Benthic cyanobacterial assemblages from ponds distributed along inland-coastal gradients in the McMurdo Sound region were studied during the 2011/12 Antarctic summer season. Twenty-five ponds were sampled in four distinct geographic locations, including the Lower and Upper Wright Valleys, Ross Island and the McMurdo Ice Shelf. For morphological identification, benthic mat samples were thawed and a subsample was directly observed by light microscopy. Remaining sample material was stored in 50 ml sterile polycarbonate bottles containing the mineral nutrient medium MLA for future studies, maintained at a temperature of 21°C. Ten morphological criteria were used to describe the morphotypes (trichome shape, number of trichomes in sheath, presence or absence of terminal attenuation of trichome, calyptra on mature apical cell, shape of apical cell, presence or absence of constrictions at transverse walls, granules, branching, range in width of trichomes and range of cell length) with reference to available identification literature. All morphospecies were documented using photomicrography. In total, 29 morphospecies were described, four assigned to the order Chroococcales, three to Nostocales and 22 to Oscillatoriales. The four geographic locations had similar taxonomic richness, sharing many morphospecies. However, each also contained distinct floristic elements that were rare or absent from the others.

Most marine biotoxins are produced by microalgae. The neurotoxin tetrodotoxin (TTX) has been reported in many seafood species worldwide but its source is unknown, making accumulation and depuration studies in shellfish difficult. Tetrodotoxin is a water-soluble toxin and cannot be directly ingested by shellfish. In the present study, a method was developed which involved binding TTX to solid particles of humic acid and encapsulating them in agar-gelatin capsules. A controlled quantity of TTX-containing microcapsules (size range 20-280 µm) was fed to Paphies australis, a bivalve known to accumulate TTX in the wild. The TTX-containing microcapsules were fed to P. australis every second day for 13 days. Ten P. australis (including five controls fed non-toxic microalgae) were harvested after 7 days and ten after 13 days. Paphies australis accumulated TTX, reaching concentrations of up to 103 µg kg-1 by day 13, exceeding the European Food Safety Authority recommended concentration of 44 µg kg-1 in shellfish. This novel method will allow future studies to explore the effects, accumulation and depuration rates of TTX in different animals and document how it is transferred through food webs.

The McMurdo Dry Valleys are the largest single ice-free area in Antarctica, and of considerable scientific and conservation value as an extreme polar desert. This is recognised through the McMurdo Dry Valleys Antarctic Specially Managed Area (ASMA), where management's goals focus on protection of its unique features, while facilitating science access. Using a mix of remote sensing and existing cartography, we have identified over 6000 lakes and ponds in the ASMA. This study develops a classification of those aquatic ecosystems to provide a framework for management. It uses a limited top-down, hierarchical classification to define 13 class separations based on physical attributes that could largely be ascribed from existing databases or remotely sensed information. The first hierarchical level was based on landscape position, separating coastal kettle holes (reflecting recent glacial history), from other "topographic" water bodies. The second level was based on endorheic vs exorheic drainage, the third on mid-summer ice condition (no-ice cap; ice capped; frozen to base) and the fourth on source of inflow (glacial or non-glacial). Kettles were sub-classed by mid-summer ice only. Classes were tested against a set of field observations and an expert workshop validation process considered management implications for the ASMA. This study shows how the classification assists our understanding of Dry Valley landscapes and addresses management issues faced by researchers, environmental managers and policy makers. The approach to classification, rather than the detailed classes that may be specific to the Dry Valleys, has potential for wider use in other polar landscapes.

Evidence of climate-driven environmental change is increasing in Antarctica, and with it comes concern that this will propagate to impacts on biological communities. Recognition and prediction of change needs to incorporate the extent and timescales over which communities vary under extant conditions. However, few observations of Antarctic microbial communities, which dominate inland habitats, allow this. We therefore carried out the first molecular comparison of Cyanobacteria in historic herbarium microbial mats from freshwater ecosystems on Ross Island and the McMurdo Ice Shelf, collected by Captain R.F. Scott's 'Discovery' Expedition (1902-1903), with modern samples from those areas. Using 16S rRNA gene surveys, we found that modern and historic cyanobacteria assemblages showed some variation in community structure but were dominated by the same genotypes. Modern communities had a higher richness, including genotypes not found in historic samples, but they had the highest similarity to other cyanobacteria sequences from Antarctica. The results imply slow cyanobacterial 16S rRNA gene genotype turnover and considerable community stability within Antarctic microbial mats. We suggest that this relates to Antarctic freshwater 'organisms requiring a capacity to withstand diverse stresses, and that this could also provide a degree of resistance and resilience to future climatic-driven environmental change in Antarctica.

Global declines of macroalgal beds in coastal waters have prompted a plethora of studies attempting to understand the drivers of change within dynamic nearshore ecosystems. Photosynthetic measurements are good tools for assessing the consequences of numerous stressors of macroalgae, but there is somewhat of a disconnection between studies that focus on organism-specific ecophysiological responses and those that address causes and consequences of shifts in macroalgal productivity. Our goal is to highlight the applications of two complementary tools for measuring photosynthesis-variable chlorophyll a fluorescence and photorespirometry-and provide guidance for the integration of physiology and ecology to understand the drivers of change in macroalgal communities. Photorespirometry can provide an integrated measure of whole-community metabolism, including an estimate of the physiological costs associated with stressors, while fluorescence-based techniques provide point measures of the efficiency of the photosynthetic apparatus within communities. Variable chlorophyll a fluorescence does not provide an estimate of carbon balance or integrated photosynthesis across either whole plants or whole communities but can be used to estimate the contribution of individual community components in the dynamic subcanopy environment to help us understand the mechanisms underlying observed responses. We highlight the importance of the highly dynamic light environment within macroalgal communities and call for better integration of physiological techniques in an ecological context to enhance our understanding of the responses of whole communities to local and global stressors.

Lake Fryxell is a perennially ice-covered lake in the McMurdo Dry Valleys, Antarctica, with a sharp oxycline in a water column that is density stabilized by a gradient in salt concentration. Dissolved oxygen falls from 20 mg liter(-1) to undetectable over one vertical meter from 8.9- to 9.9-m depth. We provide the first description of the benthic mat community that falls within this oxygen gradient on the sloping floor of the lake, using a combination of micro- and macroscopic morphological descriptions, pigment analysis, and 16S rRNA gene bacterial community analysis. Our work focused on three macroscopic mat morphologies that were associated with different parts of the oxygen gradient: (i) "cuspate pinnacles" in the upper hyperoxic zone, which displayed complex topography and were dominated by phycoerythrin-rich cyanobacteria attributable to the genus Leptolyngbya and a diverse but sparse assemblage of pennate diatoms; (ii) a less topographically complex "ridge-pit" mat located immediately above the oxic-anoxic transition containing Leptolyngbya and an increasing abundance of diatoms; and (iii) flat prostrate mats in the upper anoxic zone, dominated by a green cyanobacterium phylogenetically identified as Phormidium pseudopriestleyi and a single diatom, Diadesmis contenta. Zonation of bacteria was by lake depth and by depth into individual mats. Deeper mats had higher abundances of bacteriochlorophylls and anoxygenic phototrophs, including Chlorobi and Chloroflexi. This suggests that microbial communities form assemblages specific to niche-like locations. Mat morphologies, underpinned by cyanobacterial and diatom composition, are the result of local habitat conditions likely defined by irradiance and oxygen and sulfide concentrations.

BACKGROUND AND OBJECTIVE: Sotrovimab 500 mg administered by a single intravenous (IV) infusion has been granted special approval for emergency use in Japan for treatment of SARS-CoV-2 infection in adults and children aged ≥ 12 years weighing ≥ 40 kg. This Phase 1, single-dose study investigated the pharmacokinetics, safety, and tolerability of IV or intramuscular (IM) sotrovimab 500 mg doses versus placebo in healthy Japanese and Caucasian volunteers. METHODS: This was a two-part, Phase 1, randomized, placebo-controlled, single-blind study. In Part 1, participants received a single sotrovimab 500 mg IV infusion or matching placebo on Day 1. In Part 2, participants received a single sotrovimab 500 mg IM dose or matching placebo on Day 1, administered as two 4 mL injections. RESULTS: There was no effect of ethnicity on the peak or total serum exposure of IV sotrovimab through Week 18; after adjusting for body weight, the point estimate and 90 % confidence interval for the ratio of total exposure between Japanese and Caucasian participants fell within conventional bioavailability bounds (80-125%). Geometric mean Cmax and AUClast following a single IM administration of sotrovimab were higher in Japanese participants compared with Caucasian participants, even after adjustment for body weight. Overall, a single IV or IM dose of sotrovimab was well tolerated by both Japanese and Caucasian participants. CONCLUSIONS: After adjusting for body weight, exposures following a single IV dose of sotrovimab 500 mg were similar between Japanese and Caucasian participants, and higher in Japanese participants following IM administration. Accumulating knowledge of photo-physiological acclimation and adaptation in aquatic phototrophs to altered environmental factors are valuable for managing and conserving aquatic ecosystems. Photosynthesis-irradiance curve (PI curve) analysis is an essential technique to assess the photo-physiological states of and environmental stresses on photosystems. For PI curve analysis, replicates were rarely homogeneously illuminated, which could generate variations potentially obscuring treatment effects or lead to considerable errors. Here we present an incubation apparatus with a novel configuration of illuminating unit that supplied a gradient of irradiances with improved homogeneity. The achieved homogeneity exceeds that of other homogeneous illuminating apparatus reported for photosynthetic research. We used the elaborated apparatus to develop PI curves for S. pectinata photo-acclimated to contrasting light conditions in both greenhouse and field scenarios. Photo-acclimation to lower irradiances enhanced both maximum photosynthetic rates and light utilization efficiencies in general. And improved homogeneity for PI curve analysis most likely reduced variations of derived light utilization efficiency compared to those using conventional incubation apparatus. The elaborated incubation apparatus could provide insights into developments of illumination techniques for photosynthetic studies and has the potential to refine the subtleties of photo-acclimation studies.

Motivated by stream ecosystem degradation by eutrophication, we mimicked slow flowing lowland stream conditions with a novel experimental setup to further our understanding of aquatic plant responses to increases in nitrate and light. We conducted a mesocosm growth experiment of two species from the genus Potamogeton: P. crispus (alien) and P. ochreatus (native), grown at four nitrate and four light levels. We hypothesised that (i) internal nutrient status of the plants would scale with water column nutrient concentration, and that (ii) plant performance would reflect the nutrient status of the plant. Furthermore, we hypothesised that (iii) a low irradiance level would negate the effects of an increased nitrate level. In relation to (ii) we hypothesised that (iv) the traits of the alien species would enable it to outperform the native species where both the availability of light and nutrient resources was high. Internal tissue N content was broadly similar in the two higher (>250 µg NO3 - L-1) and the two lower nutrient treatments (<20 µg NO3 - L-1) in both species and plants were therefore collapsed into high and low N-groups. High-N individuals had higher growth rates than low-N ones regardless of species or light treatment and plants had reduced growth rates at the lowest light treatment, however this response was less evident for P. crispus. The highest growth rate was found at the high-N individuals of P. crispus at the highest light treatment, and correspondingly, in this treatment this species exhibited an increase in branching degree and lateral spread from the low-N plants. As P. crispus spreads by fragmentation, our results show it to be a highly effective competitor in anthropogenically impacted areas compared to its native counterpart. Our study exemplifies how light can influence eutrophication responses of plants and how both need to be accounted for in management decisions.

Remote sensing using satellite imagery has been promoted as a method to broaden the scale and frequency of cyanobacterial monitoring. This relies on the ability to establish relationships between the reflectance spectra of water bodies and the abundance of cyanobacteria. A challenge to achieving this comes from a limited understanding of the extent to which the optical properties of cyanobacteria vary according to their physiological state and growth environment. The aim of the present study was to determine how growth stage, nutrient status and irradiance affect pigment concentrations and absorption spectra in two common bloom forming cyanobacterial taxa: Dolichospermum lemmermannii and Microcystis aeruginosa. Each species was grown in laboratory batch culture under a full factorial design of low or high light intensity and low, medium, or high nitrate concentrations. Absorption spectra, pigment concentrations and cell density were measured throughout the growth phases. The absorption spectra were all highly distinguishable from each other, with greater interspecific than intraspecific differences, indicating that both D. lemmermannii and M. aeruginosa can be readily differentiated using hyperspectral absorption spectra. Despite this, each species exhibited different responses in the per-cell pigment concentrations with varying light intensity and nitrate exposure. Variability among treatments was considerably higher in D. lemmermannii than in M. aeruginosa, which exhibited smaller changes in pigment concentrations among the treatments. These results highlight the need to understand the physiology of the cyanobacteria and to take caution when estimating biovolumes from reflectance spectra when species composition and growth stage are unknown.

Cyanobacteria in polar environments face environmental challenges, including cold temperatures and extreme light seasonality with small diurnal variation, which has implications for polar circadian clocks. However, polar cyanobacteria remain underrepresented in available genomic data, and there are limited opportunities to study their genetic adaptations to these challenges. This paper presents four new Antarctic cyanobacteria metagenome-assembled genomes (MAGs) from microbial mats in Lake Vanda in the McMurdo Dry Valleys in Antarctica. The four MAGs were classified as Leptolyngbya sp. BulkMat.35, Pseudanabaenaceae cyanobacterium MP8IB2.15, Microcoleus sp. MP8IB2.171, and Leptolyngbyaceae cyanobacterium MP9P1.79. The MAGs contain 2.76 Mbp - 6.07 Mbp, and the bin completion ranges from 74.2-92.57%. Furthermore, the four cyanobacteria MAGs have average nucleotide identities (ANIs) under 90% with each other and under 77% with six existing polar cyanobacteria MAGs and genomes. This suggests that they are novel cyanobacteria and demonstrates that polar cyanobacteria genomes are underrepresented in reference databases and there is continued need for genome sequencing of polar cyanobacteria. Analyses of the four novel and six existing polar cyanobacteria MAGs and genomes demonstrate they have genes coding for various cold tolerance mechanisms and most standard circadian rhythm genes with the Leptolyngbya sp. BulkMat.35 and Leptolyngbyaceae cyanobacterium MP9P1.79 contained kaiB3, a divergent homolog of kaiB.

Non-native fish have been shown to have deleterious impacts on freshwater ecosystems in New Zealand. Early detection is critical for their effective management. Traditional capture-based techniques may not detect newly introduced fish, especially if they are present in low abundance. Molecular techniques that target environmental DNA (eDNA) have been shown, in many instances, to be more sensitive, cost-effective and require lower sampling effort. However, appropriate sampling strategies are needed to ensure robust and interpretable data are obtained. In this study we used droplet digital PCR assays to investigate the presence of two non-native fish in New Zealand, the European perch (Perca fluviatilis) and rudd (Scardinius erythrophthalmus) in three small lakes. Samples were collected from water and surface sediment at near-shore and mid-lake sites. Probabilistic modelling was used to assess the occupancy of fish eDNA and develop guidance on sampling strategies. Based on the detection probability measures from the present study, at least six sites and five replicates per site are needed to reliably detect fish eDNA in sediment samples, and twelve sites with eight replicates per site for water samples. The results highlight the potential of developing monitoring and surveillance programs adapted to lakes, that include the use of assays targeting eDNA. This study focused on small shallow lakes, and it is likely that these recommendations may vary in larger, deeper, and more geomorphologically complex lakes, and this requires further research.

We undertook a series of measurements of photophysiological parameters of sea ice algae over 12 days of early spring growth in a West Greenland Fjord, by variable chlorophyll fluorescence imaging. Imaging of the ice-water interface showed the development of ice algae in 0.3-0.4 mm wide brine channels between laminar ice crystals in the lower 4-6 mm of the ice, with a several-fold spatial variation in inferred biomass on cm scales. The maximum quantum yield of photosynthesis, F(v) /F(m), was initially low (~0.1), though this increased rapidly to ~0.5 by day 6. Day 6 also saw the onset of biomass increase, the cessation of ice growth and the time at which brine had reached <50 psu and >-2 °C. We interpret this as indicating that the establishment of stable brine channels at close to ambient salinity was required to trigger photosynthetically active populations. Maximum relative electron transport rate (rETR(max)), saturation irradiance (E(k)) and photosynthetic efficiency (α) had also stabilised by day 6 at 5-6 relative units, ~30 µmol photons m⁻² s⁻¹ and 0.4-0.5 µmol photons m⁻²s⁻¹, respectively. E(k) was consistent with under-ice irradiance, which peaked at a similar value, confirming that daytime irradiance was adequate to facilitate photosynthetic activity throughout the study period. Photosynthetic parameters showed no substantial differences with depth within the ice, nor variation between cores or brine channels suggesting that during this early phase of ice algal growth cells were unaffected by gradients of environmental conditions within the ice. Variable chlorophyll fluorescence imaging offers a tool to determine how this situation may change over time and as brine channels and algal populations evolve.

Understanding the historical onset of cyanobacterial blooms in freshwater bodies can help identify their potential drivers. Lake sediments are historical archives, containing information on what has occurred in and around lakes over time. Paleolimnology explores these records using a variety of techniques, but choosing the most appropriate method can be challenging. We compared results obtained from a droplet digital PCR assay targeting a cyanobacterial-specific region of the 16S rRNA gene in sedimentary DNA and cyanobacterial pigments (canthaxanthin, echinenone, myxoxanthophyll and zeaxanthin) analysed using high-performance liquid chromatography in four sediment cores. There were strong positive relationships between the 16S rRNA gene copy concentrations and individual pigment concentrations, but relationships differed among lakes and sediment core depths within lakes. The relationships were more consistent when all pigments were summed, which we attribute to different cyanobacteria species, in different lakes, at different times producing different suites of pigments. Each method had benefits and limitations, which should be taken into consideration during method selection and when interpreting paleolimnological data. We recommend this biphasic approach when making inferences about changes in the entire cyanobacterial community because they yielded complementary information. Our results support the view that molecular methods can yield results similar to traditional paleolimnological proxies when caveats are adequately addressed.

Neurons in the retrohippocampal cortices play crucial roles in spatial memory. Many retrohippocampal neurons have firing fields that are selectively active at specific locations, with memory for rewarded locations associated with reorganization of these firing fields. Whether this is the sole strategy for representing spatial memories is unclear. Here, we demonstrate that during a spatial memory task retrohippocampal neurons encode location through ramping activity that extends across segments of a linear track approaching and following a reward, with the rewarded location represented by offsets or switches in the slope of the ramping activity. Ramping representations could be maintained independently of trial outcome and cues marking the reward location, indicating that they result from recall of the track structure. When recorded in an open arena, neurons that generated ramping activity during the spatial memory task were more numerous than grid or border cells, with a majority showing spatial firing that did not meet criteria for classification as grid or border representations. Encoding of rewarded locations through offsets and switches in the slope of ramping activity also emerged in recurrent neural network models trained to solve a similar spatial memory task. Impaired performance of model networks following disruption of outputs from ramping neurons is consistent with this coding strategy supporting navigation to recalled locations of behavioral significance. Our results suggest that encoding of learned spaces by retrohippocampal networks employs both discrete firing fields and continuous ramping representations. We hypothesize that retrohippocampal ramping activity mediates readout of learned models for goal-directed navigation.

Persistent cold temperatures, a paucity of nutrients, freeze-thaw cycles, and the strongly seasonal light regime make Antarctica one of Earth's least hospitable surface environments for complex life. Cyanobacteria, however, are well-adapted to such conditions and are often the dominant primary producers in Antarctic inland water environments. In particular, the network of meltwater ponds on the 'dirty ice' of the McMurdo Ice Shelf is an ecosystem with extensive cyanobacteria-dominated microbial mat accumulations. This study investigated intact polar lipids (IPLs), heterocyte glycolipids (HGs), and bacteriohopanepolyols (BHPs) in combination with 16S and 18S rRNA gene diversity in microbial mats of twelve ponds in this unique polar ecosystem. To constrain the effects of nutrient availability, temperature and freeze-thaw cycles on the lipid membrane composition, lipids were compared to stromatolite-forming cyanobacterial mats from ice-covered lakes in the McMurdo Dry Valleys as well as from (sub)tropical regions and hot springs. The 16S rRNA gene compositions of the McMurdo Ice Shelf mats confirm the dominance of Cyanobacteria and Proteobacteria while the 18S rRNA gene composition indicates the presence of Ochrophyta, Chlorophyta, Ciliophora, and other microfauna. IPL analyses revealed a predominantly bacterial community in the meltwater ponds, with archaeal lipids being barely detectable. IPLs are dominated by glycolipids and phospholipids, followed by aminolipids. The high abundance of sugar-bound lipids accords with a predominance of cyanobacterial primary producers. The phosphate-limited samples from the (sub)tropical, hot spring, and Lake Vanda sites revealed a higher abundance of aminolipids compared to those of the nitrogen-limited meltwater ponds, affirming the direct affects that N and P availability have on IPL compositions. The high abundance of polyunsaturated IPLs in the Antarctic microbial mats suggests that these lipids provide an important mechanism to maintain membrane fluidity in cold environments. High abundances of HG keto-ols and HG keto-diols, produced by heterocytous cyanobacteria, further support these findings and reveal a unique distribution compared to those from warmer climates.

The frequency and intensity of cyanobacterial blooms is increasing worldwide. Multiple factors are implicated, most of which are anthropogenic. New Zealand provides a useful location to study the impacts of human settlement on lake ecosystems. The first humans (Polynesians) arrived about 750 years ago. Following their settlement, there were marked landscape modifications which intensified after European settlement about 150 years ago. The aims of this study were to reconstruct cyanobacterial communities in six lakes over the last 1000 years and explore key drivers of change. Cyanobacterial environmental DNA was extracted from sediment cores and analysed using metabarcoding and droplet digital PCR. Cyanobacteria, including potentially toxic or bloom forming species, were already present in these lakes prior to human arrival, however their overall abundance was low. Total cyanobacteria abundance and richness increased in all lakes after European settlement but was very pronounced in four lakes, where bloom-forming taxa became dominant. These shifts occurred concomitant with land-use change. The catchment of one deteriorated lake is only moderately modified, thus the introduction of non-native fish is posited as the key factor driving this change. The paleolimnological approach used in this study has enabled new insights into timing and potential causes of changes in cyanobacterial communities.

This review collates research into fine sediment as a stressor of seagrass and emphasizes the multiple modes of action of this contaminant. The article is based on a bibliographic database search that identified 201 articles describing sediment impacts on seagrasses. Articles were classified by one of three non-exclusive modes of action: 1) light reduction; 2) smothering (burial), and 3) effects via rhizosphere physico-chemistry. Most citations (104) investigated multi-mode impacts of sediments, but the most frequently investigated single mode was light reduction (57 citations), followed by substrate rhizosphere chemistry (31) then smothering effects (6). Mud with high organic content is particularly problematic and smaller seagrasses are particularly vulnerable. Research gaps include polyphasic approaches, and studies of interactions between smothering, rhizosphere biogeochemistry and light climate. Identifying the thresholds of seagrass health indicators under mud stress should benefit coastal resource management, enabling improved decision-making and implementation of protective actions.

Microcystis is a bloom-forming genus of cyanobacteria with some genotypes that produce highly toxic microcystin hepatotoxins. In waterbodies where biological and physical factors are relatively homogenous, toxin quotas (the average amount of toxin per cell), at a single point in time, are expected to be relatively constant. In this study we challenged this assumption by investigating the spatial distribution of microcystin quotas at a single point in time on two separate occasions in a lake with a major Microcystis bloom. Microcystis cell concentrations varied widely across the lake on both sampling occasions (730- and 137-fold) together with microcystin quotas (148- and 362-fold). Cell concentrations and microcystin quotas were strongly positively correlated (R2 = 0.89, P < 0.001, n = 28; R2 = 0.67, P < 0.001, n = 25). Analysis of Microcystis strains using high-throughput sequencing of the 16S-23S rRNA intergenic spacer region showed no relationship between microcystin quota and the relative abundance of specific sequences. Collectively, the results of this study indicate an association between microcystin production and cell density that magnifies the potential for bloom toxicity at elevated cell concentrations.
