RESUMO

The elastic coupling between plastic events is generally invoked to interpret plastic properties and the failure of amorphous soft glassy materials. We report an experiment where the emergence of a self-organized plastic flow is observed well before the failure. For this we impose an homogeneous stress on a granular material, and measure local deformations for very small strain increments using a light scattering setup. We observe a nonhomogeneous strain that appears as transient bands of mesoscopic size and a well-defined orientation, which is different from the angle of the macroscopic frictional shear band that appears at the failure. The presence and the orientation of those microbands may be understood by considering how localized plastic reorganizations redistribute stresses in a surrounding continuous elastic medium. We characterize the length scale and persistence of the structure. The presence of plastic events and the mesostructure of the plastic flow are compared to numerical simulations.

We study experimentally the dynamical heterogeneities occurring at slow shear, in a model amorphous glassy material, i.e., a 3D granular packing. The deformation field is resolved spatially by using a diffusive wave spectroscopy technique. The heterogeneities show up as localized regions of strong deformations spanning a mesoscopic size of about 10 grains and called the "hot spots." The spatial clustering of hot spots is linked to the subsequent emergence of shear bands. Quantitatively, their appearance is associated with the macroscopic plastic deformation, and their rate of occurrence gives a physical meaning to the concept of "fluidity," recently used to describe the local and nonlocal rheology of soft glassy materials.

Transcriptional memory of transient signals can be imprinted on living systems and influence their reactivity to repeated stimulations. Although they are classically ascribed to structural chromatin rearrangements in eukaryotes, such behaviors can also rely on dynamic memory circuits with sustained self-amplification loops. However, these phenomena are either of finite duration, or conversely associated to sustained phenotypic changes. A mechanism is proposed, in which only the responsiveness of the target gene is durably reset at a higher level after primary stimulation, using the celebrated but still puzzling vitellogenesis memory effect. The basic ingredients of this system are: 1), a positive autoregulation of the estrogen receptor α gene; 2), a strongly cooperative action of the estradiol receptor on vitellogenin expression; and 3), a variant isoform of the estradiol receptor with two autonomous transcription-activating modules, one of which is signal-independent and the other, signal-dependent. Realistic quantification supports the possibility of a multistationary situation in which ligand-independent activity is unable by itself to prime the amplification loop, but can click the system over a memory threshold after a primary stimulation. This ratchet transcriptional mechanism can have developmental and ecotoxicological importance and explain lifelong imprinting of past exposures without apparent phenotypic changes before restimulation and without need for persistent chromatin modifications.

RESUMO

We propose an experimental setup based on diffusive wave spectroscopy for studying deformations of a solid material. The scattered waves are measured during the deformation of the solid material at different locations of its surface. The correlations of the scattered intensities are measured. The loss of correlation can be related to the invariants of the strain tensor of the solid, giving us a spatially resolved determination of the deformation near the surface. Experiments dealing with a point stress on plate for two kinds of elastic materials are presented and compared with the theoretical predictions.

We present a thorough study of the plastic response of a granular material progressively loaded. We study experimentally the evolution of the plastic field from a homogeneous one to a heterogeneous one and its fluctuations in terms of incremental strain. We show that the plastic field can be decomposed in two components evolving on two decoupled strain increment scales. We argue that the slowly varying part of the field can be identified with the so-called fluidity field introduced recently to interpret the rheological behavior of amorphous materials. This fluidity field progressively concentrates along a macroscopic direction corresponding to the Mohr-Coulomb angle.

RESUMO

We present a one-dimensional numerical model based on elastically coupled sliders on a frictional incline of variable tilt. This very simple approach makes it possible to study the precursors to the avalanche and to provide a rationalization of different features that have been observed in experiments. We provide a statistical description of the model leading to master equations describing the state of the system as a function of the angle of inclination. Our central results are the reproduction of large-scale regular events preceding the avalanche, on the one hand, and an analytical approach providing an internal threshold for the outbreak of rearrangements before the avalanche in the system, on the other hand.

RESUMO

This article is a tutorial on the practical implementation of a method of measurement of minute deformations based on multiple scattering. This technique has been recently developed and has proven to give new insights into the spatial repartition of strain in a granular material. We provide here the basics to understand the method by giving a synthetic review on diffusing wave spectroscopy and multiple scattering in granular materials. We detail a simple experiment using standard lab equipment to pedagogically demonstrate the implementation of the method. Finally we give a few examples of measurements that have been obtained in other works to discuss the potential of the method.

RESUMO

We present a numerical implementation of an active inclusion in a granular material submitted to a biaxial test. We discuss the dependence of the response to this perturbation on two parameters: the intragranular friction coefficient on one hand, and the degree of the loading on the other hand. We compare the numerical results to theoretical predictions taking into account the change of volume of the inclusion as well as the anisotropy of the elastic matrix.

We propose a dynamical mechanism leading to the fluidization by external mechanical fluctuations of soft-glassy amorphous material driven below the yield stress. The model is based on the combination of memory effect and nonlinearity, leading to an accumulation of tiny effects over a long term. We test this scenario on a granular packing driven mechanically below the Coulomb threshold. We provide evidence for an effective viscous response directly related to small stress modulations in agreement with the theoretical prediction of a generic secular drift. We propose to extend this result more generally to a large class of glassy systems.

We present an experimental study of the deformation inside a granular material that is progressively tilted. We investigate the deformation before the avalanche with a spatially resolved diffusive wave spectroscopy setup. At the beginning of the inclination process, we first observe localized and isolated events in the bulk, with a density which decreases with the depth. As the angle of inclination increases, series of microfailures occur periodically in the bulk, and finally a granular avalanche takes place. The microfailures are observed only when the tilt angles are larger than a threshold angle much smaller than the granular avalanche angle. We have characterized the density of reorganizations and the localization of microfailures. We have also explored the effect of the nature of the grains, the relative humidity conditions, and the packing fraction of the sample. We discuss those observations in the framework of the plasticity of granular matter. Microfailures may then be viewed as the result of the accumulation of numerous plastic events.

Chaperone synthesis in response to proteotoxic stress is dependent on a family of transcription factors named heat shock factors (HSFs). The two main factors in this family, HSF1 and HSF2, are co-expressed in numerous tissues where they can interact and form heterotrimers in response to proteasome inhibition. HSF1 and HSF2 exhibit two alternative splicing isoforms, called α and ß, which contribute to additional complexity in HSF transcriptional regulation, but remain poorly examined in the literature. In this work, we studied the transcriptional activity of HSF1 and HSF2 splicing isoforms transfected into immortalized Mouse Embryonic Fibroblasts (iMEFs) deleted for both Hsf1 and Hsf2, under normal conditions and after proteasome inhibition. We found that HSF1α is significantly more active than the ß isoform after exposure to the proteasome inhibitor MG132. Furthermore, we clearly established that, while HSF2 had no transcriptional activity by itself, short ß isoform of HSF2 exerts a negative role on HSF1ß-dependent transactivation. To further assess the impact of HSF2ß inhibition on HSF1 activity, we developed a mathematical modelling approach which revealed that the balance between each HSF isoform in the cell regulated the strength of the transcriptional response. Moreover, we found that cellular stress such as proteasome inhibition could regulate the splicing of Hsf2 mRNA. All together, our results suggest that relative amounts of each HSF1 and HSF2 isoforms quantitatively determine the cellular level of the proteotoxic stress response.

We consider the propagation of a diffusive wave in a scattering medium submitted to a homogeneous expansion. The light multiply scattered by a glass spheres sample is measured. We analyze the variations of the scattered light when the material and the optical wavelength are dilated. We experimentally show that an isotropic expansion of the material is equivalent to a contraction of the wavelength. Moreover, the effect of an expansion of the material on the scattered wave may be canceled by a proportional increase of the wavelength, keeping the phase of the scattered wave unchanged. Applications to the characterization of deformation of disordered materials are outlined.

RESUMO

We show experimentally that parametric interaction can induce a cooperative oscillation of non-simultaneously resonant transverse modes in an optical parametric oscillator. More generally, this effect is expected to occur in any spatially extended system subjected to boundary conditions where nonlinear wave mixing of two nonresonant spatial modes can generate a resonant oscillation.

Two-frequency operation of a composite Er,Yb:glass-LiTaO3 monolithic microlaser is demonstrated at 1.53 microm. The thermo-optic effect of the intracavity birefringent crystal makes it possible to control the wavelengths of the two orthogonal linearly polarized eigenstates. It provides a tunable beat note from 0 to 60 GHz. Direct evaluation of the Lamb-type coupling constant C between the two eigenstates is obtained from a single intensity noise spectrum measurement, yielding values from 0.33 to 0.86, depending on transverse and axial distributions of the eigenstates. Applications to radar and telecommunication systems are discussed.

RESUMO

Although deterministic chaos has been predicted to occur in the triply resonant optical parametric oscillator (TROPO) 15 years ago, experimental evidence of chaotic behavior in this system has been lacking so far, in marked contrast with most nonlinear systems, where chaos has been actively tracked and found. This situation is probably linked to the high sensitivity of the TROPO to perturbations, which adversely affects stationary operation at high power. We report the experimental observation in this system of a burst of irregular behavior of duration 80 micros. Although the system is highly nonstationary over this time interval, a topological analysis allows us to extract a clear-cut signature of deterministic chaos from a time series segment of only nine base cycles (3 micros). This result suggests that nonstationarity is not necessarily an obstacle to the characterization of chaos.
