RESUMO

BACKGROUND: Recurrent head and neck squamous cell carcinoma (HNSCC) is associated with poor overall survival (OS). Prior studies suggested incorporation of nab-paclitaxel (A) may improve outcomes in recurrent HNSCC. METHODS: This Phase I study evaluated induction with carboplatin and A followed by concomitant FHX (infusional 5-fluorouracil, hydroxyurea and twice-daily radiation therapy administered every other week) plus A with cohort dose escalation ranging from 10-100 mg/m2 in recurrent HNSCC. The primary endpoint was maximally tolerated dose (MTD) and dose-limiting toxicity (DLT) of A when given in combination with FHX (AFHX). RESULTS: Forty-eight eligible pts started induction; 28 pts started AFHX and were evaluable for toxicity. Two DLTs occurred (both Grade 4 mucositis) at a dose level 20 mg/m2. No further DLTs were observed with subsequent dose escalation. The MTD and recommended Phase II dose (RP2D) of A was 100 mg/m2. CONCLUSIONS: In this Phase I study, the RP2D of A with FHX is 100 mg/m2 (AFHX). The role of re-irradiation with immunotherapy warrants further investigation. CLINICAL TRIAL INFORMATION: This clinical trial was registered with ClinicalTrials.gov identifier: NCT01847326.

RESUMO

This study provides evidence on the impact of physical education on child body weight, cognitive, and noncognitive achievement using data from the Early Child Longitudinal Survey Kindergarten Class of 2010-2011 (ECLS-K: 2011). Students in the 2011 cohort were exposed to increased accountability pressures by No Child Left Behind, yet average weekly physical education time has not decreased from that reported in studies using the original ECLS-K class of 1998-1999. We instrument for teacher-reported weekly PE time using state physical education laws and exploit the panel design of the data to estimate individual fixed effects models to address concerns of endogeneity. We find time spent in physical education has essentially no effect on child body weight or human capital outcomes of U.S. elementary school children.

RESUMO

Arginine vasotocin (AVT) has different effects on social behaviors across species with different social systems. However, less attention has been directed towards the way environmental stimuli interact with AVT to affect social behaviors. Previous work found that AVT affects the production of advertisement calls and the outcome of competitive interactions between male gray treefrogs (Hula versicolor). We conducted a field study to investigate how the presence of other males in a chorus influenced the production of advertisement calls in males treated with AVT or saline. After hormone treatment we placed individuals in call sites either within 50 cm of another calling male (proximate), or more than 2 m away from any calling male (distant). Males treated with AVT produced significantly longer calls with more pulses when placed at proximate call sites, but not distant call sites. Nonsignificant trends indicated that these increases in call length coincided with a decrease in call rate. There was no effect of call site distance on advertisement calls when males were treated with saline. These results indicate that the social environment can modulate the action of AVT on an important reproductive behavior, and that AVT does not influence call duration and pulse number unless another male is nearby. Understanding how social stimuli regulate the action of AVT on behavior could provide insight on the diversity of AVT function across species.
