Abstract In the past decades, an epidemic of chronic kidney disease (CKD) has been associated with environmental and occupational factors (heat stress from high workloads in hot temperatures and exposure to chemicals, such as pesticides and metals), which has been termed CKD of non-traditional origin (CKDnt). This descriptive review aims to present recent evidence about heat stress, pesticides, and metals as possible causes of CKDnt and provide an overview of the related Brazilian regulation, enforcement, and health surveillance strategies. Brazilian workers are commonly exposed to extreme heat conditions and other CKDnt risk factors, including increasing exposure to pesticides and metals. Furthermore, there is a lack of adequate regulation (and enforcement), public policies, and strategies to protect the kidney health of workers, considering the main risk factors. CKDnt is likely to be a significant cause of CKD in Brazil, since CKD's etiology is unknown in many patients and several conditions for its development are present in the country. Further epidemiological studies may be conducted to explore causal associations and estimate the impact of heat, pesticides, and metals on CKDnt in Brazil. Moreover, public policies should prioritize reducing workers´ exposure and promoting their health and safety.


Resumo Nas últimas décadas, uma epidemia de doença renal crônica (DRC) tem sido associada a fatores ambientais e ocupacionais (estresse térmico decorrente de cargas de trabalho elevadas em altas temperaturas e exposição a produtos químicos, como agrotóxicos e metais), denominada DRC de origem não tradicional (DRCnt). Esta revisão descritiva tem como objetivo apresentar evidências recentes sobre estresse térmico, agrotóxicos e metais como possíveis causas de DRCnt e fornecer uma visão geral das estratégias brasileiras de regulamentação, fiscalização e vigilância sanitária relacionadas. Os trabalhadores brasileiros são comumente expostos a condições extremas de calor e outros fatores de risco de DRCnt, incluindo o aumento da exposição a agrotóxicos e metais. Além disso, há uma falta de regulamentação e fiscalização, políticas públicas e estratégias adequadas para proteger a saúde renal dos trabalhadores em relação aos principais fatores de risco. É provável que a DRCnt seja uma causa significativa de DRC no Brasil, uma vez que a etiologia da doença é desconhecida em muitos pacientes e diversas condições para seu desenvolvimento estão presentes no país. Estudos epidemiológicos devem ser realizados para explorar associações causais e estimar o impacto do calor, dos agrotóxicos e dos metais na DRCnt no Brasil. Além disso, as políticas públicas devem priorizar a redução da exposição dos trabalhadores e a promoção de sua saúde e segurança.

Beeswaxes are used as a coating agent or as a wrapping material for food products making them potentially ingested by consumers. There is no regulation yet in Europe giving maximum levels of contaminants in this type of product. Nevertheless, being a natural product, they are exposed to environmental pollution, thus it appears necessary to establish their contamination rate in order to evaluate potential human exposure. In this study, a method of extraction of different environmental contaminants including pesticides, phthalates, PAHs and phenols was developed. Based on a hot Soxhlet extraction, followed by cleaning steps, the method was validated for the quantitation of the cited contaminants by LC-MS/MS and GC-(MS)/MS. Three different types of waxes were analyzed including typical white waxes (Cera Alba) and yellow waxes (Cera Flava). It was shown that all waxes had the presence of at least one contaminant and that phthalates, in particular DEHP, was present in all beeswax samples. Insecticides were found in majority among all the classes of pesticides screened. The yellow waxes were found to be contaminated with the highest rates of PAHs (60%), pesticides (75%) and phenols (40%). The detection frequency of PAHs, in contrast to phthalates, was the lowest for all the types of waxes combined.

The release of chemicals into the environment presents a significant threat to aquatic ecosystems dependent on the proximity to emission sources and seasonal dynamics of emission and mobilization. While spatial-temporal information on water pollution in Europe is increasing, there are substantial knowledge gaps on seasonal pollution dynamics in tropical countries. Thus, we took Lake Victoria South Basin in western Kenya as a case study to identify spatial and seasonal hot spots of contamination, quantified toxic risks to different groups of organisms, and identified seasonal risk drivers. For this purpose, we analyzed grab water samples from five rivers with agricultural and wastewater treatment plants in their catchment in four different seasons. We used liquid chromatography coupled to high resolution mass spectrometry (LC-HRMS) with a target list of 785 organic micropollutants. A total of 307 compounds were detected with concentrations ranging from 0.3 ng/L to 6.6 µg/L. Using a Toxic Unit (TU) approach based on mixture toxicity to standard test organisms, crustaceans were identified as the most affected group followed by algae and fish. For crustaceans, chronic risk thresholds were exceeded in 96 % of all the samples, while 56 % of all samples are expected to be acutely toxic, with the highest risk in February during the dry season. High toxic unit values for algae and fish were recorded in July dry season and May wet season. Diazinon, imidacloprid, clothianidin and pirimiphos-methyl were the major drivers for crustacean toxicity while triclosan and different herbicide mixtures drive risks to algae in dry and wet seasons, respectively. A total of 18 chemicals were found to exceed acute and chronic environmental risk thresholds. With this study, strong spatial-temporal patterns of pollution, risks and risk drivers could be confirmed informing prioritization of monitoring and abatement to enhance water quality and reduce toxic risks.

The ability of organophosphate pesticides to disturb immune function has been demonstrated by in vivo and in vitro studies, but evidence of such effects on humans remains scarce. To assess the association between organophosphate pesticides exposure and cytokine levels in Mexican flower workers, a cross-sectional study was carried out. A questionnaire was provided to 121 male flower workers, and urine and blood samples were collected. Using gas chromatography, urinary concentrations of dialkylphosphate metabolites were determined. The serum cytokine levels, IL-4, IL-5, IL-6, IL-8, and IL-10, were measured using multiplex analysis, and levels of INF-γ and TNF-α by ELISA. We found that a higher dialkylphosphate concentration decreased the pro-inflammatory cytokines INF-γ (ß = -0.63; 95% CI: -1.22, -0.05), TNF-α (ß= -1.18; 95% CI: -2.38, 0.02), and IL-6 (ß= -0.59; 95% CI: -1.29, 0.12), and increased IL-10 (ß=0.56; 95% CI: 0.02, 1.09), the main anti-inflammatory cytokine, suggesting an imbalance of the immune response in flower workers.

BACKGROUND AND AIMS: There is no evidence on the longitudinal and causal associations between multiple pesticides and the incidence of type 2 diabetes mellitus (T2DM) in the Chinese rural population, and whether physical activity (PA) modified these associations remains unclear. Here, we aimed to investigate the longitudinal and causal associations between pesticides mixture and T2DM, and determine whether PA modified these associations. METHODS: A total of 925 subjects with normal glucose and 925 subjects with impaired fasting glucose (IFG) were enrolled in this case-cohort study. A total of 51 targeted pesticides were quantified at baseline. Logistic regression, quantile g-computation, and Bayesian kernel machine regression (BKMR) were used to assess the individual and combined effects of pesticides on IFG and T2DM. Mendelian randomization (MR) analysis was employed to obtain the causal association between pesticides and T2DM. RESULTS: After 3-year follow-up, one-unit increment in ln-isofenphos, ln-malathion, and ln-deltamethrin were associated with an increase conversion of IFG to T2DM (FDR-P<0.05). One quartile increment in organochlorine pesticides (OCPs), organophosphorus pesticides (OPs), herbicides and pyrethroids mixtures were related to a higher incidence of T2DM among IFG patients (P<0.05). The BKMR results showed a positive trend between exposure to pesticides mixture and T2DM. The MR analysis indicated a positive association between exposure to pesticides and T2DM risk (P<0.05). No any significant association was found between pesticides and IFG. In addition, compared to subjects with high levels of PA, those with low levels of PA were related to increased risk of T2DM with the increased levels of pesticides among IFG patients. CONCLUSIONS: Individual and combined exposure to pesticides increased the incidence of T2DM among IFG patients. MR analysis further supported the causal association of pesticides exposure with T2DM risk. Our study furtherly indicated that high levels of PA attenuated the diabetogenic effect of pesticides exposure.

BACKGROUND: The continuous use of pesticides, such as dichlorvos, is a common agricultural and domestic practice. However, it is associated with shortfalls like testicular toxicity through the induction of oxidative stress-mediated signaling. On the other hand, L-arginine, a precursor of nitric oxide, has been reported to exert antioxidant activities and thus may attenuate dichlorvos-induced testicular toxicity. AIM: Hence, this study was designed to evaluate the effect of L-arginine treatment on dichlorvos-induced testicular toxicity. MATERIALS AND METHODS: Forty male Wistar rats were randomly assigned into four equal groups. The control rats were administered 0.5 mL of distilled water, dichlorvos- (DDVP-) treated rats were exposed to DDVP via inhalation for 15 min, DDVP + L-arginine-treated rats were exposed to DDVP and also received 100 mg/kg b.w/day, while L-arginine-treated rats received 100 mg/kg b.w/day. RESULTS: DDVP exposure significantly reduced testicular nitric oxide, relative testicular weight, lowered sperm count, viability, and motility, and suppressed serum FSH, LH, and testosterone levels. These findings were associated with a rise in testicular malondialdehyde, TNF-α, IL-6, and 8OHdG levels and caspase 3 activities, and a reduction in GSH and superoxide dismutase. Additionally, on histopathological examination, DDVP was observed to reduce mature sperm cells in the seminiferous tubular lumen and induce focal vascular congestion in the interstitial space. Nonetheless, L-arginine treatment significantly attenuated DDVP-induced biochemical and histological alterations. CONCLUSION: This study showed that L-arginine attenuated testicular toxicity by improving epididymal sperm variables and male sex hormones by suppressing oxidative stress, inflammation, and apoptosis in DDVP-exposed rats.

The aim of this study was to evaluate the sensitivity of the prawn Palaemon argentinus to the pyrethroid cypermethrin (CYP) and the tetramic acid spirotetramat (STM). These treatments were compared with prawns collected at a reference site to define their basal physiological state. Initially, physicochemical parameters and several pollutants at the selected site were analyzed. The LC50-96 h was determined in adult prawns. Then, prawns were exposed for 96 h to sublethal concentrations of CYP (0.0005 µg/l) and STM (0.44 mg/l) to evaluate the effects on some biochemical endpoints. A treatment combining both pesticides was also added at 5 % of these values. Controls with and without solvent (acetone) were included. The LC50-96 h values were 0.005 µg/l and 4.43 mg/l for CYP and STM, respectively. Moreover, some biomarkers linked to oxidative and energy metabolism were analyzed in the hepatopancreas and muscle of both essayed prawns and those at the basal state. The STM caused a significant decrease in total protein content (32 %) in contrast to the increase of protein carbonyl content (71 %) (p < 0.05). Also, glutathione S-transferase (52 %) and catalase (61 %) activities in the hepatopancreas of exposed prawns were higher compared to both the control and state basal groups (p < 0.05). In muscle, only a significant decrease in the lactate content (69 %) was caused by STM (p < 0.05). In addition, CYP caused a significant increase in the lactate dehydrogenase activity (110 %) in muscle and triacylglycerol content (73 %) in the hepatopancreas (p < 0.05). The integrated biomarker index (IBRv2) analysis showed that STM caused greater damage than CYP. Besides, the combined treatment showed an antagonistic interaction between both insecticides. The differential response of biomarkers to both CYP and STM exposure with respect to their basal levels shows a high sensitivity of P. argentinus demonstrating its potential role as a bioindicator organism.

BACKGROUND: Organophosphorus pesticides (OPPs) were extensively used in agriculture. Due to their adverse effect, there is a need for sensitive and reliable methods to determine these agrochemicals. Microextraction techniques (ME) afford the opportunity to substantially reduce the amount of organic solvent used in classical extraction methods for pesticide analysis. Moreover, deep eutectic solvents (DES) made of components of natural origin, have been applied in microextraction techniques as a green alternative to organic solvents. The combination of thin film microextraction and DES can be seen as an alternative for thin film microextraction of OPPs from water samples. RESULTS: We describe a thin film microextraction-GC-MS method for the determination of OPPs from water samples. The thin film was prepared by solvent casting using cellulose triacetate (CTA) as the polymer and a deep eutectic solvent as the extracting phase. Lidocaine, menthol, dodecanoic acid, and camphor were tested as the components for DES-based film. With a film containing 70 % (w) of CTA and 30 % of the DES dodecanoic acid:lidocaine, quantitative results for the extraction of an OPPs mix were achieved. Then, the elution was performed with 2 mL of ethyl acetate. The validation of the TFME method was performed with a piece of the film suspended in 20 mL of sample solution with a contact time of 1 h. Limits of detection in the low µg L-1 range were obtained using a single quadrupole mass analyser. The thin film with pipette tip configuration was tested and preliminary results for chlorpyrifos were satisfactory. SIGNIFICANCE: This represents the first approach to use polymeric films made of CTA and DES for TFME of OPPs, in two configuration the suspended film and pipette tip.

Historically used pesticides poses a threat to biodiversity while their release pathways remain unclear. Trees could be a potential release source due to their long lifespan. This study examined 38 samples to assess pesticide concentrations in pedunculate oak from Belgium. Low concentrations of procymidone were detected in two samples from one stump. Our findings suggest that accumulations of historically used pesticides in pedunculate oaks within forests are improbable. We conclude that leaving dead wood poses no risks for pesticide release to the environment. However, further research involving diverse tree species and regions is needed to refine and validate this conclusion.

The atmospheric and aqueous ozonolysis of metazachlor (MTZ) is investigated using high-level quantum chemical and kinetic calculations (M06-2X/6-311 + + G(3df,3pd)//M06-2X/6-31 + G(d,p) level of theory). The ozone (O3)-initiated degradation pathways of MTZ under three different mechanisms, namely cycloaddition, oxygen-addition, and single electron transfer (SET), are explored in the temperature range of 283-333 K and 1 atm pressure. As a result, the cycloaddition reaction at the C16C18 double bond of the benzene ring of MTZ is found to be the most dominant channel in the atmosphere with the standard Gibbs free energy of reaction (ΔrG0g) of - 129.13 kJ mol-1 and the highest branching ratio of 95.18%. In the aqueous phase, the main reaction channel turns into the SET mechanism, which owns the lowest Gibbs free energy of activation (ΔG#aq) of 73.8 kJ mol-1 and contributes 87.8% to the ktotal. Over the temperature range of 283-333 K, the total rate constant (ktotal) significantly increases from 8.42 to 5.82 × 101 M-1 s-1 in the atmosphere and from 4.10 × 102 to 2.40 × 104 M-1 s-1 in the aqueous environment. Remarkably, the ecotoxicity assessment shows that MTZ may be harmful to fish and chronically harmful to daphnia. In contrast, its main ozonolysis products exhibit no acute or chronic toxicity or mutagenic effects.

The use of chemical pesticides in agriculture contributes to soil, water and air pollution, biodiversity loss, and injury to non-target species. The European Commission has already established a Harmonized Risk Indicator to quantify the progress in reducing the risks linked to pesticides. Therefore, there is an increasing need to promote biopesticides, or so-called low-risk pesticides (LRP). Tea tree oil (TTO) is known for its antiseptic, antimicrobial, antiviral, antifungal, and anti-inflammatory properties. TTO has been extensively studied in pest management as well as in the pharmaceutical and cosmetic industry; there are already products based on its active substances on the market. This review focuses on the overall evaluation of TTO in terms of effectiveness and safety as a biopesticide for the first time. The collected data can be an added value for further evaluation of TTO in terms of the authorization extension as a fungicide in 2026.

A deep eutectic solvent (DES) with the ability to change from hydrophilic to hydrophobic was designed and synthesized and applied to the determination of organophosphorus (OPP) pesticides in honeysuckle dew samples. Choline chloride, phenol, and tetrahydrofuran (THF) were used as the hydrogen bond acceptor, hydrogen bond donor, and demulsifier, respectively. Eight OPP pesticides were extracted by DES coupled with ultrasonic-assisted extraction (UA) and then chromatographed by GC-MS. DES used as an extract solvent has the advantages of high extraction efficiency, low cost, and environmental protection. Furthermore, DES is compatible with GC-MS. The single factor experiment design and Box-Behnken design (BBD) were applied to the optimization of experimental factors, including the type and composition of extraction solvent, type of demulsifier solvent, the volume of DES and THF, pH of sample solution, and ultrasonic time. Under the optimum experimental conditions, the high degree of linearity from 0.1 to 20.0 ng mL-1 (R2 ≥ 0.9989), the limits of detection from 0.014 to 0.051 ng mL-1 (S/N = 3), and the recoveries of analytes from 81.4 to 104.4% with relative standard deviation below 8.6%. In addition, the adsorption mechanism of OPPs on DES was explored by adsorption kinetic studies. These results have demonstrated that the present method has offered an effective, accurate, and sensitive methodology for OPP pesticides in honeysuckle dew samples, and this method provides a reference for the detection of pesticide residues in traditional Chinese medicine.

Safe drinking water is essential to a healthy lifestyle and has been recognised as a human right by numerous countries. However, the realisation of this right remains largely aspirational, particularly in impoverished nations that lack adequate resources for water quality testing. Kenya, a Sub-Saharan country, bears the brunt of this challenge. Pesticide imports in Kenya increased by 144% from 2015 to 2018, with sales data indicating that 76% of these pesticides are classified as highly hazardous. This trend continues to rise. Over 70% of Kenya's population resides in rural areas, with 75% of the rural population engaged in agriculture and using pesticides. Agriculture is the country's main economic activity, contributing over 30% of its gross domestic product (GDP). The situation is further exacerbated by the lack of monitoring for pesticide residues in surface water and groundwater, coupled with the absence of piped water infrastructure in rural areas. Consequently, contamination levels are high, as agricultural runoff is a major contaminant of surface water and groundwater. The increased use of pesticides to enhance agricultural productivity exacerbates environmental degradation and harms water ecosystems, adversely affecting public health. This study proposes the development of a wireless sensor system that utilizes radio-frequency identification (RFID), Long-range (LoRa) protocol and a global system for mobile communications (GSM) for monitoring pesticide prevalence in groundwater sources. From the system design, individuals with limited literacy skills, advanced age, or non-expert users can utilize it with ease. The reliability of the LoRa protocol in transmitting data packets is thoroughly investigated to ensure effective communication. The system features a user-friendly interface for straightforward data input and facilitates broader access to information by employing various remote wireless sensing methods.

Phosphine (PH3) has been widely used as a fumigant in food storage, but increasing PH3 resistance in major pests makes finding alternative fumigants urgent. Methyl benzoate (MBe), a volatile organic compound regarded to be a food-safe natural product, has recently demonstrated significant toxicity against a variety of insect pests. This study is the first evaluation of the fumigation toxicity of three benzoate compounds, MBe, vinyl benzoate, and ethyl benzoate, against PH3-susceptible and PH3-resistant strains of Rhyzopertha dominica and Sitophilus oryzae. All strains were exposed to the compounds at concentrations up to 20 µL/1.5 L air for 24 h. Compared to vinyl benzoate and ethyl benzoate, MBe induced higher mortality rates in all strains at all concentrations. When food was made available, the lethal median concentration for MBe was 10-17-fold higher than when tested without food. Moreover, no significant differences were observed between the responses of the PH3-susceptible and PH3-resistant strains to the compounds. Notably, S. oryzae was more susceptible to MBe. In laboratory settings, MBe successfully controlled PH3-resistant strains of R. dominica and S. oryzae, making it a viable option for PH3-resistance management. Thus, MBe might be suitable for food security programs as an environmentally benign alternative fumigant.

Since ancestral times, quinoa (Chenopodium quinoa Willd.) has been cultivated in the Andean regions. Recently, this pseudocereal has received increasing international attention due to its beneficial properties, such as adaptation and resilience in the context of global change, and the nutritional value of the grains. As a result, its production areas have not only increased in the highlands of South America but have also expanded outside of its Andean origins, and the crop is currently produced worldwide. The key pests of quinoa in the Andean region are the gelechiid moths Eurysacca melanocampta and Eurysacca quinoae; in other parts of the world, new pest problems have recently been identified limiting quinoa production, including the gelechiid Scrobipalpa atripicella in North America and Europe and the agromyzid fly Amauromyza karli in North America. In this review, the status of quinoa pests in the world is presented, and different aspects of their integrated management are discussed, including sampling methodologies for pest monitoring, economic threshold levels, and various control strategies.

Pesticide residues in food pose significant risks to public health and have long been a major concern in Chinese cities. The management of these risks is influenced by various factors, including the characteristics of responsible officials. This study tests the relationship between the levels of pesticide residues and the responsible officials' interdisciplinary backgrounds and their tenure cycles, which is crucial for improving food safety governance in Chinese cities. Based on wastewater-based epidemiology (WBE) and data from 32 Chinese cities, it was found that the interdisciplinary backgrounds of officials had a significant negative relationship with urban pesticide residues in wastewater, indicating that the interdisciplinary knowledge background or working experience of officials in food safety-related agencies was associated with the supervision and control of urban pesticide residues. This study also generated evidence-based knowledge on how to improve food safety through assigning younger and interdisciplinary officials to the responsible governmental agencies, where WBE is more likely to be adopted.

Biobeds are presented as an alternative for good pesticide wastewater management on farms. This work proposes a new test for in-situ biomonitoring of pesticide detoxification in biobeds. It is based on the assessment of visually appreciable injuries to Eisenia fetida. The severity of the injury to each exposed individual is assessed from the morphological changes observed in comparison with the patterns established in seven categories and, an injury index is calculated. A linear relationship between the proposed injury index and the pesticide concentration was determined for each pesticide sprayed individually in the biomixture. The five pesticides used were atrazine, prometryn, clethodim, haloxyfop-P-methyl and dicamba. In addition, a multiple linear regression model (i.e., a multivariate response surface) was fitted, which showed a good generalization capacity. The sensitivity range of the injury test was tested from 0.01 to 630 mg kg-1 as the total pesticide concentration. This index is then used to monitor the detoxification of these pesticides in a biomixture (composed of wheat stubble, river waste, and soil, 50:25:25% by volume) over 210 days. The results are compared with standardized tests (Eisenia fetida avoidance test and Lactuca sativa seed germination test) carried out on the same biomixture. The results are also compared with data on the removal of pesticides. The injury test showed a better correlation with the removal of pesticides than the avoidance test and seed germination test. This simple and inexpensive test has proved to be useful for decontamination in-situ monitoring in biobeds.

The extensive use of chemical pesticides, such as herbicides, has resulted in significant environmental pollution. Microbial degradation represents a crucial approach for managing this pesticide-associated pollution, with enrichment culturing serving as a method for isolating pesticide-degrading microorganisms. However, the efficiency of this strategy is limited, often yielding only a few isolated strains. In this study, a new mineral salt medium (MSM) was developed, and a high-throughput method was used for screening pendimethalin-degrading bacteria by measuring the bacterial growth in the MSM. The utilization of this method resulted in the isolation of 56 pendimethalin-degrading bacteria from approximately 2000 bacterial strains, including 37 Bacillus spp., 10 Alcaligenes spp., 5 Pseudomonas spp., and other 4 strains identified for the first time as pendimethalin-degrading strains. This method may hold promise not only for isolating bacterial strains capable of degrading other pesticides but also for facilitating the utilization of the substantial bacterial strains stored in bacterial banks.

Due to the increasing importance of exposome in environmental epidemiology, feasibility and usefulness of an Environmental Data Management System (EDMS) using Open Data was evaluated. The EDMS includes data from 10 European cities (Celje (Slovenia), Lódz (Poland), Manchester (UK), Palermo (Italy), Paris (France), Porto (Portugal), Regensburg (Germany), Reus (Spain), Rijeka (Croatia), Thessaloniki (Greece)) about external non-specific and specific exposome factors at the city or country level (2017-2020). Findings showed that the highest values of life expectancy were in Reus females (86 years) and Palermo males (81 years). UK had the highest obesity rate (28%), Croatia the highest prescribed drug consumption (62%), Greece and Portugal the highest smoking rates (37%, 42%) and daily alcohol consumption (21%), respectively. The most polluted cities were Thessaloniki for PM10 (38 µg/m3), Lódz for PM2.5 (25 µg/m3), Porto for NO2 (62 µg/m3) and Rijeka for O3 (92 µg/m3). Thessaloniki had the highest grey space (98%) and Lódz the highest cumulative amount of pollen (39,041 p/m3). The highest daily noise levels ≥ 55 dB was in Reus (81% to traffic) and Regensburg (21% to railway). In drinking water, arsenic had the highest value in Thessaloniki (6.4 µg/L), boron in Celje (24 mg/L) and lead in Paris (46.7 µg/L). Portugal and Greece showed the highest pesticide residues in food (7%). In conclusion, utilizing open-access databases enables the translation of research findings into actionable strategies for public health interventions.
