RESUMO

Blood-borne extracellular vesicles (i.e., exosomes and microvesicles) carrying microRNAs (miRNAs) could make excellent biomarkers of disease and different physiologic states, including pregnancy status. We tested the hypothesis that circulating extracellular vesicle-derived miRNAs might differentiate the pregnancy status of cows that had maintained pregnancy to Day 30 from non-pregnant cows or from those that exhibited embryonic mortality between Days 17 and 30 of gestation. Cows were randomly assigned for artificial insemination with fertile semen (n = 36) or dead semen (n = 8; control group) on Day 0 (day of estrus). Blood was collected from all animals on Day 0 and on Days 17 and 24 after artificial insemination. Cows receiving live sperm were retrospectively classified as pregnant on Day 30 (n = 17) or exhibiting embryonic mortality between Days 17 and 30 (n = 19). Extracellular vesicles from Day 17 and 24 samples were isolated from serum using ultra-centrifugation, and their presence was confirmed by nanoparticle tracking and Western blot analyses (for CD81) prior to RNA extraction. MicroRNA sequencing was performed on pregnant, embryonic-mortality, and control cows (n = 4 per day), for a total of 24 independent reactions. In total, 214 miRNAs were identified in serum, 40 of which were novel. Based on differential abundance parameters, we identified 32 differentially abundant loci, representing 27 differentially abundant mature miRNA. At Days 17 and 24, specific miRNAs (e.g., miR-25, -16b, and -3596) were identified that differentiated the pregnancy status. In summary, we identified several circulating extracellular vesicles derived miRNAs that differ in abundance between embryonic mortality and pregnant cows.

Angus and Red Angus-based yearling heifers (n = 40) and lactating cows (n = 51) were each used in a complete randomized design and stratified by weight and body condition score to one of two treatments: (1) pressed supplement block containing rumen undegradable protein (RUP) and (2) pressed supplement block containing rumen degradable protein (RDP). Heifer and cow supplement intake displayed (p < 0.01) a treatment × period interaction. The RUP heifers and RDP cows consumed more in Period 2 than Period 1, whereas RDP heifers and RUP cows consumed more in Period 1 than Period 2, respectively. Intake rate demonstrated (p < 0.01) a treatment effect for heifers, with RUP consuming supplement faster than the RDP treatment. Intake rate for cows demonstrated (p < 0.01) a treatment × period interaction with RUP cows in Period 1 having faster intakes than Period 2, and RDP cows having the inverse. Cow intake variation displayed (p < 0.01) a treatment × period interaction with RUP cows having more variation in Period 2, while RDP cows had less variation in intake in Period 2. In conclusion, RDP and RUP impacted intake behavior of cows and heifers but had minimal impacts on performance.

Relationships and sexuality education (RSE) was introduced in Irish schools in 1995 to address pressures on young people specific to relationships and sexuality. RSE is part of a whole school health promotion project. Emphasis is put on personal and social development of students, cross-curricular dimensions, school ethos, school climate and partnership with families and community. However, RSE has become the preserve of female teachers with fewer male teachers attending in-service. This research explored the attitudes, beliefs and needs of male post-primary teachers with regard to RSE in their school. A qualitative design was employed utilizing focus groups. A total of 25 male post-primary teachers took part in five focus groups. Two of the groups consisted of men who had participated in RSE training; the other three groups were with men who had not. The analysis of the data suggested that there was reluctance on the part of male teachers to teach RSE, and that they feel under threat about their personal and professional identity. They felt under threat both from internal personal forces, in the form of mental models assimilated through personal history and upbringing, and from external organisational forces, communicated through the local school and wider social culture. The data indicate the need for professional development and support that is tailored specifically for male teachers. In order to support male teachers to become more involved in school health promotion and in the teaching of health curricula, pre-service education needs to prioritize key issues such as gender roles and identity.

Understanding the contribution of the whole-school staff to health education (HE) is an important goal in HE research. This study aimed to identify the views of staff (principals; teachers; school nurses and doctors; counsellors and administrative, maintenance, canteen and cleaning staff) regarding the nature of their contribution to HE. The research is based on 207 semi-structured interviews of staff in a sample of five French middle schools (grade 6-9). Content analysis was performed using Bardin's method. The results showed that staff members have different views of their role, three main roles were identified: (i) as an educator in everyday life issues (72%); (ii) individual support, listening (14%) and (iii) taking part in collective projects and facilitation (14%). Professional status has a significant influence on the view they have of their contribution to HE. These results show that in order to facilitate consistent implementation of HE, schools need to be supported to build HE policy (need analysis, definition of priorities and partnerships) and also to develop the means by which an inclusive and real sharing of common culture among all staff can happen; this is not limited to teaching staff but includes non-teaching staff also.

The effects of medroxyprogesterone acetate (MPA) on velvet antler cleaning, hypophagia, aggressive behavior and fertility were evaluated in farmed reindeer bulls during a 2-year study. Eight reindeer bulls aged 1-4 years were divided into 2 groups balanced for age. During each year, one group (MPA, n = 4) was treated with MPA 2 wk prior to the expected onset of rut while the other group (CTL, n = 4) served as untreated controls. Feed consumption, behavior and antler cleaning were recorded daily or 3 x weekly for 3 mo. Each year a dominant CTL and MPA bull were put into separate harems of estrous synchronized females for 1 wk in mid-September. Following harem breakup, semen was collected from all bulls via electroejaculation and evaluated. In Year 2 the bulls were switched such that year 1 MPA bulls received the CTL treatment and year 1 CTL bulls received MPA treatment. In Year 2 all eight bulls received MPA treatment/booster following semen collection. In both years, MPA treatment reduced rut associated body weight loss (p ≤ 0.05), rut associated hypophagia (p ≤ 0.001), interfered with velvet antler cleaning, and abolished aggressive rut related behavior. All of these changes suggest suppression of testosterone mediated effects. Alternatively, semen parameters differed little between treatment groups with the exception of reduced sperm concentration and total sperm production in MPA bulls (p ≤ 0.05). All CTL bulls in Year 2 exhibited full rut behavior with the dominant bull successfully breeding 100% of females available for breeding, suggesting no carryover effect of MPA treatment from the previous year. The MPA bull successfully bred 4 of 6 females (Year 1) but the MPA bull in Year 2 failed to sire any offspring. A single 400 mg treatment of MPA just prior to rut was sufficient to suppress rut associated aggression and hypophagia on a short-term (3 mo) basis. It did not however, completely suppress fertility.
