RESUMO

Sows and gilts of the Great White Polish (GWP) breed were classified into groups of 10 each: gilts 8 mo of age (A), sows over 1 yr old with an average litter size of five (BI) and eleven (BII), 2- to 3-yr-old sows with an average litter size of five (CI) and nine (CII), 4- to 6-yr-old sows with an average litter size of six (DI) and nine (DII). Gilts of group A were necropsied on d 10 of the estrous cycle while sows were necropsied 120 to 360 d postpartum. A blood sample obtained by vena cava puncture immediately before exsanguination was quantitated for total cholesterol, free cholesterol, cholesterol ester, total lipid, triglyceride, high and low density lipoprotein and chylomicrons. Postmortem angiograms of the uterine vasculature were evaluated for occurrence of arteriosclerotic lesions of the arteries and their branches. Sections of blood vessels from areas of restriction were examined histologically and quantitated for lipids and proteolytic and lipolytic enzyme activities. With the exception of high density lipoproteins and chylomicrons, serum concentrations of various lipids increased (P less than .01) with age. Restrictions of the lumen were found in uterine arteries and their branches of most gilts in group A and in sows in groups B, C and D, irrespective of litter size. Histology of uterine artery revealed preatherosclerotic lesions in groups B through D. No relationship between the incidence and degree of sclerotic lesions and litter size was evident. However, the incidence and degree of sclerotic lesions increased with age and parity. Results from histopathology were supported by results from measurement of lipids and enzyme activities of the uterine artery wall. Conception rates of sows in groups C and D were lower than those of group B. Many of these sows (70%) failed to conceive during 1 yr even though they displayed normal estrous cycles.

The heterogeneity of phosphoprotein phosphohydrolase--Fpf in cellular free spleen extracts of pigs may be determined by sodium fluoride inhibition. NaF concentration of 5 X 10(-3) mol/l divides Fpf into 3 groups and shows a wide inhibition range from 15 to 85% of the initial activity of the enzyme. Naf concentration of 5 X 10(-4)mol/l stops Fpf activity in the range from 10 to 65% and divides it into two groups. To classify the studied material into Fpf phenotypes, fluoride quotient index--WIF has been proposed which expresses the relationship between the percentage of the inhibition caused by NaF at a concentration 5 X 10(-3) mol/l and that of the inhibition caused by NaF at a concentration of 5 X 10(-4) mol/l in the reaction mixture and in the measurement conditions described in the methods. Five groups with the following numerical values of WIF have been distinguished: 1.15 less than or equal to WIF less than 1.40-44%--group I; 1.50 less than or equal to WIF less than 1.80-20%--group II; 1.90 less than or equal to WIF less than 2.10-16%--group III; 2.20 less than or equal to WIF less than 2.50-16%--group IV; and WIF = 3-4%--group V.

Two groups of cocks were kept for 84 days on a diet rich in fat (group D) or on a similar diet with addition of coumarin (group DK). In the plasma of these birds there occurred a distinct increase of the concentration of total fat (TC) and cholesterol (CH), in comparison with the concentration of these substances of the plasma of control birds. The concentration of TC in the plasma of group DK was higher than that observed in group D. The concentration of CH was similar in both experimental groups. The level of the lipolytic activity of plasma (ALpO) changed during the experiment and was higher in the plasma of the cocks of group DK than in that of group D. Confrontation of the results of the determinations of ALpO, TC and CH in groups D and DK demonstrates that the symptoms of a disturbance of the metabolism of fats, which occur in cocks kept on a diet rich in fats, are stimulated by coumarin. This influence is demonstrated by a speeding up, by about 42 days, of the occurrence of these symptoms, i.e. of the increase of the TC concentration and of the ALpO level. The values of TC and ALpO, determined on the 21st and 42nd day of the experiment in group DK, corresponded to the relevant values found in group D on the 63rd and 84th day of the experiment. It is highly probable that coumarin acts on the mechanisms which control the ALpO level indirectly, through the carbohydrate metabolism.

On dogs under Eunarkon anesthesia (0,03 ml Eunarkon/kg body weight) after one intramuscular lethal injection of phoschlor (500 mg/lg body weight = LD100) the proteolytic activity (APR) was estimated in the blood plasma at pH 3,5-APR-3,5 and in the hemolyzed blood at pH-7,4-APR-7,4. The results showed that heavy poisoning with phoschlor led to metabolic disturbances in the proteolytic system of the blood plasma and blood, the expression of which was increasing in the course of time a statistically important increase (alpha = 0,001) of APR-3,5 and APR-7,4. APR induction in blood is not a result of direct activation of enzymes by pesticides. It takes place outside the circulation system.
