RESUMO

Glutathione peroxidase (GPx) plays an important antioxidant role in cellular defense against environmental stress. In the present study, a novel selenium-dependent glutathione peroxidase termed McSeGPx firstly identified in thick shell mussel Mytilus coruscus. McSeGPx consists of 197 amino acid residues, characterized with one selenocysteine residue encoded by an opal stop codon TGA, one selenocysteine insertion sequence (SECIS) in the 3' untranslated region (UTR), two active site motifs and one signature sequence motif. McSeGPx transcripts were constitutively expressed in all examined tissues, and were significantly induced in gills and digestive glands with the stimulations of lipopolysaccharide (LPS), copper (Cu) and benzo[α]pyrene (B[α]P). Additionally, rough increases in McSeGPx activity were detected in both tissues under the challenge of LPS, Cu and B[α]P. Collectively, these results suggested that McSeGPx affiliate to selenocysteine dependent GPx (SeGPx) family and might play an important role in mediating the environmental stressors and antioxidant response in M. coruscus.

Interleukin-1 receptor-associated kinase-4 (IRAK4) is considered as the most upstream kinase of IRAKs and plays a vital role in Toll-like receptor/Interleukin-1 receptor (TLR/IL-1R) signal transduction. In the present study, IRAK4 from thick shell mussel Mytilus coruscus (McIRAK4) was identified and characterized. McIRAK4 showed the most similarity to its counterparts in bivalves. The conserved death domain (DD) and catalytic domain of serine/threonine kinases (STKc) were predicted in all examined IRAK4s. McIRAK4 transcripts were constitutively expressed in all examined tissues with the higher expression level in immune related tissues, and were significantly induced in haemocytes upon lipopolysaccharide (LPS) and polyinosinic-polycytidylic acid (poly I:C) challenge. Further, the expression of McIRAK4 was obviously repressed by dsRNA mediated RNA interference (RNAi), meanwhile the proinflammatory cytokines TNF-alpha and IL17 were down-regulated while the antiinflammatory cytokine TGF-ß was up-regulated. Additionally, McIRAK4 showed a global cytoplasmic localization in HEK293T cells through fluorescence microscopy. These results collectively indicated that McIRAK4 is one member of IRAK4 subfamily and might play the potential signal transducer role in inflammatory response. The present study provides supplement for TLR-mediated signaling pathway triggered by pathogenic invasions in thick shell mussel, and contributes to the clarification of the innate immune response in molluscs.

For the aim to study potential detrimental effects induced by Cu exposure at low, environmentally relevant concentrations, the in vivo activities at different levels of biological organisations of thick shell mussel Mytilus coruscus exposed to two levels of copper were assessed. Cu-induced stresses were evaluated through antioxidant responses, DNA damage and genotoxicity. The results revealed significant higher SOD and CAT activities, and MDA concentration in haemocytes of M. coruscus with Cu exposure at 8 µg/L, while only significant accumulation in CAT activity with Cu exposure at 2 µg/L and no significant changes with SOD activity and MDA concentration at this level of Cu exposure. Copper exposure induced DNA damage as induction of OTM value in a time- and concentration-dependent manner. In addition, copper exposure could significantly induced the expressions of MT-10, Hsp70, Hsp90 and C3. The present results deepen the mussels as a suitable model marine invertebrate species to study potential detrimental effects induced by possible toxicants, in combinations at different levels of biological organisations.

The NF-E2-related factor 2 (Nrf2), an ubiquitous, evolutionarily conserved transcription factor, acts as a major sensor of oxidative stress in cells. In the present study, a Nrf2 homolog was newly identified in the thick shell mussel Mytilus coruscus. Accordingly, its functional role in antioxidant defense in response to acute benzo(a)pyrene (Bap) exposure was assessed. The newly identified McNrf2 affiliated to traditional Nrf2 family through Blast, multiple alignment and phylogenetic analysis. After acute exposure to Bap, antioxidants including superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) and glutathine reductase (GR) were significantly induced in gills and digestive glands at both mRNA and enzymatic levels, and the expression of McNrf2 mRNA was also up-regulated. The analysis of correlating the expression of McNrf2 and the mRNA levels of these antioxidant genes showed positive ties, indicating that Nrf2 was needed for protracted induction of such genes. Further, the recombinant McNrf2 was produced through pET-32a prokaryotic system. After 50 µg/L Bap exposure, ROS generation and LPO level in gills of Nrf2 over-expressed mussels significantly decreased compared to Nrf2 wild-type mussels, as well as reduced ROS production in digestive glands. Collectively, these results show that Nrf2 pathway can provide protection from oxidative stress triggered by Bap in the thick shell mussel.

Toll-like receptors (TLRs) are the most well studied pattern recognition receptors (PRRs) that play a crucial role in both innate and adaptive immunity in animals. In the present study, a novel toll-like receptor (McTLRj) was identified and characterised in thick shell mussel Mytilus coruscus. McTLRj possessed a signal peptide, a transmembrane domain, leucine-rich repeats and an intracellular Toll/interleukin-1 receptor domain that were conserved in typical TLRs. McTLRj transcripts were constitutively expressed in all of the examined tissues with high expression level in immune-related tissues, and significantly induced in haemocytes upon live Vibrio alginolyticus, lipopolysaccharide, polyinosinic-polycytidylic acid and peptidoglycans challenge. The overexpression of the McTLRj TLR fragment in Drosophila S2 cells could induce the expression of Drosophila attacin A, drosomycin, cecropin A, and metchnikowin expression. The expression of McTLRj was obviously repressed by dsRNA-mediated RNA interference, and downstream TLR pathway factors, such as MyD88a, IRAK4, and TRAF6 were significantly repressed in McTLRj-silenced mussels upon LPS challenge. These results collectively indicated that McTLRj is a TLR family member that may play a potential PRR role in TLR-mediated signalling pathway. This research contributed to the clarification of innate immune response in molluscs.

Estrogen could lead to abnormal modulation or disruption of physical development, reproduction and sexual behavior in aquatic wildlife, especially in fish. Information on the toxicity of estrogens to native species in that can be used in site-specific risk assessments is scarce. In the present study, one zona pellucida 3 (ZP3) homologue termed CaZP3 was firstly identified from topmouth culter Culter alburnus, following its structural characteristics, tissue distribution and transcriptional modulation to 17α-ethinylestradiol (EE2) exposure were investigated. Meanwhile, vitellogenin (VTG) gene was employed to provide a comparison of the reactive ability to EE2 induction. The CaZP3 characterized with analogical functional domains such as ZP domain, SP, IHP, EHP, 12 cysteine residues, one N-linked glycosylation site and two conserved O-linked glycosylation sites and equal number of eight exons and seven introns with ZP3 counterparts of higher species. CaZP3 mRNA predominantly expressed in ovary, besides, highly expressed in female heart and male muscle and relatively high expressed in testis. CaZP3 has the lower reactive ability to EE2 induction in comparison with VTG, however, CaZP3 transcripts were significantly induced in gonads of both male and female culter by EE2 and could be used as an alternative biomarker to monitor EE2 activity. The present results supplement the database for toxicity of EE2, especially for fish species endemic to China and provide some useful information for the monitoring of EE2 activity in aquatic environment.

Sturgeons are euryhaline fish species that have developed specific mechanisms of osmotic and ion regulation to adapt to waters of varying salinity. For the aim to elucidate the osmoregulation strategy behind its high salinity tolerance of sturgeons, the transcriptomes of gills in Siberian sturgeon Acipenser baeri under salinity stress (30 ppt) were sequenced using deep-sequencing platform Illumina/HiSeq-2500 and differential expression genes (DEGs) were identified. A total of 167, 501, 278 clean reads were obtained and 280, 238 unigenes were composed of those clean reads with the mean length of 520nt, and the N50 of 630 bp. Unigenes Sequence alignment was implemented via KEGG, KOG, NT, NR, PFAM, Swiss-Prot, and GO databases. 62, 242 unigenes (22.21%) were annoated in at least one database. 11380 significantly differentially expressed unigenes were found, 6969 of which were up-regulated and 4411 were down-regulated by salinity stress. Amongst the top 20 KEGG pathways with the most amount of annotation sequences, some pathways such as glycerophospholipid metabolism, fatty-acid biosynthesis, glycolysis/gluconeogenesis, oxidative phosphorylation have been comprehensively proved to be relevant to osmoregulation. Despite of these, three possible osmoregulation-related signaling pathways as lipid metabolism related pathways, tight junction pathway and thyroid hormone signaling pathway have been widely analyzed in the current study. In all DEGs, some of the typical genes involved in osmoregulation, including calcium-transporting ATPase 4 (ATP2B4), Na+/K+-ATPase alpha subunit (α-NKA), potassium-transporting ATPase alpha chain 1 (ATP4A) and Ras GTPase-activating protein (RasGAP) etc were also identified. RNA-seq results were validated with quantitative real-time PCR (qPCR), the 12 selected genes showed a consistent direction in both DGE library and qPCR analysis, proving that the RNA-seq results are reliable. The present results would be helpful to elucidate the osmoregulation mechanism of aquatic animals adapting to salinity challenge.
