AIMS: As our comprehension of the intricate relationship between cellular senescence and tumor biology continues to evolve, the therapeutic potential of cellular senescence is gaining increasing recognition. Here, we identify chromobox 4 (CBX4), a Small Ubiquitin-related Modifier (SUMO) E3 ligase, as an antagonist of cellular senescence and elucidate a novel mechanism by which CBX4 promotes drug resistance and malignant progression of gastric cancer (GC). METHODS: In vitro and in vivo models were conducted to investigate the manifestation and impact of CBX4 on cellular senescence and chemoresistance. High-throughput sequencing, chromatin immunoprecipitation, and co-immunoprecipitation techniques were utilized to identify the upstream regulators and downstream effectors associated with CBX4, revealing its intricate regulatory network. RESULTS: CBX4 diminishes the sensitivity of GC cells to cellular senescence, facilitating chemoresistance and GC development by deactivating the senescence-related Hippo pathway. Mechanistically, low-dose cisplatin transcriptionally downregulates CBX4 through CEBPB. In addition, CBX4 preserves the stability and cytoplasm-nuclear transport of YAP1, the key player of Hippo pathway, by inducing SUMO1 modification at K97 and K280, which competitively inhibits YAP1-S127 phosphorylation. CONCLUSIONS: Our study highlights the anti-senescence role of CBX4 and suggests that CBX4 inhibition in combination with low-dose cisplatin has the potential to overcome chemoresistance and effectively restrict GC progression.

The ability to reversibly exhibit structural color patterns has positioned photonic crystals (PCs) at the forefront of anti-counterfeiting. However, the security offered by the mere reversible display is susceptible to illicit alteration and disclosure. Herein, inspired by the electronic message captcha, bilayer photonic crystal (BPC) systems with integrated decryption and verification modules, are realized by combining inverse opal (IO) and double inverse opal (DIO) with polyacrylate polymers. When the informationized BPC is immersed in ethanol or water, the DIO layer displayed encrypted information due to the solvent-induced ordered rearrangement of polystyrene (PS) microspheres. The verification step is established based on the different structural colors of the IO layer pattern, which result from the deformation or recovery of the macroporous skeleton induced by solvent evaporation. Moreover, through the evaporation-induced random self-assembly of PS@SiO2 and SiO2 microspheres, unclonable structurally colored identifying codes are created in the IO layer, ensuring the uniqueness upon the verification. The decrypted code in the DIO layer is valid only when the IO layer displays the pattern with the predetermined structural color; otherwise, it is a pseudo-code. This structural color-based "decryption-verification" approach offers innovative anti-counterfeiting applications in nanophotonics.

Oligoasthenoteratozoospermia (OAT) is a common type of male infertility; however, its genetic causes remain largely unknown. Some of the genetic determinants of OAT are gene defects affecting spermatogenesis. BCORL1 (BCL6 corepressor like 1) is a transcriptional corepressor that exhibits the OAT phenotype in a knockout mouse model. A hemizygous missense variant of BCORL1 (c.2615T > G:p.Val872Gly) was reported in an infertile male patient with non-obstructive azoospermia (NOA). Nevertheless, the correlation between BCORL1 variants and OAT in humans remains unknown. In this study, we used whole-exome sequencing to identify a novel hemizygous nonsense variant of BCORL1 (c.1564G > T:p.Glu522*) in a male patient with OAT from a Han Chinese family. Functional analysis showed that the variant produced a truncated protein with altered cellular localization and a dysfunctional interaction with SKP1 (S-phase kinase-associated protein 1). Further population screening identified four BCORL1 missense variants in subjects with both OAT (1 of 325, 0.31%) and NOA (4 of 355, 1.13%), but no pathogenic BCORL1 variants among 362 fertile subjects. In conclusion, our findings indicate that BCORL1 is a potential candidate gene in the pathogenesis of OAT and NOA, expanded its disease spectrum and suggested that BCORL1 may play a role in spermatogenesis by interacting with SKP1.

Researchers have investigated ways to develop optimal imaging techniques to increase the safety and effectiveness of electrophysiological (EP) procedures. Intracardiac echocardiography (ICE) is an advanced imaging tool that can directly visualize cardiac anatomical structures in high resolution, assess tissue heterogeneity and arrhythmogenic substrates, locate intracardiac catheters, monitor catheter-tissue contact and ablation injury in real-time, excluding intracardiac thrombi, and quickly detect procedural complications. Additionally, real-time imaging via ICE can be integrated with a three-dimensional (3D) electroanatomical mapping (EAM) system to reconstruct cardiac anatomy. This technique also promotes the development of zero-radiation EP procedures. Many EP studies and procedures have implemented ICE because it has several advantages over fluoroscopy and transesophageal echocardiography (TEE). ICE-guided EP procedures can be performed under conscious sedation; esophageal intubation and additional anesthesiologists are not required. Atrial fibrillation (AF) and supraventricular tachycardias (SVT) are the most common tachyarrhythmias in clinical settings. A comprehensive understanding of critical anatomical structures, such as the atrial septum, fossa ovalis (FO), and great heart vessels, is needed for the successful catheter ablation of these arrhythmias.

BACKGROUND AND OBJECTIVE: Left ventricular thrombus (LVT) formation in patients with acute myocardial infarction (AMI) or cardiomyopathies is not uncommon. The optimal oral anticoagulation therapy for resolving LVT has been under intense debate. Vitamin K antagonists (VKAs) remain the anticoagulant of choice for this condition, according to practice guidelines. Evidence supporting the use of direct oral anticoagulants (DOACs) in the management of LVT continues to grow. We performed a systematic review and meta-analysis to compare the efficacy and safety of DOACs versus VKAs. METHODS: A comprehensive literature search was carried out in PubMed, Cochrane Library, Web of Science, Embase, and Scopus databases in July 2023. The efficacy outcomes of this study were thrombus resolution, ischemic stroke, systemic embolism, stroke/systemic embolism, all-cause mortality, and adverse cardiovascular events. The safety outcomes were any bleeding, major bleeding, and intracranial hemorrhage. A total of twenty-seven eligible studies were included in the meta-analysis. Data were analyzed utilizing Stata software version 15.1. RESULTS: There was no significant difference between DOACs and VKAs with regard to LVT resolution (RR = 1.00, 95% CI 0.95-1.05, P = 0.99). In the overall analysis, DOACs significantly reduced the risk of stroke (RR = 0.74, 95% CI 0.57-0.96, P = 0.021), all-cause mortality (RR = 0.70, 95% CI 0.57-0.86, P = 0.001), any bleeding (RR = 0.75, 95% CI 0.61-0.92, P = 0.006) and major bleeding (RR = 0.67, 95% CI 0.52-0.85, P = 0.001) when compared to VKAs. Meanwhile, in the sub-analysis examining randomized controlled trials (RCTs), the aforementioned outcomes no longer differed significantly between the DOACs and VKAs groups. The incidences of systemic embolism (RR = 0.81, 95% CI 0.54-1.22, P = 0.32), stroke/systemic embolism (RR = 0.85, 95% CI 0.72-1.00, P = 0.056), intracranial hemorrhage (RR = 0.59, 95% CI 0.23-1.54, P = 0.28), and adverse cardiovascular events (RR = 0.99, 95% CI 0.63-1.56, P = 0.92) were comparable between the DOACs and VKAs groups. A subgroup analysis showed that patients treated with rivaroxaban had a significantly lower risk of stroke (RR = 0.24, 95% CI 0.08-0.72, P = 0.011) than those in the VKAs group. CONCLUSION: With non-inferior efficacy and superior safety, DOACs are promising therapeutic alternatives to VKAs in the treatment of LVT. Further robust investigations are warranted to confirm our findings.

The widespread occurrence of organic antibiotic pollution in the environment and the associated harmful effects necessitate effective treatment method. Heterogeneous electro-Fenton (hetero-EF) has been regarded as one of the most promising techniques towards organic pollutant removal. However, the preparation of efficient cathode still remains challenging. Herein, a novel metal-organic framework (MOF)-derived Fe/Ni@C marigold-like nanosheets were fabricated successfully for the degradation of oxytetracycline (OTC) by serving as the hetero-EF cathode. The FeNi3@C (Fe/Ni molar ratio of 1:3) based hetero-EF system exhibited 8.2 times faster OTC removal rate than that of anodic oxidation and possessed many advantages such as excellent OTC degradation efficiency (95.4% within 90 min), broad environmental adaptability (satisfactory treatment performance for multiple antibiotics under various actual water matrixes), good stability and reusability, and significant toxicity reduction. The superior hetero-EF catalytic performance was mainly attributed to: 1) porous carbon and Ni existence were both conducive to the in-situ generation of H2O2 from dissolved O2; 2) the synergistic effects of bimetals together with electron transfer from the cathode promoted the regeneration of ≡ FeII/NiII, thereby accelerating the production of reactive oxygen species; 3) the unique nanosheet structure derived from the precursor two-dimensional Fe-Ni MOFs enhanced the accessibility of active sites. This work presented a promising hetero-EF cathode for the electrocatalytic treatment of antibiotic-containing wastewaters.

The massive usage of phthalate esters (PAEs) has caused serious pollution. Bacterial degradation is a potential strategy to remove PAE contamination. So far, an increasing number of PAE-degrading strains have been isolated, and the catabolism of PAEs has been extensively studied and reviewed. However, the investigation into the bacterial PAE uptake process has received limited attention and remains preliminary. PAEs can interact spontaneously with compounds like peptidoglycan, lipopolysaccharides, and lipids on the bacterial cell envelope to migrate inside. However, this process compromises the structural integrity of the cells and causes disruptions. Thus, membrane protein-facilitated transport seems to be the main assimilation strategy in bacteria. So far, only an ATP-binding-cassette transporter PatDABC was proven to transport PAEs across the cytomembrane in a Gram-positive bacterium Rhodococcus jostii RHA1. Other cytomembrane proteins like major facilitator superfamily (MFS) proteins and outer membrane proteins in cell walls like FadL family channels, TonB-dependent transporters, and OmpW family proteins were only reported to facilitate the transport of PAEs analogs such as monoaromatic and polyaromatic hydrocarbons. The functions of these proteins in the intracellular transport of PAEs in bacteria await characterization and it is a promising avenue for future research on enhancing bacterial degradation of PAEs. KEY POINTS: • Membrane proteins on the bacterial cell envelope may be PAE transporters. • Most potential transporters need experimental validation.

RESUMO

Three new monoterpene phenol dimers, bisbakuchiols V-X (1-3), and two bakuchiol ethers (4 and 5), along with four known compounds (6-9) were isolated from the fruits of Psoralea corylifolia. Their structures were elucidated based on extensive spectral analysis. The absolute configurations of 1, 2, 4, and 5 were specified by quantum chemical calculations of ECD spectra.

One-step harvest of high-purity light hydrocarbons without the desorption process represents an advanced and highly efficient strategy for the purification of target substances. The separation and purification of acetylene (C2H2) from carbon dioxide (CO2) by CO2-selective adsorbents are urgently demanded yet are very challenging owing to their similar physicochemical properties. Here, we employ the pore chemistry strategy to adjust the pore environment by immobilizing polar groups into an ultramicroporous metal-organic framework (MOF), achieving one-step manufacture of high-purity C2H2 from CO2/C2H2 mixtures. Embedding methyl groups into prototype stable MOF (Zn-ox-trz) not only changes the pore environment but also improves the discrimination of guest molecules. The methyl-functionalized Zn-ox-mtz thus exhibits the benchmark reverse CO2/C2H2 uptake ratio of 12.6 (123.32/9.79 cm3 cm-3) and an exceptionally high equimolar CO2/C2H2 selectivity of 1064.9 at ambient conditions. Molecular simulations reveal that the synergetic effect of pore confinement and surfaces decorated with methyl groups provides high recognition of CO2 molecules through multiple van der Waals interactions. The column breakthrough experiments suggest that Zn-ox-mtz dramatically achieved the one-step purification capacity of C2H2 from the CO2/C2H2 mixture with a record C2H2 productivity of 2091 mmol kg-1, surpassing all of the CO2-selective adsorbents reported so far. In addition, Zn-ox-mtz exhibits excellent chemical stability under different pH values of aqueous solutions (pH = 1-12). Moreover, the highly stable framework and excellent inverse selective CO2/C2H2 separation performance showcase its promising application as a C2H2 splitter for industrial manufacture. This work paves the way to developing reverse-selective adsorbents for the challenging gas separation process.

Direct hydrogenation of CO2  to methanol using green hydrogen has emerged as a promising method for carbon neutrality, but qualifying catalysts represent a grand challenge. In2 O3 /ZrO2  catalyst has been extensively applied in methanol synthesis due to its superior activity; however, the electronic effect by strong oxides-support interactions between In2 O3  and ZrO2  at the In2 O3 /ZrO2  interface is poorly understood. In this work, abundant In2 O3 /ZrO2  heterointerfaces are engineered in a hollow-structured In2 O3 @ZrO2  heterostructure through a facile pyrolysis of a hybrid metal-organic framework precursor MIL-68@UiO-66. Owing to well-defined In2 O3 /ZrO2  heterointerfaces, the resultant In2 O3 @ZrO2  exhibits superior activity and stability toward CO2  hydrogenation to methanol, which can afford a high methanol selectivity of 84.6% at a conversion of 10.4% at 290 °C, and 3.0 MPa with a methanol space-time yield of up to 0.29 gMeOH  gcat -1  h-1 . Extensive characterization demonstrates that there is a strong correlation between the strong electronic In2 O3 -ZrO2  interaction and catalytic selectivity. At In2 O3 /ZrO2  heterointerfaces, the electron tends to transfer from ZrO2  to In2 O3  surface, which facilitates H2  dissociation and the hydrogenation of formate (HCOO*) and methoxy (CH3 O*) species to methanol. This study provides an insight into the In2 O3 -based catalysts and offers appealing opportunities for developing heterostructured CO2  hydrogenation catalysts with excellent activity.

The pore dimension and surface property directly dictate the transport of guests, endowing diverse gas selective adsorptions to porous materials. It is highly relevant to construct metal-organic frameworks (MOFs) with designable functional groups that can achieve feasible pore regulation to improve their separation performances. However, the role of functionalization in different positions or degrees within framework on the separation of light hydrocarbon has rarely been emphasized. In this context, four isoreticular MOFs (TKL-104-107) bearing dissimilar fluorination are rationally screened out and afforded intriguing differences in the adsorption behavior of C2 H6 and C2 H4 . Ortho-fluoridation of carboxyl allows TKL-105-107 to exhibit enhanced structural stabilities, impressive C2 H6 adsorption capacities (>125 cm3 g-1 ) and desirable inverse selectivities (C2 H6 over C2 H4 ). The more modified ortho-fluorine group and meta-fluorine group of carboxyl have improved the C2 H6 /C2 H4 selectivity and adsorption capacity, respectively, and the C2 H6 /C2 H4 separation potential can be well optimized via linker fine-fluorination. Meanwhile, dynamic breakthrough experiments proved that TKL-105-107 can be used as highly efficient C2 H6 -selective adsorbents for C2 H4 purification. This work highlights that the purposeful functionalization of pore surfaces facilitates the assembly of highly efficient MOF adsorbents for specific gas separation.

Industrial tobacco waste was mainly treated via a reconstituted tobacco process using the paper-making method, which involves aqueous concentrated tobacco waste extract (cTWE) fermentation (aging). The fermentation was done to improve the quality of reconstituted tobacco. However, cTWE is a multi-stress environment that is characterized by low pH (about 4), as well as high sugar (above 150 g/L) and nicotine (above 15 g/L) content. In this study, a specific selection strategy was used to successfully isolate multi-stress-resistant bacterial or fungal strains, that exhibited positive effects on cTWE fermentation, thereby improving the quality of final products. A potential strain Zygosaccharomyces parabailii MC-5K3 was used for the bioaugmentation of cTWE fermentation and it significantly influenced the microbial diversity of the fermented cTWE. Zygosaccharomyces was observed to be the only dominant fungal genus instead of some pathogenic bacterial genera, with an abundance of over 95% after four days, and still more than 80% after a week. Meanwhile, metabolomics profiling showed significant concentration decrease with regard to some flavor-improving relative metabolites, such as 3-hydroxybenzoic acid (log2FC = - 5.25) and sorbitol (log2FC = - 5.54). This finding is extrapolated to be the key influence factor on the quality of the fermented cTWE. The correlation analysis also showed that the alterations in microbial diversity in the fermented cTWE led to some important differential metabolite changes, which finally improved various properties of tobacco products.

Dimethyl phthalate (DMP), diethyl phthalate (DEP), di-n-butyl phthalate (DBP), butyl benzyl phthalate (BBP), bis(2-ethylhexyl) phthalate (DEHP), and di-n-octyl phthalate (DOP) are hazardous chemicals listed as priority pollutants that disrupt endocrine systems. According to available reports, these six priority phthalate esters (PAEs) are considered the most polluting; however, no studies have been conducted on the efficient remediation of these PAEs. We therefore designed and constructed a synthetic bacterial consortium capable of the simultaneous and efficient degradation of six priority PAEs in minimal inorganic salt medium (MSM) and soil. The consortium comprised Glutamicibacter sp. ZJUTW, which demonstrates priority for degrading short-chain PAEs; Cupriavidus sp. LH1, which degrades phthalic acid (PA) and protocatechuic acid (PCA), intermediates of the PAE biodegradation process; and Gordonia sp. GZ-YC7, which efficiently degrades long-chain priority PAEs, including DEHP and DOP. In MSM containing the six mixed PAEs (250 mg/L each), the ZJUTW + YC + LH1 consortium completely degraded the four short-chain PAEs within 48 h, and DEHP (100%) and DOP (62.5%) within 72 h. In soil containing the six mixed PAEs (DMP, DEP, BBP, and DOP, 400 mg/kg each; DBP and DEHP, 500 mg/kg, each), the ZJUTW + YC + LH1 consortium completely degraded DMP, DEP, BBP, and DBP within 6 days, and 70.84% of DEHP and 66.24% of DOP within 2 weeks. The consortium efficiently degraded the six mixed PAEs in both MSM and soil. We thus believe that this synthetic microbial consortium is a strong candidate for the bioremediation of environments contaminated with mixed PAE pollutants.

Evaporation ducts are abnormal states of the atmosphere in the air-sea boundary layer that directly affect the propagation trajectory of electromagnetic (EM) waves. Therefore, an accurate diagnosis of the evaporation duct height (EDH) is important for studying the propagation trajectory of EM waves in evaporation ducts. Most evaporation duct models (EDMs) based on the Monin-Obukhov similarity theory are empirical methods. Different EDMs have different levels of environmental adaptability. Evaporation duct diagnosis methods based on machine learning methods only consider the mathematical relationship between data and do not explore the physical mechanism of evaporation ducts. To solve the above problems, this study observed the meteorological and hydrological parameters of the five layers of the low-altitude atmosphere in the East China Sea on board the research vessel Xiangyanghong 18 in April 2021 and obtained the atmospheric refractivity profile. An evaporation duct multimodel fusion diagnosis method (MMF) based on a library for support vector machines (LIBSVM) is proposed. First, based on the observed meteorological and hydrological data, the differences between the EDH diagnosis results of different EDMs and MMF were analyzed. When ASTD ≥ 0, the average errors of the diagnostic results of BYC, NPS, NWA, NRL, LKB, and MMF are 2.57 m, 2.92 m, 2.67 m, 3.27 m, 2.57 m, and 0.24 m, respectively. When ASTD < 0, the average errors are 2.95 m, 2.94 m, 2.98 m, 2.99 m, 2.97 m, and 0.41 m, respectively. Then, the EM wave path loss accuracy analysis was performed on the EDH diagnosis results of the NPS model and the MMF. When ASTD ≥ 0, the average path loss errors of the NPS model and MMF are 5.44 dB and 2.74 dB, respectively. When ASTD < 0, the average errors are 5.21 dB and 3.46 dB, respectively. The results show that the MMF is suitable for EDH diagnosis, and the diagnosis accuracy is higher than other models.

Massive production and spread application of plastics have led to the accumulation of numerous plastics in the global environment so that the proportion of carbon storage in these polymers also increases. Carbon cycle is of fundamental significance to global climate change and human survival and development. With the continuous increase of microplastics, undoubtedly, there carbons will continue to be introduced into the global carbon cycle. In this paper, the impact of microplastics on microorganisms involved in carbon transformation is reviewed. Micro/nanoplastics affect carbon conversion and carbon cycle by interfering with biological fixation of CO2, microbial structure and community, functional enzymes activity, the expression of related genes, and the change of local environment. Micro/nanoplastic abundance, concentration and size could significantly lead to difference in carbon conversion. In addition, plastic pollution can further affect the blue carbon ecosystem reduce its ability to store CO2 and marine carbon fixation capacity. Nevertheless, problematically, limited information is seriously insufficient in understanding the relevant mechanisms. Accordingly, it is required to further explore the effect of micro/nanoplastics and derived organic carbon on carbon cycle under multiple impacts. Under the influence of global change, migration and transformation of these carbon substances may cause new ecological and environmental problems. Additionally, the relationship between plastic pollution and blue carbon ecosystem and global climate change should be timely established. This work provides a better perspective for the follow-up study of the impact of micro/nanoplastics on carbon cycle.

Heterosis refers to the superior performance of hybrid lines over inbred parental lines. Besides genetic variation, epigenetic differences between parental lines are suggested to contribute to heterosis. However, the precise nature and extent of differences between the parental epigenomes and the reprograming in hybrids that govern heterotic gene expression remain unclear. In this work, we analyzed DNA methylomes and transcriptomes of the widely cultivated and genetically studied elite hybrid rice (Oryza sativa) SY63, the reciprocal hybrid, and the parental varieties ZS97 and MH63, for which high-quality reference genomic sequences are available. We showed that the parental varieties displayed substantial variation in genic methylation at CG and CHG (H = A, C, or T) sequences. Compared with their parents, the hybrids displayed dynamic methylation variation during development. However, many parental differentially methylated regions (DMRs) at CG and CHG sites were maintained in the hybrid. Only a small fraction of the DMRs displayed non-additive DNA methylation variation, which, however, showed no overall correlation relationship with gene expression variation. In contrast, most of the allelic-specific expression (ASE) genes in the hybrid were associated with DNA methylation, and the ASE negatively associated with allelic-specific methylation (ASM) at CHG. These results revealed a specific DNA methylation reprogramming pattern in the hybrid rice and pointed to a role for parental CHG methylation divergence in ASE, which is associated with phenotype variation and hybrid vigor in several plant species.

A new polydentate tetracarboxylic acid with a benzothiadiazole unit (4',4'''-(benzo[c][1,2,5]thiadiazole-4,7-diyl)bis([1,1'-biphenyl]-3,5-dicarboxylic acid), H4BTDBA) has been used to prepare a pH-stable three-dimensional TbIII-based metal-organic framework (MOF) with the formula {[(CH3)2NH2]0.7[Tb2(BTDBA)1.5(lac)0.7(H2O)2]·solvents}n (Hlac = lactic acid, JXUST-19). JXUST-19 exhibits a new (4,4,12)-connected topology based on tetranuclear [Tb4] clusters. JXUST-19 can remain stable when soaked in water for at least 1 week and in aqueous solutions with various pH values (2-12) for 24 h. Fluorescence study indicates JXUST-19 can be employed as a rare turn-on and blue-shift MOF sensor toward benzaldehyde (BZ) and salicylaldehyde (SA). To date, JXUST-19 represents the first TbIII-based turn-on MOF sensor toward salicylaldehyde in aqueous solution, and the fluorescence enhancement and naked-eye detection of BZ have been rarely reported. In addition, JXUST-19 based fluorescent test papers, light-emitting diode lamp beads, and portable composite films were developed to realize naked-eye detection of BZ and SA, which has great potential in practical applications.

Tax1 banding protein 1 (Tax1bp1) was originally identified as an NF-κB regulatory protein that participated in inflammatory, antiviral and innate immune processes. Tax1bp1 also functions as an autophagy receptor that plays a role in autophagy. Our previous study shows that Tax1bp1 protects against cardiomyopathy in STZ-induced diabetic mice. In this study we investigated the role of Tax1bp1 in heart failure. Pressure overload-induced heart failure model was established in mice by aortic banding (AB) surgery, and angiotensin II (Ang II)-induced heart failure model was established by infusion of Ang II through osmotic minipump for 4 weeks. We showed that the expression levels of Tax1bp1 in the heart were markedly increased 2 and 4 weeks after AB surgery. Knockdown of Tax1bp1 in mouse hearts significantly ameliorated both AB- and Ang II infusion-induced heart failure parameters. On the contrary, AB-induced heart failure was aggravated in cardiac-specific Tax1bp1 transgenic mice. Similar results were observed in neonatal rat cardiomyocytes (NRCMs) under Ang II insult. We demonstrated that the pro-heart failure effect of Tax1bp1 resulted from its interaction with the E3 ligase ITCH to promote the transcription factor P73 ubiquitination and degradation, causing enhanced BCL2 interacting protein 3 (BNIP3)-mediated cardiomyocyte apoptosis. Knockdown ITCH or BNIP3 in NRCMs significantly reduced Ang II-induced apoptosis in vitro. Similarly, BNIP3 knockdown attenuated heart failure in cardiac-specific Tax1bp1 transgenic mice. In the left ventricles of heart failure patients, Tax1bp1 expression level was significantly increased; Tax1bp1 gene expression was negatively correlated with left ventricular ejection fraction in heart failure patients. Collectively, the Tax1bp1 increase in heart failure enhances ITCH-P73-BNIP3-mediated cardiomyocyte apoptosis and induced cardiac injury. Tax1bp1 may serve as a potent therapeutic target for the treatment of heart failure.• Cardiac Tax1bp1 transgene mice were more vulnerable to cardiac dysfunction under stress.• Cardiac Tax1bp1 transgene mice were more vulnerable to cardiac dysfunction under stress.• Knockout of Tax1bp1 in mouse hearts ameliorated heart failure induced by pressure overload.• Tax1bp1 interacts with the E3 ligase Itch to promote P73 ubiquitination and degradation, causing enhanced BNIP3-mediated apoptosis.• Tax1bp1 may become a target of new therapeutic methods for treating heart failure.

Nicotine is a harmful pollutant mainly from the waste of tobacco factories. It is necessary to remove nicotine via high efficient strategies such as bioremediation. So far, an increasing number of nicotine degrading strains have been isolated. However, their degrading efficiency and tolerance to high content nicotine is still not high enough for application in real environment. Thus, the modification of nicotine metabolism pathway is obligated and requires comprehensive molecular insights into whole cell metabolism of nicotine degrading strains. Obviously, the development of multi-omics technology has accelerated the mechanism study on microbial degradation of nicotine and supplied more novel strategy of strains modification. So far, three pathways of nicotine degradation, pyridine pathway, pyrrolidine pathway, and the variant of pyridine and pyrrolidine pathway (VPP pathway), have been clearly identified in bacteria. Muti-omics analysis further revealed specific genome architecture, regulation mechanism, and specific genes or enzymes of three pathways, in different strains. Especially, muti-omics analysis revealed that functional modules coexisted in different genome loci and played additional roles on enhanced degradation efficiency in bacteria. Based on the above discovery, genomic editing strategy becomes more feasible to greatly improve bacterial degrading efficiency of nicotine.

Selective-adsorption separation is an energy-efficient technology for the capture of acetylene (C2H2) from carbon dioxide (CO2) and ethylene (C2H4). However, it remains a critical challenge to effectively recognize C2H2 among CO2 and C2H4, owing to their analogous molecule sizes and physical properties. Herein, we report a new microporous metal-organic framework (NUM-14) possessing a carefully tailored pore system containing moderate pore size and nitro-functionalized channel surface for efficient separation of C2H2 from CO2 and C2H4. The activated NUM-14 (namely NUM-14a) exhibits sufficient pore space to acquire excellent C2H2 loading capacity (4.44 mmol g-1) under ambient conditions. In addition, it possesses dense nitro groups, acting as hydrogen bond acceptors, to selectively identify C2H2 molecules rather than CO2 and C2H4. The breakthrough experiments demonstrate the good actual separation ability of NUM-14a for C2H2/CO2 and C2H2/C2H4 mixtures. Furthermore, Grand Canonical Monte Carlo simulations indicate that the pore surface of the NUM-14a has a stronger affinity to preferentially bind C2H2 over CO2 and C2H4 via stronger C-H···O hydrogen bond interactions. This article provides some insights into customizing pore systems with desirable pore sizes and modifying groups in terms of MOF materials toward the capture of C2H2 from CO2 and C2H4 to promote the development of more MOF materials with excellent properties for gas adsorption and separation.
