Over evolutionary time, pathogen challenge shapes the immune phenotype of the host to better respond to an incipient threat. The extent and direction of this selection pressure depend on the local pathogen composition, which is in turn determined by biotic and abiotic features of the environment. However, little is known about adaptation to local pathogen threats in wild animals. The Gentoo penguin (Pygoscelis papua) is a species complex that lends itself to the study of immune adaptation because of its circumpolar distribution over a large latitudinal range, with little or no admixture between different clades. In this study, we examine the diversity in a key family of innate immune genes-the Toll-like receptors (TLRs)-across the range of the Gentoo penguin. The three TLRs that we investigated present varying levels of diversity, with TLR4 and TLR5 greatly exceeding the diversity of TLR7. We present evidence of positive selection in TLR4 and TLR5, which points to pathogen-driven adaptation to the local pathogen milieu. Finally, we demonstrate that two positively selected cosegregating sites in TLR5 are sufficient to alter the responsiveness of the receptor to its bacterial ligand, flagellin. Taken together, these results suggest that Gentoo penguins have experienced distinct pathogen-driven selection pressures in different environments, which may be important given the role of the Gentoo penguin as a sentinel species in some of the world's most rapidly changing environments.

BACKGROUND: Historical factors, demography, reproduction and dispersal are crucial in determining the genetic structure of seabirds. In the Antarctic marine environment, penguins are a major component of the avian biomass, dominant predators and important bioindicators of ecological change. Populations of chinstrap penguins have decreased in nearly all their breeding sites, and their range is expanding throughout the Antarctic Peninsula. Population genetic structure of this species has been studied in some colonies, but not between breeding colonies in the Antarctic Peninsula or at the species' easternmost breeding colony (Bouvetøya). RESULTS: Connectivity, sex-biased dispersal, diversity, genetic structure and demographic history were studied using 12 microsatellite loci and a mitochondrial DNA region (HVRI) in 12 breeding colonies in the South Shetland Islands (SSI) and the Western Antarctic Peninsula (WAP), and one previously unstudied sub-Antarctic island, 3600 km away from the WAP (Bouvetøya). High genetic diversity, evidence of female bias-dispersal and a sign of population expansion after the last glacial maximum around 10,000 mya were detected. Limited population genetic structure and lack of isolation by distance throughout the region were found, along with no differentiation between the WAP and Bouvetøya (overall microsatellite F ST = 0.002, p = 0.273; mtDNA F ST  = - 0.004, p = 0.766), indicating long distance dispersal. Therefore, genetic assignment tests could not assign individuals to their population(s) of origin. The most differentiated location was Georges Point, one of the southernmost breeding colonies of this species in the WAP. CONCLUSIONS: The subtle differentiation found may be explained by some combination of low natal philopatric behavior, high rates of dispersal and/or generally high mobility among colonies of chinstrap penguins compared to other Pygoscelis species.

Two main hypotheses have been debated about the biogeography of the Southern Ocean: (1) the Antarctic Polar Front (APF), acting as a barrier between Antarctic and sub-Antarctic provinces, and (2) the Antarctic Circumpolar Current (ACC), promoting gene flow among sub-Antarctic areas. The Gentoo penguin is distributed throughout these two provinces, separated by the APF. We analyzed mtDNA (HVR1) and 12 microsatellite loci of 264 Gentoo penguins, Pygoscelis papua, from 12 colonies spanning from the Western Antarctic Peninsula and the South Shetland Islands (WAP) to the sub-Antarctic Islands (SAI). While low genetic structure was detected among WAP colonies (mtDNA ФST=0.037-0.133; microsatellite FST=0.009-0.063), high differentiation was found between all SAI and WAP populations (mtDNA ФST=0.678-0.930; microsatellite FST=0.110-0.290). These results suggest that contemporary dispersal around the Southern Ocean is very limited or absent. As predicted, the APF appears to be a significant biogeographical boundary for Gentoo penguin populations; however, the ACC does not promote connectivity in this species. Our data suggest demographic expansion in the WAP during the last glacial maximum (LGM, about 20kya), but stability in SAI. Phylogenetic analyses showed a deep divergence between populations from the WAP and those from the SAI. Therefore, taxonomy should be further revised. The Crozet Islands resulted as a basal clade (3.57Mya), followed by the Kerguelen Islands (2.32Mya) as well as a more recent divergence between the Falkland/Malvinas Islands and the WAP (1.27Mya). Historical isolation, local adaptation, and past climate scenarios of those Evolutionarily Significant Units may have led to different potentials to respond to climate changes.

Phylogeographical studies are common in boreal and temperate species from the Palaearctic, but scarce in arid-adapted species. We used nuclear and mitochondrial markers to investigate phylogeography and to estimate chronology of colonization events of the trumpeter finch Bucanetes githagineus, an arid-adapted bird. We used 271 samples from 16 populations, most of which were fresh samples but including some museum specimens. Microsatellite data showed no clear grouping according to the sampling locations. Microsatellite and mitochondrial data showed the clearest differentiation between Maghreb and Canary Islands and between Maghreb and Western Sahara. Mitochondrial data suggest differentiation between different Maghreb populations and among Maghreb and Near East populations, between Iberian Peninsula and Canary Islands, as well as between Western Sahara and Maghreb. Our coalescence analyses indicate that the trumpeter finch colonized North Africa during the humid Marine Isotope Stage 5 (MIS5) period of the Sahara region 125 000 years ago. We constructed an ecological niche model (ENM) to estimate the geographical distribution of climatically suitable habitats for the trumpeter finch. We tested whether changes in the species range in relation to glacial-interglacial cycles could be responsible for observed patterns of genetic diversity and structure. Modelling results matched with those from genetic data as the species' potential range increases in interglacial scenarios (in the present climatic scenario and during MIS5) and decreases in glacial climates (during the last glacial maximum, LGM, 21 000 years ago). Our results suggest that the trumpeter finch responded to Pleistocene climatic changes by expanding and contracting its range.

Micro- and nanoplastics are widespread throughout the world. In particular, polyethylene (PE) and polyethylene terephthalate or polyester (PET) are two of the most common polymers, used as plastic bags and textiles. To analyze the toxicity of these two polymers, oligomers with different numbers of units were used as models. The use of oligomers as polymeric templates has been used previously with success. We started with the monomer and continued with different oligomers until the chain length was greater than two nm. According to the results of quantum chemistry, PET is a better oxidant than PE, since it is a better electron acceptor. Additionally, PET has negatively charged oxygen atoms and can promote stronger interactions than PE with other molecules. We found that PET forms stable complexes and can dissociate the guanine-cytosine nucleobase pair. This could affect DNA replication. These preliminary theoretical results may help elucidate the potential harm of micro- and nanoplastics.

Background: Incomplete species inventories for Antarctica represent a key challenge for comprehensive ecological research and conservation in the region. Additionally, data required to understand population dynamics, rates of evolution, spatial ranges, functional traits, physiological tolerances and species interactions, all of which are fundamental to disentangle the different functional elements of Antarctic biodiversity, are mostly missing. However, much of the fauna, flora and microbiota in the emerged ice-free land of the continent have an uncertain presence and/or unresolved status, with entire biodiversity compendia of prokaryotic groups (e.g. bacteria) being missing. All the available biodiversity information requires consolidation, cross-validation, re-assessment and steady systematic inclusion in order to create a robust catalogue of biodiversity for the continent. New information: We compiled, completed and revised eukaryotic species inventories present in terrestrial and freshwater ecosystems in Antarctica in a new living database: terrANTALife (version 1.0). The database includes the first integration in a compendium for many groups of eukaryotic microorganisms. We also introduce a first catalogue of amplicon sequence variants (ASVs) of prokaryotic biodiversity. Available compendia and literature to date were searched for Antarctic terrestrial and freshwater species, integrated, taxonomically harmonised and curated by experts to create comprehensive checklists of Antarctic organisms. The final inventories comprises 470 animal species (including vertebrates, free-living invertebrates and parasites), 306 plants (including all Viridiplantae: embryophytes and green algae), 997 fungal species and 434 protists (sensu lato). We also provide a first account for many groups of microorganisms, including non-lichenised fungi and multiple groups of eukaryotic unicellular species (Stramenophila, Alveolata and Rhizaria (SAR), Chromists and Amoeba), jointly referred to as "protists". In addition, we identify 1753 bacterial (obtained from 348117 ASVs) and 34 archaeal genera (from 1848 ASVs), as well as, at least, 14 virus families. We formulate a basic tree of life in Antarctica with the main lineages listed in the region and their "known-accepted-species" numbers.

The aim of this work is to contribute to the knowledge of gastrointestinal parasites of the Gentoo penguin (Pygoscelis papua) from 25 de Mayo/King George Island (South Shetlands, Antarctica). Gastrointestinal tracts of 37 fresh dead individuals (21 chicks, 10 juveniles, and 6 adults) were collected from December 2006 to February 2012 and examined for macroparasites. Four adult parasite species were found: one Cestoda species (Parorchites zederi), two Nematoda species (Stegophorus macronectes and Tetrameres wetzeli), and one Acanthocephalan (Corynosoma shackletoni). Two species of immature acanthocephalans, Corynosoma hamanni and Corynosoma bullosum, were found in a single host. This is the first record of Tetrameres wetzeli in Gentoo penguins. The low parasite richness observed could be related to the stenophagic and pelagic diet of this host species which feeds almost exclusively on krill.

In this Density Functional Theory study, it became apparent that astaxanthin (ASTA) may form metal ion complexes with metal cations such as Ca⁺², Cu⁺², Pb⁺², Zn⁺², Cd⁺² and Hg⁺². The presence of metal cations induces changes in the maximum absorption bands which are red shifted in all cases. Therefore, in the case of compounds where metal ions are interacting with ASTA, they are redder in color. Moreover, the antiradical capacity of some ASTA-metal cationic complexes was studied by assessing their vertical ionization energy and vertical electron affinity, reaching the conclusion that metal complexes are slightly better electron donors and better electron acceptors than ASTA.

Persistent Organic Pollutants (POPs) are a global threat, but impacts of these chemicals upon remote areas such as Antarctica remain unclear. Penguins can be useful species to assess the occurrence of POPs in Antarctic food webs. This work's aim was the evaluation of polychlorodibenzo-p-dioxins and furans (PCDD/Fs), polychlorinated biphenyls (PCBs) and polybrominated diphenyl ethers (PBDEs) in eggs of two penguin species, chinstrap (Pygoscelis antarticus) and gentoo penguins (Pygoscelis papua), breeding in the South Shetland Islands. Results showed a common pattern in POP levels regardless of the species, characterized by a major abundance of PCBs (98 %), followed by PBDEs (1-2%) and PCDD/Fs (<1 %). Concentrations of POPs in chinstrap and gentoo penguin eggs were 482 and 3250 pg/g l.w., respectively. PCBs, PBDEs and PCDD/Fs were found at higher concentrations in chinstrap penguin eggs, being these differences significant for PBDEs. Interspecies differences in POP levels agree well with potential trophic position differences among species due to changes in prey composition and foraging areas. POP profiles were dominated by congeners with a low degree of halogenation. Our results therefore suggest similar sources of POPs in the food webs exploited by both species and in both cases attributable to the long-range transportation rather than to the presence of local sources of POPs. TEQs were found between 1.38 and 7.33 pg/g l.w. and followed the pattern non-ortho dl-PCBs > PCDFs > PCDDs > mono-ortho dl-PCBs. TEQ values were lower than the threshold level for harmful effects in birds of 210 pg/g WHO-TEQ/g l.w.

Under the climate change context, warming Southern Ocean waters may allow mercury (Hg) to become more bioavailable to the Antarctic marine food web (i.e., ice-stored Hg release and higher methylation rates by microorganisms), whose biomagnification processes are poorly documented. Biomagnification of Hg in the food web of the Antarctic Peninsula, one of the world's fastest-warming regions, was examined using carbon (δ13C) and nitrogen (δ15N) stable isotope ratios for estimating feeding habitat and trophic levels, respectively. The stable isotope signatures and total Hg (T-Hg) concentrations were measured in Antarctic krill Euphausia superba and several Antarctic predator species, including seabirds (gentoo penguins Pygoscelis papua, chinstrap penguins Pygoscelis antarcticus, brown skuas Stercorarius antarcticus, kelp gulls Larus dominicanus, southern giant petrels Macronectes giganteus) and marine mammals (southern elephant seals Mirounga leonina). Significant differences in δ13C values among species were noted with a great overlap between seabird species and M. leonina. As expected, significant differences in δ15N values among species were found due to interspecific variations in diet-related to their trophic position within the marine food web. The lowest Hg concentrations were registered in E. superba (0.007 ± 0.008 µg g-1) and the highest values in M. giganteus (12.090 ± 14.177 µg g-1). Additionally, a significant positive relationship was found between Hg concentrations and trophic levels (reflected by δ15N values), biomagnifying nearly 2 times its concentrations at each level. Our results support that trophic interaction is the major pathway for Hg biomagnification in Southern Ocean ecosystems and warn about an increase in the effects of Hg on long-lived (and high trophic level) Antarctic predators under climate change in the future.

Within the Southern Ocean, the greatest warming is occurring on the Antarctic Peninsula (AP) where clear cryospheric and biological consequences are being observed. Antarctic coastal systems harbour a high diversity of marine and terrestrial ecosystems heavily influenced by Antarctic seaweeds (benthonic macroalgae) and bird colonies (mainly penguins). Primary sea spray aerosols (SSA) formed by the outburst of bubbles via the sea-surface microlayer depend on the organic composition of the sea water surface. In order to gain insight into the influence of ocean biology and biogeochemistry on atmospheric aerosol, we performed in situ laboratory aerosol bubble chamber experiments to study the effect of different leachates of biogenic material - obtained from common Antarctic seaweeds as well as penguin guano - on primary SSA. The addition of different leachate materials on a seawater sample showed a dichotomous effect depending on the leachate material added - either suppressing (up to 52%) or enhancing (22-88%) aerosol particle production. We found high ice nucleating particle number concentrations resulting from addition of guano leachate material. Given the evolution of upper marine polar coastal ecosystems in the AP, further studies on ocean-atmosphere coupling are needed in order to represent the currently poorly understood climate feedback processes.

Penguins dominate the Antarctic avifauna. As key animals in the Antarctic ecosystem, they are monitored to evaluate the ecological status of this pristine and remote region and specifically, they have been used as effective bioindicators suitable for long-term monitoring of metals in the Antarctic environment. However, studies about the role of this emblematic organism could play in the recycling of trace metals (TMs) in the Antarctic ecosystem are very limited. In this study we evaluate, using the peer review research articles already published and our own findings, the distribution of metals (i.e., Ca, Fe, Al, Na, Zn, Mg, Cu, K, Cd, Mn, Sr, Cr, Ni, Pb, Hg, V, Ba, Co, La, Ag, Rb, Hf, Sc, Au and Cs) and metalloids (As and Sb), measured in different biotic matrices, with emphasis on guano, of the Chinstrap (Pygoscelis antarcticus), Adélie (Pygoscelis adeliae) and Gentoo (Pygoscelis papua) penguins. Regarding bioactive metals, the high concentrations (µg g-1 dry weight) of Cu (2.0 ± 1.4) x 102, Fe (4.1 ± 2.9) x 102, Mn (30 ± 34) and Zn (210 ± 90) reported in the guano from all the penguin species studied including our data, are of the same order of magnitude as those reported for whale feces (µg g-1 dry weight): Cu (2.9 ± 2.4) x 102, Fe (1.5 ± 1.4) x 102, Mn (28 ± 17) and Zn (6.2 ± 4.3) x 102, and one order of magnitude higher than the metal contents in krill (µg g-1 dry weight) of Cu (10.2 ± 5.5), Fe (24 ± 29) and Zn (13.5 ± 1.7). This suggest that penguin's excretion products could be an important source of these essential elements in the surface water, with an estimated annual release on a breeding season for Cu, Fe, Mn, Zn respectively of 28, 56, 4 and 29 tons, for the Chinstrap, Adélie and Gentoo penguins. The results provide evidence on the potential influence of penguins recycling TMs in the surface layer of the water column.

Polar regions, symbols of wilderness, have been identified as potential sinks of mercury coming from natural and anthropogenic sources at lower latitudes. Changes in ice coverage currently occurring in some areas such as the Antarctic Peninsula could enhance these phenomena and their impacts on local biota. As long-lived species at the top of food chains, seabirds are particularly sensitive to this highly toxic metal with the capacity to be biomagnified. Specifically, their feathers can be useful for Hg monitoring since they mainly accumulate its most toxic and persistent form, methyl-Hg. To that end, feathers of gentoo (Pygoscelis papua), chinstrap (P. antarcticus), and Adélie penguins (P. adeliae) (n = 108) were collected by passive sampling in seven different locations throughout the Antarctic Peninsula area and analyzed by ICP-MS after microwave-digestion. More than 93% of the samples showed detectable Hg levels (range: 6.3-12,529.8 ng g-1 dry weight), and the highest ones were found in the feathers of chinstrap penguins from King George Island. Hg bioconcentration and biomagnification seem to be occurring in the Antarctic food web, giving rise to high but non-toxic Hg levels in penguins, similar to those previously found in Arctic seabirds.

Microplastics (< 5 mm in size) are known to be widespread in the marine environment but are still poorly studied in Polar Regions, particularly in the Antarctic. As penguins have a wide distribution around Antarctica, three congeneric species: Adélie (Pygoscelis adeliae), chinstrap (Pygoscelis antarcticus) and gentoo penguins (Pygoscelis papua) were selected to evaluate the occurrence of microplastics across the Antarctic Peninsula and Scotia Sea. Scat samples (used as a proxy of ingestion), were collected from breeding colonies over seven seasons between 2006 and 2016. Antarctic krill (Euphausia superba), present in scat samples, contributed 85%, 66% and 54% of the diet in terms of frequency of occurrence to the diet of Adélie, gentoo and chinstrap penguins, respectively. Microplastics were found in 15%, 28% and 29% scats of Adélie, chinstrap and gentoo penguin respectively. A total of 92 particles were extracted from the scats (n = 317) and 32% (n = 29) were chemically identified via micro-Fourier Transform Infrared Spectroscopy (µ-FTIR). From all the particles extracted, 35% were identified as microplastics, particularly polyethylene (80%) and polyester (10%). It was not possible to ascertain the identification of the remaining 10% of samples. Other anthropogenic particles were identified in 55% of samples, identified as cellulose fibres. The results show a similar frequency of occurrence of particles across all colonies, suggesting there is no particular point source for microplastic pollution in the Scotia Sea. Additionally, no clear temporal variation in the number of microplastics in penguins was observed. Overall, this study reveals the presence of microplastics across Antarctica, in three penguin species and offers evidence of other anthropogenic particles in high numbers. Further research is needed to better understand the spatio-temporal dynamics, fate and effect of microplastics on these ecosystems, and improve plastic pollution policies in Antarctica.

RESUMO

The coronavirus disease 2019 (COVID-19) pandemic is caused by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). This pathogen has spread rapidly across the world, causing high numbers of deaths and significant social and economic impacts. SARS-CoV-2 is a novel coronavirus with a suggested zoonotic origin with the potential for cross-species transmission among animals. Antarctica can be considered the only continent free of SARS-CoV-2. Therefore, concerns have been expressed regarding the potential human introduction of this virus to the continent through the activities of research or tourism to minimise the effects on human health, and the potential for virus transmission to Antarctic wildlife. We assess the reverse-zoonotic transmission risk to Antarctic wildlife by considering the available information on host susceptibility, dynamics of the infection in humans, and contact interactions between humans and Antarctic wildlife. The environmental conditions in Antarctica seem to be favourable for the virus stability. Indoor spaces such as those at research stations, research vessels or tourist cruise ships could allow for more transmission among humans and depending on their movements between different locations the virus could be spread across the continent. Among Antarctic wildlife previous in silico analyses suggested that cetaceans are at greater risk of infection whereas seals and birds appear to be at a low infection risk. However, caution needed until further research is carried out and consequently, the precautionary principle should be applied. Field researchers handling animals are identified as the human group posing the highest risk of transmission to animals while tourists and other personnel pose a significant risk only when in close proximity (< 5 m) to Antarctic fauna. We highlight measures to reduce the risk as well as identify of knowledge gaps related to this issue.

BACKGROUND: Energy landscapes provide an approach to the mechanistic basis of spatial ecology and decision-making in animals. This is based on the quantification of the variation in the energy costs of movements through a given environment, as well as how these costs vary in time and for different animal populations. Organisms as diverse as fish, mammals, and birds will move in areas of the energy landscape that result in minimised costs and maximised energy gain. Recently, energy landscapes have been used to link energy gain and variable energy costs of foraging to breeding success, revealing their potential use for understanding demographic changes. METHODS: Using GPS-temperature-depth and tri-axial accelerometer loggers, stable isotope and molecular analyses of the diet, and leucocyte counts, we studied the response of gentoo (Pygoscelis papua) and chinstrap (Pygoscelis antarcticus) penguins to different energy landscapes and resources. We compared species and gentoo penguin populations with contrasting population trends. RESULTS: Between populations, gentoo penguins from Livingston Island (Antarctica), a site with positive population trends, foraged in energy landscape sectors that implied lower foraging costs per energy gained compared with those around New Island (Falkland/Malvinas Islands; sub-Antarctic), a breeding site with fluctuating energy costs of foraging, breeding success and populations. Between species, chinstrap penguins foraged in sectors of the energy landscape with lower foraging costs per bottom time, a proxy for energy gain. They also showed lower physiological stress, as revealed by leucocyte counts, and higher breeding success than gentoo penguins. In terms of diet, we found a flexible foraging ecology in gentoo penguins but a narrow foraging niche for chinstraps. CONCLUSIONS: The lower foraging costs incurred by the gentoo penguins from Livingston, may favour a higher breeding success that would explain the species' positive population trend in the Antarctic Peninsula. The lower foraging costs in chinstrap penguins may also explain their higher breeding success, compared to gentoos from Antarctica but not their negative population trend. Altogether, our results suggest a link between energy landscapes and breeding success mediated by the physiological condition.

The antiradical capacities of 13 carotenoids (CAR) and vitamin E are explored, by assessing CAR-H bond dissociation energy. Density functional theory (DFT) calculations were performed, in order to evaluate the hydrogen atom transfer (HAT) antiradical mechanism. Results indicate that C4 or C4' is not always the reactive position when it is unsubstituted and also that CAR without H atoms in the 4 position may be as effective against free radicals as other CAR with H atoms in C4 and C4'. Lutein is the most effective antiradical for the purpose of hydrogen abstraction, whereas the least effective antiradical for this process is canthaxanthin, which is one of the reddest CAR. Vitamin E is not as effective as most of the yellow CAR but may be a better antiradical than canthaxanthin. In addition to the CAR-H bond dissociation energy, the number of reactive positions as we report in this paper represents another important aspect for consideration, when analyzing capacity for scavenging free radicals. Many additional aspects exist, which we do not consider here; thus we cannot attempt to reflect all the factors seen in vivo. However, our results provide comparative information on the relative ability of CAR to protect against free radicals, using the CAR-H bond dissociation energy, as one useful parameter. We hope that our theoretical results will contribute to the advancement of this complex research field.

Bright yellow and red colors in animals and plants are assumed to be caused by carotenoids (CAR). In animals, these pigments are deposited in scales, skin and feathers. Together with other naturally occurring and colorless substances such as melatonin and vitamins, they are considered antioxidants due to their free-radical-scavenging properties. However, it would be better to refer to them as "antiradicals", an action that can take place either donating or accepting electrons. In this work we present quantum chemical calculations for several CAR and some colorless antioxidants, such as melatonin and vitamins A, C and E. The antiradical capacity of these substances is determined using vertical ionization energy (I), electron affinity (A), the electrodonating power (omega(-)) and the electroaccepting power (omega(+)). Using fluor and sodium as references, electron acceptance (R(a)) and electron donation (R(d)) indexes are defined. A plot of R(d) vs R(a) provides a donator acceptor map (DAM) useful to classify any substance regarding its electron donating-accepting capability. Using this DAM, a qualitative comparison among all the studied compounds is presented. According to R(d) values, vitamin E is the most effective antiradical in terms of its electron donor capacity, while the most effective antiradical in terms of its electron acceptor capacity, R(a), is astaxanthin, the reddest CAR. These results may be helpful for understanding the role played by naturally occurring pigments, acting as radical scavengers either donating or accepting electrons.
