In eukaryotes, autophagy is induced as an innate defense mechanism against pathogenic microorganisms by self-degradation. Although trichinellosis is a foodborne zoonotic disease, there are few reports on the interplay between Trichinella spiralissurvival strategies and autophagy-mediated host defense. Therefore, this study focused on the association between T. spiralis and autophagy of host small intestinal cells. In this study, the autophagy-related indexes of host small intestinal cells after T. spiralis infection were detected using transmission electron microscopy, hematoxylin and eosin staining, immunohistochemistry, quantitative real-time polymerase chain reaction, and Western blotting. The results showed that autophagosomes and autolysosomes were formed in small intestinal cells, intestinal villi appeared edema, epithelial compactness was decreased, microtubule-associated protein 1A/1B-light chain 3B (LC3B) was expressed in lamina propria stromal cells of small intestine, and the expression of autophagy-related genes and proteins was changed significantly, indicating that T. spiralis induced autophagy of host small intestinal cells. Then, the effect of T. spiralis on autophagy-related pathways was explored by Western blotting. The results showed that the expression of autophagy-related pathway proteins was changed, indicating that T. spiralis regulated autophagy by affecting autophagy-related pathways. Finally, the roles of T. spiralis serine protease inhibitors (TsSPIs), such as T. spiralis Kazal-type SPI (TsKaSPI) and T. spiralis Serpin-type SPI (TsAdSPI), were further discussed in vitro and in vivo experiments. The results revealed that TsSPIs induced autophagy by influencing autophagy-related pathways, and TsAdSPI has more advantages. Overall, our results indicated that T. spiralis induced autophagy of host small intestinal cells, and its TsSPIs play an important role in enhancing autophagy flux by affecting autophagy-related pathways. These findings lay a foundation for further exploring the pathogenesis of intestinal dysfunction of host after T. spiralis infection, and also provide some experimental and theoretical basis for the prevention and treatment of trichinellosis.

BACKGROUND: The current nodal (pN) classification still has limitations in stratifying the prognosis of small cell lung cancer (SCLC) patients with pathological classifications T1-2N0-2M0. Thus. This study aimed to develop and validate a modified nodal classification based on a multicenter cohort. MATERIALS AND METHODS: We collected 1156 SCLC patients with pathological classifications T1-2N0-2M0 from the Surveillance, Epidemiology, and End Results database and a multicenter database in China. The X-tile software was conducted to determine the optimal cutoff points of the number of examined lymph nodes (ELNs) and lymph node ratio (LNR). The Kaplan-Meier method, the Log-rank test, and the Cox regression method were used in this study. We classified patients into three pathological N modification categories, new pN#1 (pN0-#ELNs > 3), new pN#2 (pN0-#ELNs ≤ 3 or pN1-2-#LNR ≤ 0.14), and new pN#3 (N1-2-#LNR > 0.14). The Akaike information criterion (AIC), Bayesian Information Criterion, and Concordance index (C-index) were used to compare the prognostic, predictive ability between the current pN classification and the new pN component. RESULTS: The new pN classification had a satisfactory effect on survival curves (Log-rank P < 0.001). After adjusting for other confounders, the new pN classification could be an independent prognostic indicator. Besides, the new pN component had a much more accurate predictive ability in the prognostic assessment for SCLC patients of pathological classifications T1-2N0-2M0 compared with the current pN classification in the SEER database (AIC: 4705.544 vs. 4731.775; C-index: 0.654 vs. 0.617, P < 0.001). Those results were validated in the MCDB from China. CONCLUSIONS: The multicenter cohort developed and validated a modified nodal classification for SCLC patients with pathological category T1-2N0-2M0 after surgery. Besides, we propose that an adequate lymph node dissection is essential; surgeons should perform and consider the situation of ELNs and LNR when they evaluate postoperative prognoses of SCLC patients.

KEY MESSAGE: OsVP1 and Sdr4 play an important role in regulating seed dormancy that involved in multiple metabolism and regulatory pathways. Seed dormancy and germination are critical agricultural traits influencing rice grain yield. Although there are some genes have identified previously, the comprehensive understanding based on transcriptome is still deficient. In this study, we generated mutants of two representative regulators of seed germination, Oryza sativa Viviparous1 (OsVP1) and Seed dormancy 4 (Sdr4), by CRISPR/Cas9 approach and named them cr-osvp1 and cr-sdr4. The weakened dormancy of mutants indicated that the functions of OsVP1 and Sdr4 are required for normal early seed dormancy. There were 4157 and 8285 differentially expressed genes (DEGs) were identified in cr-osvp1 vs. NIP and cr-sdr4 vs. NIP groups, respectively, with a large number of overlapped DEGs between two groups. The gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis of common DEGs in two groups showed that genes related to carbohydrate metabolic, nucleoside metabolic, amylase activity and plant hormone signal transduction were involved in the dormancy regulation. These results suggest that OsVP1 and Sdr4 play an important role in regulating seed dormancy by multiple metabolism and regulatory pathways. The systematic analysis of the transcriptional level changes provides theoretical basis for the research of seed dormancy and germination in rice.

Enterocytozoon bieneusi is a zoonotic parasite which is considered to be an opportunistic pathogen of humans and animals. A number of studies have reported E. bieneusi infection in various animals. However, no information is available on the occurrence of E. bieneusi in Tan sheep, a unique indigenous sheep species in the Ningxia Hui Autonomous Region, China. The objectives of the present study were to examine the prevalence and identify the genotypes of E. bieneusi in Tan sheep in China. A total of 1014 fecal specimens of Tan sheep from six farms in the Ningxia Hui Autonomous Region were examined by nested PCR amplification of the internal transcribed spacer (ITS) of nuclear ribosomal DNA. The total prevalence of E. bieneusi was 12.2% (124/1014), ranging from 0.5 to 22.2% on six farms. Sequence analysis identified 10 genotypes of E. bieneusi, including three known genotypes, BEB6, COS-I, and CHG13, and seven novel genotypes designated as NX1 to NX7, which all belonged to group 2 by phylogenetic analysis. This is the first report describing the prevalence of E. bieneusi in Tan sheep, and the new genotypes identified in the current study expand the genotype distribution of E. bieneusi. These findings provide baseline data and have implications for the epidemiology and control of E. bieneusi infection in Tan sheep.

RESUMO

Curcumin is a polyphenol extracted from turmeric rhizome and has multiple pharmacological roles. Recently,its anticancer properties have been recognized. Also,curcumin regulates autophagy in tumor cells via signaling pathways including AMP-activated protein kinase,mammalian target of rapamycin,transcription factor EB,Beclin-1,B-cell lymphoma 2,and endoplasmic reticulum stress. Considering the complicated crosstalk between autophagy and apoptosis,in this article we summaize the mechanism of curcumin-induced autophagy and its effect on apoptosis,with an attempt to provide insights on tumor therapy.

OBJECTIVE: To determine the role of MRI in the early diagnosis of tubal ectopic pregnancy (EP). METHODS: Clinical and MRI features of 27 cases of tubal pregnancy were reviewed. RESULTS: A thick-walled gestational sac (GS)-like structure was demonstrated lateral to the uterus in all cases. On T2-weighted images, the thick wall typically exhibited 3 discrete rings in 22 cases (81 %), among which 17 cases (63 %) displayed small vessels and 6 cases (33 %) exhibited small areas of fresh haemorrhage inside the thick wall. The contents demonstrated non-specific liquid in 26 %, papillary solid components in 56 %, and fresh blood or fluid-fluid level in 19 % of the cases. Dilatation of the affected fallopian tube associated with hematosalpinx was demonstrated in 18 cases (67 %) and marked enhancement of the tubal wall was observed in 22 cases (81 %). No correlation was found between the size of the GS and the estimated gestational age (r = 0.056). CONCLUSION: MRI plays an important role in the early diagnosis and management of tubal pregnancy. The characteristic MRI features include a GS-like structure with a "three rings" appearance on T2-weighted images, presence of solid components in the sac, dilatation of the affected fallopian tube with hematosalpinx, and tubal wall enhancement. KEY POINTS: • MR imaging has served as a problem-solving procedure in ectopic pregnancy. • MR imaging features can be criteria for early diagnosis of tubal pregnancy. • Detailed assessment of ectopic implantation is necessary for management decision-making.

OBJECTIVE: To determine the regulatory role and mechanism of nitric oxide (NO) in the development and hatching of mouse blastocysts. METHODS: The Kunming female mice were superovulated and then mated with mature male mice. On the day 2.5 of their pregnancy, morulae were flushed from their uterine horns with culture media. Morulae were cultured in different concentrations of N-nitro-L arginine methyl ester (L-NAME), sodium nitroprusside (SNP), or the combination of L-NAME and SNP in culture media for 48 hours. The development and hatching of blastocysts were examined on day 4 and day 5 and the total numbers of blastocyst cells and cysteinyl aspartate specific proteinase 3 (caspase 3) were observed under confocal laser scanning microscope. RESULTS: With the increase of the concentration of L-NAME or SNP, the hatching rate of blastocysts and the total number of blastocyst cells were significantly reduced. The addition of 10 nmol/L SNP in culture media with 5 mmol/L L-NAME significantly increased the development of blastocysts and promoted hatching of blastocysts. However, with increase of SNP concentration in culture media with 5 mmol/L L-NAME, the development and hatching rates of blastocysts were significantly decreased. L-NAME had no obvious effect on the expression of active caspase 3 in blastocyst cells. However,when being above 500 nmol/L,SNP significantly increased the expression of caspase 3 in blastocyst cells. CONCLUSIONS: NO plays an important role in development and hatching of mouse blastocysts. Excessively high or low NO can damage the division of blastomeres, resulting in the failure of the blastocyst development and hatching. Also, excessively high NO can lead to the apoptosis of the blastocyst cells.

Previous analysis of systems pharmacology has revealed a tendency of rational drug design in the pharmaceutical industry. The targets of new drugs tend to be close with the corresponding disease genes in the biological networks. However, it remains unclear whether the rational drug design introduces disadvantages, i.e. side effects. Therefore, it is important to dissect the relationship between rational drug design and drug side effects. Based on a recently released drug side effect database, SIDER, here we analyzed the relationship between drug side effects and the rational drug design. We revealed that the incidence drug side effect is significantly associated with the network distance of drug targets and diseases genes. Drugs with the distances of three or four have the smallest incidence of side effects, whereas drugs with the distances of more than four or smaller than three show significantly greater incidence of side effects. Furthermore, protein drugs and small molecule drugs show significant differences. Drugs hitting membrane targets and drugs hitting cytoplasm targets also show differences. Failure drugs because of severe side effects show smaller network distances than approved drugs. These results suggest that researchers should be prudent on rationalizing the drug design. Too small distances between drug targets and diseases genes may not always be advantageous for rational design for drug discovery.

Many pathological phenomena of male infertility are related to epigenetic changes in male germ cells. Epigenetic regulation during spermatogenesis plays an important role in mitotic/meiotic divisions and spermiogenesis. The histones have various post-translational modifications on different amino acid residues during spermatogenesis. These modifications are crucial to the precise regulation of spermatogenesis. Moreover, the histone-to-protamine transition will occur during spermiogenesis. Many studies have also found that abnormal changes of histone modifications during spermatogenesis may damage the sperm development, leading to male sterility. This article reviews the changes of histone modifications during spermatogenesis, the regulation of the development of male germ cells, and the relationship between histone abnormalities and male sterility.

OBJECTIVE: To determine the effects of bisphenol-A (BPA) on blastocyst development and implantation. METHODS: According to completely randomized grouping method, 90 pregnant mice were divided into 100, 300, and 600 mg/(kg·d)BPA groups and control group. BPA-treated pregnant mice were orally administered with BPA at concentrations of 100, 300 and 600 mg/(kg·d) from day 0.5 to day 3.5 of their pregnancy. Blastocyst implantation and development were studied. RESULTS: In the 300 mg/(kg·d) BPA group, the number of implantation sites and implantation rate were significantly decreased. In the 600 mg/(kg·d) group, no implantation sites were observed among pregnant mice and BPA inhibited embryo implantation. Blastocyst development on day 4 was examined, and findings showed that the development rate and total numbers of blastocysts in BPA treatment groups had no significant difference from the control group. However, BPA at 300 and 600 mg/(kg·d) significantly reduced blastocyst hatching rate and dramatically increased the number of blastocyst apoptotic cells when compared with those in the control group. CONCLUSION: BPA at a high concentration damages the blastocyst development before implantation and inhibits embryo implantation.

Frequency modulation spectroscopy (FMS) not only can be used to simultaneously measure the absorption and dispersion of atoms and molecules, but is the key technology of the noise immunity cavity enhanced optical heterodyne molecular spectroscopy (NICE-OHMS). The optical devices or the instability of output light polarization of the laser source will induce the residual amplitude modulation (RAM) in the FMS. RAM greatly limits the FMS technology application in trace gas detection, so the research on the RAM characteristics in the FMS has very important significance. Firstly, the lineshape of FMS without absorption was analyzed, and the impact factors on the RAM were acquired, then the influence of input and output polarization direction and electro-optical modulation (EOM) temperature was measured, respectively. They all have linear relationship with the RAM. The results verify the theoretical analysis and provide the basis for reducing the RAM and other related working.

In the present paper, the authors simply describe the principle of cavity ring-down spectroscopy (CRDS) measurement technology and introduce a threshold circuit, based on 555 timer IC (integrated circuit), applied to CRDS. And we verify the feasibility of the circuit. By means of changing the input resistor and input capacity, we analyzee the influence of threshold circuit characteristics on single ring-down event, and found that the greater the input capacitance, and the smaller the input impedance of circuit, the more serious the distortion of measurement of ring-down curve, especially when the resistance is less than 50 omega the ring-down time produces very big change. Then we collected and analyzed the C2H2 absorption signal at 6 531.7805 cm(-1) with different input resistance and input capacitance, and fitted the absorption line with Voigt profile. We found that the change of the input capacitance and resistance caused the distortion of fitting curve. Finally We give the optimal design of threshold circuit: the input impedance of the threshold circuit should be great, minimum of 100 omega, the capacitive reactance should be as small as possible, preferably below 1 nf, at the same time it should be ensured that the threshold of the circuit time constant is much smaller than ring-down time. The experiment research on CRDS technique application has important reference value to threshold circuit design.

In the present paper a theoretical description of the line shapes of frequency modulated noise-immune cavity-enhanced optical heterodyne molecular spectroscopy (NICE-OHMS) under saturated conditions is presented. Expressions for both Doppler broadened and Doppler free frequency modulated NICE-OHMS signals at their absorption and dispersion phases are given. The modulation frequency, saturation parameter and frequency modulation index are set to 384 MHz, 10 and 0.2, respectively in the simulation. Based on the simulated line shape curves, in the absorption detection phase there are four sub Doppler structures existing in the spectral envelope of absorption signals of two sidebands, and in the dispersion phase there are five sub-Doppler structures existing in the spectral envelope of dispersion signals of carrier and two sidebands. The sub-Doppler structure originates from the fact that two counter propagating waves interact with the same group of molecules and one can act as a detecting beam and the other can act as a pumping beam. Therefore the sub-Doppler structure can appear at laser carrier and sidebands frequency and their middle frequencies of carrier and sidebands. Since NICE-OHMS absorption signal is only related to the absorption of sidebands the small saturation parameters of two sidebands can not saturate the Doppler broadened signal too much Although the NICE-OHMS dispersion signal includes the dispersion of carrier and two sidebands, the amplitude of Doppler broadened signal does not have so much influence by saturation except for the Doppler free signal. Therefore NICE-OHMS technique is a good candidate for the high sensitive detection of gas medium but with high selectivity under the condition of low pressure. Finally the dependences of NICE-OHMS line shape on the detection phase and saturation parameter are simulated and analyzed, the results of which nicely agree with the existing experimental results. The researches on NICE-OHMS line shapes will help explain the phenomena of related experiment.

In the present paper a phase controllable waveplate model was applied to the analysis of polarization maintaining fiber. Under the temperatures of 24 degrees C and 26.8 degrees C there is no residual amplitude modulation (RAM) existing in frequency modulation spectroscopy (FMS) when performing the measurement of fiber temperature dependence on the RAM. However, the temperature setting can not reduce the background long term drift and a servo feedback suggested by N. C. Wong and J. L. Hall can be used to actively reduce the RAM. The error signal for feedback is from the dispersion background signal of FMS without gas sample. The variation of RAM induced by the temperature changing of PM fiber is the main reason for the long term background drift of dispersion signal of FMS.

The feasibility of surgery after immunotherapy for mediastinal liposarcoma remains uncertain. Besides, the case of immunotherapy for liposarcoma is still lacking. We report a case of recurrence after resection of a left mediastinal liposarcoma. After recurrence, one course of pembrolizumab plus anlotinib hydrochloride showed no tumor shrinkage, and genetic testing showed CDK4 amplification and PD-L1 TPS <1%; therefore, the plan was changed to one course of pembrolizumab plus palbociclib, but the tumor still did not shrink. Thus, second tumor resection was performed. In addition, the postoperative pathology was still well-differentiated liposarcoma. The significance of immunotherapy in liposarcoma still needs to be further explored. In the absence of surgical contraindications, secondary surgery might be feasible.

OBJECTIVE: The present study was designed to evaluate whether CP was beneficial in alleviating myocardial fibrosis following I/R injury. METHODS: Sprague-Dawley rats were subjected to 30 minutes occlusion of the LADCA, followed by reperfusion. CP (0.4 or 0.8 g/kg) was daily administered starting from three hour after reperfusion until day 6. Coronary venular diameter, RBC velocity, albumin leakage, MBF, heart function, myocardial infarction and fibrosis size, myocardium ultrastructure, MPO activity, and MDA level were evaluated. The expression of MCP-1, RP S19, TGF-ß1, P-Smad3, Smad4, MMP-9 and α-SMA, and the infiltration of leukocytes were examined. RESULTS: CP post-treatment ameliorated I/R-induced myocardial RBC velocity reduction, MBF decrease, cardiac dysfunction, and albumin leakage increase. Moreover, myocardial infarction and fibrosis size, MPO activity, MDA level, the expression of RP S19, TGF-ß1, P-Smad3, Smad4, MMP-9 and α-SMA, the number of CD68-positive cells increased significantly after I/R, and myocardium collagen deposition was observed on day 6 after reperfusion. All the alterations after I/R were significantly ameliorated by CP. CONCLUSIONS: Post-treatment with CP ameliorates I/R-induced myocardial fibrosis, suggesting that CP may be applied as an option for preventing cardiac remodeling after I/R injury.

Four recent papers by Menger et al. have questioned methods of analysis of surface tension (ST) data that use the Gibbs equation to obtain the surface excess (Γ) of a surfactant at the air-water interface. There have been two responses which challenge the assertions of Menger et al. and a response from Menger et al. We use directly determined values of Γ from a range of neutron reflectometry (NR) data to examine some of the issues that are relevant to these seven papers. We show that there is excellent agreement between NR measurements and careful ST analyses for a wide range of nonionic adsorbents, including surfactants and polymers. The reason it is possible to obtain good agreement near the critical micelle concentration (CMC) is that nonionic surfactants generally seem to saturate the surface before the CMC is reached and this makes it relatively easy to determine the limiting slope (and hence Γ) of the ST-log(concentration) plot at the CMC. Furthermore, there is also generally good agreement between ST and NR over the whole range of concentrations below the CMC until depletion effects become important. Depletion effects are shown to become important at higher concentrations than expected, which brings them into the range of many experiments, including techniques other than ST and NR. This is illustrated with new measurements on the biosurfactant surfactin. The agreement between ST and NR outside the depletion range can be regarded as a mutual validation of the two methods, especially as it is demonstrated independently of any model adsorption isotherms. In the normal experimental situation NR is less vulnerable to depletion than ST and we show how NR and a single ST measurement can be used to determine the hitherto undetermined CMC of the nonionic surfactant C18E12, which is found to be 1.3 × 10(-6) M.

Plant lipid transfer proteins (LTPs) are encoded by multigene families and play important roles in plant physiology. One full-length cDNA encoding an Arabidopsis LTP3 homologue was isolated from maize by RT-PCR and named as ZmLTP3. RT-PCR analysis indicated that the ZmLTP3 expression is induced by salicylic acid (SA), mannitol and salt. Furthermore, in different tissues the ZmLTP3 displayed different expression patterns, indicating that ZmLTP3 may play multiple roles in stress resistance. Over-expression of ZmLTP3 in wild-type Arabidopsis resulted in the increased salt tolerance. Under salt stress condition, compared to wild-type (WT) plants, transgenic Arabidopsis grew better, had higher seedling fresh (FW), dry weight (DW), seed yields, proline content and lower MDA content and relative electric conductivity level. Our results suggest that maize ZmLTP3 might encode a member of LTPs family and play roles in salt resistance.

Bisphenol A (BPA) is a commonly used phenolic environmental estrogen. Long-term exposure of female mammalians to BPA can lead to endocrine disorders, followed by the morphological and functional changes in ovary, uterus, vagina, and oviducts. The interactions of BPA with various target molecules or tissues will cause different effects. To further elucidate the effects of BPA on female reproductive system, we review the changes in the structure and functions of female reproduction system after BPA exposure and their possible mechanisms.

To facilitate the manufacturing of the thin-walled components of 2219 aluminum alloy, the quenching-forming-aging (Q-F-A) process has been increasingly utilized. However, natural aging (NA) after quenching significantly affects the cold forming performance of this alloy. In this study, experiments are conducted to investigate the effect of NA time on the cold forming performance of 2219 aluminum alloy. The results indicate that NA can weaken the Portevin-Le Chatelier (PLC) effect, thereby reducing its influence on the cold forming performance of the alloy. The PLC effect becomes indistinct when the aging time reaches 2 years. The yield strength of 2219 aluminum alloy increases monotonically with aging time, while the elongation first increases rapidly and then decreases. After an aging time of 2 years, the yield strength increases by 28.6% from that of newly quenched alloys. The strain hardening index and hardening coefficient indicate that short-term NA (less than 4 days) increases the work hardening rate, while long-term NA reduces it. Microstructural analysis shows that the strengthening effect of NA on 2219 aluminum alloy is mainly due to the growth of G.P. zones and the precipitation of θ″ phases. The NA precipitation behavior can also cause the aggregation of solute atoms and weaken the PLC effect.
