RESUMO

A 51-year-old male with previous medical history of dyslipidemia performed screening colonoscopy, which revealed a sessile polypoid lesion with a diameter of approximately 8 mm located at the proximal transverse colon, which was resected en bloc with a cold snare. Remarkably, histopathological examination revealed a proliferation of spindle cells in the lamina propria entrapping colonic crypts without evidence of nuclear pleomorphism, mitotic figures or necrosis. On immunohistochemistry, spindle cells were diffusely positive for glucose transporter-1 and negative for S100, DOG1, CD34 and smooth muscle actin. These features were consistent with a diagnosis of colonic perineurioma.

The cytotoxicity against five human tumor cell lines (THP-1, U937, Molt-4, Colo-205 and NCI-H460) of three water soluble copper(II) coordination compounds containing the ligands 3,3'-(ethane-1,2-diylbis(azanediyl))dipropanamide (BCEN), 3,3'-(piperazine-1,4-diyl)dipropanamide (BPAP) or 3,3'-and (1,4-diazepane-1,4-diyl)dipropanamide (BPAH) are reported in this work. The ligands contain different diamine units (ethylenediamine, piperazine or homopiperazine) and two propanamide units attached to the diamine centers, resulting in N2O2 donor sets. The complex containing homopiperazine unit presented the best antiproliferative effect and selectivity against lung cancer cell line NCI-H460, showing inhibitory concentration (IC50) of 58 µmol dm-3 and Selectivity Index (SI) > 3.4. The mechanism of cell death promoted by the complex was investigated by Sub-G1 cell population analysis and annexin V and propidium iodide (PI) labeling techniques, suggesting that the complex promotes death by apoptosis. Transmission electron microscopy investigations are in agreement with the results presented by mitochondrial membrane potential analysis and also show the impairment of other organelles, including endoplasmic reticulum.

Two platinum complexes [Pt(HL3)Cl]·H2O (3) and [Pt(HL4)Cl]·H2O (4) containing α- and ß-naphthyl groups, respectively, were investigated in more detail in vitro and in vivo for antineoplastic activity. The cytotoxicity activity induced by these platinum(II) compounds against breast cancer (MDA-MB-231 and MCF-7), lung (A549), prostate (PC3), pancreas (BXPC-3), and normal peripheral blood mononuclear (PBMC) cells were evaluated by MTT assay. The cell viability MTT assay showed that complex (4) was more cytotoxic to all cancer cell lines tested and less cytotoxic against human PBMC. Therefore, complex (4) was selected to further investigate the mechanism of cytotoxic effects involved against MDA-MB-231 cell line (human triple-negative breast cancer). Sub-G1 analysis of the cell cycle showed that this complex induces cell death by apoptosis due to the cell loss of DNA content detected in flow cytometry. The cytotoxic effect induced by complex (4) was associated with the capability of the complex to induce mitochondrial membrane depolarization, as well as increase ROS levels and caspase activation, as a result of the activation of both extrinsic and intrinsic apoptosis pathways. Ultrastructural alterations were observed using scanning and transmission electron microscopy (SEM and TEM), such as membrane blebbing, filopodia reduction, empty mitochondrial matrix, and DNA fragmentation. Furthermore, complex (4) was tested in an MDA-MB-231 tumor nodule xenograft murine model and demonstrated a remarkable reduction in tumor size in BALB/c nude mice, when compared to the control animals.

Sporotrichosis is a chronic and subacute mycosis causing epidemiological outbreaks involving sick cats and humans in southeastern Brazil. The systemic disease prevails in cats, and in humans, the symptoms are restricted to skin in immunocompetent individuals. Under these conditions, the prolonged treatment of animals and cases of recurrence justify the discovery of new treatments for sporotrichosis. This work addresses the antifungal activity of silver salts of Keggin-type heteropolyacid salts (Ag-HPA salts) such as Ag3[PW12O40], Ag6[SiW10V2O40], Ag4[SiW12O40] and Ag3[PMo12O40] and interactions with the antifungal drugs itraconazole (ITC), terbinafine (TBF) and amphotericin B (AMB) on the yeast and mycelia forms of Sporothrix spp. Sporothrix spp. yeast cells were susceptible to Ag-HPA salts at minimum inhibitory concentration (MIC) values ranging from 8 to 128 µg/mL. Interactions between Ag3[PW12O40] and Ag3[PMo12O40] with itraconazole and amphotericin B resulted in higher antifungal activity with a reduction in growth and melanization. Treated cells showed changes in cell membrane integrity, vacuolization, cytoplasm disorder, and membrane detachment. Promising antifungal activity for treating sporotrichosis was observed for the Ag-HPA salts Ag3[PMo12O40] and Ag3[PW12O40], which have a low cost, high yield and activity at low concentrations. However, further evaluation of in vivo tests is still required.

The aim of our study was to evaluate the in vitro and in vivo anti-proliferative potential of complex (2) [Cu (L1)Cl]Cl.2H2O, where L1 = 1-[2-hydroxybenzyl(2-pyridylmethyl)amino]-3-(1-naphthyloxy)-2-propanol on lung carcinoma cell NCI-H460. Cell viability assay determined by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) colorimetric assay demonstrated that the complex (2) exhibits higher activity against the NCI-H460 cell, with an IC50 value lower than cisplatin (26.5 µM ± 1.1 and 203 ± 1.2 µM respectively). Cell death by apoptosis was investigated by flow cytometer analysis of sub-G1 populations in the cell cycle and Annexin V/Propidium Iodide assay. Changes on the cell surface and ultrastructure were detected by scanning and transmission electron microscopy. Our work revealed that complex (2) induced changes associated with apoptosis, such as plasma membrane blebbing and a lower microvilli amount, fragmentation and condensation of chromatin, alterations in mitochondria, and enlargement of the endoplasmic reticulum. Mitochondrial function of NCI-H460 cells evaluated by 5,5',6,6'-tetrachloro 1,1',3,3' tetraethylbenzimidazolylcarbocyanine iodide (JC-1) probes showed high loss of mitochondrial membrane potential when treated with complex (2). Moreover, caspase-12 measurement showed an expressive activation level, which is related to endoplasmic reticulum stress. In vivo assay using the murine model of human lung cancer cell showed that complex (2) and cisplatin has similar antineoplastic activity.

The synthesis, physico-chemical characterization, Density functional theory (DFT) calculation and cytotoxicity against five human tumoral cell lines (THP-1, U937, Molt-4, Colo205 and H460) of four new platinum(II) coordination compounds are reported, i.e. [Pt(HL1)Cl]·H2O (1), [Pt(HL2)Cl]·H2O (2), [Pt(HL3)Cl]·H2O (3) and [Pt(HL4)Cl]·H2O (4). The ligands contain N2O donor sets. Furthermore, H2L3 and H2L4 present α and ß-naphthyl groups respectively, which are absent in HL1 and H2L2. X-ray diffraction studies were performed for complex (3), indicating the formation of a mononuclear platinum(II) complex. Complexes (3) and (4), which contain α and ß-naphthyl groups respectively, have presented lower IC50 (inhibitory concentration) values than those exhibited by complexes (1) and (2). The mechanism of cell death promoted by complexes (3) and (4) was investigated, suggesting that, toward U937 cell line, the α isomer promotes death by apoptosis and the ß isomer by necrosis. Transmission and scanning electron microscopy investigations are in agreement with the loss of mitochondrial membrane potential (ΔΨm) observed by JC-1 mitochondrial potential sensor and indicate that the activity of complex (3) against U937 cell line is mediated by an apoptotic mechanism associated with mitochondrial dysfunction. A quantification of caspases 3, 6, 8 and 9 indicated that both the intrinsic and extrinsic pathways are involved in the apoptotic stimuli. Based on DFT calculations all the Pt(II) complexes present the same coordination environment for the metal centre, indicating that the higher cytotoxic activities exhibited by complexes (3) and (4) are related to the presence of the α and ß-naphthyl groups in the ligand structure.

Headaches are characterized by a sensation of discomfort or pain in the cephalic region. Due to the large number of types and subtypes of headaches, the standardization of their approach is necessary through the International Classification of Headache Disorders (ICHD), a document that is currently in its 3rd edition (ICHD-3). The evolution of the ICHD follows the hypothetical-deductive model of science based on Karl Popper's philosophy, which has a strong presence in the health sciences. The classification of headaches has evolved over time, incorporating and removing criteria, reflecting the need to adapt the classification to constantly evolving scientific and clinical demands. However, some changes can generate discrepancies between clinical practice and the elaborated diagnostic tools. In this context, this article reflects on whether it would be advantageous to return to old principles and foundations of obsolete classifications. The criterion of osmophobia, which is no longer in use in ICHD-3, is highlighted. This reflection can be developed following Thomas Kuhn's (1922-1996) view on science. In his main work, "The Structure of Scientific Revolutions" (1962), Kuhn proposes a three-stage model for science: the adoption of a paradigm, normal science, and a period of crisis. Kuhn also proposes the principle of incommensurability to understand the limitations and complexities of science. The application of this principle allows headache specialists to recognize that different approaches to headache classification have their own limitations and complexities, encouraging them to consider a variety of perspectives, paradigms, and theories in approaching clinical cases and conducting scientific studies. An integrative approach that combines Popper's hypothetical-deductive model with Kuhn's principle of incommensurability allows headache specialists to have a broader and more critical understanding of headache classifications


As dores de cabeça são caracterizadas por uma sensação de desconforto ou dor na região cefálica. Devido ao grande número de tipos e subtipos de cefaleias, é necessária a padronização da sua abordagem através da Classificação Internacional das Cefaleias (ICHD), documento que se encontra atualmente na sua 3ª edição (ICHD-3). A evolução da ICHD segue o modelo hipotético-dedutivo de ciência baseado na filosofia de Karl Popper, que tem forte presença nas ciências da saúde. A classificação das cefaleias evoluiu ao longo do tempo, incorporando e eliminando critérios, refletindo a necessidade de adaptar a classificação às exigências científicas e clínicas em constante evolução. Contudo, algumas mudanças podem gerar discrepâncias entre a prática clínica e os instrumentos diagnósticos elaborados. Neste contexto, este artigo reflete sobre se seria vantajoso regressar aos velhos princípios e fundamentos de classificações obsoletas. Destaca-se o critério de osmofobia, que não é mais utilizado na ICHD-3. Esta reflexão pode ser desenvolvida seguindo a visão de Thomas Kuhn (1922-1996) sobre a ciência. Em sua obra principal, "A Estrutura das Revoluções Científicas" (1962), Kuhn propõe um modelo de três estágios para a ciência: a adoção de um paradigma, a ciência normal e um período de crise. Kuhn também propõe o princípio da incomensurabilidade para compreender as limitações e complexidades da ciência. A aplicação deste princípio permite que os especialistas em cefaleias reconheçam que diferentes abordagens à classificação das cefaleias têm as suas próprias limitações e complexidades, encorajando-os a considerar uma variedade de perspectivas, paradigmas e teorias na abordagem de casos clínicos e na condução de estudos científicos. Uma abordagem integrativa que combina o modelo hipotético-dedutivo de Popper com o princípio da incomensurabilidade de Kuhn permite que os especialistas em cefaleia tenham uma compreensão mais ampla e crítica das classificações das cefaleias

The synthesis, physico-chemical characterization and cytotoxicity against five human tumoral cell lines (THP-1, U937, Molt-4, Colo205 and H460) of three new cobalt(II) coordination compounds are reported (i.e. Co(HL1)Cl (1), Co(HL2)Cl (2) and [Co(HL3)Cl]0.0.5 (CH3)2CHOH (3)). H2L2 (2-{[[2-hydroxy-3-(1-naphthyloxy)propyl](pyridin-2-ylmethyl)amino]methyl}phenol) and H2L3 (2-{[[2-hydroxy-3-(2-naphthyloxy)propyl](pyridin-2-ylmethyl)amino]methyl}phenol) present α and ß-naphthyl groups respectively, which is absent in H2L1 (N-(2-hydroxybenzyl)-N-(2-pyridylmethyl)[(3-chloro)(2-hydroxy)]propylamine. These compounds were characterized by a range of physico-chemical methods. X-ray diffraction studies were performed for complex (3), indicating the formation of a mononuclear complex. Complexes (2) and (3), which contain α and ß-naphthyl groups respectively, have presented lower IC50 values than those exhibited by complex (1). Complex (3) presents IC50 values lower than cisplatin against Colo205 (90 and 196µmolL(-1), respectively) and H460 (147 and 197µmolL(-1), respectively). These human neoplastic cells under investigation were also more susceptible toward complex (3) than peripheral blood mononuclear cells. Transmission electron microscopy investigations are in agreement with the loss of mitochondrial membrane potential (ΔΨm) observed by JC-1 mitochondrial potential sensor and indicate that the activity of complex (3) against leukemic cell line (U937) is mediated by an apoptotic mechanism associated with mitochondrial dysfunction (intrinsic pathway).

The research submitted samples from stems and leaf blades from tree genotypes of Pennisetum purpureum called 93-32-02, 92-70-02, and 91-06-02 (EMBRAPA - Dairy cattle) and elephant grass cv. Napier (reference cultivar) to the chemical, anatomical evaluations, and in vitro dry matter digestibility (IVDMD) measurement. The anatomical characteristics of the stems and leaf blades, the chemical composition, and the IVDMD of these genotypes at 70 days of re-growth were correlated. Concerning IVDMD, the data highlighted differences, and the cultivar Napier presented the smallest value. Digital images obtained by light microscopy from cross-section reveal that all the stem and leaf blade have similar structural organization. Quantitative differences were verified mainly in the stem. The leaves displayed differences only in the mesophyll thickness. The genotypes showed higher potential in the rainy season since they had the largest IVDMD when compared to the cultivar Napier.

The aim of this study was to evaluate the effect of ammoniation by urea on the nutritional value of elephant grass hay harvested after flowering. A completely randomized design, in double factorial designs with an additional treatment: 4 urea doses (2, 4, 6 and 8%) x 2 treatment periods (30 and 45 days) + 1 (Control) with four replicates each, was employed. Dry matter, ash, ether extract, neutral detergent fiber, acid detergent fiber, cellulose, lignin, neutral detergent fiber corrected for the ash and the protein, total nitrogen, acid detergent insoluble nitrogen, neutral detergent insoluble nitrogen, non-protein nitrogen, in vitro gas production and carbohydrates fractionation were analyzed. The treatments influenced the contents of DM, EE, NDF, ADF, lignin, cellulose, NDFap, fraction A + B1, fraction B2, fraction C, TN, NPN, ADIN, NDIN, L and Vf2. There was a positive linear effect of urea dose for NDF, ADIN, NDIN and L, positive linear effect on fraction A + B1, NT and NPN, and positive quadratic effect for Vf2, with absolute maximum point of 4.5%. Elephant grass hay harvested after flowering has its nutritional value improved, with a minimum dose of 4.5% urea on a dry matter basis.(AU)


Objetivou-se avaliar o efeito da amonização com ureia sobre o valor nutritivo do feno de capim elefante colheitado após florescimento. O estudo foi delineado, em esquema fatorial duplo com um tratamento adicional: 4 doses de ureia (2, 4, 6 e 8%) x 2 período de tratamento (30 e 45 dias) + 1 tratamento controle, com 4 repetições cada. As variáveis analisadas foram: matéria seca (MS), cinzas, extrato etéreo (EE), fibra em detergente neutro (FDN) e ácido (FDA), FDN corrigida para cinzas e proteínas (FDNcp), celulose, lignina, nitrogênio total (NT), nitrogênio insolúvel em detergente ácido (NIDA) e neutro (NIDN), nitrogênio não proteico (NNP). Produção de gases in vitro proveniente dos carboidratos não fibrosos (Vf1) e fibrosos (Vf2), taxa de fermentação dos carboidratos não fibrosos (k1) e fibrosos (k2) e tempo de colonização bacteriana do substrato (L), além do fracionamento de carboidratos. Os tratamentos influenciaram os teores de MS, EE, FDN, FDA, lignina, celulose, FDNcp, fração A + B1, fração B2, fração C dos carboidratos, NT, NNP, NIDA, NIDN, L e Vf2. Houve efeito linear decrescente de dose de ureia para FDNcp, NIDA, NIDN e L, crescente para fração A+B1, NT e NNP, e, quadrático crescente de dose de ureia para Vf2, com ponto máximo absoluto de 4,5% de ureia obtido após derivar a equação de regressão aos 45 dias de tratamento do feno. Concluindo que feno de capim elefante colheitado após florescimento tem seu valor nutricional melhorado, com dose mínima de 4,5% de ureia em base da matéria seca.(AU)

The synthesis, physico-chemical characterization and cytotoxicity of four new ligands and their respective copper(II) complexes toward two human leukemia cell lines (THP-1 and U937) are reported (i.e. [(HL1)Cu(µ-Cl)2Cu(HL1)]Cl2·H2O (1), [(H2L2)Cu(µ-Cl)2Cu(H2L2)]Cl2·5H2O (2), [(HL3)Cu(µ-Cl)2Cu(HL3)]Cl2·4H2O (3), [(H2L4)Cu(µ-Cl)2Cu(H2L4)]Cl2·6H2O (4)). Ligands HL1 and HL3 contain two pyridines, amine and alcohol moieties with a naphthyl pendant unit yielding a N3O coordination metal environment. Ligands H2L2 and H2L4 have pyridine, phenol, amine and alcohol groups with a naphthyl pendant unit providing a N2O2 coordination metal environment. These compounds are likely to be dinuclear in the solid state but form mononuclear species in solution. The complexes have an antiproliferative effect against both leukemia cell lines; complex (2) exhibits higher activity than cisplatin against U937 (8.20 vs 16.25µmoldm(-3)) and a comparable one against THP-1. These human neoplastic cells are also more susceptible than peripheral blood mononuclear cells (PBMCs) toward the tested compounds. Using C57BL/6 mice an LD50 of 55mgkg(-1) was determined for complex (2), suggesting that this compound is almost four times less toxic than cisplatin (LD50=14.5mgkg(-1)). The mechanism of cell death promoted by ligand H2L2 and by complexes (2) and (4) was investigated by a range of techniques demonstrating that the apoptosis signal triggered at least by complex (2) starts from an extrinsic pathway involving the activation of caspases 4 and 8. This signal is amplified by mitochondria with the concomitant release of cytochrome c and the activation of caspase 9.

ABSTRACT: Find shade-tolerant species is essential to the success of silvopastoral systems, increasingly frequent in recent years. In legumes, which have potential of biological nitrogen fixation, there is a great lack of knowledge when in shaded environments.The cultivation of four tropical forage (Neonotonia wightii, Pueraria phaseoloides, Macrotyloma axilare and Arachis pintoi) was evaluated when submitted to artificial shade levels (30, 50 and 70% shade) and in full sun during water and drought seasons. The design used was in randomized complete blocks in a sub-divided plot scheme with four replications. In the Water-season the Forage Peanuts had higher forage production in full sun (11 ton ha-1 DM), and under shade did not differ from Perennial Soybean, higher than the others in all levels of shade. In Drought-season the forage production was 61% lower than in Water-season. The highest crude protein levels were reported in Forage Peanuts, Tropical Kudzu and Perennial Soybean, 19.0; 18.3 and 18.2% respectively in the Water-season. Forage Peanuts is a good option for use in silvopastoral systems although there is a small reduction in forage production (average of 23.7%). In general, species of fabaceae showed a greater reduction in forage production in the period of water deficit; however, shading at levels of 30% to 50% contribute to mitigation of water shortage. Although, there is a small reduction in forage production, withexception of perennial soybeans in dry season, it is advisable to use tropical forage legumes in silvopastoral systems, since forage quality is not affected by shade.


RESUMO: Encontrar espécies tolerantes ao sombreamento é essencial para o sucesso dos sistemas silvipastoris, cada vez mais frequentes nos últimos anos. Em se tratando de leguminosas, que possuem potencial de fixação biológica de nitrogênio, há uma grande carência de conhecimento em ambientes sombreados. Objetivou-se avaliar o cultivo de quatro leguminosas forrageiras tropicais (Neonotonia wightii, Pueraria phaseoloides, Macrotyloma axilare e Arachis pintoi) submetidas a diferentes níveis de sombreamento artificial (30, 50 e 70% de sombra) e em pleno sol durante as estações de água e seca. O delineamento utilizado foi o de blocos completos casualizados, em esquema de parcelas subdivididas, com quatro repetições. Na estação das águas, o Amendoim Forrageiro teve maior produção de forragem a pleno sol (11 ton ha-1 MS), e sob sombreamento não diferiu da Soja perene, superiores às demais em todos os níveis de sombra. Na estação de seca, as espécies apresentaram produção de forragem 61% menor do que a estação da água. Os níveis mais altos de proteína bruta foram encontrados em Amendoim Forrageiro, Kudzu tropical e Soja Perene, 19,0; 18,3 e 18,2%, respectivamente, na estação da água. O Amendoim Forrageiro é uma boa opção para uso em sistemas silvipastoris, embora haja uma pequena redução na produção de forragem (média de 23,7%). Em geral, espécies de Fabaceae apresentaram maior redução na produção de forragem no período de déficit hídrico, porém o sombreamento em níveis de 30% a 50% contribuiram para a mitigação da escassez hídrica. Embora haja uma pequena redução na produção de forragem, com exceção da Soja Perene na estação seca, é aconselhável o uso dessas leguminosas forrageiras tropicais nos sistemas silvipastoris, uma vez que a qualidade da forragem é pouco afetada pela sombra.

The longevity of the world's population is increasing, and among male patients, complaints of lower urinary tract symptoms (LUTS) are growing. Testing to diagnose LUTS and to differentiate between the various causes should be quick, easy, cheap, specific, not too bothersome for the patient, and noninvasive or minimally so. Urodynamic evaluation is the gold standard for diagnosing bladder outlet obstruction (BOO) but presents some inconveniences such as embarrassment, pain, and dysuria; furthermore, 19% of cases experience urinary retention, macroscopic hematuria, or urinary tract infection. A greater number of resources in the diagnostic armamentarium could increase the opportunity for selecting less invasive tests. A number of groups have risen to this challenge and have formulated and developed ideas and technologies to improve noninvasive methods to diagnosis BOO. These techniques start with flowmetry, an increase in the interest of ultrasound, and finally the performance of urodynamic evaluation without a urethral catheter. Flowmetry is not sufficient for confirming a diagnosis of BOO. Ultrasound of the prostate and the bladder can help to assess BOO noninvasively in all men and can be useful for evaluating the value of BOO at assessment and during treatment of benign prostatic hyperplasia patients in the future. The great advantages of noninvasive urodynamics are as follows: minimal discomfort, minimal risk of urinary tract infection, and low cost. This method can be repeated many times, permitting the evaluation of obstruction during clinical treatment. A urethral connector should be used to diagnose BOO, in evaluation for surgery, and in screening for treatment. In the future, noninvasive urodynamics can be used to identify patients with BOO to initiate early medical treatment and evaluate the results. This approach permits the possibility of performing surgery before detrusor damage occurs.

The objective of this work was to evaluate the use of citrus pulp addition and wilting on fermentative characteristics, chemical composition as well as gas and effluent losses of elephant grass silage. The elephant grass (70 growth days) was collected manually; one portion was immediately chopped and another one was allowed to wilt in the sun for a period of 6 h and subsequently chopped for the production of silage. Experimental PVC silos (10 cm diameter x 30 cm height) were used and sand bags were placed at the bottom of the silos. Citrus pulp (80 g kg-1 of MN) was added at the beginning of ensilage. The ensiled material was manually compressed to provide a specific mass of approximately 600 kg m-3 of silage. After 60 days, the silos were opened; gas and effluent losses were calculated and we determined pH, dry matter (DM), crude protein(CP), neutral detergent fibre (NDF), lignin, ammonia nitrogen, ash and in vitro dry matter digestibility (IVDMD). We added 0 or 80 g kg-1 of citrus pulp to wilted and un-wilted elephant grass. We used a completely randomised design with a 2 x 2 factorial arrangement (wilted or un-wilted) x (with or without citrus pulp), totalling treatments with five repetitions. Average values were compared using the F test with a probability of 5%. The addition of citrus pulp resulted in reduced levels of NDF, lignin, ash, N-NH3 and pH and in increased values of DM and IVDMD of silages. Wilting increased the DM, NDF and lignin values and reduced the concentrations of CP, IVDMD and N-NH3. Based on our results, citrus pulp addition improves the chemical composition of elephant grass silage and increases its in vitro dry matter digestibility.


Objetivou-se avaliar o uso de polpa cítrica e do emurchecimento sobre as características fermentativas, composição química e perdas de silagens de capim-elefante. O capim-elefante foi colhido manualmente com cerca de 70 dias de rebrota, uma parte foi picada imediatamente e outra foi emurchecida ao sol por 6 horas e posteriormente picado para confecção das silagens. Foram utilizados silos experimentais de PVC com 10 cm de diâmetro e 30 cm de altura, no fundo dos silos foram colocados sacos com areia. A polpa cítrica foi incluída na base de 80 g kg-1 da matéria natural no momento da ensilagem. O material ensilado foi compactado manualmente de forma a proporcionar massa específica de aproximadamente 600 kg m-3 de silagem. No momento da ensilagem foram pesados os silos vazios, os sacos com areia, o material ensilado e os silos cheios. Após 60 dias os silos foram pesados novamente e logo em seguida abertos. Foram calculadas as perdas por gases e efluentes e determinados o pH, conteúdo de matéria seca, teores de proteína bruta, fibra em detergente neutro (FDN), fibra em detergente ácido (FDA), celulose, hemicelulose e lignina, cinzas, nitrogênio amoniacal e digestibilidade in vitro da matéria seca (DIVMS). Foi utilizado o delineamento inteiramente casualisado em arranjo fatorial 2 (emurchecido ou não) x 2 (com polpa cítrica ou não), 4 tratamentos com 5 repetições, totalizando 20 parcelas. As médias foram comparadas pelo teste F a 5%. O uso de polpa cítrica reduziu os níveis de FDN, lignina, cinzas, N-NH3 e o pH e aumentou a MS e a DIVMS das silagens. O emurchecimento aumentou a MS, FDN e a lignina e reduziu a proteína bruta, DIVMS e N-NH3. O uso da polpa cítrica melhora a composição química e a digestibilidade in vitro da matéria seca de silagens de capim-elefante.

This survey aimed to characterize a Urochloa humidicola methanol extract regarding the presence of secondary metabolites classes and to determine its bromatological composition. U. humidicola samples were dried under shade, milled on a 2-mm sieve by a Willey mill. The solution obtained was filtered using filter paper and concentrated in a rotary evaporator under reduced pressure; the concentrated residue was then placed in an open vessel to complete solvent removal using continuous air flow dryers. Phytochemical prospection tests and bromatological composition analyses were performed on the dry methanol extract, and the results were compared to in natura U. humidicola. The methanol extract had 10.2% CP and 35% EE and in natura U. humidicola had 5.17% CP and 1.57% EE, with a difference (P < 0.05) of 5% by Fisher's test. In natura U. humidicola had 75.59% NDF, 40.77% ADF, 38.82% HEM, 29.93% CEL, and 7.19% LIG. Methanol extraction by cold maceration reduced the LIG (0.17%) and CEL (0.21%) contents as only soluble constituents were extracted. A phytochemical assay was positive for the presence of saponins, tannins, alkaloids, non-protein amino acids, carbohydrates, cardiac glycosides, steroids, tripernoids, catechins, and saccharides and was negative for the presence of flavonoids and purines. The U. humidicola methanol extract possesses traits that allow its use as a phytogenic and natural additive.


Objetivou-se neste estudo caracterizar o extrato metanólico de Urochloa humidicola, quanto à presença de classes de metabólitos secundários presentes bem como determinar a sua composição bromatológica. Para isto, amostras da parte aérea de U. humidicola foram secas à sombra, moídas em moinho tipo Willey em partículas de 2 mm, submetidas à extração por maceração a frio com metanol, a solução obtida foi concentrada em rotaevaporador e posto para termina a secagem sob fluxo de ar contínuo. O extrato metanólico seco foi submetido aos testes de prospecção fitoquímica e análises de composição bromatológica comparado com a U. humidicola in natura. O extrato apresentou 10,2% de PB e 35% de EE e U. humidicola in natura apresentou 5,17 % de PB e 1,57% de EE, diferindo entre si (P<0,05) pelo teste de Fisher a 5% de significância. A U. humidicola in natura apresentou teores de FDN (75,59%), FDA (40,77%), hemicelulose (38,82%), celulose (29,93%) e lignina (7,19%). O método de extração por maceração a frio com metanol contribuiu para a diminuição dos teores de lignina (0,17%) e celulose (0,21%), por extrair somente os constituintes solúveis. O ensaio fitoquímico apresentou presença positiva para saponina, tanino, alcaloides, aminoácidos não proteicos, carboidratos, glicosídeos cardioativos, esteroides e tripernoides, catequinas e sacarídeos, e negativa para a presença de flavonoides e purinas. O extrato metanólico de Urochloa humidicola apresenta características que permitem seu uso como aditivo natural ou fitogênico.

ß-1,3-glucanases are found in organisms as diverse as plants, animals, bacteria and fungi. In plants, such enzymes are not only associated with defense mechanisms against pathogens, but also play critical roles in physiological and developmental processes. Here we identified a new ß-1,3-glucanase in maize seeds, and named it ZmGlucA. Sequence analysis revealed that ZmGlucA belongs to the class A of ß-1,3-glucanase, a class related to defense and physiological processes in plants. mRNA and protein assays showed that zmGlucA is expressed exclusively in seeds, and it is differentially regulated during seed development. Additionally, zmGlucA expression is strongly induced in seeds of the mutant dek 827Kpro1, which is defective for embryo and endosperm development. Our data support the idea that ZmGlucA protein is relevant to seed development.

We synthesized iron(III), cobalt(II), copper(II) and zinc(II) complexes [Fe(III)(HBPClNOL)Cl(2)]·H(2)O (1), [Co(II)(H(2)BPClNOL)Cl(2)] (2), [Cu(II)(H(2)BPClNOL)Cl]Cl·H(2)O (3), and [Zn(II)(HBPClNOL)Cl] (4), where H(2)BPClNOL is the ligand (N-(2-hydroxybenzyl)-N-(2-pyridylmethyl)[(3-chloro)(2-hydroxy)]propylamine). The complexes obtained were characterized by elemental analysis, IR and UV-visible spectroscopies, electrospray ionization mass spectrometry (ESI-MS), tandem mass spectrometry (MS/MS), and cyclic voltammetry. X-ray diffraction studies were performed for complexes (3) and (4) revealing the presence of mononuclear and dinuclear structures in solid state for (3). However, the zinc complex is mononuclear in solid state. Biological studies of complexes (1)-(4) were carried out in vitro for antimicrobial activity against nine Gram-positive bacteria (Staphylococcus aureus strains RN 6390B, COL, ATCC 25923, Smith Diffuse, Wood 46, enterotoxigenic S. aureus FRI-100 (SEA+), FRI S-6 (SEB+) and SEC FRI-361) and animal strain S. aureus LSA 88 (SEC/SED/TSST-1+). The following sequence of inhibition promoted by the complexes was observed: (4)>(2)>(3)>(1), showing the effect of the metal on the biological activity. To directly observe the morphological changes of the internal structure of bacterial cells after the treatment, transmission electron microscopy (TEM) was employed. For the most active complex [Zn(II)(HBPClNOL)Cl] (4), granulation deposits around the genetic material and internal material leaking were clearly detected.

Entre os fatores determinantes da baixa utilização do consórcio gramínea-leguminosa, pode ser citado a implantação das leguminosas em pastagens já estabelecida. No entanto, animais podem servir como mecanismo de dispersão das sementes. Assim, objetivou-se neste estudo avaliar a introdução das leguminosas calopogônio, macrotiloma, kudzu tropical e soja perene em pastagem estabelecida de Brachiaria decumbens, por meio da dispersão em fezes de bovinos. Para esta finalidade, foi utilizado cinco novilhas como veiculadoras das sementes, as quais foram ofertadas 180g de sementes de cada leguminosa misturado ao concentrado de forma alternada, esperando um período de 48h para completa defecação das sementes. Todas as leguminosas foram encontradas nas placas fecais, porém o kudzu tropical apresentou maior número de exemplares encontrados em relação às demais (P<0,05). Quando avaliada a persistência após estabelecimento e pastejo, foi encontrado menor número de exemplares para o kudzu tropical em relação às outras três leguminosas (P<0,05), que não diferiram entre si. No entanto, a porcentagem de sementes germinadas nas fezes em relação à quantidade de sementes ofertadas em 180g de sementes para este mecanismo de propagação pode ser considerado baixo para as quatro leguminosas estudadas.


Among the factors to the low utilization of grass-leguminous intercropping, may be cited the deployment of leguminous in pastures already established. However, animals can serve as a seed dispersal mechanism. Thus, this study aimed to evaluate the introduction of leguminous calopogonium, macrotiloma, tropical kudzu and soybeans, in perennial pastures of Brachiaria decumbens, through dispersal in cattle dung. For this purpose we used five heifers as seeds disseminator, which were offered 180g seeds of each leguminous, alternately mixed with the concentrated, waiting a period of 48 hours to complete the seed defecation. All leguminous were found in fecal plates, but kudzu had the greatest number of specimens found in relation to others (P <.05). However, when evaluated persistence after establishment and grazing found fewer copies to kudzu in the other three leguminous (P <0.05) that did not differ among themselves. When, the percentage of seeds germinated in the stool, relative to the amount offered in 180g of seed for this propagation mechanism, can be considered low for the four leguminous studied.

The frequency of cyanobacterial blooms has been increasing all over the world. These blooms are often toxic and have become a serious health problem. The aim of this work was to search for population density control mechanisms that could inhibit the proliferation of the toxic bloom-forming genus Microcystis. Microcystis PCC 7806 cultured for long periods in liquid ASM-1 medium loses its characteristic green colour. When a medium of chlorotic cultures is added to a nutrient-replete culture, cell density increase is drastically reduced when compared with controls. Inhibition of cell proliferation occurs in Microcystis cultures from any growth stage and was not strain-specific, but other genera tested showed no response. Investigations on the mechanism of growth inhibition showed that cultures treated with the conditioned medium acquired a pale colour, with pigment concentration similar to that found in chlorotic cultures. Ultrastructural examination showed that the conditioned medium induced thylakoid membrane disorganization, typical of chlorotic cells, in nutrient-replete cultures. An active extract was obtained and investigations showed that activity was retained after heating and after addition of an apolar solvent. This indicates that activity of the conditioned medium from chlorotic cells results from non-protein, apolar compound(s).
