RESUMO

Tamarillo leaf malformation virus (TaLMV) is a potyvirus first discovered in cape gooseberry fields in Eastern, and South-western Antioquia. This virus is responsible for a very damaging disease that has resulted in significant reductions in yields and cultivated area for this crop in Colombia. Tamarillo is frequently co-cultivated with other solanaceous plants but no evidence for cross-pathogenicity of TaLMV has been found until now. In this work, we report a natural infection of cape gooseberry (Physalis peruviana L.) by TaLMV. Infection by TaLMV was detected by RNAseq screening of cape gooseberry fields and confirmed by RT-qPCR and Sanger sequencing. The sequenced genome is 99.3% identical to previously sequenced TaLMV isolates, and evidence suggests that it can accumulate at high loads in this new reported host. RT-qPCR analysis indicates that TaLMV is already widely distributed, can naturally infect other solanaceous hosts and may become an emerging threat to the cape gooseberry agroindustry, the second most important exotic fruit export in Colombia. Transcriptome analysis of a Cape gooseberry (Physalis peruviana) plant with leaf symptoms of a mild yellow mosaic typical of a viral disease revealed an infection with Potato virus X (PVX). The genome sequence of the PVX-Physalis isolate comprises 6435 nt and exhibits higher sequence similarity to members of the Eurasian group of PVX (~95 %) than to the American group (~77 %). Genome organization is similar to other PVX isolates with five open reading frames coding for proteins RdRp, TGBp1, TGBp2, TGBp3, and CP. 5' and 3' untranslated regions revealed all regulatory motifs typically found in PVX isolates. The PVX-Physalis genome is the only complete sequence available for a Potexvirus in Colombia and is a new addition to the restricted number of available sequences of PVX isolates infecting plant species different to potato.

Late blight caused by Phytophthora infestans is one of the most limiting diseases in solanaceous crops in the world. This pathogen is a main constraint in the highland Andes, where these plants are grown under high humidity conditions and continuous cropping. The aim of this research was to increase the available information on the biology of P. infestans, specifically on its level of genetic variation in south-western Colombia, an area where various solanaceous crops susceptible to this pathogen converge. The study was carried out by using AFLP molecular markers with the restriction enzymes EcoRI and MseI and different primer combinations. Results indicated a low level of genetic variation among the 26 isolates evaluated, with only 18 polymorphic bands out of 135 amplicons obtained (13.43%), a Nei's genetic diversity index of 0.04, and a Shannon's information index of 0.06.
