RESUMO

Although many prokaryotes have glycolysis alternatives, it's considered as the only energy-generating glucose catabolic pathway in eukaryotes. Here, we managed to create a hybrid-glycolysis yeast. Subsequently, we identified an inositol pyrophosphatase encoded by OCA5 that could regulate glycolysis and respiration by adjusting 5-diphosphoinositol 1,2,3,4,6-pentakisphosphate (5-InsP7) levels. 5-InsP7 levels could regulate the expression of genes involved in glycolysis and respiration, representing a global mechanism that could sense ATP levels and regulate central carbon metabolism. The hybrid-glycolysis yeast did not produce ethanol during growth under excess glucose and could produce 2.68 g/L free fatty acids, which is the highest reported production in shake flask of Saccharomyces cerevisiae. This study demonstrated the significance of hybrid-glycolysis yeast and determined Oca5 as an inositol pyrophosphatase controlling the balance between glycolysis and respiration, which may shed light on the role of inositol pyrophosphates in regulating eukaryotic metabolism.

Prostate cancer is the second most common cancer in men worldwide1. Over the past decade, large-scale integrative genomics efforts have enhanced our understanding of this disease by characterizing its genetic and epigenetic landscape in thousands of patients2,3. However, most tumours profiled in these studies were obtained from patients from Western populations. Here we produced and analysed whole-genome, whole-transcriptome and DNA methylation data for 208 pairs of tumour tissue samples and matched healthy control tissue from Chinese patients with primary prostate cancer. Systematic comparison with published data from 2,554 prostate tumours revealed that the genomic alteration signatures in Chinese patients were markedly distinct from those of Western cohorts: specifically, 41% of tumours contained mutations in FOXA1 and 18% each had deletions in ZNF292 and CHD1. Alterations of the genome and epigenome were correlated and were predictive of disease phenotype and progression. Coding and noncoding mutations, as well as epimutations, converged on pathways that are important for prostate cancer, providing insights into this devastating disease. These discoveries underscore the importance of including population context in constructing comprehensive genomic maps for disease.

RESUMO

We propose a novel, to our knowledge, step-adaptive cross-correlation algorithm tailored for distributed acoustic sensing systems based on linear frequency modulation pulses, aiming for rapid demodulation. This algorithm adjusts its step length through an adaptive "successive refinement" search strategy, which greatly improves computational efficiency by reducing the number of cross-correlation computations. Experimental results have shown that the demodulation time can be reduced by approximately 15 times compared to the conventional method, while maintaining the same demodulation result.

A novel fiber sensor for the refractive index sensing of seawater based on a Mach-Zehnder interferometer has been demonstrated. The sensor consisted of a single-mode fiber (SMF)-no-core fiber (NCF)-single-mode fiber structure (shortened to an SNS structure) with a large lateral offset spliced between the two sections of a multimode fiber (MMF). Optimization studies of the multimode fiber length, offset SNS length, and vertical axial offset distance were performed to improve the coupling efficiency of interference light and achieve the best extinction ratio. In the experiment, a large lateral offset sensor was prepared to detect the refractive index of various ratios of saltwater, which were used to simulate seawater environments. The sensor's sensitivity was up to -13,703.63 nm/RIU and -13,160 nm/RIU in the refractive index range of 1.3370 to 1.3410 based on the shift of the interference spectrum. Moreover, the sensor showed a good linear response and high stability, with an RSD of only 0.0089% for the trough of the interference in air over 1 h.

RESUMO

We found an elevation of circulating TFH13 cell subset in asthmatic children and the frequency of TFH13 cells positively correlated with the plasma dust mite-specific IgE levels. These results indicated that TFH13 cell subset may be responsible for the immunopathogenesis of excessive IgE accumulation in children with allergic asthma.

We proposed an optical fiber salinity sensor with a composite Fabry-Perot (F-P) cavity structure for simultaneous measurement of temperature and salinity based on microelectromechanical system (MEMS) technology. The sensor contains two sensing cavities. The silicon cavity is used for temperature sensing, and the seawater cavity processed by the glass microstructure is sensitive to the refractive index of seawater for salinity sensing. At the same time, the influence of the salinity-temperature cross-sensitivity error of the seawater cavity is effectively compensated by using the temperature single parameter sensitivity characteristics of the silicon cavity. The structural design of the sensor seawater cavity includes a cross-shaped groove and a cylindrical fluid cavity. The surface hydrophilicity treatment was performed on the interior of the cavity to solve the effect of no water injection in the cavity caused by the miniaturization of the sensor. The optical path difference (OPD) demodulation method is used to demodulate the two F-P cavities with large dynamic range and high resolution. In the range of 5∼40°C and 5∼ 40 ‰, the temperature and salinity sensitivity of the sensor can reach 110.25 nm/°C and 178.75 nm/‰, respectively, and the resolution can reach 5.02 × 10-3°C and 0.0138‰. It has the advantages of mass production, high stability, and small size, which give it great potential for marine applications.

Deuterium-based isotopic labeling is an important technique for tracking cellular metabolism with the Raman signals analysis of low-wavenumber (LW) C-D bonds and high-wavenumber (HW) C-H bonds. We propose and demonstrate a disposable ultra-miniature fiber probe to detect LW and HW coherent anti-Stokes Raman scattering (CARS) spectra for deuterated compounds simultaneously and bond-selectively sensing. The 10.78 µm diameter disposable fiber probe, comprised of focusing taper as fiber probe head and time-domain walk-off eliminating fiber section with designed length, realizes wide-frequency-interval dual Stokes pulse delivering and focusing. The fiber probe enables quantitative concentration determination with resolution down to 11 mM. The chemical vibration modes of LW region C-D bonds and HW region C-H bonds of the mixture samples of organic compounds and their deuterated counterparts in a simulated cell are simultaneously excited and characterized. The CARS disposable fiber probe introduces a promising handle for in vivo biochemical detection based on isotopic labeling sensing.

At present, cometabolic degradation is an extensive method for the biological removal of high molecular weight polycyclic aromatic hydrocarbons (HMW-PAHs) in the marine environment. However, due to the refractory to degradation and high toxicity, there are few studies on pyrene (PYR) cometabolic degradation with phenanthrene (PHE) as substrate. In this study, a Pseudomonas stutzeri DJP1 strain isolated from sediments was used in the cometabolic system of PHE and PYR. The biomass and the activity of key enzymes such as dehydrogenase and catechol 12 dioxygenase of strain were improved, but the enhancement of biotoxicity resulted in the inhibition of cometabolism simultaneously. Seven metabolites were identified respectively in PYR, PHE degradation cultures. It was speculated that the cometabolism of PHE and PYR had a common phthalic acid pathway, and the degradation pathway of PHE was included in the downstream pathway of PYR. The functional genes such as PhdF, NidD and CatA involved in DJP1 degradation were revealed by Genome analysis. This study provides a reference for the biodegradation of PYR and PHE in real marine environment.

OBJECTIVES: This study aimed to explore the specific pathway of family structure on adolescent health. STUDY DESIGN: This was a cross-sectional study. METHODS: We examined the influence of family structure on adolescent deviant behavior and depression, as well as the mediation roles of parental monitoring and school connectedness, using the multivariate regression and Karlson-Holm-Breen mediation models. RESULTS: Compared with adolescents in intact families, those in non-intact families showed more deviant behaviors and depression. Parental monitoring and school connectedness appeared to be two important pathways from family structure to deviant behavior and depression. In addition, urban and female adolescents in non-intact families showed more deviant behaviors and depression than their rural and male counterparts, respectively. Furthermore, adolescents in reconstituted families showed more deviant behaviors than those in single-parent families. CONCLUSIONS: The behavioral and mental health of adolescents in single-parent or reconstituted families deserves more attention, and interventions should be actively conducted at both family and school level to improve adolescent health.

In this paper, fiber sensor based on Vernier effect for simultaneous measurement of relative humidity (RH) and temperature is proposed. The sensor is fabricated by coating two kinds of ultraviolet (UV) glue with different refractive indexes (RI) and thicknesses on the end face of a fiber patch cord. The thicknesses of two films are controlled to generate the Vernier effect. The inner film is formed by a cured lower-RI UV glue. The exterior film is formed by a cured higher-RI UV glue, of which thickness is much thinner than the inner film. Through the analysis of the Fast Fourier Transform (FFT) of the reflective spectrum, the Vernier effect is formed by the inner lower-RI polymer cavity and the cavity composed of both polymer films. By calibrating the RH and temperature response of two peaks on the envelope of the reflection spectrum, simultaneous measurements of RH and temperature are realized by solving a set of quadratic equations. Experimental results show that the highest RH and temperature sensitivities of the sensor are 387.3 pm/%RH (in 20%RH to 90%RH) and -533.0 pm/°C (in 15 °C to 40 °C), respectively. The sensor has merits of low cost, simple fabrication, and high sensitivity, which makes it very attractive for applications that need to simultaneously monitor these two parameters.

Previous studies have linked poor family support and adverse childhood experiences (ACEs) to increased risk of depression; however, little is known about the interplay between the two when it comes to their effects on depression. Therefore, the current study examined if family support moderated the cumulative effect of ACEs on depression. Based on data from a migrant survey in Shiyan, Hubei Province, in 2019 (N = 1,326), this study used the ordinary least squares method to analyze the effect of ACEs on depression and evaluate whether family support moderated this effect. Higher exposure to ACEs and lower scores of family support were associated with higher depression levels in adulthood. The moderation model indicated that family support significantly moderated the relationship between ACEs and depression. Appropriate interventions to reduce depression should target internal migrants with history of ACEs. Community nurses should consider ACEs as an integral part of psychosocial assessment. Negative effects of ACEs can be reduced through teaching skills that increase effective family interaction and maintain supportive family networks. RESUMO

Developing a strategy to specifically kill cancer cells without inducing obvious damage to normal cells may be of great clinical significance for cancer treatment. In the present study, we developed a new precise personalized strategy named "i-CRISPR" for cancer treatment through adding DNA damage repair inhibitors(i) and inducing cancer cell-specific DNA double strand breaks by CRISPR. Through in vitro and in vivo experiments, we confirmed the efficacy of this strategy in multiple cancer models and revealed the mechanism of cell death. Our strategy might provide a novel concept for precise cancer therapy.

BACKGROUND: Prostate cancer is the second most frequently diagnosed cancer for males worldwide, but the spatial and temporal trends of prostate cancer burden remain unknown in Asia. This study aimed to investigate the changing spatial and temporal trends of incidence, prevalence, mortality, disability-adjusted life year (DALY), and mortality-to-incidence ratio (MIR) of prostate cancer, and their association with the Socio-Demographic Index (SDI) in 48 Asian countries from 1990 to 2019. METHODS: Data were extracted from the Global Health Data Exchange query tool, covering 48 Asian countries from 1990 to 2019. The average annual percent change was calculated to evaluate temporal trends. Spatial autocorrelation analysis was used to obtain spatial patterns, and the association between SDI and prostate cancer burden was estimated using a spatial panel model. RESULTS: In Asia, the age-standardized incidence and prevalence of prostate cancer increased in almost all countries, and its mortality and DALY also increased in over half of the countries. Significantly regional disparities were found in Asia, and the hot spots for incidence, prevalence, mortality, and DALY were all located in Western Asia, the hot spots of percent change also occurred in Western Asia for incidence and DALY. Furthermore, SDI had a positive association with mortality (coef = 2.51, 95% confidence interval [CI]: 2.13-2.90) and negative association with DALY (coef = -14.99, 95% CI: -20.37 to -9.60) and MIR (coef = -0.95, 95%CI: -0.99 to -0.92). CONCLUSIONS: Prostate cancer burden increased rapidly throughout Asia and substantial disparities had persisted between countries. Geographically targeted interventions are needed to reduce the prostate cancer burden throughout Asia and in specific countries.

In this paper, a high sensitivity pressure sensor employing an internal-external cavity Vernier effect is innovatively achieved with the microelectromechanical systems (MEMS) Fabry-Perot (FP) interferometer. The sensor consists of silicon cavity, vacuum cavity, and silicon-vacuum hybrid cavity, which is fabricated by direct bonding a silicon diaphragm with an etched cylindrical cavity and a silicon substrate. By rationally designing the optical lengths of the silicon cavity and silicon-vacuum hybrid cavity to match, the internal-external cavity Vernier effect will be generated. The proposed cascaded MEMS FP structure exhibits a pressure sensitivity of -1.028 nm/kPa by tracking the envelope evolution of the reflection spectrum, which is 58 times that of the silicon-vacuum hybrid cavity. What's more, it owns a minimal temperature sensitivity of 0.041 nm/°C for the envelope spectrum. The MEMS FP sensor based on internal-external cavity Vernier effect as the promising candidate provides an essential guideline for high sensitivity pressure measurement under the characteristic of short FP sensing cavity length, which demonstrates the value to the research community.

At present, the reconstruction error of optical fiber shape sensing is commonly represented by Euclidean distance error. However, the Euclidian error of shape reconstruction will be dependent on the shape complexity, which depends on length, curvature and torsion. In this paper, we establish a reconstruction error model of distributed shape sensing in optical frequency domain reflectometry (OFDR) based on the Frenet-Serret frame and the error delivering theory, which illustrates the relationship between the reconstruction error and parameters such as curvature, torsion, fiber length and strain measurement error. We experimentally verify the feasibility and applicability of the proposed reconstruction error model by distributed optical fiber shape sensing system based on OFDR. The proposed reconstruction error model can provide a prediction of the maximal reconstruction error when the estimated range of curvature, torsion, fiber length of a shape needs to be reconstructed and strain measurement errors of OFDR system are known. It is very useful to judge whether the shape reconstruction error meets the requirement according to the shape to be reconstructed.

Phase interrogation methods for fiber-optic Fabry-Perot (F-P) sensors may inevitably fail in the field due to the influences of irrelevant factors on signal intensity. To address this severe problem, this Letter proposes an intensity self-compensation method (ISCM) to eliminate the consecutive signal fluctuations of a polarization-based F-P interrogation system caused by multiple factors. By providing only the initial intensities of the reference signals, this attempt realizes the real-time intensity compensation of the output signals without affecting their quadrature relationship. Consecutive intensity fluctuations caused by variation of light source power, fiber loss, and polarization state are reduced to 2%-3% by the ISCM. Furthermore, the method performs ideally under dynamic modulation of the sensor. In addition, it can be applied against the inconsistent fluctuations between signals and is suitable for F-P sensors with single or multiple cavities. Owing to the high efficiency, real-time ability, and no moving parts advantage, the proposed method provides an excellent candidate for improving the accuracy and stability of F-P interrogation systems.

In this paper, an improved spectral demodulation algorithm with the ensemble empirical mode decomposition average denoising is proposed to suppress order jumps in the sapphire fiber Fabry-Perot high-temperature sensing system. It is proven that the signal-to-noise ratio of the sensor is closely related to the severity of the demodulation jumps. The proposed algorithm can reduce the fluctuations of key parameters by reducing the noise in the spectrum, thus overcoming this obstacle. The simulations and experiment demonstrate that the algorithm can effectively eliminate the order jumps in both stable and variable temperature environments. The proposed algorithm solves the order jumping problem that has long plagued the demodulation of this system, improves demodulation accuracy, ensures the reliable operation of the high-temperature sensor, and exhibits excellent demodulation performance.

Previous studies have focused on the determinants of basic public health services utilization, such as socioeconomic status and mobility characteristics, rather than on health literacy. Thus, this study aimed to estimate the effect of health literacy on the utilization of basic public health services among Chinese migrants. Based on the 2016 China Migrants Dynamic Survey data (N = 2335), this study used propensity score matching approach to estimate the effect of health literacy on the utilization of basic public health services, including the establishment of health records and receipt of health education, and to explore heterogeneity in this effect based on educational attainment and urban-rural status. The findings show that high levels of health literacy increased the probability of establishing health records and receiving more health education. Also, high levels of health literacy had a positive effect on the utilization of basic public health services among Chinese migrants, especially those with less education or living in urban areas. Policy makers should more carefully consider the actual situation and needs of migrants who are living in urban areas or have less education, constructing more targeted service programs.

The landscape and characteristics of circulating exosomal messenger RNAs (emRNAs) are poorly understood, which hampered the accurate detection of circulating emRNAs. Through comparing RNA sequencing data of circulating exosomes with the corresponding data in tissues, we illustrated the different characteristics of emRNAs compared to tissue mRNAs. We then developed an improved strategy for emRNA detection based on the features of circulating emRNAs. Using the optimized detection strategy, we further validated prostate cancer (PCa) associated emRNAs discovered by emRNA-seq in a large cohort of patients and identified emRNA signatures for PCa screening and diagnosis using logistic regression analysis. The receiver operating characteristic curve (ROC) analysis showed that the circulating emRNA-based screening signature yielded an area under the ROC curve (AUC) of 0.948 in distinguishing PCa patients from healthy controls. The circulating emRNA-based diagnostic signature also showed a great performance in predicting prostate biopsy results (AUC: 0.851). In conclusion, our study developed an optimized emRNA detection strategy and identified novel emRNA signatures for the detection of PCa.

Previous polarization underwater imaging methods based on the physical scattering model usually require background region included in the image and the prior knowledge, which hinders its practical application. In this paper, we analyze and optimize the physically feasible region and propose an improved method by degenerating intermediate variables, which can realize automatic underwater image recovery without background region or any prior. The proposed method does not need to estimate the intermediate variables in the traditional underwater imaging model and is adaptable to the underwater image with non-uniform illumination, which avoids the poor and unstable image recovery performance caused by inaccurate estimation of intermediate parameters due to the improper identification of the background region. Meanwhile, our method is effective for both images without background region and images in which the background region is hard to be identified. In addition, our method solves the significant variation in recovery results caused by the different selection of background regions and the inconsistency of parameter adjustment. The experimental results of different underwater scenes show that the proposed method can enhance image contrast while preserving image details without introducing considerable noise, and the proposed method is effective for the dense turbid medium.
