RESUMO

The placenta, as a "transit station" between mother and fetus, has functions delivering nutrients, excreting metabolic wastes and secreting hormones. A healthy placenta is essential for fetal growth and development while the melatonergic system seems to play a critical physiological role in this organ since melatonin, its synthetic enzymes and receptors are present in the placenta. In current study, Mtnr1a and Mtnr1b knockout mice were constructed to explore the potential roles of melatonergic system played on the placental function and intrauterine growth retardation (IUGR). The result showed that Mtnr1a knockout had little effect on placental function while Mtnr1b knockout reduced placental efficiency and increased IUGR. Considering the extremely high incidence of IURG in sows, the pregnant sows were treated with melatonin. This treatment reduced the incidence of IUGR. All the evidence suggests that the intact melatonergic system in placenta is required for its function. Mechanistical studies uncovered that Mtnr1b knockout increased placental oxidative stress and apoptosis but reduced the angiogenesis. The RNA sequencing combined with histochemistry study identified the reduced angiogenesis and placental vascular density in Mtnr1b knockout mice. These alterations were mediated by the disrupted STAT3/VEGFR2/PI3K/AKT pathway, i.e., Mtnr1b knockout reduced the phosphorylation of STAT3 which is the promotor of VEGFR2. The downregulated VEGFR2 and its downstream elements of PI3K and AKT expressions, then, jeopardizes the angiogenesis and placental development.

RESUMO

Beijing You Chicken, a valuable local chicken breed from Beijing, China, was once listed as an endangered breed. From the point of view of conservation, the preservation of this breed is an important task for the local researchers. Semen cryopreservation is a popular method to maintain valuable species. However, during cryopreservation, semen is susceptible to oxidative damage. Melatonin is a potent antioxidant and free radical scavenger, so it has been selected to improve the efficiency of sperm cryopreservation. In this study, the chicken semen was treated with different concentrations of melatonin in the cryopreservation solution. The results showed that melatonin at concentrations of 10-3 M and 10-5 M significantly improved sperm progressive motility and total motility, respectively, compared to the control (P < 0.05). Melatonin at 10-3 M also significantly improved the plasma membrane and acrosome integrity of spermatozoa compared to the control. The mechanisms are that melatonin significantly reduces the level of ROS and preserves sperm mitochondrial membrane potential. Most importantly, the melatonin-treated cryopreserved chicken sperm after artificial insemination significantly increased the hatching rate of chicks compared to the control (p < 0.05). The results show that melatonin has a positive effect on the quality of the cryopreserved spermatozoa. These results provide the theoretical and practical basis for using melatonin to improve Beijing You Chicken conservation, and they may also be applicable to poultry as a whole.

RESUMO

Drought stress is a crucial environmental factor that limits plant growth, development, and productivity. Autophagy of misfolded proteins can help alleviate the damage caused in plants experiencing drought. However, the mechanism of autophagy-mediated drought tolerance in plants remains largely unknown. Here, we cloned the gene for a maize (Zea mays) selective autophagy receptor, NEXT TO BRCA1 GENE 1 (ZmNBR1), and identified its role in the response to drought stress. We observed that drought stress increased the accumulation of autophagosomes. RNA sequencing and reverse transcription-quantitative polymerase chain reaction showed that ZmNBR1 is markedly induced by drought stress. ZmNBR1 overexpression enhanced drought tolerance, while its knockdown reduced drought tolerance in maize. Our results established that ZmNBR1 mediates the increase in autophagosomes and autophagic activity under drought stress. ZmNBR1 also affects the expression of genes related to autophagy under drought stress. Moreover, we determined that BRASSINOSTEROID INSENSITIVE 1A (ZmBRI1a), a brassinosteroid receptor of the BRI1-like family, interacts with ZmNBR1. Phenotype analysis showed that ZmBRI1a negatively regulates drought tolerance in maize, and genetic analysis indicated that ZmNBR1 acts upstream of ZmBRI1a in regulating drought tolerance. Furthermore, ZmNBR1 facilitates the autophagic degradation of ZmBRI1a under drought stress. Taken together, our results reveal that ZmNBR1 regulates the expression of autophagy-related genes, thereby increasing autophagic activity and promoting the autophagic degradation of ZmBRI1a under drought stress, thus enhancing drought tolerance in maize. These findings provide new insights into the autophagy degradation of brassinosteroid signaling components by the autophagy receptor NBR1 under drought stress.

RESUMO

Afforestation and reforestation are pivotal in mitigating land degradation and bolstering the carbon sink capacity of terrestrial ecosystems. However, the potential economic ramifications of afforestation and reforestation in the context of climate change remain largely unexplored. In this study, we employed an interdisciplinary methodology to establish a framework for assessing future forest potential and carbon sequestration in the Eastern Loess Plateau region of China. Our findings indicate that an estimated 17,392.99 km2 of land suitable for afforestation still existed within the region, exhibiting a propensity to aggregate around existing forests rather than being dispersed randomly. Notably, 4385.36 km2 was prioritized for afforestation initiatives. Projections suggest a significant enhancement of the forest carbon sink within the study area by 2050, ranging from 36.93 Mt to 105.38 Mt. The corresponding economic value for this enhancement is estimated to vary between US$3.25 billion and US$17.68 billion. Of significance is the observed polarization of the region's carbon sink capacity over time, with half of the total carbon sinks concentrated within 10% of the districts. Additionally, approximately 26% of the counties are expected to transition from carbon sinks to carbon sources. These findings underscore the substantial impact of climate change on forest distribution and suggest a targeted approach to combat forest degradation by circumventing ineffective afforestation activities.

RESUMO

MYB transcription factors play a vital role in response to stress in plant. MYB-CC transcription factors belong to MYB transcription factors, which contain a conserved MYB DNA-binding domain and a coiled-coil (CC) domain. MYB-CC transcription factors participate in the process of plant drought tolerance. However, the underlying molecular mechanisms of ZmMYB-CC in regulating drought tolerance are still largely unknown. Here, we found that ZmMYB-CC10 enhanced drought tolerance by reducing oxidative damage in maize. Further, ZmMYB-CC10 improves the activity of APX and decreases the content of H2O2. Overexpression of ZmMYB-CC10 increases the expression of ZmAPX4 under drought stress. Luciferase assays and Yeast one-hybrid assays (Y1H) showed that ZmMYB-CC10 activates the expression of ZmAPX4 by directly binding to its promoter. Taken together, our results demonstrate that ZmMYB-CC10 enhances tolerance to drought stress by directly activating ZmAPX4 expression, thereby reducing H2O2 content.

RESUMO

Melatonin is a pleiotropic molecule with a variety of biological functions, which include its immunoregulatory action in mammals. Brucellosis is a worldwide endemic zoonotic disease caused by the Brucella, which not only causes huge economic losses for the livestock industry but also impacts human health. To target this problem, in current study, two marker-free transgenic sheep overexpressing melatonin synthetic enzyme ASMT (acetylserotonin O-methyltransferase) gene were generated and these melatonin enrich transgenic sheep were challenged by Brucella infection. The results showed that the serum melatonin concentration was significantly higher in transgenic sheep than that of wild type (726.92 ± 70.6074 vs 263.10 ± 34.60 pg/mL, P < .05). Brucella challenge test showed that two thirds (4/6) of the wild-type sheep had brucellosis, while none of the transgenic sheep were infected. Whole-blood RNA-seq results showed that differential expression genes (DEGs) were significantly enriched in natural killer cell-mediated cytotoxicity, phagosome, antigen processing, and presentation signaling pathways in overexpression sheep. The DEGs of toll-like receptors (TLRs) and NOD-like receptors (NLRs) families were verified by qPCR and it showed that TLR1, TLR2, TLR7, CD14, NAIP, and CXCL8 expression levels in overexpression sheep were significantly higher and NLRP1, NLRP3, and TNF expression levels were significantly lower than those of wild type. The rectal feces were subjected to 16S rDNA amplicon sequencing, and the microbial functional analysis showed that the transgenic sheep had significantly lower abundance of microbial genes related to infectious diseases compared to the wild type, indicating overexpression animals are likely more resistant to infectious diseases than wild type. Furthermore, exogenous melatonin treatment relieved brucellosis inflammation by upregulating anti-inflammatory cytokines IL-4 and downregulating pro-inflammatory IL-2, IL-6, and IFN-γ. Our preliminary results provide an informative reference for the study of the relationship between melatonin and brucellosis.

RESUMO

Land use change driven by human activities plays a critical role in the terrestrial carbon budget through habitat loss and vegetation change. Despite the projections of the global population and economic growth under the framework of the Shared Socioeconomic Pathways (SSPs), little is known of land use/cover change (LUCC) at a fine spatial resolution and how carbon pools respond to LUCC under different SSPs. This study projected the future global LUCC with 1 km spatial resolution and a 10-year time step from 2010 to 2100 and then explored its direct impacts on aboveground biomass carbon (AGB) under SSPs. Scenario SSP3 yields the highest global cropland expansion, among which approximately 48% and 46% is expected to be located in the current forest land and grassland, respectively. Scenario SSP1 has the largest forest expansion and is mainly converted from grassland (54%) and cropland (30%). Due to the spatial change in land use/cover, global AGB loss is expected to reach approximately 3.422 Pg C in 2100 under scenario SSP3 and increases by approximately 0.587 Pg C under scenario SSP1. Africa is expected to lose 30% of AGB under the scenario SSP3. Aboveground biomass in Asia will fix 0.774 Pg C to reverse the AGB loss in 2100 under scenario SSP1. The global carbon loss estimated by the land use products with 10 km and 25 km resolution are less than that with 1 km by 1.5% (ranging from -11.2% in Africa to +34.0% in Oceania) and 2.9% (ranging from -11.8% in Africa to +24.0% in Oceania), respectively. These findings suggest that sufficient spatial details in the existing SSP scenario projections could reduce the uncertainties of AGB assessment, and reasonable land use development and management is a key measure to mitigate the negative impacts of LUCC on the biomass carbon pool.

RESUMO

BACKGROUND: Transgenic animal production is an important means of livestock breeding and can be used to model pharmaceutical applications. METHODS: In this study, to explore the biological activity of endogenously produced melatonin, Acetylserotonin-O-methyltransferase (ASMT)-overexpressed melatonin-enriched dairy goats were successfully generated through the use of pBC1-ASMT expression vector construction and prokaryotic embryo microinjection. RESULTS: These transgenic goats have the same normal phenotype as the wild-type goats (WT). However, the melatonin levels in their blood and milk were significantly increased (p < 0.05). In addition, the quality of their milk was also improved, showing elevated protein content and a reduced somatic cell number compared to the WT goats. No significant changes were detected in the intestinal microbiota patterns between groups. When the animals were challenged by the intravenous injection of E. coli, the ASMT-overexpressed goats had a lower level of pro-inflammatory cytokines and higher anti-inflammatory cytokines compared to the WT goats. Metabolic analysis uncovered a unique arachidonic acid metabolism pattern in transgenic goats. CONCLUSIONS: The increased melatonin production due to ASMT overexpression in the transgenic goats may have contributed to their improved milk quality and enhanced the anti-inflammatory ability compared to the WT goats.

RESUMO

Corpus luteum (CL) plays a critical role in mammalian reproductive physiology. Its dysfunction will lead to infertility or habitual abortion. In the current study, by use of melatonin specific membrane receptor 2 (MT2) knocking out (KO) mice model combined with RNA-Seq, immunohistochemistry, and immunofluorescence analyses, the genes of melatonin synthetic enzyme arylalkylamine N-acetyltransferase (AANAT) and MT2 were identified to strongly express in the CL of sows and mice. KO MT2 significantly impaired the reproductive performance in mice indicated by the reduced litter sizes. Melatonin treatment elevated the progesterone production in sows suggesting the improved CL function. Mechanistic analysis showed that melatonin upregulated a set of progesterone synthesis-related genes including cytochrome P450 family 11 subfamily A member 1 (Cyp11a1), aldo-keto reductase family 1, member C18 (Akr1c18), isopentenyl-diphosphate delta isomerase 1 (Idi1), and luteinizing hormone/choriogonadotropin receptor (Lhcgr). The upregulation of these genes directly related to the increased progesterone production. The regulatory effects of melatonin on these gene expressions were mediated by MT2 and MT2KO diminished the effects of melatonin in this respect. Thus, the presence of melatonergic system of AANAT, melatonin, and its receptor MT2 in CL is essential for reproductive success in mammals.

RESUMO

Leydig cells play a critical role in male reproductive physiology, and their dysfunction is usually associated with male infertility. Melatonin has an important protective and regulatory role in these cells. However, the lack of suitable animal models impedes us from addressing the impact of endogenous melatonin on these cells. In the current study, by using arylalkylamine N-acetyltransferase (AANAT) overexpression transgenic sheep and AANAT knockout mice, we confirmed the regulatory effects of endogenously occurring melatonin on Leydig cells as well as its beneficial effects on male reproductive performance. The results showed that the endogenously elevated melatonin level was correlated with decreased Leydig cell apoptosis, increased testosterone production, and improved quality of sperm in melatonin-enriched transgenic mammals. Signal transduction analysis indicated that melatonin targeted the mitochondrial apoptotic Bax/Bcl2 pathway and thus suppressed Leydig cell apoptosis. In addition, melatonin upregulated the expression of testosterone synthesis-related genes of Steroidogenic Acute Regulatory Protein (StAR), Steroidogenic factor 1 (SF1), and Transcription factor GATA-4 (Gata4) in Leydig cells. This action was primarily mediated by the melatonin nuclear receptor RAR-related orphan receptor alpha (RORα) since blockade of this receptor suppressed the effect of melatonin on testosterone synthesis. All of these actions of melatonin cause Leydig cells to generate more testosterone, which is necessary for spermatogenesis in mammals. In contrast, AANAT knockout animals have dysfunctional Leydig cells and reduced reproductive performance.

RESUMO

In recent years, flexible perovskite solar cells have drawn tremendous attention in the field of wearable devices, and optimization of perovskite composition plays an important role in improving film quality and photophysical properties. At present, some researchers have only studied A-site organic cations mixing or X-site halide anions mixing in the ABX3 structure of perovskite, but there are few reports on co-mixing of A-site and X-site ions in flexible perovskite solar cells. In this paper, we mainly try to study the effects of different concentrations of mixed formamidine methylamine halide (FAxMA1-xBrxClyI1-x-y) precursor solutions on the quality and photophysical properties of perovskite films under low temperature process. We conclude that the film quality and photophysical properties reached the best results when the optimized precursor solution concentration was 60:6:6. The investigation on composition optimization in this experiment laid the foundation for the improvement of the performance of flexible perovskite solar cells. We also use the results of this experiment to prepare flexible perovskite solar cells based on carbon electrodes, which are expected to be applied in other flexible optoelectronic or electro-optical devices.

RESUMO

NLRP (NACHT, LRR and PYD domain-containing proteins) family plays pivotal roles in mammalian reproduction. Mutation of NLRP7 is often associated with human recurrent hydatidiform moles. Few studies regarding the functions of NLRP7 have been performed in other mammalian species rather than humans. In the current study, for the first time, the function of NLRP7 has been explored in ovine ovary. NLRP7 protein was mainly located in ovarian follicles and in in vitro pre-implantation embryos. To identify its origin, 763 bp partial CDS of NLRP7 deriving from sheep cumulus oocyte complexes (COCs) was cloned, it showed a great homology with Homo sapiens. The high levels of mRNA and protein of NLRP7 were steadily expressed in oocytes, parthenogenetic embryos or IVF embryos. NLRP7 knockdown by the combination of siRNA and shRNA jeopardized both the parthenogenetic and IVF embryo development. These results strongly suggest that NLRP7 plays an important role in ovine reproduction. The potential mechanisms of NLRP7 will be fully investigated in the future.

RESUMO

α-Ketoglutarate (α-KG) is an intermediary metabolite in the tricarboxylic acid (TCA) cycle and functions to inhibit ATPase and maintain the pluripotency of embryonic stem cells (ESCs); however, little is known regarding the effects of α-KG on the development of preimplantation embryos. Herein, we report that α-KG (150 µM) treatment significantly promoted the blastocyst rate, the number of inner cell mass (ICM) cells and foetal growth after embryo transfer. Mechanistic studies revealed two important pathways involved in the α-KG effects on embryo development. First, α-KG modulates mitochondria function by inducing relatively low ATP production without modification of mitochondrial copy number. The relatively low energy metabolism preserves the pluripotency and competence of the ICM. Second, α-KG modifies epigenetics in embryos cultured in vitro by affecting the activity of the DNA demethylation enzyme TET and the DNA methylation gene Dnmt3a to increase the ratio of 5hmC/5mC ratio. Elevation of the 5hmC/5mC ratio not only promotes the pluripotency of the ICM but also leads to a methylation level in an in vitro embryo close to that in an in vivo embryo. All these functions of α-KG collectively contribute to an increase in the number of ICM cells, leading to greater adaptation of cultured embryos to in vitro conditions and promoting foetal growth after embryo transfer. Our findings provide basic knowledge regarding the mechanisms by which α-KG affects embryo development and cell differentiation.

RESUMO

Elucidating the key impact factors on PM2.5 concentrations is crucial to formulate effective mitigation policies. In this study, we employed an extended Stochastic Impacts by Regression on Population Affluence and Technology (STIRPAT) model to identify the socioeconomic determinants of PM2.5 concentrations for 12 different regions and across China. The evaluation was based on a balanced panel dataset integrating long-term satellite-derived PM2.5 concentrations and socio-economic data in China from 1999 to 2011. Empirical results indicate that the influencing factors can be ranked in descending order of importance as: proportion of secondary sector of the economy, GDP per capita, urbanization, population, energy intensity, and proportion of tertiary sector. Proportion of secondary sector is the greatest contribution to increasing PM2.5 concentrations, especially for heavily polluted regions. GDP per capita is secondary in importance, and its impact is weakened by the existence of an EKC relationship between GDP per capita and PM2.5 concentrations. Therefore, PM2.5 pollution is an economic development mode problem, rather than a general economic development problem. The impact of urbanization varies across regions; while promoting urbanization will be conducive to decreased PM2.5 concentrations in Northwest China and Northeast China, it will contribute to increased PM2.5 concentrations in other regions. Population and energy intensity are significant in most regions, but neither are decisive factors because of the small absolute value of their coefficients. Finally, different combinations of mitigation policies are proposed for different regions in this study to meet the mitigation targets.

RESUMO

CRISPR/Cas9 (Clustered regularly interspaced short palindromic repeats) combined with pronuclear microinjection has become the most effective method for producing transgenic animals. However, the relatively low embryo developmental rate limits its application. In the current study, it was observed that 10-7 M melatonin is considered an optimum concentration and significantly promoted the in vitro development of murine microinjected pronuclear embryos, as indicated by the increased blastocyst rate, hatching blastocyst rate and blastocyst cell number. When these blastocysts were implanted into recipient mice, the pregnancy rate and birth rate were significantly higher than those of the microinjected control, respectively. Mechanistic studies revealed that melatonin treatment reduced reactive oxygen species (ROS) production and cellular apoptosis during in vitro embryo development and improved the quality of the blastocysts. The implantation of quality-improved blastocysts led to elevated pregnancy and birth rates. In conclusion, the results revealed that the anti-oxidative and anti-apoptotic activities of melatonin improved the quality of microinjected pronuclear embryos and subsequently increased both the efficiency of embryo implantation and the birth rate of the pups. Therefore, the melatonin supplementation may provide a novel alternative method for generating large numbers of transgenic mice and this method can probably be used in human-assisted reproduction and genome editing.

RESUMO

The inferior oocytes (IOs), which are not suitable for embryo development, occupy roughly one-third or more of the collected immature bovine oocytes. The IOs are usually discarded from the in vitro bovine embryo production process. Improving the quality of the inferior oocytes (IOs) and make them available in in vitro embryo production would have important biological, as well as commercial, value. This study was designed to investigate whether melatonin could improve the quality of IOs and make them usable in the in vitro maturation (IVM) and subsequent (in vitro fertilization) IVF embryo development. The results indicated that: the maturation rate of IOs and their subsequent IVF embryo developments were impaired compared to cumulus-oocyte complexes and melatonin treatment significantly improved the quality of IOs, as well as their IVF and embryo developments. The potential mechanisms are that: (1) melatonin reduced reactive oxygen species (ROS) and enhanced glutathione (GSH) levels in the IOs, thereby protecting them from oxidative stress; (2) melatonin improved mitochondrial normal distribution and function to increase ATP level in IOs; and (3) melatonin upregulated the expression of ATPase 6, BMP-15, GDF-9, SOD-1, Gpx-4, and Bcl-2, which are critical genes for oocyte maturation and embryo development and downregulated apoptotic gene expression of caspase-3.

RESUMO

Urban landscape spatiotemporal change patterns and their driving mechanisms in China are poorly understood at the national level. Here we used remote sensing data, landscape metrics, and a spatial econometric model to characterize the spatiotemporal patterns of urban landscape change and investigate its driving forces in China between 1990 and 2005. The results showed that the urban landscape pattern has experienced drastic changes over the past 15 years. Total urban area has expanded approximately 1.61 times, with a 2.98% annual urban-growth rate. Compared to previous single-city studies, although urban areas are expanding rapidly, the overall fragmentation of the urban landscape is decreasing and is more irregular and complex at the national level. We also found a stair-stepping, urban-landscape changing pattern among eastern, central, and western counties. In addition, administrative level, urban size, and hierarchy have effects on the urban landscape pattern. We also found that a combination of landscape metrics can be used to supplement our understanding of the pattern of urbanization. The changes in these metrics are correlated with geographical indicators, socioeconomic factors, infrastructure variables, administrative level factors, policy factors, and historical factors. Our results indicate that the top priority should be strengthening the management of urban planning. A compact and congregate urban landscape may be a good choice of pattern for urban development in China.

RESUMO

Rapid economic growth, industrialization, and urbanization in China have led to extremely severe air pollution that causes increasing negative effects on human health, visibility, and climate change. However, the influence mechanisms of these anthropogenic factors on fine particulate matter (PM2.5) concentrations are poorly understood. In this study, we combined panel data and econometric methods to investigate the main anthropogenic factors that contribute to increasing PM2.5 concentrations in China at the prefecture level from 1999 to 2011. The results showed that PM2.5 concentrations and three anthropogenic factors were cointegrated. The panel Fully Modified Least Squares and panel Granger causality test results indicated that economic growth, industrialization, and urbanization increased PM2.5 concentrations in the long run. The results implied that if China persists in its current development pattern, economic growth, industrialization and urbanization will inevitably lead to increased PM2.5 emissions in the long term. Industrialization was the principal factor that affected PM2.5 concentrations for the total panel, the industry-oriented panel and the service-oriented panel. PM2.5 concentrations can be reduced at the cost of short-term economic growth and industrialization. However, reducing the urbanization level is not an efficient way to decrease PM2.5 pollutions in the short term. The findings also suggest that a rapid reduction of PM2.5 concentrations relying solely on adjusting these anthropogenic factors is difficult in a short-term for the heavily PM2.5-polluted panel. Moreover, the Chinese government will have to seek much broader policies that favor a decoupling of these coupling relationships.

RESUMO

Ensuring residents' equal access to high quality urban greenspace is vital for urban environmental justice and sustainable urban development. However, most previous studies have mainly focused on greenspace quantity, overlooking its quality. Moreover, the national-level spatial distribution pattern of residential greenspace exposure (RGE) within urban areas remains unclear. Here, we have improved the existing RGE assessment framework by integrating both the quality and quantity of urban greenspace to evaluate RGE and its associated inequality across 119,692 blocks in 334 Chinese cities in 2020. We find that the spatial distribution pattern of RGE varies with urban size. Large cities exhibit a distinct clustering of low RGE in their central areas, whereas small cities tend to show a pronounced clustering of high RGE in the central areas. RGE in Chinese cities indicates extensive inequality, as the average RGE of high-exposed people is nearly four times greater than that of low-exposed people. Moreover, residents in larger cities are more prone to experiencing greater inequalities compared to those in smaller cities. We also find that the landscape metrics (i.e., connectance index and mean Euclidean nearest-neighbor distance) of greenspace possess a strong explanatory power (R2 = 0.431) for the observed inequality. Our study underscores the importance of optimizing the landscape structure of urban greenspace and enhancing equality in the quality of greenspace. These findings provide novel insights for urban greenspace planning and promoting urban environmental justice.

RESUMO

The multiple absorber layer perovskite solar cells (PSCs) with charge transport layers-free (CTLs-free) have drawn widespread research interest due to their simplified architecture and promising photoelectric characteristics. Under the circumstances, the novel design of CTLs-free inversion PSCs with stable and nontoxic three absorber layers (triple Cs3Bi2I9, single MASnI3, double Cs2TiBr6) as optical-harvester has been numerically simulated by utilizing wxAMPS simulation software and achieved high power conversion efficiency (PCE) of 14.8834%. This is owing to the innovative architecture of PSCs favors efficient transport and extraction of more holes and the slender band gap MASnI3 extends the absorption spectrum to the near-infrared periphery compared with the two absorber layers architecture of PSCs. Moreover, the performance of the device with p-type-Cs3Bi2I9/p-type-MASnI3/n-type-Cs2TiBr6 architecture is superior to the one with the p-type-Cs3Bi2I9/n-type-MASnI3/n-type-Cs2TiBr6 architecture due to less carrier recombination and higher carrier life time inside the absorber layers. The simulation results reveal that Cs2TiF6 double perovskite material stands out as the best alternative. Additionally, an excellent PCE of 21.4530% can be obtained with the thicker MASnI3 absorber layer thickness (0.4 µm). Lastly, the highest-performance photovoltaic devices (28.6193%) can be created with the optimized perovskite doping density of around E15 cm3 (Cs3Bi2I9), E18 cm3 (MASnI3), and 1.5E19 cm3 (Cs2TiBr6). This work manifests that the proposed CTLs-free PSCs with multi-absorber layers shall be a relevant reference for forward applications in electro-optical and optoelectronic devices.
