RESUMO

The nutritive value of grass silage can be improved by harvesting herbage at a less mature growth stage, which in practice usually involves more frequent harvests. This study examined the performance of dairy cows offered grass silages produced from perennial ryegrass (Lolium perenne) based swards harvested at 2 different frequencies during the growing season (3-harvest (3H) vs. 5-harvest (5H)). Thirty-four mid-lactation (av. 147 d in milk) dairy cows (30 multiparous, 4 primiparous) were offered either 3H or 5H silages in a continuous design (21 wk) experiment. Within each treatment cows were offered silage from each harvest (in harvest number order) for a pre-determined number of days in proportion to herbage DM yield at each harvest. Silages were offered ad libitum while a common concentrate was offered to all cows at 12.0 kg per cow/d over the first 15 wk of the study, 8.0 kg per cow/d during wk 16 -19 and 6.0 kg cow/d during wk 20 - 21. Total yield of herbage harvested over the season from within 3H and 5H were 12.6 and 11.2 t DM/ha, respectively. Across all harvests the mean ME and CP concentration of silages were 10.9 MJ/kg DM and 131 g/kg DM for 3H, and 11.5 MJ/kg DM and 152 g/kg DM for 5H. Silage DMI was greater for cows offered 5H silages compared with 3H silages (14.1 vs. 11.7 kg/d, respectively). Cows offered 5H silages had a greater daily milk yield (33.5 vs. 31.9 kg) and ECM yield (37.4 vs. 35.6 kg) compared with cows offered 3H silages. Treatment had no effect on milk fat or protein concentration. Cows offered 5H silages produced milk with greater concentrations of CLA and n-3 fatty acids. Treatment had no effect on mean BW or BCS, or on efficiency metrics such as milk yield or ECM yield per kg of DMI. Molar proportions of VFA in ruminal fluid differed between the treatments, with cows offered 3H silages having higher proportion of total butyrate (15.9 vs. 14.4% of total VFA) and lower total valerate (3.2 vs. 3.7% of total VFA) compared with those offered 5H. The acetate: propionate and acetate plus butyrate: propionate ratios were unaffected by treatment. In conclusion, increasing herbage harvesting frequency from 3 to 5 times per year improved the nutritional value of the resulting silages, and this led to higher silage DMI, milk yield and ECM yield. However, overall production efficiency (ECM/DMI) was unaffected by treatment.

Bovine tuberculosis (bTB) is a chronic, infectious and zoonotic disease of domestic and wild animals caused mainly by Mycobacterium bovis. This study investigated farm management factors associated with recurrent bTB herd breakdowns (n = 2935) disclosed in the period 23 May 2016 to 21 May 2018 and is a follow-up to our 2020 paper which looked at long duration bTB herd breakdowns. A case control study design was used to construct an explanatory set of farm-level management factors associated with recurrent bTB herd breakdowns. In Northern Ireland, a Department of Agriculture Environment and Rural Affairs (DAERA) Veterinarian investigates bTB herd breakdowns using standardised guidelines to allocate a disease source. In this study, source was strongly linked to carryover of infection, suggesting that the diagnostic tests had failed to clear herd infection during the breakdown period. Other results from this study associated with recurrent bTB herd breakdowns were herd size and type (dairy herds 43% of cases), with both these variables intrinsically linked. Other associated risk factors were time of application of slurry, badger access to silage clamps, badger setts in the locality, cattle grazing silage fields immediately post-harvest, number of parcels of land the farmer associated with bTB, number of land parcels used for grazing and region of the country.

A diverse range of grassland-based milk production systems are practiced on dairy farms in temperate regions, with systems differing in relation to the proportion of grazed grass, conserved forages and concentrates in diet, calving season, duration of housing, cow genotype, and performance levels. The current study was conducted to examine performance within diverse grassland-based systems of milk production under experimental conditions. This study examined 4 milk production systems over 3 successive lactations (20 cows per system during each lactation). With winter calving-fully housed (WC-FH), Holstein cows were housed for the entire lactation and offered a complete diet consisting of grass silage, maize silage, and concentrates [approximately 50% forage on a dry matter (DM) basis]. With winter calving-conventional (WC-Con), Holstein cows were housed and offered the same diet from calving until turnout (late March) as offered with WC-FH, and thereafter cows were given access to grazing and supplemented with 5.0 kg of concentrate/cow daily. Two spring-calving systems were examined, the former involving Holstein cows (SC-H) and the latter Jersey × Holstein crossbred cows (SC-J×H). Cows on these systems were offered a grass silage-concentrate mix (70% forage on a DM basis) until turnout (late February), and thereafter cows were given access to grazing supplemented with 1.0 kg of concentrate/cow per day. The contributions of concentrates (3,080, 2,175, 722, and 760 kg of DM/cow per lactation), conserved forages (3,199, 1,556, 1,053, and 1,066 kg of DM/cow per lactation), and grazed grass (0, 2,041, 2,788, and 2,692 kg of DM/cow per lactation) to total DMI (6,362, 5,763, 4,563, and 4,473 kg of DM/cow per lactation) with WC-FH, WC-Con, SC-H, and SC-J×H, respectively, varied considerably. Similarly, milk yield (9,333, 8,443, 6,464, and 6,049 kg/cow per lactation), milk fat content (44.9, 43.3, 42.8, and 49.0 g/kg), and milk protein content (34.6, 34.9, 33.6, and 36.3 g/kg) differed between systems (WC-FH, WC-Con, SC-H, and SC-J×H, respectively). The higher milk yields with the WC systems reflect the greater concentrate inputs with these systems, whereas the greater milk fat and protein content with SC-J×H reflect the use of Jersey crossbred cows. Crossbred cows on SC-J×H produced a similar yield of milk solids as Holstein cows on SC-H. Cows on WC-FH ended the lactation with a greater body weight (BW) and body condition score than cows on any other treatment. While Jersey crossbred cows on SC-J×H had a lower BW than Holstein cows on SC-H, cows on these 2 systems were not different for any of the other BW, body condition score, or blood metabolite parameters examined. Cows on WC-FH had a greater interval from calving to conception, a greater mastitis incidence, and a greater locomotion score than cows on the spring calving systems. Whole-system stocking rates and annual milk outputs were calculated as 2.99, 2.62, 2.48, and 2.50 cows/ha, and 25,706, 20,822, 15,289, and 14,564 kg of milk/ha, with each of WC-FH, WC-Con, SC-H, and SC-J×H, respectively. Gross margin per cow was highest with WC-Con, gross margin per hectare was highest with WC-FH, and gross margin per kilogram of milk was highest with SC-J×H. This study demonstrated that diverse grassland-based milk production systems are associated with very different levels of performance when examined per cow and per hectare.

Thirty-six ewes aged 18 months were assigned to one of three groups (n = 12) on the basis of faecal egg count (FEC) for rumen fluke; C: control (FEC = 0-5 epg), T: affected (FEC ≥ 6 epg) and treated, NT: affected (FEC ≥ 6 epg) and not treated. T ewes were treated with oxyclozanide 14 days prior to the trial commencing. Ewes were fed grass silage ad libitum. Digestibility crates and respiration chambers were used to measure feed digestibility and methane production. Dry matter intake (DMI), feed digestibility and CH4/DMI did not differ (P > 0.05) between treatments. However, CH4/live weight (LW) was significantly greater (P < 0.001) for NT ewes, while that of C and T ewes were similar. This study indicates that a high rumen fluke burden can increase methane emissions but only when expressed on a body weight basis (per kg LW). As the link between rumen fluke infection and methane production has not previously been investigated within the literature, these findings demonstrate the need for further research to better understand these relationships.

The role of the Eurasian badger (Meles meles) as a wildlife host has complicated the management of bovine tuberculosis (bTB) in cattle. Badger ranging behaviour has previously been found to be altered by culling of badgers and has been suggested to increase the transmission of bTB either among badgers or between badgers and cattle. In 2014, a five-year bTB intervention research project in a 100 km2 area in Northern Ireland was initiated involving selective removal of dual path platform (DPP) VetTB (immunoassay) test positive badgers and vaccination followed by release of DPP test negative badgers ('Test and Vaccinate or Remove'). Home range sizes, based on position data obtained from global positioning system collared badgers, were compared between the first year of the project, where no DPP test positive badgers were removed, and follow-up years 2-4 when DPP test positive badgers were removed. A total of 105 individual badgers were followed over 21 200 collar tracking nights. Using multivariable analyses, neither annual nor monthly home ranges differed significantly in size between years, suggesting they were not significantly altered by the bTB intervention that was applied in the study area.

This study was designed to contribute to the understanding of the relationships between energy balance (EB) in early lactation [4 to 21 d in milk (DIM)] and fertility traits [interval to start of luteal activity (SLA), interval to first observed heat (FOH), and conception to first artificial insemination (AI)], and their associated relationships with cow performance and blood metabolites between 4 to 150 DIM. Individual cow data (488 primiparous and 1,020 multiparous lactations) from 27 experiments was analyzed. Data on cow performance, EB (on a metabolizable energy basis), and fertility traits were available for all cows, whereas milk progesterone data (to determine SLA) and periodic blood metabolite data were available for 1,042 and 1,055 lactations, respectively. Data from primiparous and multiparous cows were analyzed separately, with the data sets for the 2 parity groups divided into quartiles (Q1-Q4) according to the average EB during 4 to 21 DIM (EB range for Q1 to Q4: primiparous, -120 to -49, -49 to -24, -24 to -3, and -3 to 92 MJ/d, respectively: multiparous, -191 to -79, -79 to -48, -48 to -22, and -22 to 93 MJ/d, respectively). Differences between EB quartiles for production and fertility traits were compared. In early lactation (4 to 21 DIM), moving from Q1 to Q4 mean DMI and metabolizable energy intake increased whereas mean ECM decreased. During the same period, moving from Q1 to Q4 milk fat content, milk fat-to-protein ratio, and plasma nonesterified fatty acid and ß-hydroxybutyrate concentrations decreased, whereas milk protein content and plasma glucose concentrations increased in both primiparous and multiparous cows. When examined over the entire experimental period (4 to 150 DIM), many of the trends in intakes and milk production remained, although the magnitude of the difference between quartiles was much reduced, whereas milk fat content did not differ between quartiles in primiparous cows. The percentage of cows with FOH before 42 DIM increased from Q1 to Q4 (from 46 to 72% in primiparous cows, and from 41 to 58% in multiparous cows). Interval from calving to SLA and to FOH decreased with increasing EB during 4 to 21 DIM, with these occurring 9.8 and 10.2 d earlier, respectively, in Q4 compared with Q1 (primiparous cows), and 7.4 and 5.9 d earlier, respectively, in Q4 compared with Q1 (multiparous cows). For each 10 MJ/d decrease in mean EB during 4 to 21 DIM, FOH was delayed by 1.2 and 0.8 d in primiparous and multiparous cows, respectively. However, neither days to first AI nor the percentage of cows that conceived to first AI were affected by daily EB during 4 to 21 DIM in either primiparous or multiparous cows, and this is likely to reflect a return to a less metabolically stressed status at the time of AI. These results demonstrate that interval from calving to SLA and to FOH were reduced with increasing EB in early lactation, whereas early lactation EB had no effect on conception to the first service.

This study determined farm management factors associated with long-duration bovine tuberculosis (bTB) breakdowns disclosed in the period 23 May 2016 to 21 May 2018; a study area not previously subject to investigation in Northern Ireland. A farm-level epidemiological investigation (n = 2935) was completed when one or more Single Intradermal Comparative Cervical Test (SICCT) reactors or when one or more confirmed (positive histological and/or bacteriological result) lesion at routine slaughter were disclosed. A case-control study design was used to construct an explanatory set of management factors associated with long-duration bTB herd breakdowns; with a case (n = 191) defined as an investigation into a breakdown of 365 days or longer. Purchase of infected animal(s) had the strongest association as the most likely source of infection for long-duration bTB herd breakdowns followed by badgers and then cattle-to-cattle contiguous herd spread. However, 73.5% (95% CI 61.1-85.9%) of the herd type contributing to the purchase of infection source were defined as beef fattening herds. This result demonstrates two subpopulations of prolonged bTB breakdowns, the first being beef fattening herds with main source continuous purchase of infected animals and a second group of primary production herds (dairy, beef cows and mixed) with risk from multiple sources.

The European livestock sector has a significant deficit of high-quality protein feed ingredients. Consequently there is interest in using locally grown protein grain crops to partially or completely replace imported protein feeds in dairy cow rations. Field bean (FB; Vicia faba) has been identified as a locally grown crop with significant potential. The current study was designed to examine the effects of FB on cow performance and nutrient utilization in the diet of early-lactation dairy cows, including high levels of FB (up to 8.4 kg/cow per day). The experiment used 72 dairy cows in a 3-treatment continuous design (from calving until wk 20 of lactation). All cows were given ad libitum access to a mixed ration comprising grass silage and concentrates [45:55 on a dry matter (DM) basis]. Concentrates offered contained either 0, 349, or 698 g of FB/kg of concentrate (treatments FB0, FB-Low, and FB-High, respectively), with FB completely replacing soybean meal, rapeseed meal, maize gluten, and wheat in the concentrate for the FB-High treatment. Following completion of the 20-wk experiment, ration digestibility, nutrient utilization, and methane (CH4) production were measured using 4 cows from each treatment. Neither silage DM intake, total DM intake, nor milk yield were affected by treatment. Cows on FB0 had a higher milk fat content than those on FB-High, and cows on FB0 and FB-Low had higher milk protein contents than did those on FB-High. Field bean inclusion increased the degree of saturation of milk fat produced. Milk fat yield, milk protein yield, and milk fat plus protein yield were higher with FB0 than with either FB-Low or FB-High. Treatment had no effect on the digestibility of DM, organic matter, nitrogen (N), gross energy, or neutral detergent fiber, whereas digestibility of acid detergent fiber was higher with FB0 than with FB-High. Neither the efficiency of gross energy or N utilization, nor any of the CH4 production parameters examined, were affected by treatment. Similarly, none of the fertility or health parameters examined were affected by treatment. The reduction in milk fat observed may have been due to the higher starch content of the FB-High diet, and the reduction in milk protein may have been due to a deficit of methionine in the diet. It is likely that these issues could be overcome by changes in ration formulation, thus allowing FB to be included at the higher range without loss in performance.

Biting midges, Culicoides spp. (Diptera: Ceratopogonidae), are important vectors of viral pathogens. Following the outbreak of bluetongue serotype 8 in Europe between 2006 and 2009, many Culicoides surveillance programmes were initiated to identify vector-active periods, in accordance with European Commission regulation 2007/1266/EC. This study utilized surveillance data from 4 years of continuous light-trapping at 14 sites in Northern Ireland. The number of captured Culicoides varied from none during the vector-free period (December-April) to more than 36 000 per night during peak activity in the summer. The Obsoletus group represented 75% of Culicoides collected and the Pulicaris group represented 21%. A total of 91% of Culicoides were female, of which 42% were parous. Abundance data, sex ratios and parous rates suggested that both the Obsoletus and Pulicaris groups underwent three generations/year. The Obsoletus group was associated with cattle-rearing habitats and woodland, the Impunctatus group was found in habitats related to sheep rearing and the Pulicaris group were associated with both cattle and sheep. Housing did not reduce incursion of female Obsoletus group Culicoides but it did for males and for the Pulicaris group Culicoides. The influence of housing was strongly affected by time of year, probably reflecting the presence of livestock indoors/outdoors.

This 2 × 2 factorial design experiment was conducted to compare the performance of spring-calving Holstein dairy cows (HOL, n = 34) with Swedish Red × Jersey/Holstein crossbred (SR × J/HOL, n = 34) dairy cows within low and medium concentrate input grassland-based dairy systems. The experiment commenced when cows calved and encompassed 1 full lactation. Cows were offered diets containing grass silage and concentrates [70:30 dry matter (DM) ratio, and 40:60 DM ratio, for low and medium, respectively] until turnout, grazed grass plus either 1.0 or 4.0 kg of concentrate/d during the grazing period (low and medium, respectively), and grass silage and concentrates (85:15 DM ratio, and 70:30 DM ratio, for low and medium, respectively) from rehousing and until drying off. No significant genotype × system interactions were present for any of the feed intake or full-lactation milk production data examined. Full-lactation concentrate DM intakes were 769 and 1,902 kg/cow for the low and medium systems, respectively, whereas HOL cows had a higher total DM intake than SR × J/HOL cows in early lactation, but not in late lactation. Although HOL cows had a higher lactation milk yield than SR × J/HOL cows, the latter produced milk with a higher fat and protein content, and thus fat plus protein yield was unaffected by genotype. Milk produced by the SR × J/HOL cows had a higher degree of saturation of fatty acids than milk produced by the HOL cows, and the somatic cell score of milk produced by the former was also higher. Throughout the lactation, HOL cows were on average 30 kg heavier than SR × J/HOL cows, whereas the SR × J/HOL cows had a higher body condition score than the HOL cows. Holstein cows had a higher incidence of mastitis and ovarian dysfunction that SR × J/HOL cows.

Post-mortem examination continues to play an important surveillance role in the bovine tuberculosis (bTB) eradication programme in Northern Ireland. It is estimated that 18-28% of new bTB herd breakdowns are disclosed by the detection of bTB lesions in animals routinely slaughtered. The purpose of this study was to compare the performance of different slaughterhouses in Northern Ireland in detecting bTB-lesioned animals at routine slaughter (LRS) and to apply the findings to maximize the sensitivity of bTB slaughterhouse surveillance. Univariate statistical analysis on cattle slaughtered in Northern Ireland during 2011-2013 revealed that the risk of LRS disclosure varied between slaughterhouses, ranging from 0·08% to 0·54%. Furthermore, the risk of confirmation of these LRS as bTB varied between slaughterhouses, ranging from 57·9% to 72·4%. Logistic regression modelling of selected risk factors found that the risk of LRS disclosure increased with age, and was higher in purchased animals, during winter months, in animals coming from high bTB incidence areas and in animals slaughtered from herds with a bTB restriction in the last 2-3 years. Adjusting for these selected factors, the risk of LRS disclosure and bTB confirmation changed very little from the univariable analysis, suggesting that differences in disclosure risks between slaughterhouses were likely to be due to factors related to the slaughterhouses, rather than to the risk status of the animals presented. Examination of procedures within these slaughterhouses is recommended to identify ways that could increase the sensitivity of their bTB surveillance.

Determination of the proportion of bovine tuberculosis (bTB) breakdowns attributed to a herd purchasing infected animals has not been previously quantified using data from the Animal and Public Health Information System (APHIS) database in Northern Ireland. We used a case-control study design to account for the infection process occurring in the disclosing bTB breakdown herds. Cases (N = 6926) were cattle moving to a future confirmed bTB breakdown where they would disclose as a confirmed bTB reactor or a Lesion at Routine Slaughter (LRS). Controls (N = 303 499) were cattle moving to a future confirmed bTB breakdown where they did not become a bTB reactor or LRS. Our study showed that the cattle leaving herds which disclosed bTB within 450 days had an increased odds of becoming a confirmed bTB reactor or LRS compared with the cattle which left herds that remained free for 450 days (odds ratio (OR) = 2·09: 95% CI 1·96-2·22). Of the 12 060 confirmed bTB breakdowns included in our study (2007-2015 inclusive), 31% (95% CI 29·8-31·5) contained a confirmed bTB reactor(s) or LRS(s) at the disclosing test which entered the herd within the previous 450 days. After controlling for the infection process occurring in the disclosing bTB breakdown herd, our study showed that 6·4% (95% CI 5·9-6·8) of bTB breakdowns in Northern Ireland were directly attributable to the movement of infected animals.

RESUMO

Improving body condition score of thin cows in late lactation is necessary, because cows that are thin at drying off exhibit decreased fertility postpartum and are at increased risk of disease and of being culled in the subsequent lactation. Offering a diet low in crude protein (CP) content in late lactation may help to improve body condition score (BCS) at drying off, whereas imposing an extended dry period (EDP) has been advocated as another way to increase BCS at calving. To test these hypotheses, 65 thin cows (mean BCS 2.25 at 14 wk precalving) were managed on 1 of 3 treatments between 13 and 9 wk prepartum: normal protein control {NP; grass silage + 5 kg/d of a normal protein concentrate [228 g of CP/kg of dry matter (DM)]}, low protein [LP; grass silage + 5 kg/d of a low-protein concentrate (153 g of CP/kg of DM)], or EDP (cows dried off at 13 wk precalving and offered a grass silage-only diet). Both NP and LP cows were dried off at wk 8 prepartum, after which all cows were offered a grass silage-only diet until calving. After calving, all cows were offered a common diet (supplying 11.1 kg of concentrate DM/cow per day) for 19 wk. Between 13 and 9 wk prepartum, LP cows had lower DM intake, milk yield, and body weight than NP cows. Whereas EDP cows had lower serum ß-hydroxybutyrate and fatty acid concentrations than those of NP cows, BCS at wk 9 prepartum did not differ between treatments. Cows on the LP treatment continued to have lower DMI and BW than those of NP and EDP cows between 8 wk prepartum and calving, but only EDP cows had a higher BCS at calving. Treatment did not affect calving difficulty score or calf birth weight. Although all cows were offered a common diet postpartum, cows on the LP treatment had lower DM intake and milk fat + plus protein yield than cows on any other treatment during the 19-wk period postpartum, but we found no differences in any postpartum indicator of body tissue reserves. The treatments imposed from wk 13 to 9 prepartum had no effect on any fertility or health parameters examined postpartum. Extending the dry period for thin cows improved their BCS at calving but did not allow these cows to achieve the target BCS of 2.75, and we found no beneficial effects of this treatment on cow performance postpartum. Offering a lower-protein diet to thin cows in late lactation did not improve BCS at calving above that of cows on a normal protein diet, but had unexplained long-term negative effects on cow performance.

Biting midges (Culicoides spp.) are vectors of bluetongue and Schmallenberg viruses. Treatment of mesh barriers is a common method for preventing insect-vectored diseases and has been proposed as a means of limiting Culicoides ingression into buildings or livestock transporters. Assessments using animals are costly, logistically difficult and subject to ethical approval. Therefore, initial screening of test repellents/insecticides was made by applying treatments to mesh (2 mm) cages surrounding Onderstepoort light traps. Five commercial treatments were applied to cages as per manufacturers' application rates: control (water), bendiocarb, DEET/p-menthane-3,8-diol (PMD) repellent, Flygo (a terpenoid based repellent) and lambda-cyhalothrin. The experimental design was a 5 × 5 Latin square, replicated in time and repeated twice. Incongruously, the traps surrounded by DEET/PMD repellent-treated mesh caught three to four times more Obsoletus group Culicoides (the commonest midge group) than the other treatments. A proposed hypothesis is that Obsoletus group Culicoides are showing a dose response to DEET/PMD, being attracted at low concentrations and repelled at higher concentrations but that the strong light attraction from the Onderstepoort trap was sufficient to overcome close-range repellence. This study does not imply that DEET/PMD is an ineffective repellent for Culicoides midges in the presence of an animal but rather that caution should be applied to the interpretation of light trap bioassays.

The objective of the current study was to determine the effects of concentrate feeding method on milk yield and composition, dry matter (DM) intake (DMI), body weight and body condition score, reproductive performance, energy balance, and blood metabolites of housed (i.e., accommodated indoors) dairy cows in early to mid lactation. Eighty-eight multiparous Holstein-Friesian cows were managed on 1 of 4 concentrate feeding methods (CFM; 22 cows per CFM) for the first 21 wk postpartum. Cows on all 4 CFM were offered grass silage plus maize silage (in a 70:30 ratio on a DM basis) ad libitum throughout the study. In addition, cows had a target concentrate allocation of 11 kg/cow per day (from d 13 postpartum) via 1 of 4 CFM, consisting of (1) offered on a flat-rate basis via an out-of-parlor feeding system, (2) offered based on individual cow's milk yields in early lactation via an out-of-parlor feeding system, (3) offered as part of a partial mixed ration (target intake of 5 kg/cow per day) with additional concentrate offered based on individual cow's milk yields in early lactation via an out-of-parlor feeding system, and (4) offered as part of a partial mixed ration containing a fixed quantity of concentrate for each cow in the group. In addition, all cows were offered 1 kg/cow per day of concentrate pellets via an in-parlor feeding system. We detected no effect of CFM on concentrate or total DMI, mean daily milk yield, concentrations and yields of milk fat and protein, or metabolizable energy intakes, requirements, or balances throughout the study. We also found no effects of CFM on mean or final body weight, mean or final body condition score, conception rates to first service, or any of the blood metabolites examined. The results of this study suggest that CFM has little effect on the overall performance of higher-yielding dairy cows in early to mid lactation when offered diets based on conserved forages.

Sticky traps were mounted on heifers and sheep to assess Culicoides spp. (Diptera: Ceratopogonidae) host preference. Initially, four coloured 200-cm(2) sticky traps (white, clear, yellow and blue) were attached to the backs of each of ten Friesian heifers that were released into open pasture for 24 h, repeated on six occasions. More Obsoletus group Culicoides were caught on the white and clear traps than on the yellow and blue. Trap position on the right or left flank also affected midge catch, probably due to heifer orientation in the field. Next, six Friesian heifers and six Charollais hoggets each had one clear and one white sticky strap attached to their backs for one 24-h period per week, repeated for 24 weeks. Overall, Obsoletus group Culicoides comprised 91.8% (n = 5, 955) of the midge catch but there was no evidence of host preference, either discounting or including host live weight in the analyses. However, Pulicaris group Culicoides did demonstrate a significant host preference for sheep, providing that the analysis was adjusted for live weight. On heifers, the Pulicaris group comprised 7.5% of biting midges caught, whereas on hoggets, it comprised 12.7%.

Although the effect of nutrition on enteric methane (CH4) emissions from confined dairy cattle has been extensively examined, less information is available on factors influencing CH4 emissions from grazing dairy cattle. In the present experiment, 40 Holstein-Friesian dairy cows (12 primiparous and 28 multiparous) were used to examine the effect of concentrate feed level (2.0, 4.0, 6.0, and 8.0 kg/cow per day; fresh basis) on enteric CH4 emissions from cows grazing perennial ryegrass-based swards (10 cows per treatment). Methane emissions were measured on 4 occasions during the grazing period (one 4-d measurement period and three 5-d measurement periods) using the sulfur hexafluoride technique. Milk yield, liveweight, and milk composition for each cow was recorded daily during each CH4 measurement period, whereas daily herbage dry matter intake (DMI) was estimated for each cow from performance data, using the back-calculation approach. Total DMI, milk yield, and energy-corrected milk (ECM) yield increased with increasing concentrate feed level. Within each of the 4 measurement periods, daily CH4 production (g/d) was unaffected by concentrate level, whereas CH4/DMI decreased with increasing concentrate feed level in period 4, and CH4/ECM yield decreased with increasing concentrate feed level in periods 2 and 4. When emissions data were combined across all 4 measurement periods, concentrate feed level (2.0, 4.0, 6.0, and 8.0 kg/d; fresh basis) had no effect on daily CH4 emissions (287, 273, 272, and 277 g/d, respectively), whereas CH4/DMI (20.0, 19.3, 17.7, and 18.1g/kg, respectively) and CH4-E/gross energy intake (0.059, 0.057, 0.053, and 0.054, respectively) decreased with increasing concentrate feed levels. A range of prediction equations for CH4 emissions were developed using liveweight, DMI, ECM yield, and energy intake, with the strongest relationship found between ECM yield and CH4/ECM yield (coefficient of determination = 0.50). These results demonstrate that offering concentrates to grazing dairy cows increased milk production per cow and decreased CH4 emissions per unit of milk produced.

The development and the application of a quantitative duplex real-time PCR for the detection of Neoparamoeba perurans and the elongation factor α 1 gene (ELF) of Atlantic salmon, Salmo salar L., and rainbow trout, Oncorhynchus mykiss (Walbaum), are described. A set of primers and probe was designed to amplify a 139-bp fragment specific to the N. perurans 18S rRNA gene. The test was shown to be very sensitive, being able to detect as little as 13.4 DNA copies per µL corresponding to 0.15 fg of template DNA. In addition, the reaction that detected N. perurans was found to have a high degree of repeatability and reproducibility, to have a linear dynamic range (R(2 ) = 0.999) extending over 5 log(10) dilutions and to have a high efficiency (104%). The assay was applied to DNA samples extracted from 48 formalin-fixed, paraffin-embedded (FFPE) salmon gill tissues showing varying degrees of gill histopathology and amoebic gill disease (AGD)-type histopathology ranging from absent to severe (each scored 0-3). Neoparamoeba perurans DNA was detected in all the blocks where AGD-type histopathology was diagnosed microscopically and in 43.6% of the blocks showing signs of gill pathology. The association between parasitic load and gill histopathology and AGD-type histopathology severity was also investigated. This study also describes the development and the application of a second real-time PCR for the generic detection of Neoparamoeba spp., Page, 1987. A set of primers and probe conserved among the Neoparamoeba spp. was designed to amplify a 150-bp fragment within the 18S rRNA gene. Applied to N. perurans-negative gill tissues, the method was used to exclude the presence of other Neoparamoeba spp. in those blocks where gill pathology was observed microscopically.

The objectives of the present study were to compare the sulfur hexafluoride (SF6) and respiration chamber techniques for measuring methane (CH4) emissions from dairy cows and to determine the proportion of CH4 that is released through the rectum. Data used were derived from 20 early lactation dairy cows in a 2 × 2 factorial design study for 4 periods with 6 wk/period. The 4 treatment diets consisted of grass silage and 2 levels of concentrate (30 and 60% dry matter basis), with or without yeast supplement. At the end of each period, CH4 emissions were measured simultaneously using the SF6 and respiration chamber techniques when cows were housed in chambers. The SF6 technique was also used when cows were housed in digestibility units (barn location) before and after respiratory chamber measurements (chamber location). The simultaneous measurements in chamber location revealed that CH4 emission estimates by the SF6 technique were similar to those by the respiration chamber technique in the first 3 periods, although the SF6 estimates were significantly higher in period 4. The regression of all data from the 4 periods demonstrated a linear relationship between the SF6 and respiration chamber measurements for total CH4 emissions (g/d, R² = 0.69) and for CH4 emissions per unit of milk yield (g/kg, R² = 0.88), and a quadratic relationship for CH4 emissions per unit of dry matter intake (g/kg, R² = 0.64). The CH4 emissions from the rectum were calculated as the difference between CH4 estimates from the SF6 technique when cows were housed in respiratory chambers and barn locations, which was 3% of the total CH4 emissions from the mouth, nostrils, and rectum. The SF6 estimates in the chamber location accounted for all sources of emissions, whereas those in the barn location, like that in grazing conditions, did not include CH4 emission from the rectum. Therefore, the SF6 measurements for grazing cattle should be adjusted for CH4 emissions from the rectum (3% of total). We conclude that the SF6 technique is reasonably accurate for estimating CH4 emissions.

The development and preliminary evaluations of two TaqMan®-based, real-time reverse transcriptase-polymerase chain reaction (rRT-PCR) assays for the quantitative detection of avian nephritis virus (ANV) and chicken astrovirus (CAstV) RNAs are described. The assays used amplicons generated from the 3' untranslated region of the ANV genome and a conserved region of CAstV open reading frame 1b including its junction with open reading frame 2. High virus RNA levels (>10(5.99) viral copies) were detected for ANV and CAstV in 81% and 67% gut content samples from growth-retarded broiler flocks. Results from longitudinal surveys of two broiler flocks showed that ANV and CAstV RNAs were detected in most gut content and kidney samples collected at all time points from day 0 to day 35, with RNA levels of both astroviruses being higher in the gut contents than in the kidneys, and with the ANV RNA levels being greater than those of CAstV especially at early (days 7 and 14) time points. When the results obtained for the days 4/5 time-point samples from four broiler flocks with varying growth performances were compared, the two better-performing flocks had 100-fold to 1000-fold less ANV viral copies than the flocks that performed least well. Application of the rRT-PCR tests to samples collected from broiler chicks, which were experimentally infected with a crude gut content inoculum, demonstrated that ANV RNA could be detected in gut content and kidney samples at levels similar to those found at corresponding time points in longitudinal survey samples, whereas CAstV RNA was detected at lower levels than in the longitudinal survey samples, especially in kidney samples.
