BACKGROUND AND PURPOSE: We aimed to estimate the prevalence of perceived cognitive impairment (PCI) and explore its associations with lifestyle and disease characteristics in a large international cohort of people with multiple sclerosis (MS). METHODS: This study was a cross-sectional analysis. Participants rated their cognitive function over the preceding 4 weeks using four questions in a subscale within the Multiple Sclerosis Quality of Life questionnaire (MSQOL-54). These questions assessed perceived concentration, attention and memory by the patient and family/friends. Four definitions of PCI were derived, ranging from lowest to highest specificity. Associations with PCI were assessed by log-binomial regression. RESULTS: The prevalence of PCI in our sample ranged from 41.0% (95% confidence interval, 39.0-43.0) using the least-specific definition to 11.6% (95% confidence interval, 10.3-12.9) using the most specific definition. A number of factors were associated with PCI, increasing in magnitude as the definition specificity increased, including positive associations for smoking and body mass index, whereas physical activity, dietary quality and use of vitamin D/omega-3 supplements were inversely associated with PCI. CONCLUSIONS: Our study reports associations between healthy lifestyle behaviours and PCI in people with MS. Although reverse causality is a potential explanation for our findings, previous studies have shown comparable associations with healthy lifestyle and MS onset and progression. Subject to external validation, these results suggest benefits realized from a healthy lifestyle in people with MS.

Matter with a high energy density (>10(5) joules per cm(3)) is prevalent throughout the Universe, being present in all types of stars and towards the centre of the giant planets; it is also relevant for inertial confinement fusion. Its thermodynamic and transport properties are challenging to measure, requiring the creation of sufficiently long-lived samples at homogeneous temperatures and densities. With the advent of the Linac Coherent Light Source (LCLS) X-ray laser, high-intensity radiation (>10(17) watts per cm(2), previously the domain of optical lasers) can be produced at X-ray wavelengths. The interaction of single atoms with such intense X-rays has recently been investigated. An understanding of the contrasting case of intense X-ray interaction with dense systems is important from a fundamental viewpoint and for applications. Here we report the experimental creation of a solid-density plasma at temperatures in excess of 10(6) kelvin on inertial-confinement timescales using an X-ray free-electron laser. We discuss the pertinent physics of the intense X-ray-matter interactions, and illustrate the importance of electron-ion collisions. Detailed simulations of the interaction process conducted with a radiative-collisional code show good qualitative agreement with the experimental results. We obtain insights into the evolution of the charge state distribution of the system, the electron density and temperature, and the timescales of collisional processes. Our results should inform future high-intensity X-ray experiments involving dense samples, such as X-ray diffractive imaging of biological systems, material science investigations, and the study of matter in extreme conditions.

Solanum sisymbriifolium, the litchi tomato, is a perennial herbaceous plant from South America that is used as a trap crop to reduce soilborne populations of the pale cyst nematode Globodera pallida, an important potato pathogen. Possible interactions of soilborne potato pathogens Verticillium dahliae and Colletotrichum coccodes with litchi tomato are unknown, yet important for potato production if litchi tomato is to be planted as a trap crop. The goal of this research was to quantitatively assess if litchi tomato is a potential inoculum source for C. coccodes and V. dahliae by comparing colony forming units (CFU) observed in litchi tomato to susceptible and resistant potato cultivars. The potato cvs. Alturas (P = 0.0003), Ranger Russet (P = 0.0193), and Russet Norkotah (P = 0.0022) produced more CFUs of the potato pathotype of V. dahliae than litchi tomato the first of two years of greenhouse trials. Significantly more CFUs of the potato pathotype of V. dahliae were quantified from stems and roots of only cv. Russet Norkotah compared with litchi tomato (P = 0.0001) in the second year. The CFUs for C. coccodes varied between litchi tomato and the potato cvs., perhaps due to varying levels of resistance since litchi tomato is from a selected intermated seed source. Based on these data, the effect of litchi tomato in rotation with potato is likely to have limited effect on the proliferation of V. dahliae or C. coccodes populations in the soil when compared with a susceptible potato cultivar.

1. Hatching success of ostrich eggs is poor (50-60% of fertile eggs). The current study was designed to identify the timing of key stages in the development of the ostrich embryo. 2. Growth of both embryo and wing length during 42 d of incubation was comparable and approximately linear, with a more or less weekly doubling in size up to 35 d of incubation. 3. The embryo eye size increased more rapidly than beak length and reached a maximum of ~16.2 mm by 28 d of incubation, whereas beak length increased continuously until hatching at 42 d. 4. Linear regression equations were derived from morphometric measurements of embryos between 7 and 42 d. 5. Information stemming from these results can be used to estimate the age of dead-in-shell embryos in an attempt to identify timing of incubation problems that potentially result in low hatchability of fertile eggs.

Potato mop-top virus (PMTV; family Virgaviridae) was reported recently in the Pacific Northwestern USA. To better understand the genetic diversity of this virus, the complete genome of an isolate from Washington State (WA), USA, was characterized. Sequence comparisons of the WA isolate with other known sequences revealed that the RNA-Rep-encoded RdRp protein and the RNA-CP-encoded coat protein displayed >99 % amino acid sequence identity to those of two Nordic (RdRp) and several European and North American isolates (CP), respectively. The RNA-TGB-encoded TGB 1 and TGB 3 protein sequences had >99 % amino acid sequence identity to the corresponding proteins of Czech and Danish isolates, whereas the TGB 2 protein is identical to those of Colombian isolates. Phylogenetic analysis of the viral genes of the WA isolate reflected the close relationship between WA and European isolates. RFLP analysis of corresponding DNA of RNA TGB and RNA CP revealed that the WA isolate has the RNA TGB-II and RNA CP-B types, which are prevalent in Europe and other parts of world. This is the first report of the complete genome characterization of PMTV from the Americas.

1. Early development of ostrich embryos was investigated in relation to time of egg collection and genotype. 2. A total of 321 ostrich eggs were collected during the 2008 and 2009 breeding seasons and the development of the embryo for up to the first 168 h of incubation was described and analysed. A sample of the incubated eggs was weighed and opened daily to investigate developmental changes. 3. In fresh eggs, the blastoderm contained a round, translucent dark area pellucida (AP) in the centre, with a surrounding thin white ring, likely to be the beginning of the area opaca (AO). Fresh eggs were considered infertile if the blastoderm was absent and instead numerous white droplets were present surrounded by vacuoles. 4. The average blastoderm area of a fresh fertile egg was 15.8 mm(2), increasing to 143.3 mm(2) after 2 d of incubation. By 72 h of incubation the area vasculosa (AV) was discernible in the posterior half of the blastoderm. 5. At 48 h of incubation the blastoderm area in eggs from the South African Black genotype (SAB) × Zimbabwean Blue genotype (ZB) crosses (104.5 ± 18.6 mm(2)) was lower than the pure SAB (141.0 ± 10.5 mm(2)), ZB (161.7 ± 13.5 mm(2)) and ZB × SAB crosses (166.1 ± 14.2 mm(2)). 6. Embryo length was 5.01 mm after 72 h of incubation and 14.5 mm after 168 h of incubation. At 168 h of incubation AV lengths for both ZB × SAB (53.2 mm) and SAB × ZB crosses (54.1 mm) were longer than in embryos from the pure breeds. 7. Results from this study can be put to practical use when determining whether eggs are infertile or fertile and also in investigating the age of early embryonic mortalities.

RESUMO

Plasma turbulence plays a key role in space and astrophysical plasma systems, enabling the energy of magnetic fields and plasma flows to be transported to particle kinetic scales at which the turbulence dissipates and heats the plasma. Identifying the physical mechanisms responsible for the dissipation of the turbulent energy is a critical step in developing the predictive capability for the turbulent heating needed by global models. In this work, spacecraft measurements of the electromagnetic fields and ion velocity distributions by the Magnetospheric Multiscale (MMS) mission are used to generate velocity-space signatures that identify ion cyclotron damping in Earth's turbulent magnetosheath, in agreement with analytical modeling. Furthermore, the rate of ion energization is directly quantified and combined with a previous analysis of the electron energization to identify the dominant channels of turbulent dissipation and determine the partitioning of energy among species in this interval.

The newly commissioned Orion laser system has been used to study dense plasmas created by a combination of short pulse laser heating and compression by laser driven shocks. Thus the plasma density was systematically varied between 1 and 10 g/cc by using aluminum samples buried in plastic foils or diamond sheets. The aluminum was heated to electron temperatures between 500 and 700 eV allowing the plasma conditions to be diagnosed by K-shell emission spectroscopy. The K-shell spectra show the effect of the ionization potential depression as a function of density. The data are compared to simulated spectra which account for the change in the ionization potential by the commonly used Stewart and Pyatt prescription and an alternative due to Ecker and Kröll suggested by recent x-ray free-electron laser experiments. The experimental data are in closer agreement with simulations using the model of Stewart and Pyatt.

The extent to which fluctuating selection can maintain evolutionary stasis in most populations remains an unresolved question in evolutionary biology. Climate has been hypothesized to drive reversals in the direction of selection among different time periods and may also be responsible for intense episodic selection caused by rare weather events. We measured viability selection associated with morphological traits in cliff swallows (Petrochelidon pyrrhonota) in western Nebraska, USA, over a 14-year period following a rare climatic event. We used mark-recapture to estimate the annual apparent survival of over 26 000 individuals whose wing, tail, tarsus and bill had been measured. The fitness functions associated with tarsus length and bill dimensions fluctuated depending on annual climate conditions on the birds' breeding grounds. The oscillating yearly patterns may have slowed and occasionally reversed directional change in trait trajectories, although there was a trend over time for all traits except tarsus to increase in size. The net positive directional selection on some traits, despite periodic climate-associated fluctuations, suggests that cliff swallow morphology in the population is likely to keep changing and supports recent work contending that selection in general does not fluctuate enough to be an effective driver of stasis.

The x-ray intensities made available by x-ray free electron lasers (FEL) open up new x-ray matter interaction channels not accessible with previous sources. We report here on the resonant generation of Kα emission, that is to say the production of copious Kα radiation by tuning the x-ray FEL pulse to photon energies below that of the K edge of a solid aluminum sample. The sequential absorption of multiple photons in the same atom during the 80 fs pulse, with photons creating L-shell holes and then one resonantly exciting a K-shell electron into one of these holes, opens up a channel for the Kα production, as well as the absorption of further photons. We demonstrate rich spectra of such channels, and investigate the emission produced by tuning the FEL energy to the K-L transitions of those highly charged ions that have transition energies below the K edge of the cold material. The spectra are sensitive to x-ray intensity dependent opacity effects, with ions containing L-shell holes readily reabsorbing the Kα radiation.

We have used the Linac Coherent Light Source to generate solid-density aluminum plasmas at temperatures of up to 180 eV. By varying the photon energy of the x rays that both create and probe the plasma, and observing the K-α fluorescence, we can directly measure the position of the K edge of the highly charged ions within the system. The results are found to disagree with the predictions of the extensively used Stewart-Pyatt model, but are consistent with the earlier model of Ecker and Kröll, which predicts significantly greater depression of the ionization potential.

1. A study was conducted on ~14000 ostrich eggs to estimate genetic parameters for eggshell traits that could benefit the hatchability of ostrich eggs. Traits measured included the number of pores on the eggshell, the average diameter of these pores, the total area of pores on the eggshell, permeability (pore area/shell thickness) and eggshell thickness. 2. Heritability estimates ranged from 0·16 for total pore area to 0·41 for the natural logarithm of pore count. The heritability estimates for water loss on 21 and 35 d (WL21 and WL35) of incubation were high at 0·23 and 0·24, respectively. 3. On a genetic level, pore count was negatively correlated with average pore diameter (-0·73) and shell thickness (-0·28), whereas it was positively correlated with total pore area (0·58), WL21 (0·24) and WL35 (0·34). The direct and maternal genetic correlations of pore count with total pore area (0·58) and permeability (0·59) were high and significant. Permeability was positively correlated to WL21 and WL35, both on the direct and maternal genetic levels. 4. The estimated genetic parameters indicate that it should be possible to select for the various eggshell traits in ostrich eggs, or for permeability and water loss. However, as a trait with an intermediate optimum, direct selection for permeability and other eggshell traits would not be straightforward, and the possible application of these results to improve hatchability of ostrich eggs in the future needs consideration.

The heating of solid foils by a picosecond time scale laser pulse has been studied by using x-ray emission spectroscopy. The target material was plastic foil with a buried layer of a spectroscopic tracer material. The laser pulse length was either 0.5 or 2 ps, which resulted in a laser irradiance that varied over the range 10(16)-10(19) W/cm(2). Time-resolved measurements of the buried layer emission spectra using an ultrafast x-ray streak camera were used to infer the density and temperature conditions as a function of laser parameters and depth of the buried layer. Comparison of the data to different models of electron transport showed that they are consistent with a model of electron transport that predicts the bulk of the target heating is due to return currents.

The viral accessory protein Vpx is required for productive in vitro infection of macrophages by simian immunodeficiency virus from sooty mangabey monkeys (SIV(SM)). To evaluate the roles of Vpx and macrophage infection in vivo, we inoculated pigtailed macaques intravenously or intrarectally with the molecularly cloned, macrophage tropic, acutely pathogenic virus SIV(SM) PBj 6.6, or accessory gene deletion mutants (deltaVpr or deltaVpx) of this virus. Both wild-type and SIV(SM) PBj deltaVpx viruses were readily transmitted across the rectal mucosa. A subsequent 'stepwise' process of local amplification of infection and dissemination was observed for wild-type virus, but not for SIV(SM) PBj deltaVpx, which also showed considerable impairment of the overall kinetics and extent of its replication. In animals co-inoculated with equivalent amounts of wild-type and SIV(SM) Pbj deltaVpx intravenously or intrarectally, the deltaVpx mutant was at a strong competitive disadvantage. Vpx-dependent viral amplification at local sites of initial infection, perhaps through a macrophage-dependent mechanism, may be a prerequisite for efficient dissemination of infection and pathogenic consequences after exposure through either mucosal or intravenous routes.

During the past few years, the Orion high-resolution x-ray spectrometers have been successful tools for measuring x-ray spectra from plasmas generated in the Orion laser facility. Duplicate spectrometers also operate successfully at the Livermore EBIT-I and SuperEBIT electron beam ion traps for measuring x-ray polarization. We have recently implemented very high-quality, optically bonded, spherically bent quartz crystals to remove the structure in the x-ray image that had been observed in earlier measurements. The structure had been caused by focusing defects and limited the accuracy of our measurements. We present before and after images that show a drastic improvement. We, furthermore, have implemented a spherically bent potassium acid phthalate (KAP) crystal on one of our spectrometers. The KAP crystal was prepared in a similar fashion, and we present measurements of the N Ly-ß and Ne Lyß lines taken in first- and second-order reflections at 600 and 1200 eV, respectively. These measurements confirm that KAP crystals can be produced at a quality suitable for extending the spectral coverage to wavelengths longer than those accessible by different quartz crystals, especially those that cover the astrophysically important lines of iron.

Glucocorticosteroids are a very effective treatment for asthma and other chronic inflammatory diseases. However, a small proportion of patients is resistant to the therapeutic effects of glucocorticoids. Pharmacokinetic and ligand binding studies suggest that the molecular abnormality in steroid resistance lies distal to nuclear translocation. We have previously reported that there is a decreased ability of glucocorticoid receptors (GR) to bind to the DNA-binding site in peripheral blood mononuclear cells (PBMC) after dexamethasone treatment. This reduced DNA binding was due to a decrease in the number of receptors available rather than an alteration in affinity for DNA. To study this reduced DNA binding, we examined the ability of the nuclear translocated transcription factors activator protein 1 (AP-1), nuclear factor kappa B (NF-kappa B) and cyclic AMP response element-binding protein (CREB) to bind to their DNA-binding sites and to interact with GR in PBMC from patients with steroid-sensitive and steroid-resistant asthma. There was a significant reduction in the interaction between GR and AP-1 in these steroid-resistant patients, although interaction with other transcription factors activated in inflammation (NF-kappa B and CREB) was unaffected. An increase in the basal levels of AP-1 DNA binding was also detected in the nuclei from steroid-resistant asthmatic patients. There were no differences in the amount of messenger RNA detected for the components of AP-1, c-Fos and c-Jun, nor in the sequences of these messenger RNAs. These results suggest either that the ability of the GR to bind to glucocorticoid response elements and AP-1 is altered in steroid-resistant patients or that increased levels of AP-1 prevent GR DNA binding, and that this may be the molecular basis of resistance to the antiinflammatory effect of steroids in these cells.

The potato cyst nematode, Globodera pallida (PCN), a restricted pest in the United States, was first reported in Bingham and Bonneville counties of Idaho in 2006 (1). The U.S. government and Idaho State Department of Agriculture hope to eradicate it from infested fields. Eradicating PCN will require depriving the nematodes of their hosts over a protracted time period. Functional eradication might be achieved with relatively high, proven to be efficacious dosages of soil fumigants. The presence of host weeds of PCN can play a significant role in the success of the eradication program. To determine the host status of common weeds found in potato fields of the Pacific Northwest, host suitability tests were conducted in a secured greenhouse located at the University of Idaho at Moscow. Reproduction of PCN on nine weeds including hairy nightshade (Solanum physalifolium formerly S. sarrachoides) and cutleaf nightshade (S. triflorum) (biotypes from Idaho and Washington), black nightshade (S. nigrum) (Washington biotype), bittersweet nightshade (S. dulcamara) (Idaho biotype), redroot pigweed (Amaranthus retroflexus), kochia (Kochia scoparia), and common lambsquarters (Chenopodium album) were compared with reproduction on Desiree, Russet Burbank (known hosts), and Santé (poor host) potatoes (S. tuberosum). Plants were grown in 10-cm-diameter clay pots containing sandy loam soil previously fumigated with methyl bromide and inoculated with 10 to 150 cysts that were either collected from infested fields or raised in the secured greenhouse (ample diapause period elapsed). Treatments were replicated five times and each trial lasted 3 months. Cysts were extracted from soil with a Fenwick can, and the reproductive factor (RF = final cyst count ÷ initial inoculum) was determined. While both biotypes of hairy nightshade were suitable hosts of PCN (161-668 ÷ 150; RF = >1), cutleaf biotypes, black, and bittersweet nightshades were poor hosts (1-108 ÷ 150; RF = <1). Russet Burbank (77 ÷ 40; RF = 1.9) and Desiree (21-119 ÷ 75; RF = >1) proved to be suitable hosts and Santé (1-20 ÷ 150; RF = <1) a poor host of Idaho PCN. Although some cysts were recovered from pots containing the remaining weed species, they may have been part of the original inoculum. The significance of nightshade species (whether suitable or poor hosts) in eradication of potato cyst nematode from infested fields cannot be overemphasized. Reference: (1) A. M. Skantar et al. J. BACKGROUND: Mastery is the sense of being in control of one's life and improvement in mastery may help to enhance quality of life. Little research has explored mastery in people with multiple sclerosis (MS), including its association with quality of life. OBJECTIVE: To explore the association between mastery and health-related quality of life (HRQOL) in people with MS. METHOD: Two cross-sectional cohorts of adults with MS (n = 1401 and n = 573), recruited through convenience sampling, completed an online survey which measured mastery using the Pearlin Mastery Scale, physical and mental HRQOL via physical and mental health composite scores of MSQOL-54, along with other covariates, including demographics, clinical characteristics and lifestyle factors. Linear regression assessed associations between mastery and physical HRQOL adjusting for age, sex, education, disability and depression, and between mastery and mental HRQOL adjusting for age, sex, education, disability and clinically significant fatigue. RESULTS: Greater mastery score was associated with higher physical and mental HRQOL in both cohorts, such that a one-point increase in the PMS was associated with an increase of 2.9 (95% Confidence Interval (CI): 2.6, 3.1) and 2.8 points (95% CI: 2.4, 3.2) in the means of physical HRQOL score in the first and second cohorts respectively, and a 2.9-point (95% CI: 2.7, 3.1) and 3.1-point (95% CI: 2.7, 3.4) increase in the means of mental HRQOL score. A dose-dependent relationship was demonstrated between a quartile categorical mastery variable and both physical and mental HRQOL in both cohorts. Mastery was associated with all subscores of both physical and mental HRQOL. CONCLUSIONS: Greater mastery is associated with better physical and mental quality of life. Efforts to improve the sense of self control and agency of people with MS may have benefits for their quality of life, even despite clinical features of the illness.

RESUMO

We present the first ultrafast temporally, spectrally, and angularly resolved x-ray scattering measurements from shock-compressed matter. The experimental spectra yield the absolute elastic and inelastic scattering intensities from the measured density of free electrons. Laser-compressed lithium-hydride samples are well characterized by inelastic Compton and plasmon scattering of a K-alpha x-ray probe providing independent measurements of temperature and density. The data show excellent agreement with the total intensity and structure when using the two-species form factor and accounting for the screening of ion-ion interactions.
