RESUMO

Sewage sludge (SS) presents a high agronomic potential due to high concentrations of organic matter and nutrients, encouraging its recycling as a soil conditioner. However, the presence of toxic substances can preclude this use. To enable the safe disposal of this waste in agriculture, SS requires additional detoxification to decrease the environmental risks of this practice. Although some alternatives have been proposed in this sense, little attention is provided to eliminating endocrine-disrupting chemicals (EDCs). To fill this gap, this study aimed to develop effective and low-cost technology to eliminate EDCs from SS. For this, a detoxification process combining microorganisms and biostimulating agents (soil, sugarcane bagasse, and coffee grounds) was performed for 2, 4, and 6 months with aerobic and anaerobic SSs. The (anti-)estrogenic, (anti-)androgenic, retinoic-like, and dioxin-like activities of SSs samples were verified using yeast-based reporter-gene assays to prove the effectiveness of the treatments. A fractionation procedure of samples, dividing the target sample extract into several fractions according to their polarity, was conducted to decrease the matrix complexity and facilitate the identification of EDCs. A decrease in the abundance and microbial diversity of the SS samples was noted along the biostimulation with the predominance of filamentous fungal species over yeasts and gram-positive bacteria and non-fermenting rods over enterobacteria. Among the 9 EDCs quantified by LC-ESI-MS/MS, triclosan and alkylphenols presented the highest concentrations in both SS. Before detoxification, the studied SSs induced significant agonistic activity, especially at the human estrogen receptor α (hERα) and the human aryl hydrocarbon receptor (AhR). The raw anaerobic sludge also activated the androgen (hAR), retinoic acid (RARα), and retinoid X (RXRα) receptors. However, no significant endocrine-disrupting activities were observed after the SS detoxification, showing that the technology applied here efficiently eliminates receptor-mediated toxicity.

Background: The discovery of cryptic species complexes within morphologically established species comes with challenges in the classification and handling of these species. We hardly know to what extent species within a species complex differ ecologically. Such knowledge is essential to assess the vulnerability of individual genetic lineages in the face of global change. The abiotic conditions, i.e., the Grinnellian niche that a genetic lineage colonizes, provides insights into how diverse the ecological requirements of each evolutionary lineage are within a species complex. Material and Methods: We sampled the cryptic species complex of the amphipod Gammarus roeselii from Central Germany to Greece and identified genetic lineages based on cytochrome c oxidase subunit I (COI) barcoding. At the same time, we recorded various abiotic parameters and local pollution parameters using a series of in vitro assays to then characterize the Grinnellian niches of the morphospecies (i.e., Gammarus roeselii sensu lato) as well as each genetic lineage. Local pollution can be a significant factor explaining current and future distributions in times of increasing production and release of chemicals into surface waters. Results: We identified five spatially structured genetic lineages in our dataset that differed to varying degrees in their Grinnellian niche. In some cases, the niches were very similar despite the geographical separation of lineages, supporting the hypothesis of niche conservatism while being allopatrically separated. In other cases, we found a small niche that was clearly different from those of other genetic lineages. Conclusion: The variable niches and overlaps of different dimensions make the G. roeselii species complex a promising model system to further study ecological, phenotypic and functional differentiation within this species complex. In general, our results show that the Grinnellian niches of genetically distinct molecular operational taxonomic units (MOTUs) within a cryptic species complex can differ significantly between each other, calling for closer inspection of cryptic species in a conservational and biodiversity context.

This study describes the chemical composition and in vitro toxicity of the organic fraction of fine particulate matter (PM2.5) at an urban background site, which receives emissions either from Frankfurt international airport or the city centre, respectively. We analysed the chemical composition of filter extracts (PM2.5) using ultrahigh-performance liquid chromatography coupled to a high-resolution mass spectrometer, followed by a non-target analysis. In parallel, we applied the bulk of the filter extracts to a Microtox and acetylcholinesterase-inhibition assay for in vitro toxicity testing. We find that both the chemical composition and toxicity depend on the prevailing wind directions, and the airport operating condition, respectively. The occurrence of the airport marker compounds tricresyl phosphate and pentaerythritol esters depends on the time of the day, reflecting the night flight ban as well as an airport strike event during November 2019. We compared the organic aerosol composition and toxicity from the airport wind-sector against the city centre wind-sector. We find that urban background aerosol shows a higher baseline toxicity and acetylcholinesterase inhibition compared to rural PM2.5 that is advected over the airport. Our results indicate that the concentration and individual composition of PM2.5 influence the toxicity. Suspected drivers of the acetylcholinesterase inhibition are i.e. organophosphorus esters like triphenyl phosphate and cresyldiphenyl phosphate, and the non-ionic surfactant 4-tert-octylphenol ethoxylate. However, further research is necessary to unambiguously identify harmful organic air pollutants and their sources and quantify concentration levels at which adverse effects in humans and the environment can occur.

Plastics can release numerous chemicals and thereby, contribute to the chemical pollution in aquatic systems. To which extent environmental degradation processes influence the release of plastic chemicals, is currently unknown and subject of research. We therefore evaluated aqueous leachates of 12 differently formulated plastics (e.g., pre-production, post-industrial and recycled pellets as well as final products) using in vitro bioassays and chemical analysis via LC-HRMS nontarget approach. We weathered these plastics by UV irradiation (UV-C and UV-A/B) under laboratory conditions in dryness and a subsequent leaching period in ultrapure water ('atmospheric' weathering) or directly in water ('aquatic' weathering, UV-A/Baq). A dark control (DC) without UV light served as a reference treatment. Some plastics triggered several toxicological endpoints (low-density polyethylene recyclate (LDPE-R), starch blend (SB), bio-based polybutylene succinate (Bio-PBS) and polyvinyl chloride (PVC)), whereas others caused little to no effects (polyethylene terephthalate (PET), polystyrene (PS), polypropylene (PP) and LDPE). UV irradiation enhanced the plastics' toxicity, even for samples initially evaluated as toxicologically inconspicuous. The plastic samples caused oxidative stress (85%), baseline toxicity (42%), antiestrogenicity (40%) and antiandrogenicity (27%). Positive findings were measured after UV-C (63%) and UV-A/Baq (50%) treatments, followed by UV-A/B (48%) and DC (33%). Overall, we detected between 42 (DC) and 2896 (UV-A/Baq) chemical compounds. Our study demonstrates that differently formulated plastics leach toxic chemicals. UV exacerbates the plastics' toxicity by either generating active compounds and/or by facilitating their release. UV light even leads to the release of bioactive compounds from plastics of low chemical complexity. To prevent the exposure to plastic-associated chemicals, the application of chemicals could be reduced to a minimum, while on a regulatory level the evaluation of plastic eluates could be another focal point next to singular compounds.

In the past two decades, an increasing body of studies has been published on the intersex phenomenon in separate-sexed crustaceans from marine and freshwater ecosystems. Various causes are being considered that could have an influence on the occurrence of intersex. Besides genetic factors, environmental conditions such as photoperiodicity, temperature, salinity and parasitism, but also environmental pollution with endocrine disrupting chemicals (EDCs) are discussed. As part of a long-term monitoring (2012 - 2020) in north-west Brittany, we recorded the occurrence of intersex in the marine amphipod Echinogammarus marinus. We quantified the intersex incidence at marine and estuarine sites and analyzed the incidence in relation to the endocrine potential of the sediments. Intersex occurred with mean frequencies between 0.87% and 12%. It was striking that the incidence of intersex increased with increasing distance from the sea. Since the highest incidence was observed at the range boundary of this stenohaline species, we assume that intersex is triggered by endocrine potential and increasing stress due to increasing freshwater content - and thus an interplay of different environmental factors.

Plastics contain a complex mixture of known and unknown chemicals; some of which can be toxic. Bioplastics and plant-based materials are marketed as sustainable alternative to conventional plastics. However, little is known with regard to the chemicals they contain and the safety of these compounds. Thus, we extracted 43 everyday bio-based and/or biodegradable products as well as their precursors, covering mostly food contact materials made of nine material types, and characterized these extracts using in vitro bioassays and non-target high-resolution mass spectrometry. Two-third (67%) of the samples induced baseline toxicity, 42% oxidative stress, 23% antiandrogenicity and one sample estrogenicity. In total, we detected 41,395 chemical features with 186-20,965 features present in the individual samples. 80% of the extracts contained >1000 features, most of them unique to one sample. We tentatively identified 343 priority compounds including monomers, oligomers, plastic additives, lubricants and non-intentionally added substances. Extracts from cellulose- and starch-based materials generally triggered a strong in vitro toxicity and contained most chemical features. The toxicological and chemical signatures of polyethylene (Bio-PE), polyethylene terephthalate (Bio-PET), polybutylene adipate terephthalate (PBAT), polybutylene succinate (PBS), polylactic acid (PLA), polyhydroxyalkanoates (PHA) and bamboo-based materials varied with the respective product rather than the material. Toxicity was less prevalent and potent in raw materials than in final products. A comparison with conventional plastics indicates that bioplastics and plant-based materials are similarly toxic. This highlights the need to focus more on aspects of chemical safety when designing truly "better" plastic alternatives.

Wastewater treatment plants are major point sources of (micro)pollutant emissions and advanced wastewater treatment technologies can improve their removal capacity. While abundant data on individual advanced treatment technologies is available, there is limited knowledge regarding the removal performance of ozonation combined with multiple post-treatments and stand-alone membrane bioreactors. This is especially true for the removal of in vitro and in vivo toxicity. Therefore, we investigated the removal of 40 micropollutants and toxicity by a pilot-scale ozonation with four post-treatments: non-aerated and aerated granular activated carbon and biological filtration. In addition, two stand-alone membrane bioreactors fed with untreated wastewater and one MBR operating with ozonated partial flow recirculation were analysed. Aqueous and extracted samples were analysed in vitro for (anti)estrogenic, (anti)androgenic and mutagenic effects. To assess in vivo effects, the mudsnail Potamopyrgus antipodarum was exposed in an on-site flow-through system. Multiple in vitro effects were detected in conventionally treated wastewater including estrogenic and anti-androgenic activity. Ozonation largely removed these effects, while anti-estrogenic and mutagenic effects increased suggesting the formation of toxic transformation products. These effects were significantly reduced by granular activated carbon being more effective than biological filtration. The membrane bioreactor performed similarly to the conventional treatment while the membrane bioreactor with ozonation had a comparable removal performance like ozonation. Conventionally treated wastewater increased the growth of P. antipodarum. Ozonation reduced the reproduction indicating a potential formation of toxic transformation products. In the post-treatments, these effects were compensated or remained unaffected. The effluents of the membrane bioreactors induced reproductive toxicity. Our results show that ozonation is effective in further reducing toxicity and micropollutant concentrations. However, the formation of toxicity requires a post-treatment. Here, ozonation coupled to granular activated carbon filtration seemed the most promising treatment process.

BACKGROUND: Patients receiving dialysis are thought to have increased energy needs due to stress from the dialysis process or uraemic toxins. However, certain conditions may induce hypometabolic states potentially leading to unwanted weight gains when increased calorie intake is recommended. OBJECTIVES: Since there is conflicting research, this study aims to assess total energy expenditure (TEE) of patients receiving dialysis through analysis of resting energy expenditure (REE) and physical activity levels. DESIGN: Comprehensive review of the current literature on REE and physical activity levels. METHODS: Two electronic databases (PubMed and CINAHL) were searched using keywords to find papers published within the last 10 years for physical activity studies and within the last five years for REE. INCLUSION CRITERIA: Adults undergoing long-term dialysis treatments who do not have comorbidities that influence energy expenditure such as inflammation, hyperthyroidism or cancers. Participants were also required to ambulate without assistance. RESULTS: Only seven of the 325 studies found were included in this review. Most studies were controlled trials with one being a prospective study. Patients receiving dialysis had rest energy expenditures comparable to healthy adults. There was not a consensus between studies as to which predictive energy equation produced accurate energy recommendations. Overall, patients receiving dialysis were significantly less active compared with healthy adults and this related to muscle mass. CONCLUSION: Patients receiving dialysis tend to be sedentary or lightly active and do not have increased energy expenditure compared with healthy adults. Therefore, stable patients should not be prescribed increased calorie intake.
