber interactions in the final transparent paper. The obtained paper (CM-paper) exhibits an optical transparency of 91 % and a folding endurance of 37,466 times, comparable to that of commercial PET (39,955 folds). Furthermore, the underlying mechanism for the superior foldability of the transparent paper was unveiled at three scales is explored. Finally, the potential application of CM-paper in electromagnetic induction electrodes was demonstrated, where the CM-paper electrodes exhibit satisfactory conductivity after repeated rubbing. This work offers an innovative approach to produce transparent papers with outstanding foldability, which is expected to plastic replacement.

The development of a direct method for preparing regenerated cellulose (RC) ultrafiltration membranes from cellulose is urgently needed. In this study, refined cotton was used as the raw material to successfully prepare RC ultrafiltration membranes at room temperature using a ZnCl2/AlCl3 solvent system combined with a nonsolvent-induced phase separation (NIPS) method. This solvent system effectively degrades cellulose, producing RC ultrafiltration membranes with varying degrees of polymerization (DP). The research results indicate that reducing the DP of cellulose significantly decreases the viscosity of the solution, facilitating the formation of an asymmetric, finger-like pore structures in the membrane. Furthermore, a decrease in DP slightly enlarges the surface pore size and significantly thickens the dense layer. At a DP of 250, the water flux of the DP250-ET membrane reached 630 L·m-2·h-1·bar-1, with a molecular weight cut-off (MWCO) of ~300 kDa, enabling efficient separation of viruses (LRV > 3.91) and IgG. The exposure of more hydroxy groups on the RC enhances the membrane's hydrophilicity, indicated by a water contact angle (WCA) of 39.5°. Compared to commercial polyethersulfone (PES) membranes, the DP250-ET membrane exhibited lower protein adsorption and excellent anti-fouling performance in practical applications (FRR > 80 %). Overall, this work confirms the significant potential of the eco-friendly ZnCl2/AlCl3 solvent system in the fabrication of RC ultrafiltration membranes, where the structure and performance of the membrane can be tailored by adjusting the DP of cellulose.

The direct enzymatic conversion of untreated waste shrimp and crab shells has been a key problem that plagues the large-scale utilization of chitin biological resources. The microorganisms in soil samples were enriched in two stages with powdered chitin (CP) and shrimp shell powder (SSP) as substrates. The enrichment microbiota XHQ10 with SSP degradation ability was obtained. The activities of chitinase and lytic polysaccharide monooxygenase of XHQ10 were 1.46 and 54.62 U/mL. Metagenomic analysis showed that Chitinolyticbacter meiyuanensis, Chitiniphilus shinanonensis, and Chitinimonas koreensis, with excellent chitin degradation performance, were highly enriched in XHQ10. Chitin oligosaccharides (CHOSs) are produced by XHQ10 through enzyme induction and two-stage temperature control technology, which contains CHOSs with a degree of polymerization (DP) more significant than ten and has excellent antioxidant activity. This work is the first study on the direct enzymatic preparation of CHOSs from SSP using enrichment microbiota, which provides a new path for the large-scale utilization of chitin bioresources.

Anthelmintic resistance in gastrointestinal nematodes produces substantial challenges to agriculture, and new strategies for nematode control in livestock animals are called for. Natural compounds, including tannins, with proven anthelmintic activity could be a functional option as structurally diverse complementary compounds to be used alongside commercial anthelmintics. However, the dual use of two anthelmintic components requires an understanding of the pharmacological effects of the combination, while information concerning the interactions between plant-based polyphenols and commercial anthelmintics is scarce. We studied the direct interactions of proanthocyanidins (PAs, syn. condensed tannins) and a commercial anthelmintic thiabendazole, as a model substance of benzimidazoles, by isothermal titration calorimetry (ITC). Our results show evidence of a direct interaction of an exothermic nature with observed enthalpy changes ranging from 0 to -30 kJ/mol. The strength of the interaction between PAs and thiabendazole is mediated by structural characteristics of the PAs with the strongest positive correlation originating from the presence of galloyl groups and the increased degree of polymerization.

Cellulose nanofibrils (CNFs) are particles with a high aspect ratio. Typically, chemically pre-treated CNFs (containing anionic or cationic charged groups) consist of long fibrils (up to 2 µm) with very low thickness (less than 10 nm). Derived from their high aspect ratio, CNFs form strong hydrogels with high elasticity at low concentrations. Thus, CNF suspensions appear as an interesting rheology modifier to be applied in cosmetics, paints, foods, and as a mineral suspending agent, among other applications. The high viscosity results from the strong 3D fibril network, which is related to the good fibrillation of the material, allowing the nanofibrils to overlap. The overlap concentration (c*) was found to vary from ca. 0.13 to ca. 0.60 wt.% depending on the type and intensity of the pre-treatment applied during the preparation of the CNFs. The results confirm the higher tendency for the fibres treated with (3-chloro-2-hydroxypropyl) trimethylammonium chloride (CHPTAC) and 2,2,6,6-tetramethylpiperidine 1-oxyl (TEMPO) to form a 3D network, resulting in the lowest c*. For the TEMPO-oxidised CNF suspensions, it was also found that aggregation is improved at acidic pH conditions due to lower charge repulsion among fibrils, leading to an increase in the suspension viscosity as well as higher apparent yield stresses. TEMPO CNF suspensions with a low content of carboxylic groups tend to precipitate at moderately acidic pH values.

Prebiotic oligosaccharides are naturally occurring nondigestible carbohydrates with demonstrated health benefits. They are also a chemically diverse class of nutrients, offering an opportunity to investigate the impact of molecular structure on oligosaccharide taste perception. Accordingly, a relevant question is whether these compounds are detected by the human gustatory system, and if so, whether they elicit sweet or "starchy" taste. Here, in 3 psychophysical experiments, we investigated the taste perception of 3 commercially popular prebiotics [fructooligosaccharides (FOS), galactooligosaccharides (GOS), xylooligosaccharides (XOS)] in highly pure form. Each of these classes of prebiotics differs in the type of glycosyl residue, and position and type of bond between those residues. In experiments I and II, participants were asked to discriminate a total of 9 stimuli [FOS, GOS, XOS; degree of polymerization (DP) of 2, 3, 4] prepared at 75 mM in the presence and absence of lactisole, a sweet receptor antagonist. We found that all 9 compounds were detectable (Pâ <â 0.05). We also found that GOS and XOS DP 4 were discriminable even with lactisole, suggesting that their detection was not via the canonical sweet receptor. Accordingly, in experiment III, the taste of GOS and XOS DP 4 were directly compared with that of MOS (maltooligosaccharides) DP 4-6, which has been reported to elicit "starchy" taste. We found that GOS and MOS were perceived similarly although narrowly discriminable, while XOS was easily discriminable from both GOS and MOS. The current findings suggest that the molecular structure of oligosaccharides impacts their taste perception in humans.

This study aimed to comprehensively characterize chemical profiles of proanthocyanidins (PACs) from grape seed extract (GSE), examine their interactions with proteins in a cream system, and define the mechanisms mediating PAC-protein interactions. GSE PACs were fractionated and characterized by thiolysis followed by liquid chromatography-high resolution mass spectrometry (LC-HRMS) analysis. New PACs with a degree of polymerization (DP) up to 16 were identified by improved HRMS data processing methods. In the model cream system, high-DP PACs exhibited greater precipitation capacity and protein binding than low-DP PACs. Low-DP PACs primarily engaged in hydrogen bonding, while high-DP PACs predominantly utilized multiple hydrophobic interaction sites to form cream protein aggregates. Furthermore, particle size and viscosity measurement of cream revealed a progressively DP-dependent increase in aggregated fat globules and cream viscosity. These findings enhanced our understanding of PACs' structural intricacies and highlighted their functional role as PAC-rich natural ingredients in creating structured cream systems.

This study examined the impact of grape flesh polysaccharide, protein, and amino acid contents on polyphenol retention from skins and seeds in Pinot noir (Vitis vinifera) and cold-hardy interspecific cultivars Marquette and Frontenac (Vitis spp.). After isolating grape tissues (skin, seed, and flesh), they were soaked either individually or combined with other tissues in a wine-like solution for up to 7 days. Findings revealed that flesh significantly reduces the concentration of condensed tannin, and mono- and diglucoside forms of anthocyanins in the supernatants, due to its rich content in polysaccharides and proteins. Frontenac skin and flesh tissues were the main sources of soluble proteins, amino acids, and soluble polysaccharides. Surprisingly, Marquette exhibited a higher retention of skin tannin than Pinot noir, likely due to its smaller tannin molecular mass, and a potential competitive effect with anthocyanins for the binding sites of flesh.

Oil-immersed transformers are expensive equipment in the electrical system, and their failure would lead to widespread blackouts and catastrophic economic losses. In this work, an elaborate diagnostic approach is proposed to evaluate twenty-six different transformers in-service to determine their operative status as per the IEC 60599:2022 standard and CIGRE brochure. The approach integrates dissolved gas analysis (DGA), transformer oil integrity analysis, visual inspections, and two Back Propagation Neural Network (BPNN) algorithms to predict the loss of life (LOL) of the transformers through condition monitoring of the cellulose paper. The first BPNN algorithm proposed is based on forecasting the degree of polymerization (DP) using 2-Furaldehyde (2FAL) concentration measured from oil samples using DGA, and the second BPNN algorithm proposed is based on forecasting transformer LOL using the 2FAL and DP data obtained from the first BPNN algorithm. The first algorithm produced a correlation coefficient of 0.970 when the DP was predicted using the 2FAL measured in oil and the second algorithm produced a correlation coefficient of 0.999 when the LOL was predicted using the 2FAL and DP output data obtained from the first algorithm. The results show that the BPNN can be utilized to forecast the DP and LOL of transformers in-service. Lastly, the results are used for hazard analysis and lifespan prediction based on the health index (HI) for each transformer to predict the expected years of service.

Vegetables, cereals and fruit are foods rich in fibre with beneficial and nutritional effects as their consumption reduces the onset of degenerative diseases, especially cardiovascular ones. Among fibres, inulin, oligofructose or fructooligosaccharide (FOS) are the best-studied. Inulin is a generic term to cover all linear ß(2-1) fructans, with a variable degree of polymerization. In this review a better understanding of the importance of the degree of polymerization of inulin as a dietary fibre, functions, health benefits, classifications, types and its applications in the food industry was considered in different fortified foods. Inulin has been used to increase the nutritional and healthy properties of the product as a sweetener and as a substitute for fats and carbohydrates, improving the nutritional value and decreasing the glycemic index, with the advantage of not compromising taste and consistency of the product. Bifidogenic and prebiotic effects of inulin have been well established, inulin-type fructans are fermented by the colon to produce short-chain fatty acids, with important local and systemic actions. Addition of inulin with different degrees of polymerization to daily foods for the production of fortified pasta and bread was reviewed, and the impact on sensorial, technological and organoleptic characteristics even of gluten-free bread was also reported.

Degradation of insulation paper is a key contributor to the failure of power transformers. Insulation degradation accelerates at elevated temperatures, which highlights the potential for better thermal management to prolong life. While several studies have analyzed the benefits of high thermal conductivity oil for reducing temperatures inside a transformer, this study is an initial assessment of the benefits of high thermal conductivity paper on transformer life. Blending particulates with cellulosic fibers offers a pathway for high thermal conductivity paper (with good dielectric properties), which can reduce internal temperatures. Presently, life extensions that can be achieved by the use of such thermally conducting papers were estimated, with the thermal conductivity of the paper being the key parameter under study. The analytical-numerical thermal model used in this study was validated against experimental measurements in a distribution transformer, adding confidence to the utility of the model. This model was then used to provide estimates of hot-spot temperature reduction resulting from the use of papers with higher thermal conductivity than baseline. Transformer life was predicted conventionally by tracking the degree of polymerization of paper over time, based on an Arrhenius model. Results indicate that increasing the thermal conductivity of paper from 0.2 W/mK (baseline) to 1 W/mK reduces the hot spot temperature by 10 °C. While degradation significantly depends on the moisture and oxygen content, the model shows that such a temperature reduction can increase life for all conditions, by as much as a factor of three.

The European Green Deal aims to reduce the pesticide use, notably by developing biocontrol products to protect crops from diseases. Indeed, the use of significant amounts of chemicals negatively impact the environment such as soil microbial biodiversity or groundwater quality, and human health. Grapevine (Vitis vinifera) was selected as one of the first targeted crop due to its economic importance and its dependence on fungicides to control the main damaging diseases worldwide: grey mold, downy and powdery mildews. Chitosan, a biopolymer extracted from crustacean exoskeletons, has been used as a biocontrol agent in many plant species, including grapevine, against a variety of cryptogamic diseases such as downy mildew (Plasmopara viticola), powdery mildew (Erysiphe necator) and grey mold (Botrytis cinerea). However, the precise molecular mechanisms underlying its mode of action remain unclear: is it a direct biopesticide effect or an indirect elicitation activity, or both? In this study, we investigated six chitosans with diverse degrees of polymerization (DP) ranging from low to high DP (12, 25, 33, 44, 100, and 470). We scrutinized their biological activities by evaluating both their antifungal properties and their abilities to induce grapevine immune responses. To investigate their elicitor activity, we analyzed their ability to induce MAPKs phosphorylation, the activation of defense genes and metabolite changes in grapevine. Our results indicate that the chitosans with a low DP are more effective in inducing grapevine defenses and possess the strongest biopesticide effect against B. cinerea and P. viticola. We identified chitosan with DP12 as the most efficient resistance inducer. Then, chitosan DP12 has been tested against downy and powdery mildews in the vineyard trials performed during the last three years. Results obtained indicated that a chitosan-based biocontrol product could be sufficiently efficient when the amount of pathogen inoculum is quite low and could be combined with only two fungicide treatments during whole season programs to obtain a good protection efficiency. On the whole, a chitosan-based biocontrol product could become an interesting alternative to meet the chemicals reduction targeted in sustainable viticulture.

Chitosan oligosaccharides (COS), derived from chitin, have garnered considerable attention owing to their diverse biological activities and potential applications. Previous investigations into the bioactivity of COS often encountered challenges, primarily stemming from the use of COS mixtures, making it difficult to discern specific effects linked to distinct degrees of polymerization (DP). Recent progress underscores the significant variation in the biological activities of COS corresponding to different DPs, prompting dedicated research towards synthesizing COS with well-defined polymerization. Among the available methods, enzymatic preparation stands out as a viable and environmentally friendly approach for COS synthesis. This article provides a comprehensive overview of emerging strategies for the enzymatic preparation of single COS, encompassing protein engineering, enzymatic membrane bioreactors, and transglycosylation reactions. Furthermore, the bioactivities of single COS, including anti-tumor, antioxidant, antibacterial, anti-inflammatory, and plant defense inducer properties, exhibit close associations with DP values. The potential applications of single COS, such as in functional food, food preservation, and crop planting, are also elucidated.

Glycoside hydrolases (GHs) are known to depolymerize polysaccharides into oligo-/mono-saccharides, they are extensively used as additives for both animals feed and our food. Here we reported the characterization of IDSGH5-14(CD), a weakly-acidic mesophilic bifunctional mannanase/glucanase of GH5, originally isolated from sheep rumen microbes. Biochemical characterization studies revealed that IDSGH5-14(CD) exhibited preferential hydrolysis of mannan-like and glucan-like substrates. Interestingly, the enzyme exhibited significantly robust catalytic activity towards branched-substrates compared to linear polysaccharides (P < 0.05). Substrate hydrolysis pattern indicated that IDSGH5-14(CD) predominantly liberated oligosaccharides with a degree of polymerization (DP) of 3-7 as the end products, dramatically distinct from canonical endo-acting enzymes. Comparative modeling revealed that IDSGH5-14(CD) was mainly comprised of a (ß/α)8-barrel-like structure with a spacious catalytic cleft on surface, facilitating the enzyme to target high-DP or branched oligosaccharides. Molecular dynamics (MD) simulations further suggested that the branched-ligand, 64-α-D-galactosyl-mannohexose, was steadily accommodated within the catalytic pocket via a two-sided clamp formed by the aromatic residues. This study first reports a bifunctional GH5 enzyme that predominantly generates high-DP oligosaccharides, preferentially from branched-substrates. This provides novel insights into the catalytic mechanism and molecular underpinnings of polysaccharide depolymerization, with potential implications for feed additive development and high-DP oligosaccharides preparation.

Xylooligosaccharides (XOS) have been employed as prebiotics containing oligomers of varying sizes or molecular ratios. XOS with a low degree of polymerization (DP) has been demonstrated to have high prebiotic potential. However, there is limited information regarding the specific chain length of XOS required to elicit distinct responses in the gut microbiota. In this study, we aimed to explore whether variations in XOS DP could alter the fate of colonic fermentation. Five XOS fractions (BWXFs) with DP ranges of >40, 20-40, 10-20, 5-10, and 2-4 were prepared by beechwood xylan autohydrolysis and tested on human gut microbiota. Extracellular XOS degradation was observed for molecules with a DP exceeding 5. BWXF treatments altered the microbial community structures, and substrate size-dependent effects on the microbial composition and metabolic outputs were observed. Bacteroidaceae were specifically enriched by xylan. Lachnospiraceae were particularly stimulated by XOS with a DP of 20-40 and 2-4. Bifidobacteriaceae were notably enriched by XOS with a DP of 5-20. High butyrate yields were obtained from cultures containing long-chain BWXFs. Microbiota responses differed with XOS DP composition changes, and microbial competition with XOS with a DP of 2-4 requires further exploration.

This study compares the physicochemical and prebiotic properties of inulin isolated from five botanical sources. The average degree of polymerization (DP) for inulin ranged from 5.00 to 13.33. Notably, inulin from Dahlia tubers (DP = 13) and Platycodonis Radix (DP = 8) demonstrated granular, clustered morphology under SEM, semi-crystalline structures via X-ray diffraction, and exhibited shear-thinning behaviors from shear rate 1 s-1 to 500 s-1. In contrast, inulin from Jerusalem artichoke (DP = 5), chicory root (DP = 7), and Asparagi Radix (DP = 5) showcased rough flake morphologies under SEM, amorphous structures in X-ray patterns, and similar shear-thinning behaviors. All inulin types showed acid stability at pH levels below 2.0, with a reducing sugar conversion ratio (RRS) under 1 %. Furthermore, the isolated inulin from the different sources presented prebiotic capacity when added as a sole carbon source in the culture media of the probiotics Lactobacillus paracasei and Bifidobacterium longum. This study provides the properties of inulin from various sources, thereby offering a reference for the selection of appropriate inulin in industrial applications based on the desired characteristics of the final product.

Power transformers are essential apparatuses used to transfer electrical energy from one voltage-level circuit to another. For reliable systems, preventive maintenance of the transformers is required to ensure good services of all mechanical, electrical, and insulation parts. Oil-immersed paper is most often used for transformer insulation. To ensure such good insulation performance and for assessing insulation conditions, advanced transformer sensing, monitoring, and effective assessment techniques are required. This paper introduces an effective technique for assessing the insulation conditions in power transformers, which are crucial for ensuring reliable energy transfer. The method utilizes advanced transformer sensing and monitoring, focusing on oil-immersed paper insulation commonly used in transformers. The technique employs dielectric response sensing, obtained from frequency-domain spectroscopy tests, to estimate degrees of polymerization (DP) and percentages of moisture content (PMCs) in the oil-immersed paper insulation. These parameters are well-known indicators of insulation performance. The approach is based on the weighted k-nearest neighbor regression, using a database of dielectric loss factors at low frequency and oil conductivities. To overcome limited data availability, linear interpolation and extrapolation techniques are applied to enlarge the database. Experimental verification and comparison with a previously developed method demonstrate the proposed technique's superiority in accuracy and complexity. The maximum deviations of DP and PMC in the validation cases are 6.2% and 18.7%, respectively. In addition, to evaluate the validity of our proposed method in the case of a real power transformer, a comparative analysis of the DP and PMC values determined by the proposed method with those obtained through a previously developed and complicated approach was performed. The predicted results indicate that the DP and PMC values of the oil-immersed insulation fall within the ranges of 800 to 1000 and 1.5 to 2.0, respectively, which agree with the results determined by the complicated approach and closely align with real conditions. By offering a reliable and advanced means of assessing insulation conditions, this technique contributes to the preventive maintenance and overall efficiency of power transformers.

The global shortage of cotton for textile production, forces the exploitation of forests´ lignocellulosic biomass to produce man-made cellulosic fibres (MMCF). This has a considerable environmental impact, pressing the textile industry to search for new sustainable materials and to the development of sustainable recycling processes. Bacterial cellulose (BC), an exopolysaccharide produced by fermentation, could represent such an alternative. In particular, we tested the possibility of improving the mechanical properties of cellulose filaments with a low degree of polymerization (DP) by combining them with high DP from BC, so far exploited to little extent in the textile field. In this work, BC with different degrees of polymerization (DPcuaxam) (BCneat: 927; BCdep:634 and BCblend: 814) were dissolved in N-methylmorpholine-N-oxide (NMMO) and their spinnability was studied. The rheological behaviour of the dopes was assessed and all were found to be spinnable, at suitable concentrations (BCneat:9.0 %; BCdep:12.2 %; BCblend:10.5 %). A continuous spinning was obtained and the resulting filaments offered similar mechanical performance to those of Lyocell. Further, the blending of BC pulps with different DPs (BCblend, obtained by combining BCneat and BCdep) allowed the production of fibres with higher stiffness (breaking tenacity 56.4 CN.tex-1) and lower elongation (8.29 %), as compared to samples with more homogeneous size distribution (neat BC and depolymerized BC).

Konjac glucomannan (KGM) is a high-molecular-weight polysaccharide that was originally extracted from the corms (underground storage organs) of Amorphophallus konjac. KGM and its oligomers have been reported as dietary fibers that exhibit an array of health benefits. The depolymerization of KGM via enzymatic hydrolysis at different conditions gives products of low viscosity and can be used for coating materials in microencapsulation. In the present study, konjac glucomannan hydrolysates (KGMHs) were produced by enzymatic hydrolysis using commercial mannanase at pH 4.5 at 70 °C for 5-120 min, then KGMHs' molecular weight (Mw), Degree of Polymerization (DP) and their bioactivities were determined. A longer hydrolysis time resulted in KGMH of a lower DP. Oligoglucomannans (Mw < 10,000) could be obtained after hydrolysis for 20 min. The DP of KGMH rapidly decreased during an early stage of the hydrolysis (first 40 min); DP reached around 7 at the end of the hydrolysis. Antioxidant activities were determined by the DPPH radical scavenging and FRAP assays of KGMHs prepared at pH 4.5 and evaluated at pH 2.0-8.0 depending on pH. KGMH having lower Mw exhibited higher antioxidant activities. KGMHs having the smallest molecular weight (Mw = 419) exhibited the highest DPPH radical scavenging activity. Mw and pH have a greater impact on KGMHs' bioactivities which can be useful information for KGMHs as functional ingredients.
