RESUMO

The thin flyer is a small-scale flying object, which is well known as the core functional element of the initiator. Understanding how flyers perform has been a long-standing issue in detonator science. However, it remains a significant challenge to explore how the flyer is formed and functions in the barrel of the initiator via tabletop devices. In this study, we present dynamic and unprecedented images of flyer in barrel via high intensity short-pulse laser. Advanced radiography, coupled with a high-intensity picosecond laser X-ray source, has enabled the provision of state-of-the-art radiographs in a single-shot experiment for observing micron-scale flyer formation in a hollow cylinder in nanoseconds. The flyer was clearly visible in the barrel and was accelerated and restricted differently from that without the barrel. This first implementation of a tabletop X-ray source provided a new approach for capturing dynamic photographs of small-scale flying objects, which were previously reported to be accessible only via an X-ray phase-contrast imaging system at the advanced photon source. These efforts have led to a significant improvement of radiographic capability and a greater understanding of the mechanisms of "burst" of exploding foil initiators for this application.

Holographic display faces the trade-off between image quality and focus cues, resulting from the specific choice of phase distribution. In this Letter, we propose a speckle-free holographic display with enhanced focus cues by multiple directional light reconstruction. The uniform phase hologram is first generated by the gradient descent optimization algorithm. The blazed grating phase is used to steer the object light to a specific direction. Multiple sub-holograms with different blazed gratings are refreshed fast to reconstruct the images from different directions. Thus, the defocus blur is improved due to the separation of multiple spots on the defocus plane. The multi-plane reconstruction is also realized by pre-processing the depth images to eliminate image separation. The proposed method provides apparent focus cues while maintaining high image qualities, which is expected to realize comfortable holographic near-eye display in the near future.

The spotted hyena (Crocuta crocuta) is a large and unique terrestrial carnivore. It is a particularly fascinating species due to its distinct phenotypic traits, especially its complex social structure and scavenging lifestyle, with associated high dietary exposure to microbial pathogens. However, the underlying molecular mechanisms related to these phenotypes remain elusive. Here, we sequenced and assembled a high-quality long-read genome of the spotted hyena, with a contig N50 length of ∼13.75 Mb. Based on comparative genomics, immunoglobulin family members (e.g., IGKV4-1) showed significant adaptive duplications in the spotted hyena and striped hyena. Furthermore, immune-related genes (e.g., CD8A, LAG3, and TLR3) experienced species-specific positive selection in the spotted hyena lineage. These results suggest that immune tolerance between the spotted hyena and closely related striped hyena has undergone adaptive divergence to cope with prolonged dietary exposure to microbial pathogens from scavenging. Furthermore, we provided the potential genetic insights underlying social complexity, hinting at social behavior and cognition. Specifically, the RECNE-associated genes (e.g., UGP2 and ACTR2) in the spotted hyena genome are involved in regulation of social communication. Taken together, our genomic analyses provide molecular insights into the scavenging lifestyle and societal complexity of spotted hyenas.

The Siberian roe deer (Capreolus pygargus) is a widely distributed ungulate in northeast China. Due to a series of human disturbance activities such as large-scale forest cutting, deforestation and reclamation, road construction in the past, the appearance and internal structure of forest vegetation in the habitat of Siberian roe have changed significantly. At the same time, Siberian roe population had a series of ecological adaptation responses in the face of such habitat changes. Therefore, two typical vegetation types with differences were selected in the Muling Forest, China. We used nutritional ecology and microbial metagenomic analysis techniques to compare the nutritional selection strategy and the structure and functional characteristics of faecal microbiota of Siberian roe groups in two vegetation types. The results showed that the α diversity of dietary and gut microbes of deer in Natural Forest was higher than that in Plantation Forest. However, the gut microbes of the Plantation Forest group contained more unique enzymes in the functional pathways of carbon metabolism and biosynthesis of amino acids. This study suggests that habitat type is associated with plant community composition, and contributes to changes in the intake proportions of major macronutrients by altering the availability, quality, and composition of certain edible plants. Feeding behaviour may be an important regulatory factor of gut microbiota structure and function of deer. The metabolic function of gut microbiota to different nutrients may affect the microbial community structure. Therefore, our results suggest that the gut microbes of Siberian roe may have coevolved with their diets, and reflect the adaptability of deer populations to environmental changes (e.g., vegetation type). Our study provides new insights into how spatial heterogeneity affects nutrition and microecosystems by describing the interactions among the environment, diet, and symbiotic gut microbes in wild ungulates.

Multi-plane reconstruction is essential for realizing a holographic three-dimensional (3D) display. One fundamental issue in conventional multi-plane Gerchberg-Saxton (GS) algorithm is the inter-plane crosstalk, mainly caused by the neglect of other planes' interference in the process of amplitude replacement at each object plane. In this paper, we proposed the time-multiplexing stochastic gradient descent (TM-SGD) optimization algorithm to reduce the multi-plane reconstruction crosstalk. First, the global optimization feature of stochastic gradient descent (SGD) was utilized to reduce the inter-plane crosstalk. However, the crosstalk optimization effect would degrade as the number of object planes increases, due to the imbalance between input and output information. Thus, we further introduced the time-multiplexing strategy into both the iteration and reconstruction process of multi-plane SGD to increase input information. In TM-SGD, multiple sub-holograms are obtained through multi-loop iteration and then sequentially refreshed on spatial light modulator (SLM). The optimization condition between the holograms and the object planes converts from one-to-many to many-to-many, improving the optimization of inter-plane crosstalk. During the persistence of vision, multiple sub-hologram jointly reconstruct the crosstalk-free multi-plane images. Through simulation and experiment, we confirmed that TM-SGD could effectively reduce the inter-plane crosstalk and improve image quality.The proposed TM-SGD-based holographic display has wide applications in tomographic 3D visualization for biology, medical science, and engineering design, which need to reconstruct multiple independent tomographic images without inter-plane crosstalk.

In near-eye displays (NEDs), retinal projection display (RPD) is one kind of promising technology to alleviate the vergence-accommodation conflict (VAC) issue due to its always-in-focus feature. Viewpoint replication is widely used to enlarge the limited eyebox. However, the mismatch between viewpoint interval and eye pupil diameter will cause the inter-viewpoint cross talk when multiple viewpoints enter the pupil simultaneously. In this Letter, a holographic complementary viewpoint method is proposed to solve this cross talk problem. Instead of avoiding observing multiple viewpoint images simultaneously, it is designed that multiple complementary viewpoints jointly project the complete image on the retina without cross talk. To do this, the target image is segmented into multiple sub-images, each multiplied with a corresponding partial spherical phase to converge to a specific complementary viewpoint. A group of complementary viewpoint enter the eye pupil simultaneously, and each viewpoint project a corresponding sub-image on a specific area of the retina and splice to a complete image. All of the complementary viewpoints are duplicated to an interlaced two-dimensional array to extend the eyebox in both horizontal and vertical directions. Optical experiment verifies that the proposed method could present smooth transition between viewpoints to avoid both inter-viewpoint cross talk and blank image issues.

Conventional spatial light modulator (SLM)-based holographic 3D display faces limited field of view (FOV) and eyebox, due to its limited pixel number. In this paper, a lens array is used to expand the FOV and eyebox of an SLM-based holographic display. The hologram is calculated to reconstruct a 3D sub-image array, each sub-image corresponding to a specific perspective of the 3D object. Then, the 3D sub-image array is imaged and magnified by the lens array to integrate to the original 3D image. The FOV is expanded due to the large numerical aperture of the lens, and the eyebox is expanded because the lens array generates multiple viewpoints with a large pitch. The optical experiment realizes a 17.6° FOV and 50 mm eyebox, which contains 4 × 4 viewpoints. Apparent motion parallax is observed through the viewpoint array, which is usually hard to observe in a conventional SLM-based holographic display. The proposed method provides a novel, to the best of our knowledge, way to expand the FOV and eyebox of holographic 3D display without increasing the total pixel number of the SLM.

Ferroptosis, triggered by discoordination of iron, thiols and lipids, leads to the accumulation of 15-hydroperoxy (Hp)-arachidonoyl-phosphatidylethanolamine (15-HpETE-PE), generated by complexes of 15-lipoxygenase (15-LOX) and a scaffold protein, phosphatidylethanolamine (PE)-binding protein (PEBP)1. As the Ca2+-independent phospholipase A2ß (iPLA2ß, PLA2G6 or PNPLA9 gene) can preferentially hydrolyze peroxidized phospholipids, it may eliminate the ferroptotic 15-HpETE-PE death signal. Here, we demonstrate that by hydrolyzing 15-HpETE-PE, iPLA2ß averts ferroptosis, whereas its genetic or pharmacological inactivation sensitizes cells to ferroptosis. Given that PLA2G6 mutations relate to neurodegeneration, we examined fibroblasts from a patient with a Parkinson's disease (PD)-associated mutation (fPDR747W) and found selectively decreased 15-HpETE-PE-hydrolyzing activity, 15-HpETE-PE accumulation and elevated sensitivity to ferroptosis. CRISPR-Cas9-engineered Pnpla9R748W/R748W mice exhibited progressive parkinsonian motor deficits and 15-HpETE-PE accumulation. Elevated 15-HpETE-PE levels were also detected in midbrains of rotenone-infused parkinsonian rats and α-synuclein-mutant SncaA53T mice, with decreased iPLA2ß expression and a PD-relevant phenotype. Thus, iPLA2ß is a new ferroptosis regulator, and its mutations may be implicated in PD pathogenesis.

To further study the aminoalcohol-diterpenoid alkaloids (ADAs) in Fuzi (Aconiti Lateralis Radix Praeparata), a simple and sensitive UFLC-MS/MS method was established and validated for the determination of five ADAs, aconine, mesaconine, hypaconine, deoxyaconine and fuziline, in rat plasma to compare the pharmacokinetic characteristics of pure ADAs and Fuzi decoction. After precipitating protein with methanol, plasma samples were isolated at 0.5 mL/min flow rate on Waters Acquity UPLC BEH C18 column (100 mm × 2.1 mm, 1.7 µm). The mobile phase was composed of 0.1% formic acid-water and methanol with gradient elution. Mass spectrometric inspection was conducted on a 5500 UFLC-MS/MS system with an electrospray ionization source in patterns of positive ion and multiple reaction-monitoring (MRM). All calibration curves were proved to have acceptable linearity (r2 > 0.99) in linear ranges. Intra-day and inter-day precision and the accuracy met the requirements. The matrix effects of all analytes were between 85% and 115% of three concentration levels. This method has been under verification for comparative pharmacokinetic research after oral administration between aqueous extract of Fuzi and single pure ADAs. The results demonstrated that there are evident pharmacokinetic discrepancies between them, and administration in the extract form instead of pure form may contribute to higher absorption.

Uric acid (UA) is the final product of purine metabolism in human body,and its metabolic disorder will induce hyperuricemia (HUA).The occurrence and development of HUA are associated with a variety of pathological mechanisms such as oxidative stress injury,activation of inflammatory cytokines,and activation of renin-angiotensin-aldosterone system.These mechanisms directly or indirectly affect the bioavailability of endogenous nitric oxide (NO).The decrease in NO bioavailability is common in the diseases with high concentration of UA as an independent risk factor.In this review,we summarize the mechanisms by which high concentrations of UA affect the endogenous NO bioavailability,with a focus on the mechanisms of high-concentration UA in decreasing the synthesis and/or increasing the consumption of NO.This review aims to provide references for alleviating the multisystem symptoms and improving the prognosis of HUA,and lay a theoretical foundation for in-depth study of the correlations between HUA and other metabolic diseases.

Holographic retinal projection display (RPD) can project images directly onto the retina without any lens by encoding a convergent spherical wave phase with the target images. Conventional amplitude-type holographic RPD suffers from strong zero-order light and conjugate. In this paper, a lensless phase-only holographic RPD based on error diffusion algorithm is demonstrated. It is found that direct error diffusion of the complex Fresnel hologram leads to low image quality. Thus, a post-addition phase method is proposed based on angular spectrum diffraction. The spherical wave phase is multiplied after error diffusion process, and acts as an imaging lens. In this way, the error diffusion functions better due to reduced phase difference between adjacent pixels, and a virtual image with improved quality is produced. The viewpoint is easily deflected just by changing the post-added spherical phase. A full-color holographic RPD with adjustable eyebox is demonstrated experimentally with time-multiplexing technique.

The Artificial Intelligence of Things (AIoT) turns passive fiber sensors into learning machines. It can be used to integrate intelligent nodes into a multi-dimensional sensing system. In this study, the temperature measurement system based on Brillouin Gain Spectrum (BGS) test setup is creatively implemented with the AIoT architecture; the training and testing stages of the neural network are divided into different layers of equipment to improve performance and reduce the network traffic. To enable the lightweight and low-power consumption edge computing device to extract accurate temperature from the BGS during testing, we have integrated the post-processing method inspired by curve fitting into the machine learning and proposed the efficient digital resampling filter. The resampling filter approach is achieved by the peak range algorithm with Gauss differential operator and digital band-pass filter. The evaluation results for different methods on the BGS datasets show the superior performance of our approach. Notably, the approach can increase temperature extraction accuracy and compress the sampling data. The RMSEA of the extraction temperature is 0.5635, which can bring an almost 21% accuracy increase over the classic method. Compared with the classic method of processing the original data on the same hardware platform, the amount of data post-processed by the filter is reduced by 75%; the time consumption is reduced by 22%.

Augmented reality (AR) near-eye displays (NEDs) are emerging as the next-generation display platform. The existing AR NED only present one single video channel at a time, same as traditional media such as TVs and smartphones. In this Letter, to the best of our knowledge, we propose for the first time a multi-channel holographic retinal projection display (RPD), which can provide multi-channel image sources simultaneously, thus greatly increasing the information content. Due to the superposition capacity of a hologram, multiple images are projected to different viewpoints simultaneously through multiple spherical wave encoding, so that the viewer can switch among playing channels very fast through eye rotation. A full-color dynamic multi-channel holographic near-eye display is demonstrated in the optical experiment. The proposed method provides a good prospect that the future AR glasses can play dozens of video channels in parallel, and the user can switch among channels freely and efficiently just through a simple eye rotation.

Enhancer of Zeste Homologue 2 (EZH2) as a histone methyltransferase epigenetically regulates laryngeal carcinoma (LGC) progression. The present study sought to explore the role and mechanism of EZH2 in the epithelial-mesenchymal transition (EMT) of LGC cells. Expressions of EZH2, secreted frizzled-related protein 1 (SFRP1), and trimethylation of lysine 27 on histone H3 (H3K27me3) in LGC tissues or cells were detected via reverse transcription quantitative polymerase chain reaction (qRT-PCR) and western blotting. Upon transfection of si-EZH2, si-SFRP1, oe-SFRP1, or H3K27me3 upregulation, cell viability was assessed via cell counting kit-8, protein levels of E-cadherin, N-cadherin, ß-catenin, c-Myc, and Cyclin D1 were determined via western blotting, and Vimentin expression was determined via immunofluorescence. The enrichment level of H3K27me3 in the SFRP1 promoter was measured via chromatin immunoprecipitation-PCR. EZH2 was highly expressed in LGC tissues and cells. Silencing EZH2 repelled the EMT of LGC cells. Mechanically, EZH2 upregulated H3K27me3 in the SFRP1 promotor to inhibit SFRP1 expression, and SFRP1 overexpression inactivated the Wnt pathway. H3K27me3 upregulation or SFRP1 downregulation reversed the inhibition of silencing EZH2 in the EMT of LGC cells. Overall, EZH2 upregulated H3K27me3 in the SFRP1 promoter to inhibit SFRP1 expression and activate the Wnt pathway, thereby facilitating the EMT of LGC cells.

PURPOSE: This work was to identify the function and mechanism of miR-219a-5p in regulating knee osteoarthritis (KOA). METHODS: Rat fibroblast-like synoviocytes (FLSs) were isolated to construct KOA cell model by lipopolysaccharide and adenosine triphosphate treatment. miR-219a-5p and FBXO3 expression in FLSs was modulated by transfection. Flow cytometry was executed to research FLSs apoptosis. Caspase-1 and IL-1ß expression in FLSs was researched by immunofluorescence. The binding between miR-219a-5p and FBXO3 was identified by dual luciferase reporter gene assay. KOA rat model and miR-219a-5p up-modulation KOA rat model were constructed. Step size of rats was analyzed. Knee joints of rats were experienced Safranin O-fast green staining to evaluate the knee joint injury. FBXO3, pyroptosis-associated proteins, and IL-1ß and IL-18 expression in FLSs and articular cartilage tissues of rats were assessed by Western blot, qRT-PCR and Enzyme-linked immunosorbent assay. RESULTS: KOA cell model had higher apoptosis percentage, expression of pyroptosis-associated proteins, and IL-1ß and IL-18 level. miR-219a-5p up-modulation decreased the above indicators, whereas miR-219a-5p down-modulation increased the above indicators. FBXO3 expression was directly repressed by miR-219a-5p. Loss of FBXO3 suppressed the above indicators. FBXO3 counteracted the suppression of miR-219a-5p on the above indicators. miR-219a-5p agomir attenuated knee joint injury, increased step size of KOA rats, and reduced FBXO3, pyroptosis-associated proteins and level of IL-1ß and IL-18 in the articular cartilage tissues of KOA rats. CONCLUSION: miR-219a-5p suppressed the pyroptosis in KOA by inactivating the NLRP3 signaling via targeting FBXO3, which might be a promising target for ameliorating KOA in the clinic.

This study aims to explore anti-obesity and lipid-lowering mechanism of Corydalis Bungeanae Herba(CB) based on intestinal microflora and metabolomics. Specifically, high-fat high-sugar diet(HFHS, 10 weeks) was used to induce obesity in rats. Then the model rats were randomized into the model group, low-dose(0.18 g·kg~(-1)), medium-dose(0.9 g·kg~(-1)), and high-dose(1.8 g·kg~(-1)) CBH groups, and orlistat group(0.03 g·kg~(-1)), 12 in each group. Rats which received normal diet were used as control. The body weight and feed intake of rats were recorded every week. After 6 weeks of administration, rats were killed and gastric emptying and small intestinal propulsion were examined. Enzyme-linked immunosorbent assay(ELISA) was employed to analyze serum indexes, and liver and perirenal fat were collected for haematoxilin-eosin(HE) staining. Rat feces and serum were gathered for 16 S rDNA sequencing and metabolomics analysis and Spearman's correlation analysis was performed to explore the correlation between differential microflora and differential metabolites. The result showed that CBH extract decreased body weight, feed intake, and serum cholecystokinin(CCK), triglyceride(TG), and total cholesterol(TC), delayed gastric emptying, and reduced fat accumulation in liver and perirenal adiposity as compared with rats in the model group. In addition, Lachnospiraceae and Sutterellaceaecan significantly decreased in the model group, but CBH extract up-regulated their abundance. Moreover, the abundance of Prevotellaceae was significantly raised by HFHS, but CBH decreased it. Glutaric acid, glyceric acid, hippuric acid, malic acid, glyceric acid, oxoglutaric acid, fumaric acid/succinic acid, oxoglutaric acid/isocitric acid, D-glucuronic acid, cholic acid were the main deferentially expressed metabolites and significantly correlated with Sutterellaceae and Prevotellaceae. These key metabolites and microbiota mainly involved in tricarboxylic acid(TCA) cycle, glucose metabolism, amino acid metabolism, and energy metabolism. This study proved that CBH can efficiently improve body weight and blood lipids, reduce adipocyte volume, and positively regulate the intestinal microflora and serum metabolites, thereby achieving the anti-obesity and lipid-owering effect.

The holographic Maxwellian display is a promising technique for augmented reality (AR) display because it solves the vergence-accommodation conflict while presenting a high-resolution display. However, conventional holographic Maxwellian display has the inherent trade-off between depth of field (DOF) and image quality. In this paper, two types of holographic Maxwellian displays, the spherical wave type and the plane wave type, are proposed and analyzed. The spherical wavefront and the plane wavefront are produced by a spatial light modulator (SLM) for Maxwellian display. Due to the focusing properties of different wavefronts, the two types of display have complementary DOF ranges. A hybrid approach combining the spherical wavefront and plane wavefront is proposed for a large DOF with high image quality. An optical experiment with AR display is demonstrated to verify the proposed method.

The development of optical fiber sensors has led to the possibility of accumulating vast, real-time databases of acoustic and other measurements throughout fiber networks, which brings even more widespread concern on improving the sampling effectiveness. In this paper, we present two kinds of sweep frequency methods based on using a neural network to extract temperature from the Brillouin gain spectrum (BGS). Gauss centralization and variance weight probability methods are proposed to compare with the uniform sweep frequency method. By analyzing formulas of the ideal BGS model, we find the gain near the peak of Brillouin gain spectrum has greater correlation with temperature extraction than other positions. Therefore, the Gaussian centralized sweep method is proposed. We further investigate the variation of the weights in the neural network and Brillouin data distribution in different positions and find that the variance is positively correlated with the weights in hidden layers. So, we propose the sweep frequency method based on variance weight probability and make a complement to interpret the rationality of this method in neural network. In all the aforementioned approaches, 281 points are obtained between the 9.07 GHz to 9.35 GHz range under the same condition. The data of each method is trained ten times and tested through the same neural network structure. All the RMSE of each test stage covers all data collecting the passage. The result shows that the RMSE of variance weight probability sweep frequency method is 0.5277, which is superior to the Gauss centralization sweep frequency method that was 0.6864 and the uniform sweep frequency method that was 0.9140.

Conventional holographic display suffers from the conjugate light issue. In this Letter, we propose to efficiently extend the eyebox of holographic Maxwellian near-eye display by encoding the conjugate wavefront as the multiplication of plane wave phase with the target image. It is interesting that after being focused by the lens, the generated conjugate viewpoints also present erect virtual images with the same image quality as the signal viewpoints. Multiple plane wave encoding is used for eyebox extension, and, because of the utilization of conjugate light, the effect of eyebox extension is doubled. That is, the space bandwidth of the amplitude-type hologram is fully used. A speckless holographic image is produced in mid-air with high quality within a large depth range. The proposed display is compact and promising for the augmented reality near-eye display. Furthermore, it may inspire better solutions for the conjugate light issue of amplitude-type holography.

A lensless full-color holographic Maxwellian near-eye display using a single amplitude-type spatial light modulator is proposed in this Letter. The color holographic image is directly projected onto the retina without any eyepiece. The color crosstalk is clearly separated from the signal in the space owing to the encoded spherical wave and carrier wave. An aperture numerical filter and a real polarized filter are used at the pupil plane to accurately stop the crosstalk light. A high-quality dynamic speckless color holographic image was produced in the mid-air within a specific depth range. The horizontal eyebox expansion is achieved simply through multiple spherical wave encoding and verified through an optical experiment. The proposed display is compact and promising as the augmented reality near-eye display.
