RESUMO

Much like passive materials, active systems can be affected by the presence of imperfections in their microscopic order, called defects, that influence macroscopic properties. This suggests the possibility to steer collective patterns by introducing and controlling defects in an active system. Here we show that a self-assembled, passive nematic is ideally suited to control the pattern formation process of an active fluid. To this end, we force microtubules to glide inside a passive nematic material made from actin filaments. The actin nematic features self-assembled half-integer defects that steer the active microtubules and lead to the formation of macroscopic polar patterns. Moreover, by confining the nematic in circular geometries, chiral loops form. We find that the exact positioning of nematic defects in the passive material deterministically controls the formation and the polarity of the active flow, opening the possibility of efficiently shaping an active material using passive defects.

RESUMO

The development of accurate and efficient image reconstruction algorithms is a central aspect of quantitative photoacoustic tomography (QPAT). In this paper, we address this issues for multi-source QPAT using the radiative transfer equation (RTE) as accurate model for light transport. The tissue parameters are jointly reconstructed from the acoustical data measured for each of the applied sources. We develop stochastic proximal gradient methods for multi-source QPAT, which are more efficient than standard proximal gradient methods in which a single iterative update has complexity proportional to the number applies sources. Additionally, we introduce a completely new formulation of QPAT as multilinear (MULL) inverse problem which avoids explicitly solving the RTE. The MULL formulation of QPAT is again addressed with stochastic proximal gradient methods. Numerical results for both approaches are presented. Besides the introduction of stochastic proximal gradient algorithms to QPAT, we consider the new MULL formulation of QPAT as main contribution of this paper.

Praziquantel (PZQ), long-used in veterinary and human medicine for the treatment of helminth parasites, is known to enhance humoral and cellular immune responsiveness in mammals but has unknown direct immunomodulatory capabilities in fish. In the present study, we examined the ability of PZQ to induce gene transcriptional changes in immune-competent primary tissue/organ cultures of two highly important yet evolutionarily discrete fish species--Southern bluefin tuna Thunnus maccoyii and Atlantic salmon Salmo salar. These cultures consisted of mixed blood cell population for both species, as well as intestinal explants from bluefin. Although expression profiles varied between species and tissue/organ type, PZQ induced both T-cell receptor (more than twofold) and IL-8 transcriptional expression (more than fourfold). Additionally, increased expression of other inflammatory cytokines including IL-1ß was detected in blood cell cultures from both species, and a general pattern of heightened antiviral signaling was observed. Specifically, elevated transcription of Type I (IFNα) and Type II (IFNγ) interferon in Atlantic salmon blood cultures along with elevated expression of MHC class I in blood cultures of both species. These findings provide preliminary evidence for direct immunomodulation by PZQ in fish and insight into its potential capacity as an immune stimulant/adjuvant in the rapidly expanding aquaculture industry.

RESUMO

Image reconstruction in multispectral computed tomography (MSCT) requires solving a challenging nonlinear inverse problem, commonly tackled via iterative optimization algorithms. Existing methods necessitate computing the derivative of the forward map and potentially its regularized inverse. In this work, we present a simple yet highly effective algorithm for MSCT image reconstruction, utilizing iterative update mechanisms that leverage the full forward model in the forward step and a derivative-free adjoint problem. Our approach demonstrates both fast convergence and superior performance compared to existing algorithms, making it an interesting candidate for future work. We also discuss further generalizations of our method and its combination with additional regularization and other data discrepancy terms.

Melanomacrophage centres (MMCs) are aggregates of macrophages accumulating various pigments. They have been proposed as an indicator of fish immune response. Blood flukes are common parasites in farmed fish. Two cohorts of wild Southern Bluefin Tuna (Thunnus maccoyi) were examined at transfer, before treatment against blood flukes (pre-treatment) and at harvest. MMCs were assessed in histological sections using image analysis, while Cardicola forsteri and Cardicola orientalis infection severity was determined using qPCR, count of adult flukes in heart flushes and count of eggs in gill filaments. Fish from both cohorts showed the same pattern in the changes in the surface area of MMCs. The surface area of splenic MMCs increased over the ranching duration and was positively correlated to the PCR determined copy numbers of Cardicola forsteri ITS2 rDNA in the gills of those fish. However, the infection with blood fluke was more variable, both between cohorts and individuals within the same cohort. Eggs of blood fluke were detected in renal MMCs using histology. Cardicola forsteri had a higher prevalence than Cardicola orientalis. This study contributes to our understanding of blood fluke infections in Southern Bluefin Tuna and their interactions with MMCs.

Cardiolipin (CL) is a phospholipid specific for mitochondrial membranes and crucial for many core tasks of this organelle. Its acyl chain configurations are tissue specific, functionally important, and generated via post-biosynthetic remodeling. However, this process lacks the necessary specificity to explain CL diversity, which is especially evident for highly specific CL compositions in mammalian tissues. To investigate the so far elusive regulatory origin of CL homeostasis in mice, we combine lipidomics, integrative transcriptomics, and data-driven machine learning. We demonstrate that not transcriptional regulation, but cellular phospholipid compositions are closely linked to the tissue specificity of CL patterns allowing artificial neural networks to precisely predict cross-tissue CL compositions in a consistent mechanistic specificity rationale. This is especially relevant for the interpretation of disease-related perturbations of CL homeostasis, by allowing differentiation between specific aberrations in CL metabolism and changes caused by global alterations in cellular (phospho-)lipid metabolism.

An end-to-end real-time text localization and recognition method is presented. Its real-time performance is achieved by posing the character detection and segmentation problem as an efficient sequential selection from the set of Extremal Regions. The ER detector is robust against blur, low contrast and illumination, color and texture variation. In the first stage, the probability of each ER being a character is estimated using features calculated by a novel algorithm in constant time and only ERs with locally maximal probability are selected for the second stage, where the classification accuracy is improved using computationally more expensive features. A highly efficient clustering algorithm then groups ERs into text lines and an OCR classifier trained on synthetic fonts is exploited to label character regions. The most probable character sequence is selected in the last stage when the context of each character is known. The method was evaluated on three public datasets. On the ICDAR 2013 dataset the method achieves state-of-the-art results in text localization; on the more challenging SVT dataset, the proposed method significantly outperforms the state-of-the-art methods and demonstrates that the proposed pipeline can incorporate additional prior knowledge about the detected text. The proposed method was exploited as the baseline in the ICDAR 2015 Robust Reading competition, where it compares favourably to the state-of-the art.

The effect of oral vaccines against bacterial fish diseases has been a topic for debate for decades. Recently both M-like cells and dendritic cells have been discovered in the intestine of rainbow trout. It is therefore likely that antigens reaching the intestine can be taken up and thereby induce immunity in orally vaccinated fish. The objective of this project was to investigate whether oral and anal vaccination of rainbow trout induces protection against an experimental waterborne infection with the pathogenic enterobacteria Yersinia ruckeri O1 biotype 1 the causative agent of enteric redmouth disease (ERM). Rainbow trout were orally vaccinated with AquaVac ERM Oral (MERCK Animal Health) or an experimental vaccine bacterin of Y. ruckeri O1. Both vaccines were tested with and without a booster vaccination four months post the primary vaccination. Furthermore, two groups of positive controls were included, one group receiving the experimental oral vaccine in a 50 times higher dose, and the other group receiving a single dose administered anally in order to bypass the stomach. Each group was bath challenged with 6.3 × 10(8) CFU/ml Y. ruckeri, six months post the primary vaccination. The challenge induced significant mortality in all the infected groups except for the groups vaccinated anally with a single dose or orally with the high dose of bacterin. Both of these groups had 100% survival. These results show that a low dose of Y. ruckeri bacterin induces full protection when the bacterin is administered anally. Oral vaccination also induces full protection, however, at a dose 50 times higher than if the fish were to be vaccinated anally. This indicates that much of the orally fed antigen is digested in the stomach before it reaches the second segment of the intestine where it can be taken up as immunogenic antigens and presented to lymphocytes.

RESUMO

An efficient enantioselective synthesis of the chiral polycyclic cholesteryl ester transfer protein (CETP) inhibitor 1 has been developed. The synthesis was rendered practical for large scale via the development of a modified Hantzsch-type reaction to prepare the sterically hindered pyridine ring, enantioselective hydrogenation of hindered ketone 6 utilizing novel BIBOP-amino-pyridine derived Ru complex, efficient ICl promoted lactone formation, and a BF3 mediated hydrogenation process for diastereoselective lactol reduction. This efficient route was successfully scaled to produce multikilogram quantities of challenging CETP drug candidate 1.
