RESUMO

BACKGROUND: Gliomas are the deadliest malignant tumors of the adult central nervous system. We previously discovered that beta2-microglobulin (B2M) is abnormally upregulated in glioma tissues and that it exerts a range of oncogenic effects. Besides its tissue presence, serum B2M levels serve as biomarkers for various diseases. This study aimed to explore whether serum B2M levels can be used in the diagnosis and prognosis of gliomas. METHODS: Medical records from 246 glioma patients were retrospectively analyzed. The relationship between preoperative serum B2M levels and clinicopathological features was examined. Kaplan-Meier analysis, alongside uni- and multivariate Cox regression, assessed the association between B2M levels, systemic inflammatory markers, and glioma patient prognosis. Receiver operating characteristic (ROC) curve analysis evaluated the diagnostic significance of these biomarkers specifically for glioblastoma (GBM). RESULTS: Patients with malignant gliomas exhibited elevated preoperative serum B2M levels. Glioma patients with high serum B2M levels experienced shorter survival times. Multivariate Cox analysis determined the relationship between B2M levels (hazard ratio = 1.92, 95% confidence interval: 1.05-3.50, P = 0.034) and the overall survival of glioma patients. B2M demonstrated superior discriminatory power in distinguishing between GBM and non-GBM compared to inflammation indicators. Moreover, postoperative serum B2M levels were lower than preoperative levels in the majority of glioma patients. CONCLUSIONS: High preoperative serum B2M levels correlated with malignant glioma and a poor prognosis. Serum B2M shows promise as a novel biomarker for predicting patient prognosis and reflecting the therapeutic response.

RESUMO

Central nervous system (CNS) disorders represent the leading cause of disability and the second leading cause of death worldwide, and impose a substantial economic burden on society. In recent years, emerging evidence has found that beta2 -microglobulin (B2M), a subunit of major histocompatibility complex class I (MHC-I) molecules, plays a crucial role in the development and progression in certain CNS diseases. On the one hand, intracellular B2M was abnormally upregulated in brain tumors and regulated tumor microenvironments and progression. On the other hand, soluble B2M was also elevated and involved in pathological stages in CNS diseases. Targeted B2M therapy has shown promising outcomes in specific CNS diseases. In this review, we provide a comprehensive summary and discussion of recent advances in understanding the pathological processes involving B2M in CNS diseases (e.g., Alzheimer's disease, aging, stroke, HIV-related dementia, glioma, and primary central nervous system lymphoma).

RESUMO

Adult gliomas are divided into isocitrate dehydrogenase (IDH) wild-type and IDH mutant subtypes according to the new 2021 World Health Organization classification system. However, the local and systemic effects of IDH mutations on primary glioma patients are not well illustrated. Retrospective analysis, immune-cell infiltration analysis, meta-analysis, and immunohistochemistry assay were applied in the present study. The results from our cohort showed that IDH mutant gliomas own a lower proliferating rate compared to that in wild-type gliomas. Patients with mutant IDH exhibited a higher frequency of seizures in both our cohort and the cohort from the meta-analysis. Mutations in IDH result in lower levels of intra-tumour but higher levels of circulating CD4+ and CD8+ T lymphocytes. Levels of neutrophils in both intra-tumour and circulating blood were lower in IDH mutant gliomas. Moreover, IDH mutant glioma patients receiving radiotherapy in combination with chemotherapy exhibited better overall survival with respect to radiotherapy alone. Mutations in IDH alters the local and circulating immune microenvironment, and increases the sensitivity of tumour cell to chemotherapy.

RESUMO

BACKGROUND: Glioma is the most common and aggressive tumor in the adult brain. Recent studies have indicated that Zinc finger DHHC-type palmitoyltransferases (ZDHHCs) play vital roles in regulating the progression of glioma. ZDHHC15, a member of the ZDHHCs family, participates in various physiological activities in the brain. However, the biological functions and related mechanisms of ZDHHC15 in glioma remain poorly understood. METHODS: Data from multiple glioma-associated datasets were used to investigate the expression profiles and potential biological functions of ZDHHC15 in glioma. Expression of ZDHHC15 and its association with clinicopathological characteristics in glioma were validated by quantitative reverse transcription PCR (RT-qPCR) and immunohistochemical experiments. GO enrichment analysis, KEGG analysis, GSEA analysis, CCK-8, EdU, transwell, and western blotting assays were performed to confirm the functions and mechanism of ZDHHC15 in glioma. Moreover, we performed Kaplan-Meier analysis and Cox progression analysis to explore the prognostic significance of ZDHHC15 in glioma patients. RESULTS: ZDHHC15 expression was significantly up-regulated in glioma and positively associated with malignant phenotypes. Results from the GO and KEGG enrichment analysis revealed that ZDHHC15 was involved in regulating cell cycle and migration. Knockdown of ZDHHC15 inhibited glioma cell proliferation and migration, while overexpression of ZDHHC15 presented opposite effects on glioma cells. Besides, results from GSEA analysis suggested that ZDHHC15 was enriched in STAT3 signaling pathway. Knockdown or overexpression of ZDHHC15 indeed affected the activation of STAT3 signaling pathway. Additionally, we identified ZDHHC15 as an independent prognostic biomarker in glioma, and higher expression of ZDHHC15 predicted a poorer prognosis in glioma patients. CONCLUSION: Our findings suggest that ZDHHC15 promotes glioma malignancy and can serve as a novel prognostic biomarker for glioma patients. Targeting ZDHHC15 may be a promising therapeutic strategy for glioma.

RESUMO

Revealing how aquatic organisms respond to dam impacts is essential for river biomonitoring and management. Traditional examinations of dam impacts on macroinvertebrate assemblages were frequently conducted within single rivers (i.e., between upstream vs. downstream locations) and based on taxonomic identities but have rarely been expanded to level of entire basins (i.e., between dammed vs. undammed rivers) and from a functional trait perspective. Here, we evaluated the effects of dams on macroinvertebrate assemblages at both the within-river and basin scales using functional traits in two comparable tropical tributaries of the Lancang-Mekong River. At different scales, maximum body size, functional feeding groups (FFG), voltinism and occurrence in drift respond significantly to dam impact. Armoring categories varied significantly between downstream sites and upstream sites, and oviposition behavior, habits and adult life span significantly differed between rivers. The key traits at the within-river scale resembled to those at the between-river scale, suggesting that within-river trait variation could further shape functional trait structure at the basin scale in dammed rivers. Furthermore, water nutrients and habitat quality induced by dams showed the most important role in shaping trait structure, although trait-environment relationships varied between the two different scales. In addition, the trait-environment relationships were stronger in the dry season than in the wet season, suggesting a more important role of environmental filtering processes in the dry season compared with the wet season. This study highlights the utility of the trait-based approach to diagnose the effects of damming and emphasizes the importance of spatial scale to examine dam impacts in riverine systems.

RESUMO

Malignant tumor cells can obtain proliferative benefits from deviant metabolic networks. Emerging evidence suggests that lipid metabolism are dramatically altered in gliomas and excessive fatty acd accumulation is detrimentally correlated with the prognosis of glioma patients. Glioma cells possess remarkably high levels of free fatty acids, which, in turn, enhance post-translational modifications (e.g. palmitoylation). Our and other groups found that palmitoylational modification is essential for remaining intracellular homeostasis and cell survival. Disrupting the balance between palmitoylation and depalmitoylation affects glioma cell viability, apoptosis, invasion, self-renew and pyroptosis. In this review, we focused on summarizing roles and relevant mechanisms of protein palmitoylational modification in gliomas.

RESUMO

The safety of two-wheelers is a serious public safety issue nowadays. Two-wheelers usually have severe conflict interaction with vehicles at intersections, such as running red lights, which is very likely to cause traffic accidents. Therefore, a model of two-wheeler driving behavior in conflicting interactions can provide guidance for traffic safety management on one hand, and can be used for the development and testing of autonomous vehicles on the other. However, the existing models perform poorly when interacting with vehicles. To address the problems, this paper proposes a modeling method (an improved social force model, ISFM) for two-dimensional two-wheeler driving simulation for conflict interaction at intersections. Based on analysis of naturalistic driving study data, when two-wheelers encounter with vehicles, their driving intentions and trajectories can be categorized into two groups, which are yielding and overtaking. Therefore, the vehicle-related social forces are designed to be a set of two forces rather than a repulsion force in original SFM, which is a yielding force based on the relative distance between the two-wheeler and the vehicle, and an overtaking force based on the velocity of the two-wheeler itself. This opens up the possibilities for modeling the multi-modal driving intention of two-wheelers encountering with cross traffic. Based on ISFM, a bicycle model, a powered two-wheeler (PTW) model and a model of a group of PTWs, are then constructed. Compared to the original SFM, ISFM increases the precision of driving intention prediction by 19.7 % (yielding situation) and 25.0 % (overtaking situation), and reduces the root mean square error between simulated and actual trajectories by 7.8 % and 14.8 % on the bicycle model and the PTW model, respectively. Meanwhile, the model of a group of PTWs also performs well. Finally, the results of ablation experiments also validate the effectiveness of the social force designed based on velocity.

RESUMO

AIMS: The purpose of this study is to explore the potential biological role and prognostic significance of chromatin regulators (CRs) in low-grade gliomas (LGGs). MAIN METHODS: CRs were obtained from the FACER database. Transcription profiles of LGG patients were collected from the TCGA and CGGA databases. Differentially expressed CRs (DECRs) between LGGs and normal controls were identified using DESeq2. The consensus clustering algorithm was employed to distinguish subtypes of LGGs based on prognosis-related DECRs. The differences in clinical and molecular characteristics between different subtypes were explored. R packages, GSVA, ssGSEA, and ESTIMATE were utilized to elucidate the tumor microenvironment and activated pathways in different subtypes. Subsequently, a CRs-related signature was developed using LASSO Cox regression. Its performance was evaluated by Kaplan-Meier curve and ROC curve analyses. In vitro experiments were performed to explore the function of JADE3 in LGGs, which predominantly expressed in glioma cells. KEY FINDINGS: We identified 43 DECRs and two CRs-related subtypes of LGGs. The subtype characterized by shorter survival displayed significant enrichment for pathways associated with DNA damage response and repair, along with heightened immune cell infiltration. Furthermore, the CRs-based signature exhibited excellent prognostic performance in both the TCGA and CGGA databases. Knockdown of JADE3 significantly increased the invasion, migration, and proliferation abilities of Hs683. SIGNIFICANCE: Our study reveals the aberrant expression and prognostic value of CRs in LGGs. It emphasizes the potential regulatory role of CRs in the microenvironment and DNA damage repair in LGGs. JADE3 could be a possible therapeutic target for LGGs.

RESUMO

OBJECTIVE TERT: is the most frequently mutated gene in adult glioblastomas (GBMs) defined by the 2021 World Health Organization classification system. The present study aims to explore differences in clinical characteristics and immune microenvironment between TERT mutant and wild-type GBM. METHODS: Three GBM-related cohorts consisting of 205 GBM patients in our cohort, 463 GBM patients without immune checkpoint inhibitor(ICI) therapy and 1465 tumour patients (including 92 GBM cases) receiving ICI treatment in the MSK cohort were included. Retrospective analysis and immunohistochemistry assay were used for investigating the local (including tumour cells, local immune cells, and seizures) and systemic (including circulating immune cells, coagulation-related functions, and prognosis) effects of TERT mutations. Besides, differences in genetic alterations and immunotherapy responses between TERT mutant and wild-type GBMs were also explored. RESULTS: We found that TERT mutant and wild-type GBMs possessed similar initial clinic symptoms, circulating immune microenvironment and immunotherapy response. With respect to that in TERT wild-type GBMs, mutations in TERT resulted in higher levels of tumour-infiltrating neutrophils, prolonged coagulation time, worse chemotherapy response and poorer overall survival. CONCLUSION: Mutations in TERT alter the local immune environment and decrease the sensitivity of GBM to chemotherapy.

RESUMO

Due to the high biological toxicity, the concurrent elimination of lead (Pb (II)) and methylene blue (MB) has become a challenging problem. Therefore, a newly ß-cyclodextrin (ß-CD) modified magnetic alginate/biochar (ß-CD@MBCP) material was developed. Comprehensive characterizations proved the successful coating of ß-CD onto MBCP surface by microwave-aided fabrication. The ß-CD@MBCP achieved high-efficiency uptake for contaminants under a wide pH scope. In the dual system, Pb (II) elimination was facilitated with the presence of MB, due to the active sites provided by MB. In the presence of Pb (II), MB uptake was inhibited due to the electrostatic repulsion between positively charged MB and Pb (II). Electrostatic attraction and complexation contributed to capturing Pb (II), while π-π interactions, host-guest effect, and H-bonding were important in MB elimination. After four cycles, ß-CD@MBCP maintained comparatively good renewability. Findings demonstrated that ß-CD@MBCP could be an effective remediation material for Pb (II)/MB adsorption from aqueous environments.

RESUMO

Although the benefits of biochar amendment for heavy metal(loid) immobilization in soil have been widely recognized, its migration in soil and the resultant effects on the risk of downward migration of metal(loid)s are still poorly understood. In this study, based on biochar derived dissolved organic matter (DOM), excitation-emission matrix coupled with parallel factor analysis (EEM-PARAFAC) technique was employed to trace biochar migration within one year in 0-100 cm soil profiles in the field. The vertical co-migration of Pb, Cu and As was also analyzed. With biochar amended, DOM, humification index (HIX) and biological index (BIX) in 0-60 cm soil profiles increased significantly, while pH only increased in the topsoil. The identified water-extracted DOM components showed that biochar could enhance the content of fulvic acids and humic acids in soil DOM and biochar might migrate downward to 60 cm soil profiles. Furthermore, toluene/methanol-extracted DOM also confirmed the migration extent of biochar, which was more suitable to trace biochar migration because of its high resistance to the long-term ageing in the field. Moreover, we found that biochar reduced the content of Cu in 0-60 cm soil profiles, but increased the available Pb and As in the 20-40 cm soil layers. The Pearson's correlation study confirmed a strong correlation (0.568**≤R ≤ 0.803**) between the content of heavy metal(loid)s and humic-like components of soil DOM, which suggested that biochar co-migrated with Pb, Cu and As, and the potential environmental risks of biochar should be fully evaluated while it was applied for soil metal(loid) remediation.

RESUMO

It is highly significant for the fabrication of rechargeable metal-air batteries to develop cost-efficient and high-performance electrocatalysts of bifunctionality for both oxygen evolution reaction (OER) and oxygen reduction reaction (ORR). Herein, we demonstrate a hybrid composed of CeO2-decorated Co nanoparticles supported on three-dimensionally porous carbon aerogels (Co-CeO2/C aerogels) as a superior bifunctional electrocatalyst. The preparation of Co-CeO2/C aerogels depends on the formation of a novel CeCl3/K3Co(CN)6-chitosan (CS) hydrogel, during which the cyanide groups of K3Co(CN)6 combines the hydroxyls in CS by hydrogen bridges, accompanying with the substitution of chloride groups in CeCl3 by cyanide groups in K3Co(CN)6. The electron spin resonance offers a convincing proof that numerous oxygen vacancies were found in Co-CeO2/C aerogels after the introduction of CeO2. The developed Co-CeO2/C aerogels showed an outstanding electrochemical performance for both OER and ORR in comparsion with RuO2 and Pt/C catalysts in 0.1 M KOH solution. A small overpotential (380 mV) and a low Tafel slope (99 mV dec-1) were observed for OER, while the half-wave potential (0.75 V) and the onset potential (0.92 V) were high for ORR. The superior performance could be put down to the multihole heterostructure, multiple components and abundant oxygen vacancies. It was very helpful for the adsorption and the catalyzation of the reactants and the efficient mass transport of reagent/product. This work paves a neoteric method to synthesize a bifunctional hybrid catalyst with a highly efficient performance of energy conversion and storage.

RESUMO

Lithium metal batteries (LMBs) are a promising candidate for next-generation energy storage devices. However, the high irreversibility and dead Li accumulation of the lithium metal anode caused by its fragile original solid electrolyte interface (SEI) seriously hinder the practical application of LMBs. Herein, a facile slurry-coating and one-step thermal fluorination reaction method is proposed to construct the 3D structural LiF-protected Li/G composite anode. The existence of a 3D LiF protection layer is convincingly confirmed and the function of the Li/G skeleton is discussed in detail. The 3D structural LiF protection layer results in superior electrochemical performance by improving the utilization of Li and suppressing the accumulation of dead Li in symmetric and full coin cells. Moreover, a 0.85 Ah pouch cell strictly following the parameters of the practical battery industry can work stably for 140 cycles with a gradual internal resistance increase. This novel Li/G composite anode indicates a promising strategy in lithium/carbon composite anodes for LMBs, and the facile thermal fluorination reaction method presented in this paper offers a new method for the construction of a 3D structural protection layer for lithium metal anodes.

RESUMO

Metal-free biochar to activate persulfate and degrade organic contaminants has attracted great attention in advanced oxidation processes, while the role of biochar adsorption in the activation and oxidative decomposition process still needed to be further clarified. In this study, nitrogen-doped porous biochar derived from a waste litchi branch was prepared as a peroxydisulfate (PDS) activator for bisphenol A (BPA) degradation, in which the regulation behavior of biochar adsorption was evaluated on the basis of phase distribution and PDS activation mechanism. N-doped biochar obtained at 800 °C with urea and sodium bicarbonate added presented a high specific surface area (821 m2/g), abundant nanopores, and a graphitic structure, and showed the best adsorption capacity and catalytic activity toward BPA. At a dosage of 0.15 g/L NBC-800, 95% BPA can be completely degraded within 60 min with an apparent rate constant (kobs) of 0.0483 min-1. The identified active sites and reactive oxygen species as well as electrochemical tests suggested that both free radicals O2•- and •OH and nonradical pathways including 1O2 originated from C = O and surface electron-transfer mechanisms were involved in BPA decomposition. The experiments and activation mechanisms all confirmed that BPA adsorption on the NBC-800 surface was an extremely crucial step for BPA oxidative degradation.

RESUMO

Isocitrate dehydrogenase (IDH) is an essential metabolic enzyme in the tricarboxylic acid cycle (TAC). The high mutation frequency of the IDH gene plays a complicated role in gliomas. In addition to affecting gliomas directly, mutations in IDH can also alter their immune microenvironment and can change immune-cell function in direct and indirect ways. IDH mutations mediate immune-cell infiltration and function by modulating immune-checkpoint gene expression and chemokine secretion. In addition, IDH mutation-derived D2-hydroxyglutarate can be absorbed by surrounding immune cells, also affecting their functioning. In this review, we summarize current knowledge about the effects of IDH mutations as well as other gene mutations on the immune microenvironment of gliomas. We also describe recent preclinical and clinical data related to IDH-mutant inhibitors for the treatment of gliomas. Finally, we discuss different types of immunotherapy and the immunotherapeutic potential of IDH mutations in gliomas.

RESUMO

Conversion of forests to urban land-use in the processes of urbanization is one of the major causes of biotic homogenization (i.e., decline in beta diversity) in freshwater ecosystems, threating ecosystem functioning and services. However, empirical studies exploring urban land-use shaping patterns of taxonomic and functional beta diversities and their components in subtropical urban rivers are limited. Here, by leveraging data for 43 sampling sites from urban and forest rivers in Shenzhen, a megacity showing rapid urbanization, we determined the spatio-temporal dynamics and associated drivers of taxonomic and functional beta diversities of river macroinvertebrates. Our results showed that, from the forest to urban rivers, taxonomic beta diversity (wet: 32.9%; dry: 17.1%) declined more significantly than functional beta diversity (wet: 17.4%; dry: 9.5%) in different seasons. We further found that these compositional changes were largely driven by decreased roles of species/traits replacement. Although replacement was also dominant for taxonomic beta diversity (60.4%-68.4%) in two sets of rivers, richness difference contributed more to functional beta diversity in the urban river (52.6%-60.5%). Both deterministic and stochastic processes simultaneously affected beta diversity, with stochastic processes being more important in the urban (3.0-19.0%) than forest rivers (0.0%-3.0%). Besides, db-RDA and variation partitioning results showed that local-scale environmental variables explained considerably large fractions of variation in beta diversity. We hence recommended that biodiversity conservation should focus on improving and restoring local environmental conditions. Despite no significant seasonal differences in beta diversity were detected in this study, we found that the roles of deterministic (i.e., local-scale and land-use variables) and stochastic processes varied considerately across seasons. This result highlights the viewpoint that urban river biodiversity monitoring should go beyond one-season snapshot surveys. As the ongoing trend of urbanization in developing countries, the findings of this study are relevant in guiding urban river environmental monitoring, biodiversity conservation and land-use planning.

RESUMO

Here, we investigated the effects of straw returning combined with biochar application on summer maize yield and soil nitrous oxide (N2O) emissions, based on a field location trial in the Guanzhong Plain from 2019 to 2020. The soil N2O emission rates were monitored using the static chamber-chromatography method. A comprehensive analysis of summer maize yields, soil N2O emissions, and soil labile nitrogen components was conducted to clarify the effects of straw returning combined with biochar application on improving soil fertility, increasing summer maize yield, and reducing greenhouse gas emissions. The three treatments were no straw returning (S0), straw returning (S), and straw returning combined with biochar application (SB). The results showed that the peak of N2O emissions from each treatment occurred 10 d after the straw return, and the rate of soil N2O emissions remained at a low level after 30 d of straw return. The rate of soil N2O emissions showed highly significant positive correlations (P<0.05) with ammonium nitrogen (NH4+-N), inorganic nitrogen (SIN), microbial nitrogen (MBN), and dissolved organic nitrogen (DON) contents. S significantly increased summer maize yield, cumulative N2O emissions, yield-scaled N2O intensity, and total nitrogen (TN) content by 7.4%-13%, 65.8%-132.2%, 54.6%-103%, and 27.8%-33%, respectively, compared to those in S0. Although the trend for SB to increase summer maize yield (2.5%-3.3%) compared to that in S was not significant (P>0.05), SB significantly reduced cumulative N2O emissions and yield-scaled N2O intensity by 24.0%-27.3% and 26.4%-29.2%, respectively, compared to that in S. SB significantly reduced the rate of soil N2O emissions by 45.1%-69.6% at the peak of N2O emissions compared to that in S. Biochar application mitigated soil N2O emissions induced by straw return and had a peak-shaving effect. SB significantly increased soil total N by 9.1%-12.2% compared to that in S. Combining summer maize yield, soil N2O emissions, and TN content, SB not only improved soil fertility and summer maize yield but also reduced yield-scaled N2O intensity, making it a suitable management practice that can be replicated to balance crop yield and environmental friendliness.

RESUMO

It is important to develop cost-efficient electrocatalysts used in the oxygen reduction reaction (ORR) for widespread applications in fuel cells. Palladium (Pd) is a promising catalyst, due to its more abundant reserves and lower price than platinum (Pt), and doping an earth-abundant 3d-transition metal M into Pd to form Pd-M bimetallic alloys may not only further reduce the use of expensive Pd but also promote the electrocatalytic performance of ORR, owing to the synergistic effect between Pd and M. Here we report a cyanogel-derived synthesis of PdFe alloys with porous nanostructure via a simple coinstantaneous reduction reaction by using K2PdIICl4/K4FeII(CN)6 cyanogel as precursor. The synthesized PdFe alloys possess hydrangea-like morphology and porous nanostructure, which are beneficial to the electrochemical performance in ORR. The onset potential of the porous PdFe nanohydrangeas is determined to be 0.988 V, which is much more positive than that of commercial Pt/C catalyst (0.976 V) and Pd black catalyst (0.964 V). Resulting from the unique structural advantages and synergetic effect between bimetals, the synthesized PdFe nanohydrangeas with porous structure have outstanding electrocatalytic activity and stability for ORR, compared with the commercial Pd black and Pt/C.

RESUMO

Expansion of agricultural and urban areas and intensification of catchment land-use increasingly affect different facets of biodiversity in aquatic communities. However, understanding the responses of taxonomic and functional diversity to specific conversion from natural forest to agriculture and urban land-use remains limited, especially in subtropical streams where biomonitoring programs and using functional traits are still under development. Here, we conducted research in a subtropical stream network to examine the responses of macroinvertebrate taxonomic and functional diversity to different types of land-use in central China. Our results showed that medium body size, univoltine, gill respiration, and slow seasonal development were much higher in natural forest sites, while certain traits related to strong resilience and resistance (e.g., small body size, fast seasonal development, bi-or multivoltine, abundant occurrence in drift, sprawler) dominated in high-intensity agriculture and urbanization sites. We further found that land-use compromised water quality (e.g., increases in total phosphate, conductivity and water temperature) and habitat conditions (e.g., high proportion of sand and silt, gravel, and channel width) accounted for the changes in trait composition based on a combination of RLQ and fourth-corner analysis. Moreover, natural forest sites presented relatively high values of functional richness than other land-use, demonstrating the importance of natural forest maintenance to promote high levels of functional diversity. However, taxonomic diversity indexes showed higher sensitivity to distinguish different types of land-use compared to functional diversity measures. Even so, given that certain trait categories showed significant relationships with specific local environmental stressors, trait-based approaches can provide reliable evidence to diagnose the cause of impairment and complement the results of the taxonomic-based approaches. Our findings support the idea that taxonomic and functional approaches should be integrated in river restoration and land-use management.

RESUMO

Effects of commercial sand mining on aquatic diversity are of increasing global concern, especially in parts of some developing countries. However, understanding of this activity on the diversity of macroinvertebrates remains focused on the α component of species diversity, rather than community functioning. Thus, there remains much uncertainty regarding how each component of taxonomic (TD) and functional (FD) diversity respond to the activity both in freshwater and marine environments. Here, we assessed the effect of sand dredging on α, ß and γ components of TD and FD during different dredging periods based on the response of macroinvertebrate communities over 4 years in the second largest freshwater lake in China. After three years of active dredging, substantial reductions in each component (α, ß and γ) of TD and FD were observed within the dredged area. Moreover, after one year of natural recovery, a distinct restoration was observed with an obvious return in multiple facets of TD and FD indices. No such changes were observed within the adjacent and reference areas. Decreases in the multiple components of TD and FD within the dredged area were most likely associated with the direct extraction of substrate and associated benthic fauna and indirect variations of the water and sediment environment (e.g., increases in water depth and decreases in %Clay). Furthermore, dispersal processes and mass effects mainly contributed to the maintenance of TD and FD during the dredged and recovery stages. In addition, the fast recovery of TD and FD was also related to the simple taxonomic structure and highly connected nature of the study area. Our results suggest that a more precise experimental design (BACI) should be pursued to avoid potentially confounding effects (e.g., natural disturbance) because the sensitivity of diversity indices depends upon different experimental designs. Moreover, measurement of the impacts of sand dredging on macroinvertebrate diversity can be undertaken within a rigorous framework for better understanding the patterns and processes of each component of TD and FD under the sand dredging disturbance.
