In this study, potential toxic element (PTEs) including lead (Pb), arsenic (As), cadmium(Cd), iron (Fe) and zinc (Zn) in traditional and industrial edible vegetable oils (peanut, sunflower, olive and sesame) collected from Hamadan, west of Iran were determined using Inductivity Coupled Plasma Optical Emission Spectrometry (ICP-OES). Besides, probabilistic health risk assessment (non-carcinogenic and carcinogenic risks) was identified via total target hazard quotient (TTHQ) and cancer risk (CR) by the Monte Carlo Simulation (MCS) model. The ranking of concentration PTEs in traditional and industrial edible vegetable oils was Fe > Zn > As > Pb > Cd. The in all samples, content of PTEs in industrial oils were upper than traditional oils (p < 0.001). The level of PTEs in most of vegetable oils was lower than permissible concentration regulated by Codex and national standard. In term of non-carcinogenic, consumers were at acceptable range (TTHQ < 1) due to ingestion both traditional and industrial vegetable oils content of PTEs. In term of carcinogenic, CR the both adults and children was higher than acceptable range (CR < 1E-6), Hence consumer are at unacceptable risk due to ingestion industrial vegetable oils content of inorganic As. Therefore, it is recommended to implement control plans for PTEs in vegetable oils consumed in Hamadan, Iran.

Potato is one of the essential food products whose health quality is greatly influenced by soil contamination and properties. In the current study, we have investigated the physicochemical characteristics of agricultural areas and the accumulation of nitrite/nitrate and metals in potato products in Hamedan, Iran. After determining the physicochemical characteristics of soil samples from four agricultural regions of Hamedan, 48 potato samples were collected from these regions. The heavy metals and nitrate/nitrite content were determined by ICP-OES and calorimetric methods, respectively. A negative correlation was observed between soil pH changes with nitrite/nitrate content and the accumulation of some heavy elements in potatoes. Furthermore, a positive correlation was found between soil phosphorus content and lead accumulation in potato. In present study, the amounts of lead, nitrate, and nitrite in 83.3%, 56%, and 12% of the collected samples were higher than the permissible limit reported by the World Health Organization (WHO), respectively. The EDI range for nitrate and nitrite was determined to be 130-260 and 1.4-2.7 µg/kg/day, respectively, which is much lower than the RfD set by the US Environmental Protection Agency (USEPA) for nitrite and nitrate. Among metal pollutants, the toxic risk caused by lead in potato consumers was higher than the threshold limit. In conclusion, our findings showed that the physicochemical characteristics of the soil could effectively increase the availability of metal pollutants and nitrite/nitrate to the potato product and significantly reduce its health quality. Therefore, monitoring these pollutants in the soil-potato system, preventing the entry of industrial wastewater, and managing the use of agricultural fertilizers can effectively improve the health of this product for consumers.

Coffee is the most popular beverage after water in the world, which has an important role in health as a result of various minerals and vitamins but it may be pollution source of potentially toxic elements (PTEs) that can threate the health. Thus, the current study intended to detect the level of PTEs such as Cadmium (Cd), Copper (Cu), Lead (Pb), Nickel (Ni), Znc (Zn) and Iron (Fe), in various coffee and coffee-based products (powder, ground, processed, infusion and bean). Considering the databases of Scopus, Google scholar, PubMed, and Web of Science, the concentration of PTEs in coffee and coffee-based products was retrieved and meta-analyzed. Additionally, the non-carcinogenic risks in terms of total hazard quotient (TTHQ) were assessed using Monte Carlo simulated (MCS) model. According to the findings of 23 articles, the ranking of metal concentration in different coffees was Fe > Zn > Cu> Ni > Pb > Cd in powder, Fe > Cu > Zn> Ni in ground, Fe > Zn > Ni> Cu> Pb > Cd in processed and infusion and Fe > Zn > Ni> Cs > Pb in bean. Moreover, based on WHO regions, the highest concentrations of Cd and Pb (0.742 mg/kg) were related to the South-East Asia Region (SEARO) and European region (EURO) respectively. However, the highest concentrations of Fe (81.161 mg/kg), Zn (33.392 mg/kg), Cu (9.408 mg/kg), and Ni (18.064 mg/kg) were related to Pan American health organization (PAHO), PAHO, PAHO and Eastern Mediterranean Region (EMRO), respectively. On the other hand, the risk pattern was different in different countries. Moreover, consumers in some countries were not at significant non-carcinogenic risks because of ingestion of PTEs via coffee and consumption of coffee-based products.

An effervescent tablet-assisted switchable polarity solvent-based homogeneous liquid-phase microextraction combined with gas chromatography with flame ionization detection has been conducted for the separation, preconcentration, and detection of permethrin and deltamethrin in the river water specimens. Triethylamine (TEA) was utilized as the switchable polarity solvent in this method. The switching process was carried out by the dissolution of an effervescent tablet including an effervescency agent (sodium carbonate) and a proton donor agent (citric acid). Changing the pH of the specimen solution enhanced the conversion of TEA into protonated triethylamine carbonate through the tablet that generated carbon dioxide bubbles in situ. Finally, the addition of sodium hydroxide changed the ionization state of TEA and separated the two phases. Influential factors in the extraction were investigated. According to optimal situations, the limit of detection and the limit of quantification were 0.16 and 0.5 µg L-1 for permethrin and 0.03 and 0.1 µg L-1 for deltamethrin, respectively. The preconcentration factor was 194 in river water samples and inter- and intra-day precision (relative standard deviation %; n = 5) was <5%. The extraction recovery was obtained in the range of 93.0%-97% for permethrin and deltamethrin in water samples.

This study aimed to investigate the acute and chronic hazard quotients of chlorpyrifos and diazinon for tomatoes in preharvest and postharvest conditions, and to evaluate the relationship between the main variables, including temperature and contact time and pesticide dissipation at different conditions using response surface methodology for the first time. The qualification analyses were conducted by a gas chromatography-tandem mass spectrometry. The Monte Carlo simulation technique was utilized to evaluate the variability and uncertainty and achieve more accurate results in the health risk assessment process. A quadratic model and the second-order polynomial analysis were employed to investigate the mutual effect of time and temperature on removing diazinon and chlorpyrifos. Based on findings, the chronic hazard quotient values of chlorpyrifos and diazinon residues ranged from 0.43 - 1.33 to 0.13 - 2.27 for boiling, 0.65 - 1.49 to 3.05 - 7.15 for room condition, and 0.63 - 1.92 to 3.28 - 7.47 for refrigerator condition, respectively. According to the Monte Carlo simulation, the hazard quotient and estimated daily intake values were more affected by the consumption rate, pesticide concentration, and body weight. The results of response surface methodology showed that the effect of temperature variations on the dissipation of both pesticides was more than that of contact time variations.

Paraquat (PQ) is a widely used herbicide all over the world, which is highly toxic for animals and humans. Its cytotoxicity is based on reactive radical generation. The aim of this study is to evaluate and compare the hepatoprotective effects of curcumin and nanocurcumin against liver damage caused by sub-acute exposure with PQ via modulation of oxidative stress and genes expression of nuclear factor erythroid 2-related factor 2 (Nrf2) pathway. Rats were exposed to PQ (5 mg/kg/day, orally) + curcumin or nanocurcumin (100 mg/kg/day, orally) for 7 days. Then rats were anesthetized and serum and liver samples were collected. Next, serum enzymatic activities, liver histopathology, oxidative stress, and expression of genes involved in Nrf2 signaling pathway were assessed by biochemical and enzyme-linked immunosorbent assay methods, hematoxylin and eosin staining, and real-time polymerase chain reaction analysis. PQ significantly increased malondialdehyde, alanine transaminase, aspartate aminotransferase, alkaline phosphatase levels, and Kelch-like ECH-associated protein 1 gene expression and also decreased total antioxidant capacity, total thiol group levels, Glutathione S-transferases, heme oxygenase 1, Nrf2, and NAD(P)H:quinone oxidoreductase 1 genes expression, causing histological damages to liver tissue. These changes were significantly modulated by curcumin and nanocurcumin treatments. Our findings showed that nanocurcumin had better hepatoprotective effect than curcumin in liver damage after PQ exposure most likely through modulation of oxidative stress and genes expression of Nrf2 pathway.

BACKGROUND: Traditional drug discovery approaches are mainly relied on the observed phenotypic changes following administration of a plant extract, drug candidate or natural product. Recently, target-based approaches are becoming more popular. The present study aimed to identify the cellular targets of crocin, the bioactive dietary carotenoid present in saffron, using an affinity-based method. METHODS: Heart, kidney and brain tissues of BALB/c mice were homogenized and extracted for the experiments. Target deconvolution was carried out by first passing cell lysate through an affinity column prepared by covalently attaching crocin to agarose beads. Isolated proteins were separated on a 2D gel, trypsinized in situ and identified by MALDI-TOF/TOF mass spectrometry. MASCOT search engine was used to analyze Mass Data. RESULTS: Part of proteome that physically interacts with crocin was found to consist of beta-actin-like protein 2, cytochrome b-c1 complex subunit 1, ATP synthase subunit beta, tubulin beta-3 chain, tubulin beta-6 chain, 14-3-3 protein beta/alpha, V-type proton ATPase catalytic subunitA, 60 kDa heat shock protein, creatine kinase b-type, peroxiredoxin-2, cytochrome b-c1 complex subunit 2, acetyl-coA acetyltransferase, cytochrome c1, proteasome subunit alpha type-6 and proteasome subunit alpha type-4. CONCLUSION: The present findings revealed that crocin physically binds to a wide range of cellular proteins such as structural proteins, membrane transporters, and enzymes involved in ATP and redox homeostasis and signal transduction.

In this study, gluten-free pancakes were prepared using rice flour and potato starch at a ratio of 50:50. Due to a lack of gluten networks in these ingredients, the hydrocolloid gums including carboxymethyl cellulose (CMC) at 0.5%, 1%, and 1.5% and hydroxypropyl methylcellulose (HPMC) at 1%, 2%, and 3% were added to improve the quality of the final products. The effects of these hydrocolloid gums on the physicochemical, textural, and sensory properties of the gluten-free pancakes were evaluated. Pancakes prepared with wheat flour were used as a control sample. The results showed that the addition of both gums decreased the hardness and chewiness of the gluten-free pancakes while increasing the springiness and their moisture content. Increasing the concentration of the gums resulted in an L* value (lightness) reduction, which produced a darker crust on the pancakes. Moreover, the gluten-free pancakes containing CMC and HPMC had higher specific volumes than the gluten-free samples made without CMC and HPMC. From a sensory point of view, the samples containing 2%, 3% HPMC and 1% CMC received the highest overall acceptance score. Thus, CMC and HPMC can be used as improvers in gluten-free pancakes.

Exposure to heavy metals can endanger the health of exposed people in the long term. The consumption of fruit juice is increasing; it is important to estimate the health risk of consumers due to heavy metals. The current study was carried out for the analysis of toxic metals (lead (Pb), arsenic (As), and cadmium (Cd)) and essential elements (copper (Cu) and zinc (Zn)) in 60 samples of traditional and industrial fruit juices (10 samples of different brands of apple, orange, grape, peach, mango, and pineapple) in Hamadan, West Iran, using inductivity coupled plasma optical emission spectrometry (ICP-OES) method. The validation protocol included precision of the analytical method; recovery, the determination of the limit of detection (LOD), the limit of quantification (LOQ), and linearity were measured. Moreover, risk assessment was detected using target hazard quotient (THQ) and cancer risk (CR) by the Monte Carlo simulation (MCS) model. The ranking of metal concentration in traditional and industrial fruit juices was Zn > Cu > As > Pb > Cd. In all samples, concentrations of heavy metals in industrial fruit juices were higher than traditional fruit juices p < < 0.001. The level of metals in all samples was lower of the US Environmental Protection Agency (USEPA), the World Health Organization (WHO), and the Iran Standard (IS) permissible limit set for drinking water. In terms of non-carcinogenic, values of toxic elements for children and adult in traditional and industrial fruit juices were 1.6E-3 and 1.72E-3 and 2.6E-3 and 1.85E-3, respectively. The 95th percentile of CR in adults and children due to both industrial and traditional fruits juices was higher than 1E-6; hence, reducing the concentration of As in fruit juices should be conducted. Consumption of fruit juice can increase carcinogenic risk of consumers. Therefore, it is recommended to consume it with caution.

In the present study, six potentially toxic elements (PTEs), including chromium (Cr), arsenic (As), cadmium (Cd), lead (Pb), copper (Cu), and nickel (Ni), were determined in 41 domestic rice samples collected from Tehran using ICP-OES (inductively coupled plasma-optical emission spectrometry). The mean concentration of Cd, As, Cu, Pb, Cr, and Ni was found as 0.014 ± 0.01, 0.018 ± 0.005, 2.15 ± 1.84, 0.42 ± 0.31, 0.1 ± 0.16, and 0.48 ± 0.36 mg kg-1, respectively. Possible risks due to ingestion of PTEs via rice consumption for children and adults were assessed by Monte Carlo simulation. The 50th percentile of estimated Cr intake for children through domestic rice consumption exceeded the maximum tolerable daily intake. There was only a potential non-carcinogenic risk for single Cr exposure for children. The 95th percentile of the estimated hazard index (HI) for children and adults was 4.34 and 1.05, indicating a potential non-carcinogenic risk related to multiple PTE exposure. The lifetime cancer risk (ILCR) values derived from Cr, Ni, As, and Cd exposure exceeded the threshold value, indicating a carcinogenic risk due to PTEs' exposure. The deterministic assessment demonstrates that the Tehran population may be at risk through domestic rice consumption. This study indicates that risk related to the exposure to multiple PTEs through the consumption of rice in adults and children in Tehran is recognized as an important issue, thus supporting the importance of cumulative analysis of the risk of exposure to PTEs through food. Finally, national strategic environmental assessment and technological solutions for monitoring and protecting freshwater, soil, waste management, and chemicals as a global concern policy are needed for public health.

This study focuses on measuring potentially toxic elements (PTEs) including mercury (Hg), lead (Pb), cadmium (Cd), arsenic (As), iron (Fe), copper (Cu), zinc (Zn), manganese (Mn) in farmed and wild carp by inductively coupled plasma-optical emission spectrometry (ICP-OES) and their intake amount and risk assessment. Estimated Daily Intake (EDI), Target Hazard Quotient (THQ) and the total THQ (TTHQ) were calculated for each element. In the present research, the concentration of As in farmed and wild carp samples was below the detection limit of ICP-OES (< 0.005 mg/kg). The mean concentrations of Pb, Cd, Fe, and Cu in farmed carp samples were significantly higher than that in wild carp samples, while Zn level was higher in the wild carp samples (P < 0.05). There is no significant difference between Hg and Mn in both fish (P > 0.05). The Monte Carlo simulation (MCS) results showed that the ranking order of PTEs based on their THQ was Hg > Pb > Zn > Cu > Fe > Cd > Mn. In the worst-case scenario (in the top 95 percentile) for both wild and farmed carp, the THQ of measured metals was less than one except Hg in children. Overall, this study demonstrated that the levels of PTEs in farmed and wild common carp had a potential non-carcinogenic risk for children (TTHQ > 1).

This study aims to produce fat-reduced cream cheese using the different levels (0.25%-0.5%) of basil seed and xanthan gum by a RSM method. The basil seed, xanthan gum, and fat levels did not significantly influence the cream cheese's pH and acidity. With the fat reduction, textural properties were lost; for example, hardness, gumminess, and adhesiveness increased, and cohesiveness decreased. In addition, low-fat cream cheese's sensory score (taste, mouthfeel, and overall acceptance score) was lower. However, adding basil seed and xanthan gum could improve water holding capacity (WHC), hardness, gumminess, cohesiveness, adhesiveness and scores of mouthfeel, and overall acceptance. Basil seed gum had a better impact than xanthan on fat-reduced cream cheese properties among the two gums. In general, results showed that adding 0.5% basil and 0.5% xanthan into cream cheese could manufacture a product with a reduced-fat level (19.04%). At the same time, its physicochemical, sensory, and textural attributes were similar to cream cheese with high fat (24%). In addition, the price of the obtained product was lower.

This study achieved maximum removal of ochratoxin A (OTA) during the grape juice clarification process with minimal reduction in antioxidant compounds (phenolic acid, flavonoids, and antioxidant capacity by FRAP) by the RSM method. Independent variables included three types of clarifiers-gelatin, bentonite, and diatomite (diatomaceous earth)-at a concentration level of 0.25-0.75% and clarification time of 1-3 h. OTA was measured by high-performance liquid chromatography with fluorescence detection. Clarifying agent concentration and clarification time affected the reduction amount of OTA and antioxidant compounds in grape juice. There was a direct linear correlation between the reduction amounts of OTA and antioxidant compounds and capacity with the concentration of bentonite, gelatin, and diatomite, and the clarification time. The reduction amount of OTA and antioxidant capacity followed the linear mode. However, the decreased phenolic acid and flavonoid values followed the quadratic model. The study results showed that if the concentrations of bentonite, gelatin, and diatomite and clarification time were 0.45, 0.62, 0.25%, and 1 h, respectively, the maximum amount of OTA reduction (41.67%) occurred. Furthermore, the phenolic acid, flavonoid, and antioxidant activity decrease amounts were at their lowest levels, i.e., 23.86, 7.20, and 17.27%, respectively.

Sunflower oil is a common edible oil in the world, which is highly prone to oxidative degradation during the frying process. The present study aimed to investigate the effects of products obtained from the thermal oxidation process of sunflower oil on metabolic indices, and the secretion status of leptin and ghrelin in rats. In vivo studies were designed after determining the rate of formation of active aldehydes and peroxide value in sunflower oil following 300°C in a period of 30-240 min. To this end, 36 rats in 6 separate groups were fed with 2 ml of normal saline, fresh sunflower oil, and heated oils at 30, 60, 120, and 240 min for 45 days. Finally, lipid profile changes and leptin/ghrelin secretion were examined, along with histological changes in the liver tissue. The results indicated a significant increase in serum LDL, VLDL and triglycerides, and a decrease in HDL, in the groups treated with heated oils. These changes were associated with a higher accumulation of triglycerides, active aldehydes, and histological changes in the hepatic tissue. Although the serum ghrelin level in the groups receiving heated oil did not change significantly compared to the fresh oil, the serum leptin level increased significantly in the groups receiving heated oil. According to our findings, increasing the time of sunflower oil heating enhanced the formation of active aldehydes, so that daily consumption of such oxidized oils might be associated with the occurrence of dyslipidemia, fatty liver and the development of leptin resistance. PRACTICAL APPLICATIONS: Sunflower oil is highly prone to oxidative degradation during the frying process. Increasing time of sunflower oil heating enhanced the formation of active aldehydes. Daily consumption of oxidized oils might be associated with the occurrence of dyslipidemia, fatty liver and the development of leptin resistance.

The present study aimed to develop a probiotic rose petal jam containing microencapsulated L. plantarum. The attributes of L. plantarum microcapsules and bacteria viability in simulated gastrointestinal conditions and jam were assessed. In addition, L. plantarum effects on physicochemical, textural and sensorial properties of jam were studied. The microencapsulation yield, diameter, and zeta potential value of the microcapsules ranged from 90.23 to 92.75%, 14.80-35.02 µm, and - 16.83 to - 14.71 mV, respectively. The microencapsulation process significantly increases the survival of L. plantarum in simulated gastrointestinal tract and jam. In jam samples containing L. plantarum microencapsulated with 2% sodium alginate and 3.5% or 5% Arabic gum and stored for 90 days, the bacterial count was higher than the acceptable level (106 CFU/g). While there was no significant difference (P > 0.05) between physicochemical characteristics of non-probiotic and probiotic jams, taste and overall acceptance scores of microencapsulated probiotic jams were higher. The microencapsulation of L. plantarum in sodium alginate (2%) and Arabic gum (5%) and its inoculation into rose petal jam could yield a new probiotic product with increased health benefits.

In this study, a novel colorimetric bioassay method was developed for the sensitive determination of tobramycin (TOB). To detect TOB, silver nanoparticles (AgNPs) were decorated with TOB-specific aptamers (apt), and positively charged poly diallyl dimethyl ammonium chloride (PDDA) was used. As long as tobramycin is not present in the assay system, PDDA can coalesce with the aptamer, and AgNPs would remain stable (λmax = 400 nm) in the dispersed system against PDDA-induced aggregation. When TOB is added, aptamer can bind to the compound, which leads to release of PDDA and subsequent aggregation of AgNPs (λmax = 540 nm). This remarkable change, as a colorimetric analytics signal, can be used for quantitative analysis of TOB. TOB can be detected by this highly sensitive colorimetric aptasensor with a limit of detection (LOD) of 70 pM. Furthermore, TOB can be detected with the naked eye at concentrations above 1 nM.

OBJECTIVE: Aluminum phosphide (AlP) is widely used to protect stored food products and grains from pests and rodents. The availability of AlP, especially in Asian countries it has become a desirable factor to commit suicide. The phosphine produced from ALP is a very reactive radical and a respiratory inhibitor that causes oxidative damage. There is no dedicated antidote or effective drug to manage AlP-induced lung toxicity. The present study aims to evaluate and compare the protective effects of curcumin and nanocurcumin on ALP­induced subacute lung injury and determine the underlying mechanism. METHODS: Rats were exposed to AlP (2 mg/kg/day, orally)+curcumin or nanocurcumin (100 mg/kg/day, orally) for 7 days. Then rats were anesthetized and lung tissues were collected. Oxidative stress biomarkers, genes expression of antioxidant enzymes, participated genes in the SIRT1/FOXO3 pathway, and lung histopathology were assessed by biochemical and ELISA methods, Real-Time PCR analysis, and H&E staining. RESULTS: Curcumin and nanocurcumin produced a remarkable improvement in AlP-induced lung damage through reduction of MDA, induction of antioxidant capacity (TAC, TTG) and antioxidant enzymes (CAT, GPx), modulation of histopathological changes, and up-regulation of genes expression of SIRT1, FOXO3, FOXO1 in lung tissue. CONCLUSION: Nanocurcumin had a significantly more protective effect than curcumin to prevent AlP-induced lung injury via inhibition of oxidative stress. Nanocurcumin could be considered a suitable therapeutic choice for AlP poisoning.

This study aimed to investigate the concentration of 33 pesticide residues in 60 black tea samples collected from Iran, determine their transfer rate, and assess their health risk during brewing. Pesticide extraction and analysis were performed by using a quick, easy, cheap, effective, rugged, and safe (QuEChERS) method and gas chromatography/tandem mass spectrometry (GC-MS/MS), respectively. The limits of detection (LOD) and the limits of quantification (LOQ) of pesticides were ranged 0.1-7.26 and 0.8-24 µg/kg for dried tea leaves and 0.03-3.1 and 0.09-10 µg/L for the tea infusion, respectively. The levels of pesticide residue in 52 (86.67%) out of 60 tea samples were above the LOD (0.1-7.26 µg/kg). Twenty four (40%) of the samples contained pesticides in a concentration higher than the maximum residue limit (MRL) set by the European Commission (EC). Seven out of 33 validated pesticides were detected in dried tea leaf samples that only four of seven, including buprofezin, chlorpyrifos, hexaconazole, and triflumizole, were transferred into tea infusion, demonstrating that the concentrations of pesticides in infusion were raised during brewing. The risk assessment study for detected pesticides in the tea infusion samples indicated that this beverage consumption was safe for consumers, while the mean residue of some pesticides in positive samples was higher than the MRL; therefore, periodic control of these pesticides should be regularly implemented.

The response surface method (RSM) was used to produce sour cherry jam containing stevia sweetener with favorable physicochemical, textural, and rheological properties. The experiments were designed based on RSM using a central composite design (CCD) with three independent variables: sugar, pectin, and stevia. Sample processing was performed at five levels of sucrose (10%-50%), pectin (0.1%-0.5%), and stevia (0.2%-0.6%) at a constant fruit weight of 300 g. To evaluate the jam, the physicochemical, textural, and rheological properties of the jam were determined and optimized. The concentrations of sucrose, pectin, and stevia had a significant effect on the textural and rheological properties of the jam. The results showed that increasing the concentration of sucrose is the main factor in increasing the soluble solids and the total sugar content of the jam. Pectin had a positive effect on textural characteristics such as hardness and adhesiveness of the jam, while the cohesiveness of the jam decreased with increasing pectin. Herschel-Bulkley model was appropriate for describing the rheological behavior of the stable state of sour cherry jam containing stevia. Decreasing sucrose concentration is accompanied by a decrease in yield stress and consistency index of jam samples due to the TSS reduction. Pectin also had a positive effect on the amount of yield stress and consistency index. Dynamic rheological tests indicated that the sour cherry jam is a weak gel. The predominant viscose behavior and the formation of a dilute solution were evident in the samples with 10% and 20% sucrose concentrations. The most favorable condition for the production of the jam was obtained at a 36.5% sugar, 0.277% pectin, and stevia 0.30%.
