This study aims to investigate the level of genetic variability of Fasciola hepatica flukes isolated from cattle in Algeria and to determine the phylogenetic and phylogeographic relationships with sequences isolated worldwide. Mitochondrial (Cytochrome c Oxidase subunit I gene - COI) and nuclear markers (Internal Transcribed Spacers of nuclear ribosomal DNA - ITS) for 24 F. hepatica flukes isolated from 12 cattle in North Algeria were characterised. Only two haplotypes were obtained for the COI gene, resulting in a low level of genetic variation. The analysis of variation among the COI sequences isolated from around the world did not show high levels of genetic divergence, and the phylogenetic analysis revealed a genetic similarity among F. hepatica isolates from different areas of the world. The analysis of the ITS region showed a low level of variability, which prevented obtaining informative phylogenetic and phylogeographic results. The present study also revealed that specimens of F. hepatica are genetically similar in different hosts, indicating that the genetic structure among populations of this parasite is not influenced by the host species. The low levels of genetic variation for COI and ITS regions among fluke isolates from all continents are consistent with a common origin for the flukes' worldwide distribution.

This study provides updated information on the distribution of the phlebotomine sand fly species and their genetic characterization in Sardinia, a Mediterranean island where leishmaniasis is endemic. From April to November 2017, sand flies were trapped in five different capture sites using sticky traps and light traps as collection methods, operated nearby sheep, poultry, cat, and dog shelters. Phlebotomine specimens (n = 513) collected were morphologically and molecularly identified as Phlebotomus perniciosus (249/513, 48.5%), Phlebotomus perfiliewi (236/513; 46%), and Sergentomyia minuta (28/513, 5.5%). Sand flies were collected from the second half of May to October confirming the well-defined seasonal activity, which peaks in August in Sardinia. Overall, correlation analyses indicated a significant positive association between the monthly number of sand flies collected and the mean temperature (r = 0.88, rho = 0.87, and tau = 0.69, P < 0.05), while there was non-significant, moderately negative correlation between the monthly number of sand flies collected and the monthly mean relative humidity and wind (r = - 0.22, rho = 0.02, and tau = 0.04, P > 0.05). This study provides the first data on the molecular characterization of phlebotomine sand flies in this region and confirms the presence of three sand fly species. Molecular results suggest that the morphological features used for analysis represent synapomorphic-derived characters which are shared among descendant taxa and the common ancestor.

The aim of this study was to examine the effect of chemical cationization on the structure and function of antifreeze protein III (AFP III) over an extreme temperature range (-40°C to +90°C) using far-UV synchrotron radiation circular dichroism (SRCD) and ice recrystallization inhibition (IRI) assays. Chemical cationization was able to produce a modified AFP III with a net cationic charge at physiological pH that had enhanced resistance to denaturation at elevated temperatures, with no immediate negative impact on protein structure at subzero temperatures. Furthermore, cationized AFP III retained an IRI activity similar to that of native AFP III. Consequently, chemical cationization may provide a pathway to the development of more robust antifreeze proteins as supplementary cryoprotectants in the cryopreservation of clinically relevant cells.

Mast cell tumor (MCT) is a common canine cutaneous neoplasm with variable biological behavior. A 2-tier histologic grading system was recently proposed by Kiupel et al to reduce interobserver variation and eliminate prognostic uncertainty of the Patnaik system. This study compared the ability of these 2 grading systems to predict survival in a cohort of dogs with MCTs. However, surgical margins were unknown, and the risk of developing new/metastatic MCTs was not studied. Histologic grade was assessed according to both systems for 137 surgically resected cutaneous MCTs. The relationship between grade and survival was evaluated. According to the Patnaik system, 18 MCTs (13.1%) were classified as grade I, 83 (60.6%) as grade II, and 36 (26.3%) as grade III. Grade III was associated with a poorer prognosis (P < .001), but no significant difference between grades I and II was detected. Grading according to the Patnaik system was based on consensus grading among 3 pathologists, and interobserver variability was not considered. All grade I MCTs were low grade in the Kiupel system, and all grade III were high grade. Among grade II, 71 (85.6%) were low grade, and 12 (14.4%) were high grade, with a 1-year survival probability of 94% and 46%, respectively (P < .001). The 2-tier system had a high prognostic value and was able to correctly predict the negative outcomes of some grade II MCTs. Data also confirm that histologic grading cannot predict biological behavior of each MCT and should be supplemented with molecular methods for more accurate prognostication.

First of all, this article aimed to evidence the role of a modified printer developed for continuous carbon fibre reinforced PolyAmide (cCF/PA6-I) together with the use of a fully open slicing step on the printing quality and the longitudinal/transverse tensile and in-plane shear properties. A comprehensive assessment of the microstructure and properties with a similar material (cCF/PA6-I), but produced with a commercial printer (i.e., Markforged® MarkTwo) has been achieved. Our customised printer and the open slicer used have made possible to better control the print conditions (i.e., layer height and distance between filaments), to reduce the porosity from more than 10% to about 2% and improve the mechanical properties. Moreover, the understanding of the behaviour of these 3D printed composites with wide-ranging external temperatures is mandatory for future use in a severe environment and/or development of new thermally active 4D printed composites. The 3D printed cCF/PA6-I composites have been then thermomechanically characterised along different printing directions (0, 90 and ± 45°) from -55 to +100 °C. Unlike the longitudinal properties that hardly change with temperature, the transverse and in-plane shear stiffness and strength of these 3D printed composites were particularly sensitive to temperature variations, with decreases of 25-30% and 30-55%, respectively. This was due to the high sensitivity of the polymer matrix, the fibre/matrix and interfilament interfaces when the composites were loaded along those directions, because damages induced by internal thermal stresses. Fractography has also been carried out to reveal damage mechanisms.

We propose an analytical formulation to extract from energy equivalence principles the equivalent thickness and in-plane mechanical properties (tensile and shear rigidity, and Poisson's ratio) of hexagonal boron nitride (h-BN) nanosheets. The model developed provides not only very good agreement with existing data available in the open literature from experimental, density functional theory (DFT) and molecular dynamics (MD) simulations, but also highlights the specific deformation mechanisms existing in boron nitride sheets, and their difference with carbon-based graphitic systems.

We evaluate the vibrational properties of single-wall carbon nanotube (SWCNT) hetero-junction (HJ) oscillators using a hybrid atomistic-continuum approach validated by molecular mechanics/molecular dynamics simulations. The SWCNT-HJs show a broken symmetry topology of their mode shapes, with striction effects caused on the bending and radial modes by the combined effect of the HJ and the tube with the thinner radius. The single-wall nanotube HJs also show selective mass sensing properties based solely on the geometry and type of the boundary conditions of the specific nanostructure. This unusual behaviour has not been observed so far in classical SWCNT systems.

The potent anti-tumour activities of gammadelta T cells have prompted the development of protocols in which gammadelta-agonists are administered to cancer patients. Encouraging results from small Phase I trials have fuelled efforts to characterize more clearly the application of this approach to unmet clinical needs such as metastatic carcinoma. To examine this approach in breast cancer, a Phase I trial was conducted in which zoledronate, a Vgamma9Vdelta2 T cell agonist, plus low-dose interleukin (IL)-2 were administered to 10 therapeutically terminal, advanced metastatic breast cancer patients. Treatment was well tolerated and promoted the effector maturation of Vgamma9Vdelta2 T cells in all patients. However, a statistically significant correlation of clinical outcome with peripheral Vgamma9Vdelta2 T cell numbers emerged, as seven patients who failed to sustain Vgamma9Vdelta2 T cells showed progressive clinical deterioration, while three patients who sustained robust peripheral Vgamma9Vdelta2 cell populations showed declining CA15-3 levels and displayed one instance of partial remission and two of stable disease, respectively. In the context of an earlier trial in prostate cancer, these data emphasize the strong linkage of Vgamma9Vdelta2 T cell status to reduced carcinoma progression, and suggest that zoledronate plus low-dose IL-2 offers a novel, safe and feasible approach to enhance this in a subset of treatment-refractory patients with advanced breast cancer.

Functional near-infrared spectroscopy (fNIRS) is a neuroimaging technique that measures changes in oxy-hemoglobin (ΔHbO) and deoxy-hemoglobin (ΔHbR) concentration associated with brain activity. The signal acquired with fNIRS is naturally affected by disturbances engendering from ongoing physiological activity (e.g., cardiac, respiratory, Mayer wave) and random measurement noise. Despite its several drawbacks, the so-called conventional averaging (CA) is still widely used to estimate the hemodynamic response function (HRF) from noisy signal. One such drawback is related to the number of trials necessary to derive stable HRF functions adopting the CA approach, which must be substantial (N >> 50). In this work, a pre-processing procedure to remove artifacts followed by the application of a non-parametric Bayesian approach is proposed that capitalizes on a priori available knowledge about HRF and noise. Results with the proposed Bayesian approach were compared with CA and with a straightforward band-pass filtering approach. On simulated data, a five times lower estimation error on HRF was obtained with respect to that obtained by CA, and 2.5 times lower than that obtained by band pass filtering. On real data, the improvement achieved by the present method was attested by an increase in the contrast to noise ratio (CNR) and by a reduced variability in single trial estimation. An application of the present Bayesian approach is illustrated that was optimized to monitor changes in hemodynamic activity reflecting variations in visual short-term memory load in humans, which are notoriously hard to detect using functional magnetic resonance imaging (fMRI). In particular, statistical analyses of HRFs recorded during a memory task established with high reliability the crucial role of the intraparietal sulcus and the intra-occipital sulcus in posterior areas of the human brain in visual short-term memory maintenance.

The unique features of axial, torsional, transverse and radial breathing vibrations are captured for armchair and zigzag singlewalled boron nitride nanotubes (BNNTs) based on molecular mechanics simulations and continuum mechanics theories. Equivalent Young's modulus 1 TPa and shear modulus 0.4 TPa are obtained independent of the chirality of BNNTs. In particular, a distorted optimized structure is observed for the first time for BNNTs with sufficiently large diameter and length. It is found that the deformed structures result in behaviours of BNNTs deviating from those of classical columns/beams. Such symmetry-breaking could also exert significant impacts on the structural instability (buckling) and electronic properties of BNNTs that are sensitive to the structural symmetry.

The out-of-plane bending behaviour of single layer graphene sheets (SLGSs) is investigated using a special equivalent atomistic-continuum model, where the C-C bonds are represented by deep shear bending and axial stretching beams and the graphene properties by a homogenization approach. SLGS models represented by circular and rectangular plates are subjected to linear and nonlinear geometric point loading, similar to what is induced by an atomic force microscope (AFM) tip. The graphene models are developed using both a lattice and a continuum finite element discretization of the partial differential equations describing the mechanics of the graphene. The minimization of the potential energy allows us to identify the thickness, elastic parameters and force/displacement histories of the plates, in good agreement with other molecular dynamic (MD) and experimental results. We note a substantial equivalence of the linear elastic mechanical properties exhibited by circular and rectangular sheets, while some differences in the nonlinear geometric elastic regime for the two geometrical configurations are observed. Enhanced flexibility of SLGSs is observed by comparing the nondimensional force versus displacement relations derived in this work and the analogous ones related to equivalent plates with conventional isotropic materials.

The elastic moduli of single layer graphene sheet (SLGS) have been a subject of intensive research in recent years. Calculations of these effective properties range from molecular dynamic simulations to use of structural mechanical models. On the basis of mathematical models and calculation methods, several different results have been obtained and these are available in the literature. Existing mechanical models employ Euler-Bernoulli beams rigidly jointed to the lattice atoms. In this paper we propose truss-type analytical models and an approach based on cellular material mechanics theory to describe the in-plane linear elastic properties of the single layer graphene sheets. In the cellular material model, the C-C bonds are represented by equivalent mechanical beams having full stretching, hinging, bending and deep shear beam deformation mechanisms. Closed form expressions for Young's modulus, the shear modulus and Poisson's ratio for the graphene sheets are derived in terms of the equivalent mechanical C-C bond properties. The models presented provide not only quantitative information about the mechanical properties of SLGS, but also insight into the equivalent mechanical deformation mechanisms when the SLGS undergoes small strain uniaxial and pure shear loading. The analytical and numerical results from finite element simulations show good agreement with existing numerical values in the open literature. A peculiar marked auxetic behaviour for the C-C bonds is identified for single graphene sheets under pure shear loading.

OBJECTIVES: To evaluate the diagnostic utility of individual cytological criteria and their best combination to differentiate benign from malignant perianal gland proliferative lesions in dogs. MATERIALS AND METHODS: Retrospective study of cytological samples of canine perianal gland proliferative lesions that had subsequent histopathological confirmation. RESULTS: Seventy-seven perianal gland nodules from 56 dogs were included. Histologically, lesions were diagnosed as hyperplasia (n=2), adenoma (n=53), epithelioma (n=6) and carcinoma (n=16). Of the 28 cytological criteria assessed, 13 showed promise for distinguishing benign from malignant lesions. A diagnostic algorithm with an 87% accuracy (sensitivity, 90.9%; specificity, 85.4%) was developed from these data. CLINICAL SIGNIFICANCE: Cytological evaluation can provide useful information for presurgical differentiation between benign and malignant hepatoid gland proliferative lesions. The proposed algorithm must be validated and tested for reproducibility in further, preferably larger, series of cases.

We present a new class of TiBw/Ti6Al4V composites with a network reinforcement architecture that exhibits a significant creep resistance compared to monolithic Ti6Al4V alloys. Creep tests performed at temperatures between 773 K and 923 K and stress range of 100 MPa-300 MPa indicate both a significant improvement of the composites creep resistance due to the network architecture made by the TiB whiskers (TiBw), and a decrease of the steady-state creep rates by augmenting the local volume fractions of TiBw in the network region. The deformation behavior is driven by a diffusion-controlled dislocation climb process. Moreover, the activation energies of these composites are significantly higher than that of Ti6Al4V alloys, indicating a higher creep resistance. The increase of the activation energy can be attributed to the TiBw architecture that severely impedes the movements of dislocation and grain boundary sliding and provides a tailoring of the stress transfer. These micromechanical mechanisms lead to a remarkable improvement of the creep resistance of these networked TiBw/Ti6Al4V composites featuring the special networked architecture.

Corneal images acquired by in-vivo specular and confocal microscopy provide clinical information on the cornea endothelium health state. Indeed, the normal hexagonal shape of endothelial cells is usually affected by age and pathologies. At present, the analysis is based on manual or semi-automatic methods and the segmentation of a large number of endothelial cells is required for a meaningful estimation of the clinical parameters. We propose an automated method that detects the centers of endothelial cells by convolving the original image with customized two-dimensional kernels, derives a structure made by connected vertices from the recognized centers using the Euclidean distance, and fine-tunes the derived structure through a genetic algorithm, which combines information about the typical regularity of endothelial cells shape with the pixels intensity of the actual image. The final structure of connected vertices forms a set of polygons that fit the underlying cells contours. From these contours the morphometric parameters of clinical interest can be easily computed. The procedure was applied to 15 images acquired with the SP-3000P (Topcon, Japan) specular microscope and 15 images acquired with the Confoscan4 (Nidek Technologies, Italy) confocal microscope, from both healthy and pathological subjects. Ground truth values for the morphometric parameters were obtained from manually carefully drawn cell contours. Results show that the mean percent absolute difference between the automated and the manual estimate of the clinical parameters is between 2 and 6%, and no statistically significant difference was found between them. The proposed totally automatic method appears capable of detecting contour of hundreds of cells covering a large area, and of obtaining a reliable estimation of the relevant morphometric parameters used in clinical practice.

Rubber band ligation is a commonly employed office procedure for the eradication of symptomatic internal hemorrhoids. Since 1980 increasingly frequent reports of an often fatal complication--"pelvic cellulitis"--have appeared. Death has been avoided for some of these patients by early recognition and treatment. One such survivor is reported here, in a case report that illustrates the value of early recognition of symptoms and appropriate diagnostic and therapeutic intervention.

The authors report the results obtained on 22 selected cases of tubaric pregnancy in evolution and treated by reconstructive plastic surgery. They point out the necessity of a very precocious laparoscopic diagnosis in order to obtain a better functional recovery of the fallopian tube. On the basis of the results, the authors suggest a more frequent reconstructive treatment of the tubaric pregnancy.

Canine nodular dermatofibrosis is a rare skin disease associated with renal cystadenoma or cystadenocarcinoma and uncommonly with uterine leiomyoma. It is generally seen in German shepherd dogs, but has been also reported in other breeds, and a relationship has been suggested with mutation of the gene encoding folliculin (FLCN), which is located on chromosome 5. A 10-year-old female golden retriever was presented because of numerous firm cutaneous nodules up to 4 cm in diameter over the entire body surface. Cytological and histopathological examinations confirmed generalized cutaneous nodular dermatofibrosis, but ultrasonography of both kidneys ruled out renal neoplasia. Ovariohysterectomy was performed because of prolonged oestrus periods. Microscopical examination of the excised tissues confirmed the absence of uterine neoplasia, but identified rete adenoma of the right ovary. Abdominal ultrasound performed repeatedly over a 5-year follow-up period did not identify any alteration in the renal parenchyma. Molecular studies excluded the presence of any mutation in the FLCN gene.
