concepts like mental state concepts lack a physical referent, which can be directly perceived. Classical theories therefore claim that abstract concepts require amodal representations detached from experiential brain systems. However, grounded cognition approaches suggest an involvement of modal experiential brain regions in the processing of abstract concepts. In the present functional magnetic resonance imaging study, we investigated the relation of the processing of abstract mental state concepts to modal experiential brain systems in a fine-grained fashion. Participants performed lexical decisions on abstract mental state as well as on verbal association concepts as control category. Experiential brain systems related to the processing of mental states, generating verbal associations, automatic speech as well as hand and lip movements were determined by corresponding localizer tasks. Processing of abstract mental state concepts neuroanatomically overlapped with activity patterns associated with processing of mental states, generating verbal associations, automatic speech and lip movements. Hence, mental state concepts activate the mentalizing brain network, complemented by perceptual-motor brain regions involved in simulation of visual or action features associated with social interactions, linguistic brain regions as well as face-motor brain regions recruited for articulation. The present results provide compelling evidence for the rich grounding of abstract mental state concepts in experiential brain systems related to mentalizing, verbal communication and mouth action.

Scientific concepts typically transcendent our sensory experiences. Traditional approaches to science education therefore assume a shift towards amodal or verbal knowledge representations during academic training. Grounded cognition approaches, in contrast, predict a maintenance of grounding of the concepts in experiential brain networks or even an increase. To test these competing approaches, the present study investigated the semantic content of scientific psychological concepts and identified the corresponding neural circuits using functional magnetic resonance imaging (fMRI) in undergraduate psychology students (beginners) and in graduated psychologists (advanced learners). During fMRI scanning, participants were presented with words denoting scientific psychological concepts within a lexical decision task (e.g. "conditioning", "habituation"). The individual semantic property content of each concept was related to brain activity during abstract concept processing. In both beginners and advanced learners, visual and motor properties activated brain regions also involved in perception and action, while mental state properties increased activity in brain regions also recruited by emotional-social scene observation. Only in advanced learners, social constellation properties elicited brain activity overlapping with emotional-social scene observation. In line with grounded cognition approaches, the present results highlight the importance of experiential information for constituting the meaning of abstract scientific concepts during the course of academic training.

Refined grounded cognition accounts propose that abstract concepts might be grounded in brain circuits involved in mentalizing. In the present event-related potential (ERP) study, we compared the time course of neural processing in response to semantically predefined abstract mental states and verbal association concepts during a lexical decision task. In addition to scalp ERPs, source estimates of underlying volume brain activity were determined to reveal spatio-temporal clusters of greater electrical brain activity to abstract mental state vs. verbal association concepts, and vice versa. Source estimates suggested early (onset 194 ms), but short-lived enhanced activity (offset 210 ms) to verbal association concepts in left occipital regions. Increased occipital activity might reflect retrieval of visual word form or access to visual conceptual features of associated words. Increased estimated source activity to mental state concepts was obtained in visuo-motor (superior parietal, pre- and postcentral areas) and mentalizing networks (lateral and medial prefrontal areas, insula, precuneus, temporo-parietal junction) with an onset of 212 ms, which extended to later time windows. The time course data indicated two processing phases: An initial conceptual access phase, in which linguistic and modal brain circuits rapidly process features depending on their relevance, and a later conceptual elaboration phase, in which elaborative processing within feature-specific networks further refines the concept. This study confirms the proposal that abstract concepts are based on representations in distinct neural circuits depending on their semantic feature content. The present research also highlights the importance of investigating sets of abstract concepts with a defined semantic content.

Conceptual knowledge is central to cognition. Previous neuroimaging research indicates that conceptual processing involves both modality-specific perceptual-motor areas and multimodal convergence zones. For example, our previous functional magnetic resonance imaging (fMRI) study revealed that both modality-specific and multimodal regions respond to sound and action features of concepts in a task-dependent fashion (Kuhnke P, Kiefer M, Hartwigsen G. 2020b. Task-dependent recruitment of modality-specific and multimodal regions during conceptual processing. Cereb Cortex. 30:3938-3959.). However, it remains unknown whether and how modality-specific and multimodal areas interact during conceptual tasks. Here, we asked 1) whether multimodal and modality-specific areas are functionally coupled during conceptual processing, 2) whether their coupling depends on the task, 3) whether information flows top-down, bottom-up or both, and 4) whether their coupling is behaviorally relevant. We combined psychophysiological interaction analyses with dynamic causal modeling on the fMRI data of our previous study. We found that functional coupling between multimodal and modality-specific areas strongly depended on the task, involved both top-down and bottom-up information flow, and predicted conceptually guided behavior. Notably, we also found coupling between different modality-specific areas and between different multimodal areas. These results suggest that functional coupling in the conceptual system is extensive, reciprocal, task-dependent, and behaviorally relevant. We propose a new model of the conceptual system that incorporates task-dependent functional interactions between modality-specific and multimodal areas.

INTRODUCTION: There are reports of an increase in depressive symptoms and fear during the COVID-19 pandemic, in particular in patients with depression. This study investigates factors related to fear of COVID-19 in former inpatients suffering from depression and healthy controls by assessing variables typically associated with depression and anxiety disorders, i.e. stressful life events (SLEs), the primary emotions SADNESS, PLAY and SEEKING as well as dysfunctional emotion regulation strategies with respect to suppression and reappraisal. METHODS: Data of n = 44 former inpatients suffering from depression and n = 49 healthy controls were collected. The study had a longitudinal design with two measurement points. Before the pandemic, SLEs, primary emotions, emotion regulation and depression severity were assessed. During the pandemic, COVID-19 associated stressors and life events, emotion regulation, depression severity and fear of COVID-19 were assessed. RESULTS: Fear of COVID-19 and depression severity during the pandemic were significantly higher in former inpatients than in healthy controls. Depression diagnosis, SLEs and depression severity before the pandemic were significant positive predictors of fear of COVID-19. The primary emotion PLAY was a significant negative predictor of fear of COVID-19. Depression severity did not change significantly in healthy controls. CONCLUSION: The results show that risk factors for depression might be risk factors for high fear of COVID-19. In addition, a playful personality could help preventing mental stress in pandemic situations. Thus, positivity based interventions could counteract elevated fear scores during a pandemic.

Grounded cognition theories assume that conceptual processing depends on modality-specific brain systems in a context-dependent fashion. Although the relation of abstract concepts to modality-specific systems is less obvious than for concrete concepts, recent behavioral and neuroimaging studies indicated a foundation of abstract concepts in vision and action. However, due to their poor temporal resolution, neuroimaging studies cannot determine whether sensorimotor activity reflects rapid access to conceptual information or later conceptual processes. The present study therefore assessed the time course of abstract concept processing using event-related potentials (ERPs) and compared ERP responses to abstract concepts with a strong relation to vision or action. We tested whether possible ERP effects to abstract word categories would emerge in early or in later time windows and whether these effects would depend on the depth of the conceptual task. In Experiment 1, a shallow lexical decision task, early feature-specific effects starting at 178 ms were revealed, but later effects beyond 300 ms were also observed. In Experiment 2, a deep conceptual decision task, feature-specific effects with an onset of 22 ms were obtained, but effects again extended beyond 300 ms. In congruency with earlier neuroimaging work, the present feature-specific ERP effects suggest a grounding of abstract concepts in modal brain systems. The presence of early and late feature-specific effects indicates that sensorimotor activity observed in neuroimaging experiments may reflect both rapid conceptual and later post-conceptual processing. Results furthermore suggest that a deep conceptual task accelerates access to conceptual sensorimotor features, thereby demonstrating conceptual flexibility.

Conceptual knowledge is central to cognitive abilities such as word comprehension. Previous neuroimaging evidence indicates that concepts are at least partly composed of perceptual and motor features that are represented in the same modality-specific brain regions involved in actual perception and action. However, it is unclear to what extent the retrieval of perceptual-motor features and the resulting engagement of modality-specific regions depend on the concurrent task. To address this issue, we measured brain activity in 40 young and healthy participants using functional magnetic resonance imaging, while they performed three different tasks-lexical decision, sound judgment, and action judgment-on words that independently varied in their association with sounds and actions. We found neural activation for sound and action features of concepts selectively when they were task-relevant in brain regions also activated during auditory and motor tasks, respectively, as well as in higher-level, multimodal regions which were recruited during both sound and action feature retrieval. For the first time, we show that not only modality-specific perceptual-motor areas but also multimodal regions are engaged in conceptual processing in a flexible, task-dependent fashion, responding selectively to task-relevant conceptual features.

BACKGROUND: An early onset of depression is associated with higher chronicity and disability, more stressful life events (SLEs), higher negative emotionality as described by the primary emotion SADNESS and more severe depressive symptomatology compared to depression onset later in life. Additionally, methylation of the serotonin transporter gene (SLC6A4) is associated with SLEs and depressive symptoms. METHODS: We investigated the relation of SLEs, SLC6A4 methylation in peripheral blood, the primary emotions SADNESS and SEEKING (measured by the Affective Neuroscience Personality Scales) as well as depressive symptom severity to age at depression onset in a sample of N = 146 inpatients suffering from major depression. RESULTS: Depressed women showed higher SADNESS (t (91.05) = - 3.17, p = 0.028, d = - 0.57) and higher SLC6A4 methylation (t (88.79) = - 2.95, p = 0.02, d = - 0.55) compared to men. There were associations between SLEs, primary emotions and depression severity, which partly differed between women and men. The Akaike information criterion (AIC) indicated the selection of a model including sex, SLEs, SEEKING and SADNESS for the prediction of age at depression onset. SLC6A4 methylation was not related to depression severity, age at depression onset or SLEs in the entire group, but positively related to depression severity in women. CONCLUSIONS: Taken together, we provide further evidence that age at depression onset is associated with SLEs, personality and depression severity. However, we found no associations between age at onset and SLC6A4 methylation. The joint investigation of variables originating in biology, psychology and psychiatry could make an important contribution to understanding the development of depressive disorders by elucidating potential subtypes of depression.

Conceptual knowledge is central to human cognition. The left posterior inferior parietal lobe (pIPL) is implicated by neuroimaging studies as a multimodal hub representing conceptual knowledge related to various perceptual-motor modalities. However, the causal role of left pIPL in conceptual processing remains unclear. Here, we transiently disrupted left pIPL function with transcranial magnetic stimulation (TMS) to probe its causal relevance for the retrieval of action and sound knowledge. We compared effective TMS over left pIPL with sham TMS, while healthy participants performed three different tasks-lexical decision, action judgment, and sound judgment-on words with a high or low association to actions and sounds. We found that pIPL-TMS selectively impaired action judgments on low sound-low action words. For the first time, we directly related computational simulations of the TMS-induced electrical field to behavioral performance, which revealed that stronger stimulation of left pIPL is associated with worse performance for action but not sound judgments. These results indicate that left pIPL causally supports conceptual processing when action knowledge is task-relevant and cannot be compensated by sound knowledge. Our findings suggest that left pIPL is specialized for the retrieval of action knowledge, challenging the view of left pIPL as a multimodal conceptual hub.

BACKGROUND: The angiosperm family Bromeliaceae comprises over 3.500 species characterized by exceptionally high morphological and ecological diversity, but a very low genetic variation. In many genera, plants are vegetatively very similar which makes determination of non flowering bromeliads difficult. This is particularly problematic with living collections where plants are often cultivated over decades without flowering. DNA barcoding is therefore a very promising approach to provide reliable and convenient assistance in species determination. However, the observed low genetic variation of canonical barcoding markers in bromeliads causes problems. RESULT: In this study the low-copy nuclear gene Agt1 is identified as a novel DNA barcoding marker suitable for molecular identification of closely related bromeliad species. Combining a comparatively slowly evolving exon sequence with an adjacent, genetically highly variable intron, correctly matching MegaBLAST based species identification rate was found to be approximately double the highest rate yet reported for bromeliads using other barcode markers. CONCLUSION: In the present work, we characterize Agt1 as a novel plant DNA barcoding marker to be used for barcoding of bromeliads, a plant group with low genetic variation. Moreover, we provide a comprehensive marker sequence dataset for further use in the bromeliad research community.

Recent studies using an induction task paradigm indicated that unconscious automatic processes underlying masked semantic priming are susceptible to cognitive control influences. In this paradigm, participants first perform different induction tasks (semantic decision vs. perceptual decision), which serve to activate a corresponding task set. Thereafter, the masked prime and the target for the lexical decision task is presented. Previously, perceptual and semantic induction tasks were presented in separate blocks, and the response to the induction task was given immediately after the inducing stimulus. The present study, therefore, tested two possible boundary conditions, flexibility of cognitive control and completeness of task set execution, for the emergence of task set effects on masked semantic priming. In the first experiment, perceptual and semantic induction tasks were presented in a randomized fashion, to assess whether task set influences on masked semantic priming can occur on a trial-by-trial basis. The other two experiments tested whether task set effects on masked priming survive, when the response to the induction task is delayed. The present study yielded the same pattern of results irrespective of the variations in the induction task paradigm: When the masked prime was shortly presented after the induction task, masked semantic priming was larger subsequent to the semantic than subsequent to the perceptual induction task. The present study shows that task sets can configure unconscious processing streams rapidly on a trial-by-trial basis and demonstrates the generalizability of cognitive control effects on masked semantic priming across variations of the induction task paradigm.

BACKGROUND: At the end of the Pliocene and the beginning of Pleistocene glaciation and deglaciation cycles Ginkgo biloba went extinct all over the world, and only few populations remained in China in relict areas serving as sanctuary for Tertiary relict trees. Yet the status of these regions as refuge areas with naturally existing populations has been proven not earlier than one decade ago. Herein we elaborated the hypothesis that during the Pleistocene cooling periods G. biloba expanded its distribution range in China repeatedly. Whole plastid genomes were sequenced, assembled and annotated, and sequence data was analyzed in a phylogenetic framework of the entire gymnosperms to establish a robust spatio-temporal framework for gymnosperms and in particular for G. biloba Pleistocene evolutionary history. RESULTS: Using a phylogenetic approach, we identified that Ginkgoatae stem group age is about 325 million years, whereas crown group radiation of extant Ginkgo started not earlier than 390,000 years ago. During repeated warming phases, Gingko populations were separated and isolated by contraction of distribution range and retreated into mountainous regions serving as refuge for warm-temperate deciduous forests. Diversification and phylogenetic splits correlate with the onset of cooling phases when Ginkgo expanded its distribution range and gene pools merged. CONCLUSIONS: Analysis of whole plastid genome sequence data representing the entire spatio-temporal genetic variation of wild extant Ginkgo populations revealed the deepest temporal footprint dating back to approximately 390,000 years ago. Present-day directional West-East admixture of genetic diversity is shown to be the result of pronounced effects of the last cooling period. Our evolutionary framework will serve as a conceptual roadmap for forthcoming genomic sequence data, which can then provide deep insights into the demographic history of Ginkgo.

Unconscious visuomotor priming defined as the advantage in reaction time (RT) or accuracy for target shapes mapped to the same (congruent condition) when compared with a different (incongruent condition) motor response as a preceding subliminally presented prime shape has been shown to modulate activity within a visuomotor network comprised of parietal and frontal motor areas in previous functional magnetic resonance imaging (fMRI) studies. The present fMRI study investigated whether, in addition to changes in brain activity, unconscious visuomotor priming results in a modulation of functional connectivity profiles. Activity associated with congruent compared with incongruent trials was lower in the bilateral inferior and medial superior frontal gyri, in the inferior parietal lobules, and in the right caudate nucleus and adjacent portions of the thalamus. Functional connectivity increased under congruent relative to incongruent conditions between ventral visual stream areas (e.g., calcarine, fusiform, and lingual gyri), the precentral gyrus, the supplementary motor area, posterior parietal areas, the inferior frontal gyrus, and the caudate nucleus. Our findings suggest that an increase in coupling between visuomotor regions, reflecting higher efficiency of processing, is an important neural mechanism underlying unconscious visuomotor priming, in addition to changes in the magnitude of activation.

BACKGROUND: The present study investigated individual differences in the Affective Neuroscience Personality Scales (ANPS), representing measures of primary emotional systems, and depressive tendencies in two independent samples. METHODS: In order to be able to find support for a continuum model with respect to the relation of strength in the cross-species "affective neuroscience" taxonomy of primary emotional systems, we investigated ANPS measured personality traits in a psychologically mostly healthy population (n=614 participants) as well as a sample of clinically depressed people (n=55 depressed patients). RESULTS: In both normal and depressed samples robust associations appeared between higher FEAR and SADNESS scores and depressive tendencies. A similar - albeit weaker - association was observed with lower SEEKING system scores and higher depressive tendencies, an effect again seen in both samples. LIMITATIONS: The study is of cross-sectional nature and therefore only associations between primary emotional systems and depressive tendencies were evaluated. CONCLUSIONS: These results show that similar associations between ANPS monitored primary emotional systems and tendencies toward depression can be observed in both healthy and depressed participants. This lends support for a continuum of affective changes accompanying depression, potentially reflecting differences in specific brain emotional system activities in both affectively normal as well as clinically depressed individuals.

Recent evidence suggests an interaction between the ventral visual-perceptual and dorsal visuo-motor brain systems during the course of object recognition. However, the precise function of the dorsal stream for perception remains to be determined. The present study specified the functional contribution of the visuo-motor system to visual object recognition using functional magnetic resonance imaging and event-related potential (ERP) during action priming. Primes were movies showing hands performing an action with an object with the object being erased, followed by a manipulable target object, which either afforded a similar or a dissimilar action (congruent vs. incongruent condition). Participants had to recognize the target object within a picture-word matching task. Priming-related reductions of brain activity were found in frontal and parietal visuo-motor areas as well as in ventral regions including inferior and anterior temporal areas. Effective connectivity analyses suggested functional influences of parietal areas on anterior temporal areas. ERPs revealed priming-related source activity in visuo-motor regions at about 120 ms and later activity in the ventral stream at about 380 ms. Hence, rapidly initiated visuo-motor processes within the dorsal stream functionally contribute to visual object recognition in interaction with ventral stream processes dedicated to visual analysis and semantic integration.

Men have been frequently found to perform more accurately than women in mental rotation tasks. However, men and women also differ with regard to the habitual use of emotion regulation strategies, particularly with regard to expressive suppression, i.e., the suppression of emotional expression in behavior. As emotional suppression is more often used by men, emotion regulation strategies might be a variable modulating gender differences in mental rotation performance. The present study, therefore, examined the influences of gender and emotion regulation strategies on mental rotation performance accuracy and feedback processing. Twenty-eight men and 28 women matched for relevant demographic variables performed mental rotation tasks of varying difficulty over a prolonged time. Emotional feedback was given immediately after each trial. Results showed that women reported to use expressive suppression less frequently than men. Women made more errors in the mental rotation task than men confirming earlier demonstrations of gender differences. Furthermore, women were more impaired by the negative feedback as indicated by the increased likelihood of subsequent errors compared with men. Task performance of women not habitually using expressive suppression was most inferior and most strongly influenced by failure feedback compared with men. Women using expressive suppression more habitually did not significantly differ in mental rotation accuracy and feedback processing from men. Hence, expressive suppression reduces gender differences in mental rotation accuracy by improving cognitive performance following failure feedback.

Evaluative priming by masked emotional stimuli that are not consciously perceived has been taken as evidence that affective stimulus evaluation can also occur unconsciously. However, as masked priming effects were small and frequently observed only for familiar primes that there also presented as visible targets in an evaluative decision task, priming was thought to reflect primarily response activation based on acquired S-R associations and not evaluative semantic stimulus analysis. The present study therefore assessed across three experiments boundary conditions for the emergence of masked evaluative priming effects with unfamiliar primes in an evaluative decision task and investigated the role of the frequency of target repetition on priming with pictorial and verbal stimuli. While familiar primes elicited robust priming effects in all conditions, priming effects by unfamiliar primes were reliably obtained for low repetition (pictures) or unrepeated targets (words), but not for targets repeated at a high frequency. This suggests that unfamiliar masked stimuli only elicit evaluative priming effects when the task set associated with the visible target involves evaluative semantic analysis and is not based on S-R triggered responding as for high repetition targets. The present results therefore converge with the growing body of evidence demonstrating attentional control influences on unconscious processing.

Classical theories of semantic memory assume that concepts are represented in a unitary amodal memory system. In challenging this classical view, pure or hybrid modality-specific theories propose that conceptual representations are grounded in the sensory-motor brain areas, which typically process sensory and action-related information. Although neuroimaging studies provided evidence for a functional-anatomical link between conceptual processing of sensory or action-related features and the sensory-motor brain systems, it has been argued that aspects of such sensory-motor activation may not directly reflect conceptual processing but rather strategic imagery or postconceptual elaboration. In the present ERP study, we investigated masked effects of acoustic and action-related conceptual features to probe unconscious automatic conceptual processing in isolation. Subliminal feature-specific ERP effects at frontocentral electrodes were observed, which differed with regard to polarity, topography, and underlying brain electrical sources in congruency with earlier findings under conscious viewing conditions. These findings suggest that conceptual acoustic and action representations can also be unconsciously accessed, thereby excluding any postconceptual strategic processes. This study therefore further substantiates a grounding of conceptual and semantic processing in action and perception.
